Preparation of the Proteus vulgaris Bacterial Electrodes for the Determination of Urea and Their Application (요소 정량을 위한 Proteus vulgaris 박테리아 전극의 개발과 그 응용)

  • The bacteria containing urease convert each molecule of urea into two molecules of ammonia and one molecule of carbon dioxide gas. Bacterial electrodes have been constructed by immobilizing the Proteus vulgaris on an ammonia and a carbon dioxide gas-sensors, and were investigated for the effects of pH, temperature, buffer solution, bacterial amounts and interferences, and life time. NH3-bacterial electrode based on ammonia gas-sensor had linearity in the range of $7.0{\times}10^{-4}\;-\;3.0{\times}10^{-2}$M urea in pH 7.4, 0.05M phosphate buffer at $25^{\circ}C$ with a slope of 116.7 mV/decade. While $CO_{2-}$bacterial electrode based on carbon dioxide gas-sensor bad linearity in the range of $7.0{\times}10^{-4}\;-\;5. 0{\times}10^{-2}$M urea in pH 7.0, 0.1M phosphate buffer at $30^{\circ}C$with a slope of $45.4{\times}45.7mV/decade$. As the clinical application, urea in urine was determined by these devices and this result was compared with spectrophotometric method. Consequently, these electrodes could be used for the analysis of many samples because of simplicity, rapidity and convenience of the experimental procedure.

Distribution and Leaching of Basal Nitrogen in Direct Seeding Rice on Dry Paddy (벼 건답직파 재배에서 기비질소의 토층간 분포와 용탈)

  • Urea, which is the major nitrogenous fertilizer used in Korea, has been used inefficiently in direct-seeding on dry soil by farmers. This study was conducted to investigate changes in concentrations of basal N within soil layers and its loss during early stage of rice growth. Urea fertilizer was applied in the rates of 7, 5.25, 3.5, 1.75, 0kg- N /10a under direct-seeded rice in dry paddy soil. The concentrations of ammonium and nitrate were determined in soil samples with different depths during period from seeding to the 3rd leaf stage. Futhermore, N leaching was measured in lysimeter designed with pot in greenhouse. ${NH_4}^+ \; and\; {NO_3}^-$ adsorption by soil increased with increasing concentration of added urea and decreased as deeper in soil layers. ${NH_4}^+$ concentration reached its peak at 7 days after urea application (DAA) and disappeared almostly at 14 DAA. ${NO_3}^-$ reached its peak at 10 DAA and decreased slowly until 14 DAA. ${NO_3}^-$N leaching started next day after urea application and completed until 11 DAA. We concluded that most of basal N applied to direct-seeded paddy was lost by leaching and not useful for rice plant which was in stage of germination. It is urgent need to develop new nitrogen application method for direct-seeding rice on dry soil.

Investigation on the Preparation Method of TiO2-mayenite for NOx Removal (질소산화물 제거를 위한 TiO2-mayenite 제조 방법에 관한 연구)

  • In order to apply a photocatalyst (TiO2) to various building materials, TiO2-mayenite was prepared in this study. The TiO2 was synthesized using the sol-gel method by fixing titanium isopropoxide (TTIP) and urea at a ratio of 1 : 1. Later, they were calcined in a temperature range of 400-700 ℃ to analyze the properties according to temperature. BET, TGA, and XRD were used to analyze the physical and chemical properties of TiO2. The nitrogen oxide removal test was confirmed by measuring the change in the concentration of NO for 1 h according to KS L ISO 22197-1. The prepared TiO2 samples exhibited an anatase crystal structure below 600 ℃, and TiO2 (urea)-400 showed the highest nitrogen oxide removal rate at 2.35 µmol h-1. TiO2-mayenite was prepared using two methods: spraying TiO2 dispersion solution (s/s) and sol-gel solution (g/s). Through BET and XRD analysis, it was found that 5-TiO2 (g/s) prepared by spraying a sol-gel solution has maintained its crystallinity even after heat treatment. Also, 5-TiO2 (g/s)-500 showed the highest removal rate of 0.55 µmol h-1 in the nitrogen oxide removal test. To prepare TiO2-mayenite, it was confirmed that mayenite should be blended with TiO2 in a sol-gel state to maintain the crystal structure and exhibit a high nitrogen oxide removal rate.

  • Lithium cobalt oxide $(LiCoO_2)$ cathode powders for rechargeable battery have been successfully prepared using urea hydrolysis method. The obtained hydrolysis-derived precursors with different Li/Co molar ratio were calcined at various temperatures. Low temperature phase $(LT-LiCoO_2)$ and high temperature phase $(HT-LiCoO_2)$ were obtained after calcination at $500^{\circ}C$ for 2 hr, and phase transformation from $LT-LiCoO_2{\;}to{\;}HT-LiCoO_2$ was completely occurred over $700^{\circ}C$. The layered structure of $LiCoO_2$ was well developed with a rise in the calcination temperature. Charge-discharge test show that the lithium cobalt oxide with 1.2 molar ratio prepared at $800^{\circ}C$ has an initial discharge capacity as high as 152 mAh/g, and the relatively stable cycling characteristic with 9.2 % of capacity fading was obtained after 40th charge-discharge test.

Influence of Initial Molar Ratios on the Performance of Low Molar Ratio Urea-Formaldehyde Resin Adhesives

  • In this paper, the influence of initial formaldehyde/urea (F/U) molar ratios on the performance of low molar ratio (1.0) urea-formaldehyde (UF) resin adhesives has been investigated. Two initial F/U molar ratios, i.e., the first and second initial molar ratios were used for the alkaline addition reaction. Three levels of the first initial F/U molar ratios (2.0, 3.0, and 4.0) and two levels of the second initial molar ratios (2.0 and 1.7) were employed to prepare a total of six UF resins with an identical final molar ratio (1.0). The basis properties, functional groups, molecular weight, crystallinity, and thermal curing properties of the UF resins were characterized in detail. Higher levels (3.0 and 4.0) of the first initial F/U molar ratio provided the UF resins with better properties (non-volatile solids content, viscosity, gelation time, pH, and specific gravity) than those of the resins prepared with the conventional level F/U molar ratio of 2.0. Statistical analysis suggested that combining the first and second initial molar ratio of 4.0 with 1.7 would result in UF resins with greater adhesion strength and lower formaldehyde emission than those of the resins prepared with other molar ratios. The results showed that higher levels of the first initial molar ratio resulted in a more branched structure, as indicated by GPC, FTIR, DSC, XRD, and greater adhesion strength than those of the other UF resins with an identical final molar ratio of 1.0.

Effects of a Telephone Follow-up Program on Patient Compliance, Urea Kinetic Modeling and Incidence of Infection in Patients with Continuous Ambulatory Peritoneal Dialysis (CAPD) (전화추후관리 프로그램이 복막투석환자의 치료지속이행, 투석적절도와 감염발생도에 미치는 효과)

  • Purpose: The objective of this study was to identify the effects of a telephone follow-up program on patient compliance, urea kinetic modeling and incidence of infection in patients with continuous ambulatory peritoneal dialysis (CAPD). Method: A pretest-posttest design with a nonequivalent control group was used to examine the effects of a telephone follow-up program for 12 weeks in a group of 39 CAPD patients. The telephone follow-up group of 17 patients received telephone follow-up 10 min once a week for 1 month and 10 min twice a month for 2 months during 12 weeks compared to control group of 22 who patients received usual care. Results: The telephone follow-up group showed no significant changes in patient compliance and urea kinetic modeling, although patients with CAPD receiving a telephone follow-up showed reduced incidence of exit site infection during the intervention compared with the control group. Conclusion: Telephone follow-up program for CAPD patients may decrease the incidence of exit site infection. Future studies with a larger sample be done utilizing diet and exercise reinforcement program combined with telephone follow-up as a nursing intervention for patients with CAPD.

Decomposition and N Release of Hairy Vetch Applied as a Green Manure and its Effects on Rice Yield in Paddy Field

  • Decomposition of green manure is necessary for the nutrient supply in farm soil. Hairy vetch as a green manure is superior to other winter legumes in terms of wintering ability and N accumulation. This experiment was carried out to investigate the decomposition and N release of hairy vetch and its effect on rice production as the following crop in paddy field. Decomposition of hairy vetch placed by soil depth of 0, 10 and 20cm at transplanting time was investigated by mesh bag method, which was enclosed chopped residue in mesh bags. The fate of $^{15}$ N derived from $^{15}$ N-labeled hairy vetch was investigated at harvest in three levels of N fertilization. Grain yield of the transplanted paddy rice cultured with hairy vetch as starter N were compared with that of applying urea as starter N in the field. Hairy vetch residue decomposed very rapidly both in transplanted and dry-seeded paddy field. In transplanted paddy field, hairy vetch residue lost 72-81 % and 86-90% of its weight after one and five month, respectively, as affected by incorporation depth. The C/N ratio of the decomposing vetch residue increased sharply during the early stages and after then, decreased slowly. The amounts of N and P released from the vetch were about 90% and 97% of initial content after one month, respectively. Recoveries of hairy vetch-$^{15}$ N by rice plant were 30.6, 34.6 and 35.7% in 0, 6 and 12 kg urea-N 10 $a^{-l}$ application, respectively, indicating that N fertilization increased the recovery of hairy vetch. $^{15}$ N. Hairy vetch residue incorporated as starter maintained significant N $H_4$$^{+}$-N concentration in soil water of plow layer until effective tillering stage. Grain yield in the plot applied with hairy vetch was not significantly different from that in the plot with urea. We concluded that hairy vetch incorporation could substitute starter N fertilization and showed possibility to reduce N amount of top-dressing.g.g.

Evaluation of Invasive and Noninvasive Methods for the Diagnosis of Helicobacter Pylori Infection

  • Objective: The present study was conducted to evaluate invasive and noninvasive diagnostic methods for detection of Helicobacter pylori (H. pylori) in patients admitted with dyspeptic complaints and to compare sensitivities and specificities. Method: Sets of four gastric biopsy specimens were obtained from a total of 126 patients included in the study. The presence of H. pylori was determined by invasive tests including culture, rapid urease test, polymerase chain reaction (PCR) and histopathology. Among noninvasive tests, urea breath test, serological tests and enzyme-linked immunosorbent assay (ELISA) were performed. Results: H. pylori was isolated in 79 (62.7%) gastric biopsy cultures, whereas positivity was concluded for 105 (83.3%) patients by rapid urease test, for 106 (84.1%) by PCR, for 110 (87.3%) by histopathology, for 119 (94.4%) by urea breath test, and for 107 (84.9%) by ELISA. In the present study, the culture findings and histopathological examination findings were accepted as gold standard. According to the gold standard, urea breath test had the highest sensitivity (96.5%) and the lowest specificity (30%), whereas culture and histopathology had the highest specificities (100%). Conclusion: The use of PCR invasively with gastric biopsy samples yielded parallel results with the gold standard. PCR can be recommended for routine use in the diagnosis of H. pylori.

Experimental and theoretical investigation of micellization behavior of sodium dodecyl sulfate with cetyltrimethylammonium bromide in aqueous/urea solution at various temperatures

  • Mixed micelle formation behavior of cationic surfactant-cetyltrimethylammonium bromide (CTAB) and anionic surfactant sodium dodecyl sulfate (SDS) in aqueous as well as in urea medium from 303.15 K to 323.15 K at 5 K interval was carried out by conductometric method. The differences between the experimental values of critical micelle concentrations (cmc) and ideal critical micelle concentrations ($cmc^{id}$) illustrate the interaction between the amphiphiles studied. The values of micellar mole fraction ($X_1^{Rub}$ (Rubingh), $X_1^M$ (Motomura), $X_1^{Rod}$ (Rodenas) and $X_1^{id}$(ideal) of surfactant CTAB determined by different proposed models and outcome indicate high involvement of CTAB in SDS-CTAB mixed micellization, which enhance by means of the augment of mole fraction of CTAB. The negative value of interaction parameter (${\beta}$) showed an attractive interaction involving CTAB and SDS. Activity coefficients were less than unity in all case, which also reveals the presence of interaction between CTAB & SDS. The negative ${\Delta}G^0_m$ values imply the spontaneous mixed micellization phenomenon. The attained values of ${\Delta}H^0_m$ were positive at inferior temperature, while negative at superior temperature. The negative ${\Delta}H^0_m$ values in urea ($NH_2CONH_2$) medium illustrate exothermic micellization process. The magnitudes of ${\Delta}S^0_m$ were positive in almost all cases. The excess free energy of mixed micelle formation (${\Delta}G_{ex}$) was found to be negative, which indicates the stability of mixed micelle as compared to the individual's components micelles.

Luminescence behaviour of rare earth doped alkaline earth aluminates synthesized by combustion method (연소법에 의한 rare earth doped doped alkaline earth aluminates 형광체의 발광특성)

  • A new method for the preparation of lanthanide ions activated strontium aluminates phosphor by combustion method has been proposed. Combustion method consist of the redox reactions between the respective metal nitrates and urea in a preheated funace at $500^{\circ}C$. The luminescence behavior of the phosphor was studied and compared with corresponding phosphor prepared by conventional method. Effect of $Mg^{2+}$ ion concentration in strontium aluminate phosphor was investigated and the maximum luminescence of about $100cd/m^2$ was obtained. This method gave better brightness and small size to the phosphor than made by conventional method.
