운향과(芸香科) 식물(植物) 종실(種實)의 항성화성(抗醒化性) (Studies on the Antioxidative Substances in the Seeds of the Rutaceae Family)

  • Some seeds of the Rutaceae family, Zanthoxylum piperitum, Z. schinifolium officinalis, Poncirus trifoliata, Citrus unshin, were investigated to clarify their antioxidative components. Finely powdered samples were extracted by hexane, followed by dichioromethane and then 70% methanol in a hot bath. Its unsaponifiables containing X-and Y-tocopherol with trace amount of ${\beta}-and$\;{\delta}-tocopherol$. also showed comparatively weak activity, although the hexane fraction itself had no significant antioxidative effect on lard. Levels of total tocopherols in the samples averages 42. 24-154. 11 mg/lOOg total extractives. The dichloromethane-and 70% methanol extractives showed strong antioxidative activity, from which antioxidative substances were purified with benzene-acetone(6:5, V/V) on a silica gel column, and with a solvent mixture of acetonitrile-methanol-$H_2O$(40:40:20, V/V/V) on a Sep-Pak $C_{18}$ hydrolyzed by 5% KOH-ethanol. The recovered unsaponifiables were, then, separated on a column of high performance liquid chromatography. The unsaponifiables produced by hydrolysis of the isolates from dichloromethane extractives has epi-catechin(40.0-57.1%) and (+)-catechin<$l9.1{\sim}24.4%$ to total phenolic substances, on area base) as major component, accompanied by chlorogenic acid, gallic acid(?), trans-p-coumaric acid and tralls-p-ferulic acid including some unknown components, and those derived from 70% methanol extractives also comprise (+)-catechin($31.3{\sim}39.6%$ to total components, on area base), epi-catechin($2O.2{\sim}36.4%$), trans-p-cournaric acid(8.4-15.3%) and trans-p-ferulic acid($7.7{\sim}14.1%$) as predominant component with some minor coponents, but the fraction supposed to be gallic acid(?) is not present. The antioxidative activities of the phenolic components isolated in this work were in order of epi-catechin>catechin>chlorogenic acid>trans-p-ferluic acid>trans-p-coumaric acid.

유지 불검화물과 식물체 추출물이 식용유의 $180^{\circ}C$ 가열 산화에 미치는 효과 (Effects of Oil Unsaponifiables and Plant Extracts on the Thermal Oxidation of Oils at $180^{\circ}C$)

  • 여러 종류(8종)의 유지로 부터 추출한 불검화물과, 강력한 자동산화억제력이 있다고 알려진 53종의 식물체(농산물 및 생약) 메탄을 추출물을 유지에 첨가한후 $180^{\circ}C$에서 14 혹은 16시간 연속 가열하는 동안 유지의 지방산조성을 측정하여 이들 추출물들이 유지의 고열 가열 산화에 미치는 영향을 연구하였다. 추출된 유지의 불검화물들은 모두 $180^{\circ}C$의 고온 가열산화 조건에서 항산화력을 나타내지 못하였다. 그러나 53종의 농산물 및 생약중에서는 일부 종들은 강력한 가열산화 억제력을 나타내었다. 특히, 대황, 백굴체, 정향 등의 메탄올 추출물은 강력한 가열산화 억제력을 나타내었다. 또한 이들 대황, 백굴체 및 정향의 메탄올 추출물들이, 면실유를 $180^{\circ}C$에서 1일 7시간씩 8일간 가열 처리하는 과정에서 면실유의 linoleic acid 파괴 및 중합체 생성에 미치는 영향을 연구하였다. 이 결과 정향 및 백굴체추출물은 $180^{\circ}C$에서 장기간의 가열조건에서도 linoleic acid 파괴 억제 및 중합체 생성을 억제하는 효과가 크게 나타낸 반면, 대황의 경우에는 장기 가열산화 조건에서 지방산의 파괴에 대한 억제력은 미미하였으나, 중합체 생성억제력은 비교적 양호하였다.

참깨 저장중(貯藏中) 지질(脂質)의 특성(特性) 및 Sterol의 조성(組成) 변화(變化) (Changs in Characteristics and Sterol Composition of Sesame Seed Lipid during Storage)

  • 참깨종실(種實)을 56개월(個月) 저장(貯藏)한 후 함유량(含油量)은 참깨 100g당(當) 수확 직후(直後) 50.3g이 45.5g으로 감소(減少)되었고 산가(酸價)는 1.8이 13.4로 크게 증가(增加)하였으나 sterylester는 9.9mg이 6.7mg으로 sterylglycoside가 3.8mg에서 8.3mg으로 크게 변화(變化)하였고 불(不)감화물(化物)은 24.3mg이 20.9mg로 감소(減少)되였으며 분회별(分劃別) sterol은 4,4-dimethylsterol 및 4-monomethylsterol이 감소(減少)한 반면(反面) 4-desmethylsterol은 78.5%가 90.7%로 증가(增加)되었다. 4-desmethylsterol의 sterol 조성변화(組成變化)는 campesterol 및 stigmasterol은 거의 변화(變化)가 없으나 sitosterol은 51.0%에서 48.2%로 감소(減少)한 반면(反面) ${\Delta}^5-avenasterol$은 15.2%에서 18.4%로 증가(增加)되었다.

  • An attempt was made to produce by using Rh. gracilis (NRRL Y-1091) in the same wav as currently considered siglecell protein production. The lipid content of Rh. gracilis were 56.9% of dry weight. Economic coefficient (E. C.) and fat coefficient (F. C.) were 31.3 and 17.8, respectivelv. The lipid and fats extracted from Rh. gracilis were characterized as following: Specific gravity 0.875, A.V. 42.5, S.V. 220, I.V. 57.7, Phospholipid 1.58, Unsaponifiables 25.5, Volatile matter 18.7. Fatty acid composition of the lipids were analvzed by HPLC. In addition to the lipid analvsis, proteins were extracted from defatted Rh. gracilis and their amino acid compositions were examined by amino acid analyzer.

  • 오이, 수박, 수세미, 박, 하늘수박의 다섯가지 박과식물 종자유에서 세가지 스테롤 플랙숀을 박층 크로마토그래피로 나누고 각 플랙숀의 조성을 가스크로와 가스크로에 연결된 질량분석법으로 밝혔다. 이중 4-데스메틸스테롤 플랙숀에서 ${\alpha}$-Spinasterol, ${\Delta}^{7,22,25}$-stigmastatrienol 및 ${\Delta}^{7,25}$-stigmastadienol를 단리(單離)하여 IR, NMR 및 질량분석법으로 동정(同定)하였다.

옥수수 종실 및 속대의 Phytosterol 동정과 함량 변이 (Identification and Quantification of Phytosterols in Maize Kernel and Cob)

  • 옥수수 종실(kernel)과 속대(cob)의 불검화물(ZML) 중 phytosterol의 조성과 함량의 변이를 구명하여 고품질 옥수수 신품종 육성 및 생리활성물질의 유용 소재화를 위한 기초자료로 활용하고자 본 연구를 실시하여 얻어진결과를 요약하면 다음과 같다. 1. 옥수수 종실과 속대의 포화지방산은 phytosterol 함량과 부의 상관관계의 경향이었고, 불포화지방산 중 stearic 및 linoleic acid는 정상관 경향을 보였으나, linolenic acid는 종실에서 정상관($r=0.652^*$), 속대는 부상관($r=-0.505^*$) 관계를 보였다. 2. 옥수수 종실의 불검화물을 TLC로 분리한 결과 band I (campesterol, stigmasterol, ${\beta}$-sitosterol), band II (${\Delta}^5$-avenasterol), band III (${\Delta}^7$-stigmastenol), 및 band IV (${\Delta}^7$-avenasterol)로 뚜렷하게 분리되었고, 속대는 band I~IV 이외에도 3종 이상의 band가 추가적으로 분리되었다. 3. 옥수수 종실과 속대에 함유된 phytosterol의 GC 분리 패턴을 확인한 결과 campesterol, stigmasterol 및 ${\beta}$-sitosterol의 분리능이 좋았으나, ${\Delta}^7$-avenasterol (RT 22.846), ${\Delta}^7$-stigmastenol (RT 22.852) 및 ${\Delta}^5$-avenasterol (RT 22.862)은 혼합물질 상태로는 분리가 되질 않았다. 4. 옥수수 종실의 평균 phytosterol 함량은 635.9 mg/100g, 속대는 273.0 mg/100 g으로 종실이 속대에 비해 약 2.4배 정도 phytosterol 함량이 높았다. 옥수수 종실의 phytosterol 조성은 ${\beta}$-sitosterol 80.05% > campesterol 10.5% > stigmasterol 9.46% 순이었으나, 속대는 ${\beta}$-sitosterol 59.43% > stigmasterol 31.72% > campesterol 10.98%으로 종실과 속대의 phytosterol 조성비는 다소 상이하였다. 5. 본 연구 결과를 토대로 판단할 때 옥수수 종실에 함유된 ${\beta}$-sitosterol, campesterol 및 stigmasterol의 생합성 경로에서 전구물질이 되는 ${\Delta}^7$-avenasterol, ${\Delta}^7$-stigmastenol 및 ${\Delta}^5$-avenasterol이 옥수수 속대에서 검출되는 것으로 보아 옥수수 종실의 phytosterol은 속대에서 합성되어 종실로 전이되는 물질인 것으로 추정되었다.

한국산 식용패류중의 지방질성분에 관한 연구 (Studies on the Lipid Components of Edible Shellfishes in Korea)

  • This study was designed to elucidate the lipid contents, neutral lipids components and fatty acid composition in fresh shellfishes, produced in Korea. Four kinds of shellfishes including sea mussel, short-necked clam, corb shell and and ark shell were selected according to the higher sales order and cheaper retail price at fish markets in Seoul in July 1985. The results abtained were as follows; 1. The average total lipid contents in four shellfishes were 2.3% by wet weight basis. The ratios of neutrial lipid, glycolipid and phospholipid in the total lipid were 51.1 : 4.9 : 44.0 in sea mussel, 66.0 : 3.2 : 30.8 in ark shell, 37.8 : 2.2 : 60.0 in short-necked clam and 54.5 : 2.0 : 53.5 in corb shell, 2. The average value of acid value, iodine value and unsaponifiables contents of total lipids were 1.3, 217.8, 92.0 and 20.3%, respectively. 3. The composition of the neutral lipids were triglycerides, esterified fatty acids, sterylesters, free sterols and monoglycerides in four shellfishes. 4. The major fatty acid composition of total lipids were palmitic, eicosapentaenoic, docosahexaenoic and palmitoleic acids in four shellfishes. The average total unsaturated fatty acids of total lipid were 64.5%, and $\omega$-3 highly unsaturated fatty acids were 27.0%. The average p/s Ratiio were 1.3.

참깨의 품질평가 현황과 전망 (Current Status and Prospects of Quality Evaluation in Sesame)

  • Sesame (Sesamum indicum L.) is probably the most ancient oilseed crop known in the world. Sesame seed is known for its high nutritional value and for having oil (51%) and protein (20%) content. The fatty acid composition of sesame oil is palmitic acid (7.8%), stearic acid (3.6%), oleic acid (45.3%), and linoleic acid (37.7%). Sesame oil is characterized by a very high oxidative stability compared with other vegetable oils. Two lignan-type compounds, sesamin and sesamolin, are the major constituents of sesame oil unsaponifiables. Sesamol (a sesamolin derivative) can be present in sesame seeds and oils in very small amount. Other lignans and sesamol are also present in sesame seeds and oils in very small amount as aglycones. Lipid oxidation activity was significantly lower in the sesamolin-fed rats, which suggests that sesamolin and its metabolites contribute to the antioxidative properties of sesame seeds and oil and support that sesame lignans reduce susceptibility to oxidative stress. Sesaminols strongly inhibit lipid peroxidation related to their ability to scavenge free radical. The sesame seed lignan act synergistically with vitamin I in rats fed a low $\alpha$-tocopherol diet and cause a marked increase in a u-tocopherol concentration in the blood and tissue of rats fed an $\alpha$-tocopherol containing diet with sesame seed or its lignan. The authors are reviewed and discussed for present status and prospects of quality evaluation and researched in sesame seeds to provide and refers the condensed informations on their quality.

동백(冬柏)을 위시한 차나무과(科) 식물(植物) 종실(種實)에 함유(含有)된 항산화제(抗酸化劑)에 관한 연구(硏究) (Studies on the Antioxidative Substances in the Seeds of Some Theaceae Family)

  • Dried seeds of Camellia japonica and Thea sinensis were investigated to determine the nature of their antioxidative activity. Activity was measured by the induction period in the coupled oxidation of a substrate lard and extracts or isolates to be tested. 70% methanol and dichloromethane extracts were found to be antioxidative abilities. Their unsaponifiables revealed weak antioxidative activity, although hexane extracts did not show antioxidative effect on lard. Column chromatography for dlchloromethane extracts gave 4 fractions(only 2 fractions were potent). HPLC was used in isolating potent antioxidative components from the column fractions and the precolumn-passed methanol extracts. They were separated into 7 and 8 components, respectively. The column fractions obtained from both seeds comprised trans-p-coumaric acid. trans-p-ferulic acid and an unknown component with minor components such as chlorogenic acid and catechin. On the other hand, the most prominent components in the methanol extracts were an unidentified component. trans-pcoumaric acid, trans-p-ferulic acid, catechin and chlorogenic acid. The unknown compound isolated from the column fractions and methanol extracts was identified as epicatechin by $^1H-and\;^{13}C-NMR$. The antioxidative activities of these components were epicatechin > catechin > chlorogenic acid > trans-p-ferulic acid > trans-p-coumaric acid.
