DNA Fingerprinting Analysis of the Genus Phytophthora in Korea

  • In order to investigate biodiversity and establish identification system for Phytophthora spp. in Korea, a variety of band pattern was produced by using the URP(universal rice primer). The fingerprint patterns of Phytophthora spp. showed many common and variable fragments according to their isolates in distinct genotypes. In particular, P. drechsleri was classified into four distinct types(I to IV). P. drechsleri(KACC 40498 and KACC 40499) and P. cryptogea(KACC 40413) appeared to have almost equal bands despite their being different species. Ninety isolates of Phytophthora spp. were clustered into 13 groups based on UPGMA(unweighted pair group method with arithmetic means) analysis. These DNA fingerprinting data would be helpful for inter- and intra-species identification of Phytophthora species.

느티만가닥버섯 균주의 형태 및 유전적 유연관계 분석 (Analysis of Morphological and Genetic Relationships amomg Isolates of the Artificially Cultivated Mushroom, Hypsizygusmarmoreus)

  • Hypsizygus marmoreus의 형태적 특성과 유전적 유연관계를 조사하기 위해 한국 및 다른 국가에서 111개의 균주를 수집하였다. RAPD (Random amplified polymorphic DNA) 및 ITS rDNA 염기서열 분석을 이용하여 H. marmoreus 균주간의 유전적관계를 확인하였다. URP-PCR을 이용한 RAPD 분석결과, H. marmoreus의 모든 균주는 크게 3개의 그룹으로 분류되었으나, 90% 이상의 높은 유사성을 보였다. 또한 형태적 및 지리적으로 분류가 가능하였으나 갈색 균주 및 흰색 균주는 구별되지 않았다. 따라서 형태적, 지리적 차이가 있는 16개의 균주를 선별하여, 640 bp 길이의 ITS 염기서열을 정렬하고 비교하였다. ITS 염기서열의 유사성은 공시균주와 GenBank의 H. marmoreus 균주 사이에서 94.8-99.1 %였다. 수집지역과 형태에 차이가 있었지만, 실험에 사용된 공시균주는 모두 H. marmoreus에 가까운 유연관계를 형성하였다.

Genetic Differentiation of Pseudomonas syringae Pathovar tomato from Other P. syringae Pathovars using REP-PCR and URP-PCR

  • For the genetic differentiation of $Pseudomonas$ $syringae$ pathovar $tomato$, a total of 51 $P.$ $syringae$ pv. strains infecting 33 different host plants were analyzed using repetitive element PCR(REP-PCR) and universal rice primer PCR(URP-PCR). The entire DNA fingerprint profiles were analyzed using unweighted pair-group method with arithmetic averages (UPGMA). The 51 $P.$ $syringae$ pv. strains could be divided into five clusters based on 65% similarity by Rep-PCR using BOX, ERIC, and REP primers. $P.$ $syringae$ pv. $tomato$ cluster was well separated from other 31 $P.$ $syringae$ pathovars. $P.$ $syringae$ pv. $tomato$ cluster included only $P.$ $syringae$ pv. $maculicola$ and $P.$ $syringae$ pv. $tomato$. $P.$ $syringae$ pv. $tomato$ strains could be divided into two genetic groups. Meanwhile, the Pseudomonas pv. strains could be divided into four clusters based on 63% similarity by URP-PCR using 2F, 9F, and 17R primers. $P.$ $syringae$ pv. $tomato$ cluster was also well separated from 30 other $P.$ $syringae$ pathovars. In this case, $P.$ $syringae$ pv. $tomato$ cluster included $P.$ $syringae$ pv. $maculicola$, $P.$ $syringae$ pv. $berberidi$, and $P.$ $syringae$ pv. $tomato$. $P.$ $syringae$ pv. $tomato$ strains was also separated into two genetic groups by URP-PCR analysis. Overall, our work revealed that $P.$ $syringae$ pv. $tomato$ can be genetically differentiated from other $P.$ $syringae$ pathovars by the DNA fingerprint profiles of REP-PCR and URP-PCR. We first report that there are two genetically diverged groups in $P.$ $syringae$ pv. $tomato$ strains.

가지과 작물에서 분리한 Alternaria 속 균의 형태적, 분자생물학적 특징 (Morphological and Molecular Characterization of Alternaria Isolates from Solanaceous Crops)

  • 국내의 가지과 작물에서 분리한 Alternaria 25 균주를 공시하여 분생포자의 형태적 특징을 조사하였고 A. solani와 A. tomaotphilad의 대표균주와 비교하였다. 분리균주 중 형태적으로 구별되는 몇 균주들과 대표균주를 공시하여 감자, 토마토, 가지, 고추에 대한 병원성 검정을 실시하였다. 대표균주를 포함한 Alternaria 17개 균주를 공시하여 ITS 영역과 histone h3 유전자의 염기서열분석, URP (universal rice primer)에 의한 핵산지문분석을 실시하였다. Alternaria 균주들은 분생포자의 형태적 특징에 의하여 A. tomaotophila(ATO), A. solani(ASO) 및 미동정의 Alternaria sp.(ASP)의 group으로 나눌 수 있었고 A. solani(ASO)는 분생포자 부리(beak)의 특징에 의하여 다시 ASO(1)과 ASO(2)의 2 type으로 나눌 수 있었다. ATO와 ASO 균주들은 실험에 사용한 감자, 토마토, 가지, 고추에 모두 병원성이 있었고 ATO 균주는 특히 토마토에 강한 병원성이 있었으나, ASP 균주는 감자에만 병원성이 있었다. 이 연구에서 사용한 molecular marker중 ITS와 histon H3 유전자의 염기서열 분석은 4개의 형태적 group을 명확히 구분할 수 없었지만, URP-PCR 핵산지문분석법은 이들 group을 명확히 구분할 수 있었다. 분생포자의 형태, 병원성검정, 분자생물학적 특징을 종합할 때 토마토 겹무늬병에 관여하는 A. tomatophila는 감자겹둥근무늬병에 관여하는 A. solani와 뚜렷이 구분할 수 있었다. 또한 감자에서 분리한 Alternaria sp.(ASP)는 A. solani(ASO)와 형태적으로 유사하였지만 분생포자의 폭이 두껍고 부리(beak)가 작다는 점에서 ASO와 구별되었으며 가지과 작물에서 보고된 바 없는 신종으로 추정되었다.

PCR 다형성에 의한 양송이(Agaricus bisporus) 계통의 유전적 다양성 분석 (Genetic Diversity of Agaricus bisporus Strains by PCR Polymorphism)

  • 본 연구에서는 국내외에서 수집한 A. bisporus 45계통과 Agaricus spp. 19균주를 포함한 64균주로부터 genomic DNA를 추출하고 12종류의 UFPF primer (JK Biotech., Ltd.)를 사용하여 PCR 다형성 분석을 실시한 바 양송이 계통간 PCR다양성이 관찰되었다. 7종류의 UFPF primer에 의해 생성된 A. bisporus계통의 95 PCR다형성 밴드가 유전적 유사도 산출에 이용되어 UPGMA cluster분석을 적용 dendrogram을 작성한 결과 A. bisporus의 계통군은 3 cluster 내에 8개의 세부 group으로 분류되었으며 유사성 함수가 0.75에서 0.90의 유연관계를 보였다. 한국에서 최근 개발된 새아, 새정, 새연, 새도는 같은 group에 포함되었으며 0.96%에서 1.0%의 밀접한 근연관계에 있었으며 그밖에 한국품종은 미국 유럽의 양송이 품종과 유전적 유연관계를 보였다.

Characterization of Newly Bred Cordyceps militaris Strains for Higher Production of Cordycepin through HPLC and URP-PCR Analysis

  • Cordyceps militaris, a member of Ascomycota, a mushroom referred to as caterpillar Dong-chung-ha-cho, is commercially valuable because of its high content of bioactive substances, including cordycepin, and its potential for artificial cultivation. Cordycepin (3'-deoxyadenosine) is highly associated with the pharmacological effects of C. militaris. C. militaris is heterothallic in that two mating-type loci, idiomorph MAT1-1 and MAT1-2, exist discretely in two different spores. In this study, nine C. militaris strains were mated with each other to prepare newly bred strains that produced a larger amount of cordycepin than the parent strains. Nine strains of C. militaris were identified by comparing the internal transcribed spacer sequence, and a total of 12 single spores were isolated from the nine strains of C. militaris. After the MAT idiomorph was confirmed by PCR, 36 mating combinations were performed with six single spores with MAT1-1 and the others with MAT1-2. Eight mating combinations were successfully mated, producing stroma with perithecia. Cordycepin content analysis of all strains by high-performance liquid chromatography revealed that the KASP4-bred strain produced the maximum cordycepin among all strains, regardless of the medium and stroma parts. Finally, universal rice primer-PCR was performed to demonstrate that the bred strains were genetically different from the parental strains and new C. militaris strains. These results may be related to the recombination of genes during mating. The newly produced strains can be used to meet the industrial demand for cordycepin. In addition, breeding through mating suggests the possibility of producing numerous cordycepin-producing C. militaris strains.

Molecular Identification and Effects of Temperature on Survival and Growth of Hybrids between Haliotis gigantea Gmelin (♀) and Haliotis discus hannai Reeve (♂)

  • In abalones, interspecific hybridization has been suggested as a possible means to increase production and desired traits for the industry. In Korea, Haliotis gigantea is considered a species with a larger size and higher temperature tolerance than H. discus hannai. However, H. discus hannai is considered the most valuable and popular fishery resource due to its better acceptance and higher market prices. Thus, viable interspecific hybrids have been produced by artificial inseminating H. gigantea eggs with H. discus hannai sperm. However, the reciprocal hybrid cross was not successful. In this study, the hybridity and the growth and thermal tolerance performance of the interspecific hybrids were examined. A combination of various assays revealed maximum growth occurrence at 21℃ and the higher growth rate in the hybrids than that of H. discus hannai parent. In addition, the growth and survival at high-temperature (28℃) of the hybrids was equivalent to that of the highly tolerant H. gigantea parent, suggesting new possibilities to overcome the mass mortality in H. discus hannai during high temperature periods of summer season in Korea. Furthermore, the induced interspecific hybrid status was confirmed by the presence of species-specific bands for each parental species of the random amplified polymorphic DNA (RAPD) profiles using universal rice primer (URP), which could be used as speciesspecific markers to distinguish the hybrids and their parental species.
