Universal-, Genus-specific, Species-specific Probes and Primers Design for Microbial Identification

  • MIPROBE is a web-based tool for design of universal, genus-specific, and species-specific primers and probes. The main functions of MIPROBE are collection of target gene sequences, construction of consensus sequences, collection of candidate primers and probes, and evaluation of candidates by BLAST. Biologists with little computer skills can easily use MIPROBE to design large-scale universal, genus-, and species-specific primers and probes. This software is available at http://www.miprobe.com. Also detailed descriptions of how to use the program are found at this site.

The study on Zhong-lie-xiao-wu-yi(『忠烈小五義』)'s transmission and the story (『충렬소오의(忠烈小五義)』의 국내유입과 스토리 연구)

  • Zhong-lie-xiao-wu-yi(忠烈小五義), whose author was Shiyukun(石玉昆), is a Xia-Yi-Gong-An(俠義公案) novel in the late Qing Dynasty. This work published in 1890 when Emperor Guangxu(光緖) governed China. This work's author is Shiyukun, distribution books has an amender. The amender will be a shuoshuyiren (說書藝人). Zhong-lie-xiao-wu-yi is Zhong-lie-xia-yi-zhuan(忠烈俠義傳)'s a sequel, the story leads from Zhong-lie-xia-yi-zhuan. It is just the beginning of Zhong-lie-xiao-wu-yi is redundant. Zhong-lie-xiao-wu-yi was introduced to the late Chosun(朝鮮) Dynasty. This work was translated in Hangeul, Chosun's readers read Zhong-lie-xiao-wu-yi. This work's circulation is not clear, But this work's exciting story is interested in the readers. This work is characterized as follows: First of all, Zhong-lie-xia-yi-zhuan's charaters appear equally, the readers feels familiar. The readers like the familiar characters, because the readers read the book. The familiar characters can have a sense of speed in reading. Second, the story is continuous. Zhong-lie-xiao-wu-yi is narrated by connecting Zhong-lie-xia-yi-zhuan's story. Third, Zhong-lie-xiao-wu-yi was seeking an open ending. Classical novels prefer happy ending, this work is open ending, the expectations for the sequel became more doubled. The fourth, this work took advantage of the colloquial expressions. Zhong-lie-xiao-wu-yi is Huabenti(話本體) novel, took advantage of the spoken language. Suyu(俗語) and xiehouyu(歇後語) was represented in this work. Fifth, this work is formed a universal consensus. Ordinary people must empathize about xia-yi(俠義) and retribution, this work was well represented. Because the readers would have liked to this story.

  • The Broadcasting Law amended in January 2007 declared to adopt universal right to view(known more widely as Universal Access Right, UAR), the right to access broadcasting programs on such major sports or other events that are likely to catch the gaze of the public television viewers. Then its implementation rule was issued in February 2008, and under the regulations the Committee of Ensuring Universal Access Right has been established, where detailed action measures and guidelines are currently in the process of preparing. However, it is no question that an effectiveness of the implementation system of Universal Access Right presupposes sufficient amount of discussion and social consensus. At present, the major focus in this issue is on the matters including which type of events is subject to UAR and what criteria are desirable in determining which broadcasting company has priority. In this context, this study aims at identifying implications for policy by examining the precedent cases of Europe, Australia, and other countries, where UAR is enacted and implemented. Further, this study tries to draw up a specific scheme for ensuring universal right to view through conducting a survey on public television viewers. We will include specific guidelines for selecting events of public attention, criteria for selecting broadcasting companies regarding priority, and relevant operating rules regarding relayed broadcasting.

  • The power of the internet community as a social network exerts great influence on all areas of our society not only on knowledge creation and knowledge sharing but also on politics, economy, and culture. This study aims to investigate how the diffusion process of information evolves on the internet in the new environment. The focus of the study is particularly on aspects of using information and the process of information production and the types and methods of online communities. It is the goal of the study to clarify aspects of the online community as an important mediator among universal means of communication in our society. Ways of social communication are changing rapidly and thus the reasoning for this change is very important policy issues to form a social consensus.

  • To gain competitive advantage in a fast changing environment, the higher education institution (HEI) must continuously adjust the strategies to that environment. One important strategy is how to determine appropriate practical actions based on what students really need and want. Despite the abundance of research on service quality management, there is no universal consensus on how best to determine appropriate practical actions in HEIs. The aim of this paper is to develop an integrated model to be used to accurately acquire the most critical service attributes and determine appropriate actions that promote student satisfaction. Drawing on relevant literature, an integrated model is proposed which is based on students' perspective by integrating the fuzzy SERVQUAL, refined Kano, and Blue Ocean model. Subsequently, an empirical case study in the higher education sector is described that illustrates the value of the model in determining the most critical attributes and how to improve them.

  • The March 2022 presidential election held at the peak of the COVID-19 pandemic drew flak for undermining the principle of universal suffrage by failing to guarantee properly the voting rights of confirmed and quarantined persons. Guaranteeing their voting rights requires thinking about e-voting that can fundamentally overcome the temporal and spatial limitations of current paper voting polling stations. The question is how to deal with the increased possibility of contradicting or violating the principles of equality and direct and secret suffrage due to the expansion of universal suffrage. In order to obtain implications for this, we looked at the case of Estonia, which has been holding 11 national elections without any problems since the introduction of e-voting in 2005. Estonia was successfully building trust in the system, government, and society through the institutionalization and routinization of the overall socio-technical system of e-voting, along with political and constitutional agreements on the principles of elections. Therefore, we should not only consider the possibility of e-voting in terms of technological development and level but also discuss the establishment of trust by mediating conflicts between election principles from a normative point of view to reach a social consensus.

  • Although providing universal coverage for health care through the National Health Insurance(NHI) is a remarkable achievement, the issue of limited benefit coverage of the NHI has been at the core of national debate over how to improve its coverage. This study aims to evaluate benefit extension strategies and implemented policies with regard to the NHI since 1989 using 'policy window theory' proposed by John W. Kingdon. Understanding problem stream, policy stream, political stream, and coupling streams regarding the NHI, in particular benefit extension, would contribute to broaden policy debates and to develop more effective strategies for the future. Historically, political stream had opened policy window in the past two decades and policy streams can be characterized by three waves. Three streams have been coupled since 2003 and the government had a strong will to fulfill better performance of NHI coverage. Study findings indicate that identification of problem structure regarding NHI benefit was not connected with policy stream tightly. In addition, there has been limited discussion on policy goal and principles for extension coverage of the NHI. Policy strategies to improve coverage of the NHI should be linked to characteristics of problem and sought solutions under the principle which is expected to be sustainable through consensus in the society.

Development of Genus- and Species-Specific Probe Design System for Pathogen Detection Based on 23S rDNA

  • Amplification by universal consensus sequences in pathogenic bacterial DNA would allow rapid identification of pathogenic bacteria, and amplification of genus-specific and species-specific sequences of pathogenic bacterial DNA might be used for genotyping at the genus and species levels. For design of probes for molecular diagnostics, several tools are available as stand-alone programs or as Web application. However, since most programs can design only a few probe sets at one time, they are not suitable for large-scale and automatic probes design. Therefore, for high-throughput design of specific probes in diagnostic array development, an automated design tool is necessary. Thus, we developed a Web-based automatic system for design of genus-specific and species-specific probes for pathogen detection. The system is available at http://www.miprobe.com.

Analysis of Health Promoting Schools: Focusing on Small-sized Elementary School in Rural Area (건강증진모델학교 분석: 읍면 지역 소규모 초등학교를 중심으로)

  • The purpose of the study was to present a model of WHO Health Promoting School easily applicable to small-sized schools in rural areas. Methods: The study analyzed data of 11 small-sized schools in rural areas selected from 85 health promoting schools, an initiative led by the Ministry of Education from 2012 to 2014. Results: Through the analysis, the study found out the operation process of health promoting schools consists of five stages: system development, needs survey & survey on current status, school health policy development, program development & execution, and evaluation. In addition, the study was able to discover three key factors in operating health promoting schools: connection with curriculum, connection with community, and consensus among members. While it turned out the schools were following operation manuals faithfully, the biggest problem with operation was that the stages of needs survey, priority setting, and core task development were not closely coordinated. Conclusion: Although the model suggested in the study fails to reflect the characteristics of small-sized schools in rural areas, it is close to a universal model which could serve as a guideline when regular schools adopt the system of health promoting schools.

A Universal Analysis Pipeline for Hybrid Capture-Based Targeted Sequencing Data with Unique Molecular Indexes

  • Hybrid capture-based targeted sequencing is being used increasingly for genomic variant profiling in tumor patients. Unique molecular index (UMI) technology has recently been developed and helps to increase the accuracy of variant calling by minimizing polymerase chain reaction biases and sequencing errors. However, UMI-adopted targeted sequencing data analysis is slightly different from the methods for other types of omics data, and its pipeline for variant calling is still being optimized in various study groups for their own purposes. Due to this provincial usage of tools, our group built an analysis pipeline for global application to many studies of targeted sequencing generated with different methods. First, we generated hybrid capture-based data using genomic DNA extracted from tumor tissues of colorectal cancer patients. Sequencing libraries were prepared and pooled together, and an 8-plexed capture library was processed to the enrichment step before 150-bp paired-end sequencing with Illumina HiSeq series. For the analysis, we evaluated several published tools. We focused mainly on the compatibility of the input and output of each tool. Finally, our laboratory built an analysis pipeline specialized for UMI-adopted data. Through this pipeline, we were able to estimate even on-target rates and filtered consensus reads for more accurate variant calling. These results suggest the potential of our analysis pipeline in the precise examination of the quality and efficiency of conducted experiments.
