Ultraviolet A Induces Immunosuppression, Protection or Memory Enhancement Depending on Dose, while Ultraviolet B is Immunosuppressive and Tolerogenic over a Large Dose Range

  • UVR-induced immunosuppression contributes to skin cancer. The aim was to construct accurate dose response curves for primary and secondary contact sensitivity for solar-simulated UVR (ssUVR; 290-400nm), UVA and UVB as the role of UVA in immunosuppression is controversial. We used a xenon arc source. The mice were immobilised, enabling accurate dosing. C57BL/6 mice were immunosuppressed at half the dose of ssUVR required to cause sunburn but not by higher doses (up to the sunburn dose). Thus, ssUVR causes systemic immunosuppression only over a narrow, low dose range. UVA caused suppression at low but not high doses whereas UVB induced immunosuppression at all doses tested. 8 weeks later the mice were resensitised to assess tolerance. Mice exposed to the minimum immunosuppressive dose of ssUVR prior to primary sensitisation were tolerant to re-sensitisation. However, at higher doses of ssUVR, these mice were protected from tolerance. Interestingly, while low doses of UV A caused immunosuppression, even lower doses enhanced the response to the second sensitisation. Higher doses of UVA had no affect. UVB induced tolerance in a dose related manner. Thus, ssUVR only induces immunosuppression and tolerance over a narrow dose range. Both UVA and UVB are immunosuppressive at this dose, while higher doses of UVA protect from the suppressive effects of UVB. Surprisingly very low doses of UVA enhanced memory development. Thus UVR has complex effects on the immune system depending on dose and spectrum.

마우스에서 보중익기탕이 자외선 B 조사에 의한 표피멜라닌세포 변화에 미치는 영향 (The Effect of Bu-Zhong-Yi-Qi-Tang on Epidermal Melanocytes in Ultraviolet B-irradiated Mice)

  • C57BL/6 마우스에서 자외선(Ultraviolet, UV) B 조사에 의한 표피 멜라닌세포의 변화에 대한 보중익기탕(Bu-Zhong-Yi-Qi-Tang, BZYQT)의 효과를 관찰하였다. 마우스에 UVB를 매일 $80\;mJ/cm^2$ (0.5 mW/sec)씩 7일간 조사하고, BZYQT를 UV 조사전 또는 조사 후에 복강내주사 또는 피부에 도포하여, 멜라닌세포 형성 억제효과 및 형성된 멜라닌세포에 대한 미백 효과를 dihydroxyphenylalanine (DOPA) 염색으로 관찰하였다. 마우스의 귀등쪽 표피를 분리하여 관찰한바, 정상대조군에서는 $mm^2$ 당 11-16개의 멜라닌세포가 관찰되었으며, UV 조사 일주일 후 발달된 가지를 가진 DOPA양성 멜라닌세포는 급격히 증가하였다. 멜라닌세포 형성 억제 실험에서는 평균치를 기준으로 복강 내 주사군에서 16.3%, 피부도포군에서 26.6%(p<0.01)의 효과가 관찰되었으며, 형성된 멜라닌세포의 감소 효과 실험에서는 평균치를 기준으로 복강 내 주사군의 경우 3주에 24.0%(p<0.01), 6주에 26.0%(p<0.01)의 효과가 관찰되었고, 피부도포군의 경우 3주에 5.2%, 6주에 12.5%(p<0.05)의 효과를 보였다. 이상의 결과는 BZYQT가 UV에 의한 멜라닌세포 형성 억제제 및 미백제로서의 적용 가능성을 제시하였다.

우엉 뿌리 추출물의 항산화 활성 및 세포 독성 평가 (Antioxidant Activity and Cytotoxicity on cell of Arctium lappa L. root extract)

  • 본 연구는 우엉 뿌리 추출물의 항산화 활성 및 피부에 대한 안전성을 평가하기 위하여 총 폴리페놀 함량, 총 플라보노이드 함량, DPPH radical 소거 활성을 통하여 항산화 활성을 살펴보고, B16F10 melanoma 세포에 대한 세포 독성 및 자외선 A에 대한 피부 세포 보호 효과를 확인하였다. 또한 화장품 소재로서의 활용을 검증하기 위하여 1차 피부 첩포 테스트를 실시하였다. 본 실험 결과 우엉뿌리 추출물의 함량이 증가됨에 따라 높은 폴리페놀과 플라보노이드의 함량이 확인되었으며, DPPH radical 소거 활성을 확인하였다. B16F10세포에 대한 세포 독성을 확인한 결과 B16F10 melanoma 세포에 대해 독성이 낮고, 자외선 A에 대해 80% 이상의 세포 보호 효과를 확인하였다. 또한 1차 피부 첩포 테스트를 통해 우엉 뿌리 추출물이 피부에 자극이 거의 없음을 확인하였다. 이러한 결과를 통하여 우엉뿌리 추출물이 항산화 활성과 자외선에 대한 피부 보호 효과가 뛰어나고 피부 세포에 대한 독성이 낮으며, 피부에 대한 안전성이 확인됨에 따라 화장품 소재로서의 가능성을 확인하였다.

UV-B조사로 유도된 접촉 과민반응에 대한 홍삼과 홍삼+EGb 761 혼합물의 억제 효과 비교 (Combined Effect of Korean Red Ginseng and EGb 761 on Ultraviolet B-induced Contact Hypersensitivity)

  • Exposure of skin to UV-B radiation can cause inflammatory response and immunosuppression. It has been reported that Korean Red Ginseng (KRG) has several pharmacological and physiological effects such as antioxidant, anticancer and improving immune function. In this study, we investigated that topical KRG and KRG + EGb 761 (Ginkgo biloba extract) combination prevented UV-B induced inflammation and inhibition of contact hypersensitivity response. Topical application of KRG, f days prior to or 5 days after exposure to 1MED and 2MED of UV-B, reduced skin thickness compared to non -treated group and resulted in protection against immunosuppression. However, KRG+EGb 761 combination has a little protection against the only 1MED UV-B. In conclusion. Topical application of KRG was more effective than combination in protection against UV-B induced inflammation and immune suppression. Also, we suggest that KRG can provide protection from inflammation and immunosuppression by UV-B radiation.

Effects of Egg Shell Membrane Hydrolysates on Skin Whitening, Wound Healing, and UV-Protection

  • This study was conducted to examine the effects of egg shell membrane hydrolysates (ESMH) on skin whitening, wound healing, and UV-protection. ESMH was divided into three groups by molecular weight (Fraction I: above 10 kDa of ESMH, Fraction II: 3 kDa-10 kDa of ESMH, Fraction III: below 3 kDa of ESMH). As a result, all of ESMHs showed over 90% of protein contents. The wound healing experiment using HaCaT cells showed that the fraction I was slightly superior to other fractions depending on the concentration though it was not significantly different. In the experiments of inhibition of tyrosinase and L-3,4-dihydroxyphenylalanine (L-DOPA) oxidation to verify the L-DOPA whitening effect, the whole ESMH (before fractioning) showed a similar amount of inhibition effect with arbutin (control). In the inhibition of melanin formation in B16-F1 melanoma cells, the fraction I showed a high inhibitory effect. In the experiment for protecting the skin from ultraviolet rays using HaCaT cells, all the fractions showed a higher rate of cell viability than the control. In conclusion, this study confirmed that the cosmetic effects of ESMHs such as skin whitening, wound healing, and UV-protection, which were divided depending on the molecule weight. We could confirm that the possibility of ESMHs as a material for functional cosmetics.

Formulation of Mamestra brassicae Nucleopolyhedrovirus-K1 as Viral Insecticide

  • The objective of our study was the formulation of a local strain of Mamestra brassicae nucleopolyhedrovirus-K1 (MabrNPV-K1) for the development of viral insecticide to control M. brassicae. To formulate MabrNPV-K1, feeding toxicities of various supplements and ultraviolet (UV)-protection were investigated. Optical brightener Tinopal UNPA-GX (Tinopal) as UV protectant and Bentonite had some toxicity themselves to increase the mortality. The protection of polyhedra from UV light radiation was observed only by Tinopal. The MabrNPV-K1 was formulated as a wettable powder form. The mortality of the formulation was higher and rapid than that of the un-formulated. This suggested the possibility of MabrNPV-K1 formulation as an effective biological control agent for M. brassicae.

자외선 흡수제 처리 면직물의 소비성능 개선(제1보) - 자외선 차단성능에 관한 연구 - (A Study on the UV-cut Properties of Cotton Fabrics Treated with UV-absorber)

  • The influence of ultraviolet(UV)-ray in sun light on human skin has been noted. Textiles can provide protection against harmful UV-radiation. Normally UV-absorbing finishes are used to get better protection. The purpose of this study is to evaluate the UV-cut properties of cotton fabrics treated with UV-absorber. 2,2-dihydroxy-4,4-dimethoxbenzophenone, as UV-absorber was applied to 100% cotton fabric. Reagents added in finishing solution were Triton X-100, polyethylene glycol 400, and $MgCl_2{\cdot}6H_2O$, and C.I. Direct Red 81. Both untreated and treated cotton fabrics were exposed to a xenon arc lamp for 20 and 80 hours. UV absorption spectra of finishing solutions and UV transmission spectra of fabrics were measured by the UV/VIS spectrophotometer. The results of this study can be summarized as follows. The results of this study can be summarized as follows. Absorption and the related transmission spectra were modified in a controlled way with UV-absorber. Absorption effect of UV-absorber was improved by adding Triton X-100, PEG 400, and $MgCl_2{\cdot}6H_2O$ in finishing solution. The UV absorption of finishing solution was in the following order: U/D/T/P/M>D/T/P/M> D/T> D/P, D>U/T/P/M>U/T>T/P/M>T. The UV transmittance of cotton fabrics was remarkably decreased by the application of UV-absorber and additives. The UV-cut properties were most improved by the application of U/D/T/P/M.

Agricultural Safety Clothing: Implications for Making and Selecting Clothes

  • Objective: The study was performed to suggest factors to consider when making or selecting protective clothing to enhance farmers' health. The purpose of this study was to review and summarize the agricultural safety technology on clothing including hat, gloves, and shoes. Background: The farmers' work clothing has to meet a multitude of requirement. Risk factors in farm-work were identified by previous researches. Agricultural safety clothing needs to be introduced. Method: We reviewed previous papers regarding the agricultural workers' health, safety clothing to protect body from excessive heat and humid environment, ultraviolet(UV) protection, and hazardous body working postures. Also the UV cut effect, pesticideproof clothing, and preventable farm work related musculoskeletal disorders were discussed. Results: The agricultural safety clothing needs to be developed to provide comfort and to be resistant against heat. The UV protection function on skin and eyes has not been fully researched in safety clothing studies yet. Conclusions and Application: The farmers' work-related body posture needs to be studied in order to make and design agricultural safety clothing. The safety issues on agricultural work shoes are also needed to be included in future studies.

Photoprotective Effect of Lotus (Nelumbo nucifera Gaertn.) Seed Tea against UVB Irradiation

  • Lotus (Nelumbo nucifera Gaertn.) seed is widely used as a traditional medicine in countries of Asia. Among many functions of the lotus seed, one interesting activity is its skin protection from the sunlight and scar. In this study, we focused on the skin protective property of lotus seed tea against ultraviolet B (UVB) irradiation. Two groups of a hairless mouse model, water as control (water group) and lotus seed tea (LST group), were administrated a fluid drink water for six months. After 6 month of administration, UVB exposure was carried out to both groups for another 3 months. During and after the administration, the skin moisture content and the morphological and histopathological analyses through biopsy were carried out. Prior to UVB irradiation, no significant difference was discovered in the skin moisture content for the water group and LST group (P<0.05). However, drastic changes were observed after the UVB treatment. The LST group showed a clear evidence of skin protection compared to the control group (P<0.05). The moisture content, epidermal and horny layer thickness, and protein carbonyl values all revealed that the intake of the lotus seed tea enhanced protection against UVB exposure. As a result, the long-term intake of the lotus seed tea showed the effect of preventing loss of skin moisture, mitigating the formation of abnormal keratinocytes, and contributing to protein oxidation inhibition.
