Ultrastructure of the Androgenic Gland of the Freshwater Prawn, Macrobrachium nipponense (징거미새우, Macrobrachium nipponense의 Androgenic Gland 미세구조)

  • The androgenic gland secretes a hormone, androgenic gland hormone, which is believed to act on the differentiation of the primary, secondary, and behavioral sex characteristics in most malacostracan crustaceans. This report presents the ultrastructural morphology of the androgenic gland in the freshwater prawn Macrobrachium nipponense. This gland, located in the coxopodite of the last pair of walking legs, was attached to the subterminal region of the sperm duct. The gland was composed of simple cellular strands, encased by a fibrous sheath. Microvilli were situated in the fibrous sheath, especially at the edge of each cellular strand. The androgenic gland cells had the large and round nucleus and rough endoplasmic reticulum arranged either in spirals or in concentric circles throughout the cytoplasm of the cell. They also had the well-developed Golgi complex and long mitochondria with flat and transverse cristae. The Golgi complex was similar to microvesicular cluster, but usually in the shape of typical dictyosomes, These features of androgenic gland cells coincides well with the protein/peptide secretion in their function. However, despite the apparent ultastructual equipment for protein/peptide secretion, no accumulation of materials secreted were noticed in the cytoplasm. Therefore it is strongly suggested that the transient transportation of the materials into the hemocoel has occurred just after synthesis.

A Study on the Effects of Sodium Salicylate on the Spiral Ganglion Cells (살리실산 나트륨이 백서 와우의 나선신경절에 미치는 영향에 대한 연구)

  • The ototoxic effects of salicylate on the ultrastructure of spiral ganglion cells were examined. Sodium salicylate($50{\sim}60\/kg$ body weight, once a day for 7 days) were injected subcutaneously to $5{\sim}6$ week-old fifteen Sprague-Dawley rats. Animals were sacrificed 24 hours (group 1), 6 weeks (group 2) or 10 weeks (group 3) after the last injection. In group 1 animals, distention of membranous cisternae was found in the cytoplasm of ganglion cells, satellite cells and Schwann cells in which enlargement or multicystic cytosome formation of the mitochondria were shown. In group 2 animals, membranous cisternae became larger or fused to form larger vacuoles or cysts. Shrinkage of spiral ganglion cell cytoplasm and loosening of myelin sheath were seen. In group 3 animals, extensive swelling or loss of nerve fibers were shown along with the folding or partial loss of myelin sheath which caused leakage of ganglion cell cytoplasm. It was concluded that the ototoxicity of salicy-late caused the ultrastructral changes of the spiral ganglion cells which became more severe in group 2 and 3 animals. The possibility of retrograde degeneration following the sensory cell changes was suggested.

  • The study on the nerve cells in the pars intercerebralis(IP) of 5-day-old cabbage butterfly Pieris rapae L. was performed to observe their ultrastructures and classify them on the basis. of the differences in size, shape and relative distribution cf cell organelles. The brain-subesophageal ganglion complex was fixed in 1% paraformaldehyde-1% gluaraldehyde mixture and embedded in araldite mixture. The transverse thin sections of IP were stained with uranyl acetate and lead citrate and examined by Hitachi 500 and ]EM 100B electron microscope. Five distinct types. of nerve cells are recognized and are arbitrarily designated as Type I, Type II Type III, Type IV and Type V. Type I neurone: These neurones are neurosecretory cells. Several neurosecretory cells are. recognized in the pars intercerebralis. They are roughly round or peach-shaped cells measuring $13{\sim}25{\mu}m$ in diameter. The rounded nucleus shows about $5{\sim}10{\mu}m$ in diameter. The chromatin is predominantly diffused with only occasional dense patches. The perikaryon contains numerous. mitochondria, free polyribosomes and neurosecretory granules. The neurosecretory granules are relatively uniform in electron density, and each one is about $100{\sim}400{\mu}m$ in diameter and surrounded by a single membrane. The granules are also observed mostly as in groups. In one group of neurones the cisternae of endoplasmic reticulum are distended or in other group of neurones are not distended. Golgi saccules are slightly dilated at their lateral extremities and contains. frequenty dense rounded materials. Type II neurone: Thes have the largest soma in the pars intercerebralis about $30{\sim}35{\mu}m$ in diameter. They also show roughly polygonal in shape. The nucleus is elongated or sickle-shaped. The chromatin is mainly in the euchromatin form. The perikarya in these cells are well populated with populated with free ribosomes and contain numerous mitochondria and Golgi bodies. The cisternae of granular endoplasmic reticulum are also well distributed. Type III neurone: They are oval or spindle-shaped and also medium-sized. neurones approximately $15{\sim}17{\mu}m$ in length. The nucleus is oval or slightly elongated in shape and $8{\sim}9{\mu}m$ in length. The chromatin occurs in diffused form. The cytoplasm contains many filamentous or oval mitochondria. The perikaryon has also numerous free polyribosomes and cisternae of granular endoplasmic reticulum. Type VI neurone: They are roughly polygonal in shape probably due to the close approximation of the adjacent cells. The soma is about $7{\sim}8{\mu}m$ in diameter. The nucleus is round or oval in shape and $5.0{\sim}5.8{\mu}m$ in diameter. The necleus also occupies a large proprion of the cell body. The perikaryon is well populated with free ribosomes and contains several mitochondria and cistenae of granular endoplasmic reticulum. Type V neurone: These neurones are similar to Type VI neurones in various respects such as cell size and cell inclusion, but they differ from Type IV neurones in shape. The soma is oval or slightly elongated. The cell body contains several filamentous and oval mitochondria.

An Ultrastructural Study on the Effect of Exogenous $Ca^{2+}$ Stimulation to Ischemic Myocardium during Post-ischemic Reperfusion (재관류시 외인성 칼슘 자극이 허혈 심근에 미치는 영향에 대한 전자현미경적 연구)

  • The effects of exogenous $Ca^{2+}$ stimulation on the post-ischemic myocardial cells were studied using isolated Langendorff-perfused guinea pig hearts. At the starting point of reperfusion, Tyrode solutions, each containing 2.0mM, 4.0mM and 8.0mM $CaCl_2$ respectively, were administered for 2 minutes apart by descending, ascending, or by combined sequences followed by standard Tyrode solution containing 1.0mM $CaCl_2$. The minutes of global ischemia produced reversible but moderate to severe degree of myocardial ultrastructrual changes including focal destruction of sarcolemma, loss of nuclear matrix, clumping and margination of chromatins, mitochondrial swelling, destruction of mitochondrial cristae, shortening of sarcomeres, focal loss of myofibrils, and separation of cell junctions. In spite of reperfusion, the ultrastructure was more severely damaged and irreversible changes such as intracellular fluid accumulation, contracted sarcomeres, mitochondrial destruction, disruption of sarcolemma, loss of nuclear matrix, and separation of cell junction were observed in a large number of cells. In contrast, Tyrode-perfused $Ca^{2+}$-stimulated myocardial cells showed relatively well preserved ultrastucture, except slight changes including focal mitochondrial swelling, widening of T-tubule, and widening of cell junctions, especially at fasciae adherentes. The post-ischemic $Ca^{2+}$-stimulated reperfused myocardial cells produced focal changes such as mitochondrial destruction, disintegration of sarcolemma, widening of T-tubule, and intracellular fluid accumulation with slight variation in degree of changes by the method of $Ca^{2+}$ administration sequence. However, in a large number of the myocardial cells, chromatins were redistributed relatively evenly in the nuclear matrix, mitochondrial cristae were tightly packed, and a considerable number of intramitochondrial granules and glycogen granules reap-pealed. These results indicate that exogenous $Ca^{2+}$ stimulation in the initial period of reperfusion may be beneficial to salvage or to reduce the post-ischemic myocardium from further deleterious changes, and that the beneficial effects may be derived from the reserves of the function of the intracellular $Ca^{2+}$ regulating organelles and/or from the responsiveness of contractile apparatus to $Ca^{2+}$ stimulation.

Comparative Ultranstructures of the Fertilized Egg Envelopes from Three-spot gourami, Pearl gourami and Marble gourami, Belontiidae, Teleost (경골어류 등목어과 Three-spot gourami, Pearl gourami 및 Marble gourami의 수정란 난막 미세구조 비교)

  • The structures of the fertilized egg envelope from three species, three-spot gourami (Trichogaster trichopterus), pearl gourami (Trichogaster leeri) and marble gourami (Trichogaster trichopterus trichopterus) belong to Belontiidae were observed, utilizing light, scanning and transmission electron microscopes. In all three species, the fertilized eggs were the colorless, transparent, spherical, adhesive and pelagic type. A large oil droplet was located in vitelline membrane of the fertilized egg. The egg envelopes have a single micropyle, which is thought to the pathway of sperm in the area of the animal pole. Specially, the micropyle of three-spot gourami was similar to that of marble gourami which is subspecies of three-spot gourami. An outer surface of the fertilized egg envelope was arranged by grooves in all three species. The fertilized egg envelopes consists of two distinct layers; an adhesive outer layer and an inner layer with high electron density. In conclusion, the morphological similarity of the fertilized egg, micropyle, outer surface and transverse section of the fertilized e9g envelope seems to be an indication of the Belontiidae.

The Differentiation and Ultrastructural Characteristics of Type II Pneumocyte in Early Human Fetal Lungs (태아 허파의 초기 발달 중 과립허파꽈리세포의 분화와 미세구조의 특징)

  • The differentiation and ultrastructural characteristics of type II pneumocyte was investigated using 7 cases of human fetal lungs from 9 to 20 weeks of gestation by transmission electron micropscopy. The result obtained were as follows. 1. From the pattern of epithelium, type II pneumocyte was not discernable, but with the gradual development of gestation, the epithelium of the future pulmonary alveoli was transformed from pseudostratified columnar into simple cuboidal epithelium after 15 weeks of geatation. 2. The multilamellar body very specific to type II pneumocyte was observed at first at 9 weeks of gestation. Besides, another characteristics of the cell were also observed such as cytoplasmic inclusion body, granular inclusionbody,multivesicularinclusionbodyanddensebody. 3. The number and size of multilamellar body increased, but those of other inclision bodies decreased at 19 and 20 weeks of gestation. In summary, type II pneumocytes (or precursor cells) with multilamellar body and another characteristic inclusion bodies were observed in the human fetal lungs from 9 weeks of gestation. And so, it is suggested that the differentiation of type II pneumocyte starts at or before 9 weeks of gestation.

  • The purpose of this study is to investigate the hemp (Cannabis sativa cv. Chungsam) seed structure and ultrastructure of food reserves by scanning and transmission electron microscopy. We examined the seed coat and embryo consisting of a hypocotyl-radicle axis and two cotyledons. The seed coat consisted of exotesta and endotesta. The exotesta was a mechanical layer with lignified and elongated cells, while endotesta of the underlying layers of the exotesta was consisted of two separated cell layers. The collapsed outer layer of endotesta showed the unique reticulate structures. In cotyledon cells, protein and lipid bodies occupied most of cytoplasm. Protein bodies varied in diameter from 1.8 to $5.0{\mu}m$ and possessed a protein matrix containing electron-dense globoid crystals. Numerous lipid bodies ranged from 0.8 to $3.0{\mu}m$ in diameter were distributed around the protein bodies. During the early stages of breakdown, protein bodies rapidly changed their shape into the granular feature, however, lipid bodies were gradually degradated and fused each other. The degeneration process of protein bodies and lipid bodies of cotyledon cells might be correlated with the reports which hemp seeds rapidly lose their ability to germinate.

The Effects of Lycii fructus and Lycii folium on the Liver in Ovariectomized Rat (구기자와 구기엽이 난소적출 흰쥐의 간에 미치는 영향)

  • We aimed to investigate whether the extracts of Lycii fructus, Lycii folium, and estradiol can attenuate on the level of cholesterol on serum and ultrastructure alteration of hepatocytes isolated from ovariectomized rats. Experimental groups are divided into five: Sham (sham-operated), OVX (ovariectomized animal), LCF (OVX, Lycii fructus), LCL (OVX, Lycii folium), and $E_2$ (OVX, estradiol). The body weights of OVX group showed increase compared with Sham, LCF, and LCL groups. The levels of total cholesterol and low-density lipoprotein (LDL) cholesterol were significantly decreased in the LCF and LCL groups compared with those of OVX group. Moreover, the High-density lipoprotein (HDL) cholesterol level was increased in the LCL group. As the results of electron microscopical observation, lipid droplets in the OVX group showed increase compared with Sham group. But in the LCF group not showed lipid droplets. In conclusion, administration of Lycii fructus or Lycii folium lowers the serum cholesterol content of the ovariectomized rats and also the damage of the hepatocytes and formation of lipid droplets.

The Shape of Synaptic Vesicles in the Tooth Pulp Afferent Terminals and P-endings in the Rat Trigeminal Nucleus Principalis (흰쥐의 삼차신경주감각핵에서 치수유래 일차들신경종말 및 이와 연접하는 연접이전신경종말의 연접소포 형태 비교)

  • In order to investigate shape of synaptic vesicles of the tooth pulp afferent boutons and their presynaptic endings (p-endings), and the neuroactive substance of the p-endings in the trigeminal nucleus principalis, rat incisor tooth pulp afferents were labeled by the horseradish peroxidase (HRP) and quantitative ultrastructural analysis and postembedding immunogold labeling were performed. Labeled tooth pulp afferent boutons contained clear, spherical synaptic vesicles (diameter: $45{\sim}55\;nm$) and occasionally dense core vesicles(diameter: $80{\sim}120\;nm$). They formed symmetrical synapses with unlabeled axon terminals (p-endings) containing pleomorphic synaptic vesicles. The ratio of short to long diameter (form factor) of synaptic vesicles of pulp afferent boutons was 0.6 to 0.99, whereas that of p-endings was 0.25 to 0.99. In addition, most of the p-endings showed GABA-like immunoreactivity. These results indicate that the shape of synaptic vesicles is quite different between the tooth pulp afferent boutons and p-endings, and the p-endings may contain GABA as a neuroactive substance in the trigeminal nucleus principalis.

Fine Structure of Blue-green Algae, Microcystis aeruginosa Kutzing (남조(藍藻) Microcystis aeruginosa Kutzing의 미세구조(微細構造)에 관(關)한 연구(硏究))

  • In order to understand the morphological differences between two different organic loadings by its upstream, and to compare with other algal groups with references, the fine structure of blue-green algae, Microcystis aeruginosa Kitzing, taken from two branches, Tongbok and Bosung stream of Lake Chuam, Korea pennisula was examined. It showed extinct differences in most physicochemical factors between both branches, except water temperature and pH values. The concentrations of total phosphorus in Tongbok branch were twice as those of Bosung. M. aeruginosa cells were enumerated totally $1.2X10^4cells/ml$ and these individuals in branch of Tongbok were close to two times as much as Bosung. In light and electron microscopy, natural M. aeruginosa colonies formed irregular shape and non-directional array in amorphous matrix. They were consisted of many kinds of cells, youngs or olds in cell division, solitary, and various size of cells. Each cell ranged from 2.61 to $5.40{\mu}m$ in diameter, and averaged as $3.54{\pm}0.19{\mu}m$. In cytoplasm, they contained a number of inclusions in various size, shape and appearances. Among them, polyhedral bodies or carboxysomes, a structured granules, photosynthetic lamellae or thylakoids, and gas vacuoles were prominent and easy to recognize. Although it was failed to find the definable morphological variations in the ultrastructure of M. aeruginosa in terms of algal habitual environments, some useful characters were founded, outer layer of cell wall, polyhedral bodies and gas vacuoles, in blue-green algal classification and taxonomy.
