A Study on the Fine Structural Changes of Porcine Ovarian Follicles during Atresia (돼지 난소내 여포의 폐쇄에 따른 미세구조의 변화에 관한 연구)

  • This experiment has been done in order to study the correlation between the ultrastructure changes and the atresia phenomenon of the follicles in porcine ovary. The ovaries were assorted according to the presence or absence of corpus luteum. Thereafter, the follicles were classified into normal, pyknotic, necrotic and cystic groups by atretic characteristics on the histological observation, and then their ultrastructures were examined with an electron microscope. The results were as followings. 1. In normal group, granulosa cells represented the ultrastructural characteristics of protein-synthesizing cells. Since the initiation of atresia, the line structure of granulosa cells showed many of the characteristic features of steroid-secreting cells, followed by gradual pyknosis. 2. In necrotic group of the ovary without corpus luteum, the theca interns became hypertrophic and displayed the ultrastructural features of active steroidsecreting cells. But this phenomenon was not seen in the follicles of the ovary with corpus luteum. 3. Degenerative changes of cumulus cells were similar to those of granulosa cells, and the degenerating oocytes showed the degeneration of cellular organelles, cytoplasmic vacuolization and disappearance of microvilli on the surface. The degeneration of granulosa cells tended to procede that of oocytecumulus complex in the follicles of the ovary having no corpus luteum, but this tendency was reversed in the case of presence of corpus luteum.   • Mussel byssus is a bundle of threads used to attach mussels to wet substrates. Recently, a thin cuticle layer on the byssus has attracted public attentions due to its remarkable toughness - stiff as epoxy resin and extensible as rubber. Here, we observed ultrastructure of the cuticle layer in a far eastern mussel (Mytilus coruscus) to understand underlying mechanisms for the mechanical properties. The cuticle layer observed by TEM was composed of submicron-sized granular inclusions in a continuous matrix phase. In addition, ultrastructural study in the presence of tertiary amine (Tetraethylammonium, TEA) showed an evidence that the cuticle is stabilized by cation-${\pi}$ interaction.

Observation of Histochemical Ultrastructure in Regenerating Rat Liver (재생중인 흰쥐 간세포의 조직화학적 미세구조 관찰)

  • An ultrastructural study of hepatocyte proliferation in the regenerating rat liver has been made by means of the partial hepatectomy. And electron microscopic histochemistry of hepatocyte in the regenerating rat liver is studied through alkaline phosphatase reaction. The results are as follows: 1. When the regeneration of rat liver is induced by the partial hepatectomy, the prominent ultrastructural characteristics of hepatocyte are changes of the distribution of chromatin in nucleus, increase of the number of mitochondria and decrease of the size of them, development of rough endoplasmic reticulum, and transient decrease of glycogen granules in cytoplasm. 2. Alkaline phosphatase reaction products are appeared in the nucleus or rough endoplasmic reticulum of hepatocyte during the initial regeneration of liver as 24, 48 and 72 hour groups after partial hepatectomy. And these positive reaction are mainly increased in cytoplasm and plasma membrane of hepatocytes during 1, 2 and 3 week groups after partial hepatectomy. As 4 weeks passed after partial hepatectomy, these positive reaction is located in the sinusoidal epithelial cells or erythrocytes. With above results, we concluded that alkaline phosphatase was synthesized in the rough endoplasmic reticulum bounded ribosomes of regenerating hepatocyte, was transported to the plasma membrane of them, and then was transported in blood by the way sinusoidel epithelial cells.

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.

  • To investigate the ultrastructural features of rat back skin tumor induced by 7, 12-Dimethylbenz[a]anthracene(DMBA), morphological changes during carcinogenesis were undertaken by the light and the electron microscopes. On the light microscopic levels, dysplasia was observed at the 6th week of treatment with 0.5% DMBA, hyperplasia at the 8th week, and carcinoma in sitv or invasive carcinoma at the 12th week. In addition to those characteristics, papillomatosis was observed at the 18th week. Under the electron microscopy, intercellular spaces were enlarged in the early phase. Nucleolar enlargement, the increase of free ribosomes, the condensation of tonofibrils, and the irregular arrangement of desmosomes appeared at the 6th week of application. Thereafter, the degradation of desmosomes and the discontinuity of basal lamina were characterized. And then, epidermal cell processes were protruded toward the connective tissue at the site of discontinuity of basal lamina at the 12th week. At the 14th to 18th week, DMBA-induced fibrosarcomas or papillomas were observed.

  • The internal ultrastructural changes of germ cells and external morphology of spermatozoon during the spermatogenesis in the rockfish, Sebastes inermis were studied using transmission and scanning electron microscope. The testis is seminiferous tubule type in internal structure. Seminiferous tubule consist of many cyst which contain numerous germ cells in same developmental stage. Spermatogonium contained a large nucleus with single nucleolus in interphase. Primary spermatocyte identified by the presence of synaptonemal complex in nucleus and the contained a number of mitochondria, endoplasmic reticula and Golgi bodies in cytoplasm. The nucleoplasm of secondary spermatocyte was more concentrated than that of the previous phase. Spermatids were more condensed in nucleus and cytoplasm, and show the long-spherical shape. In the cytoplasm of spermatid mitochondria located to lower portion of the nucleus and Golgi bodies located to upper portion, but proacrosomal granule is not appeared. The spermatozoon consist of the head and tail. No acrosome could be found in the head. The cytoplasmic collar of posterior part in sperm head contained mitochondria which surrounded axial filament. The well developed axonemal lateral fins were identified in sperm flagellum, and the axial filament of the flagellum consist of nine pairs of peripheral microtubules and one pair of central microtubules.

An Ultrastructural Investigation of Infection Threads in Sesbania rostrata Stem Nodules Induced by Sinorhizobium sp. Strain MUS10

  • Sinorhizobium sp. strain MUS10 forms nitrogen-fixing stem nodules on Sesbania rostrata, a tropical green manure crop. In this study, the ultrastructural events associated with the formation of stem nodules were investigated. Sinorhizobium sp. strain MUS10 entered the host tissue through cracks created by the emerging adventitious root primordia and multiplied within the intercellular spaces. During early phases of infection, host cells adjacent to invading bacteria revealed cellular damage that is typical of hypersensitive reactions, while the cells at the inner cortex exhibited meristematic activity. Infection threads were numerous in S-day-old nodules and often were associated with the host cell wall. In several cases, more than one infection thread was found in individual cells. The junction at which the host cell walls converged was often enlarged due to fusion of intracellular branches of infection threads resulting in large infection pockets. The infection threads were made up of a homogeneous, amorphous matrix that enclosed the bacteria. Several finger-like projections were seen radiating from these enlarged infection threads and were delineated from the host cytoplasm by the plasma membrane. As in Azorhizobium caulinodans induced root nodules, the release of Sinorhizobia from the infection threads into the plant cells appears to be mediated by 'infection droplets'. A 15-day­old Sesbania stem nodule revealed typical ultrastructure features of a determinate nodule, containing several bacterioids within symbiosomes.

An Experimental Study on Growth Pattern and Ultrastructure of Human Gingival Fibroblasts Treated by Low Level Laser (저출력레이저가 성인의 치은섬유아세포의 성장양상과 미세구조에 미치는 영향에 관한 실험적 연구)

  • In order to verify the acceleration effect of low level laser (LLL) on oral mucosal wound healing process at cell biological level, the author studied growth pattern and ultrastructure in human gingival fibroblasts flashed by GaAlAs LLL-830 nm, 15mW for 10 minutes/flash one to three times at interval of 3-4 days through the evaluation of cell growth rate, protein conent/cell, DNA content/cell and ultrastructural changes for 14 days. The results were as follows : 1. The growth rate in gingival fibloblasts treated by LLL showed 4 orderly stages-decreasing stage after LLL treatment, acute increasing stage 3 days after LLL treatment, restring stage and recovering stage. 2. The effect of multiple flashes on LLL at interval of 3 days more or less was not proportional to times of flash on acceleration of growth in gingival fibroblasts. 3. The total protein content per gingival fibroblast was not significantly changed by LLL treatment in comparison with control group. But some kinds of protein which might be cell growth promoting factors were decreased immediately after LLL treatment, thereafter were acutely increased in cellular protein profile. 4. In ultrastructural changes of gingival fibroblasts treated by LLL, more prominent rough endoplasmic reticulum, mitochondrial hyperplasia/hypertrophy and increased extracellular fibrillar matrix were observed in comparison with control group under same experimental period.

  • Ultrastructural and histochemical studies were carried out on the prostate gland of a Korean slug (Incilaria fruhstorferi) with light and electron microscopes. The results are as follows. The endothelial tissue of the prostate gland is constituted with tall and narrow ciliated columnar cells, irregularly shaped cells and gland cells in which are included the elongated oval or ellipsoid nucleus those are surrounded by curved membrane. The endothelial tissue of the prostate gland is composed by type-A, B, C and type-D gland cells, and the glanules of type-A, B, and D gland cells are certified to neutral mucopolysaccharide because are related by PAS-alcian blue (pH 2.5) , but the other hand type-C gland cell is only weakly reacted to PAS, but strongly reacted by Million reaction. The glanules of type-A gland cell are small size (about $0.4{\mu}m$) and are seen high electron density, but the glanules of type-B gland cell are large size (about $0.7{\mu}m$) and glanule density is same type-A glanules. Long ellipsoid type-C gland cell contained round nucleus which is well developed beterochromatin in, and that small oval glanules (size, about $0.9{\mu}m$) of moderate high electron density which are formed a group of large glanule together with $4\sim5ea$, but type-D gland cell possessed round small nucleus are seen high electron dense glanules (size, $0.8{\mu}m$).

Ultrastructural Changes in Cryptosporidium parvum Oocysts by Gamma Irradiation

  • Cryptosporidium parvum is known as one of the most highly resistant parasites to gamma irradiation. To morphologically have an insight on the radioresistance of this parasite, ultrastructural changes in C. parvum sporozoites were observed after gamma irradiation using various doses (1, 5, 10, and 25 kGy) following a range of post-irradiation incubation times (10 kGy for 6, 12, 24, 48, 72, and 96 hr). The ultrastructures of C. parvum oocysts changed remarkably after a 10-kGy irradiation. Nuclear membrane changes and degranulation of dense granules were observed with high doses over 10 kGy, and morphological changes in micronemes and rhoptries were observed with very high doses over 25 kGy. Oocyst walls were not affected by irradiation, whereas the internal structures of sporozoites degenerated completely 96 hr post-irradiation using a dose of 10 kGy. From this study, morphological evidence of radioresistance of C. parvum has been supplemented.
