Surface Imaging of Barley Aleurone Cell by Atomic Force Microscopy

  • To observe and analysis ultra-microscopically barley aleurone cell surface, atomic force microscope (AFM) was used. Seed coat of early maturing germplasm, eam9, was dehulled and scanned by non-contact mode. We have obtained the high resolution topographic 3-dimensional image of barley aleurone layer with high resolution. These images showed the membrane proteins in barley aleurone cell. One channel protein and numerous peripheral or integral proteins were detected in a area of 100 $\mu\textrm{m}^2$. Furthermore, we found that their widths were ranged from 50 to 750nm and lengths from 0 to 66 $\mu\textrm{m}$. The thickness of aleurone layer was measured by scanning electron microscope. The thickness at early developmental stage was about 16 and then the aleurone cell enlarged upto 57 $\mu\textrm{m}$${\mu}{\textrm}{m}$ at least until 42 days after anthesis. In this study, we firstly reported on the ultrastructural AFM analysis of living aleurone cell as a biological specimen. It was clearly suggested that AFM will become an powerful tool for probing both the structural properties of biological samples.

Spermiogenesis and Taxonomical Values of Sperm Ultrastructures in Male Crassostrea ariakensis (Fujita & Wakiya, 1929) (Pteroirmorphia: Ostreidae) in the Estuary of the Seomjin River, Korea

  • Characteristics of the developmental stages of spermatids during spermiogenesis and phylogenetic classicfication of the species using sperm ultrastructures in male Crassostrea ariakensis were investigated by transmission electron microscope observations. The morphology of the spermatozoon of this species has a primitive type and is similar to those of Ostreidae. Ultrastructures of mature sperms are composed of broad, modified cap-shaped acrosomal vesicle and an axial rod in subacrosomal materials on an oval nucleus, four spherical mitochondria in the sperm midpiece, and satellite fibres which appear near the distal centriole. The axoneme of the sperm tail shows a 9+2 structure. Accordingly, the ultrastructural characteristics of mature sperm of C. ariakensis resemble to those of other investigated ostreids in Ostreidae in the subclass Pteriomorphia. In this study, particularly, two transverse bands (stripes) appear at the anterior region of the acrosomal vesicle of this species, unlike two or three transverse bands (stripes) in C. gigas. It is assumed that differences in this acrosomal substructure are associated with the inability of fertilization between the genus Crassostrea and other genus species in Ostreidae. Therefore, we can use sperm ultrastructures and morphologies in the resolution of taxonomic relationships within the Ostreidae in the subclass Pteriomorphia. These spermatozoa, which contain several ultrastructures such as acrosomal vesicle, an axial rod in the sperm head part and four mitochondria and satellite fibres in the sperm midpiece, belong to the family Ostreidae in the subclass Pteriomorphia.

An Ultrastructural Study on Endocrine Cells in the Pyloric Region of the Korean Hedgehog(Erinaceus koreanus) (한국산 고슴도치 유문부에 있어서 내분비세포의 전자현미경적 연구)

  • Endocrine cells in the pyloric region of the hedgehog were studied ultrastructurally. 4 types of endocrine cells classified as gastric-type EC, ELC, $D_1$, and G cells were observed in this region. The gastric-type EC cells contained pleomorphic granules with high electron density and highly dense bodies in a dense matrix. ECL cells were characterized by the presence of round or oval granules with high electron density. Some granules of ECL cells showed a small amount of content or empty. $D_1$ cells contained round and small granules with low electron density. They occasionally showed a narrow halo between the limiting membrane and the dense materials. G cells were characterized by the presence of round or oval granules with low electron density. Some granules of these cells showed a narrow halo between the limiting membrane and the core.

Electron Microscopic Study of Osseointegration between Bone and Smooth Machined Implants (선반가공 임플란트와 골조직의 유착에 관한 전자현미경적 연구)

  • This paper reports that the ultrastructural nature of the interface process between the implants and surrounding bone has been studied after in vivo 1, 4, 8, 12 weeks of implantation of smooth machined implants into rabbit tibias. There was no indication of the fibrous connective tissue formation around the implant that imply intolerance of the bone tissue towards the implant after 1 week of implantation. The regions showing direct bone tissue bonding to the smooth machined implant contained osteoblast activating across the interface in the direction after 4 weeks of implantation. The reaction of a smooth machined implant caused in the first instance formation of an amorphous woven bone, which transformed into a mineralized bone containing collagen fibers. After 8 weeks of implantation, the activities of osteoblast initiated osseointegration forming bone matrix at the interface. During this period, the osteoblast surrounded with a matrix consisting of collagen bundles running in various directions. In the interface area between newly formed bone tissue and implants which has been inserted in rabbit tibias for 12 weeks, the implant and mineralized bone was separated by an amorphous electron dense material layer about $1{\sim}1.5{\mu}m$ in thickness.

Accumulation of Heavy Metals in the Antarctic Clam, Laternula elliptica, and in the Korean coastal Clam, Ruditapes philppinarum

  • Immunohistochemical and ultrastructural experiments were conducted to find out heavy metal accumulation in some selected organs such as the kidney, the digestive gland, and the gill of the Antarctic clam Laternula elliptica and R. philippinarum. According to the immunohistochemical study the subject organs of the clam showed reactions indicating the presence of MT (metallothionein), a metal-binding protein involved in metal detoxifying process. Examination under the transmission electron microscope also revealed that other ligands may play a role in metal accumulating and detoxifying process in L. elliptica and R. philippinarum. In the artificial exposure of the clam to Cd, the clams showed immediate subcellular responses. The level of the anti-MT reactions became higher in the proportion to the degree of pollution of their habitat and to the period of Cd exposure. These suggest that the two species can be used as efficient biomarkers for Cd exposure in the natural environment.

Morphological Study of Acute Lung Injury Induced by Interleukin-1$\alpha$ Intratracheally in Young and Old Rats (젊은 흰쥐와 늙은 흰쥐에서 인터루킨-1$\alpha$로 유도된 급성폐손상에 관한 형태학적 연구)

  • In order to investigate the effect of aging and the $H_2O$$_2$ localization in association with histological, ultrastructural, and cytochemical studies in lung tissue after interleukin-1$\alpha$(IL-1) induced lung injury, an acute lung injury was induced by instillation of IL-1 into the trachea. Both of 4- and 20-months-old male rats, protein contents in IL-1 treated branchoalveolar lavage increased significantly compared to each control rats. Acute lung injury occured by oxidative stress because neutrophils accumulated in vascular lumen and formed the adhesion with endothelial cells. As these cause, tissue proteins were exuded and leukocytes migrated into the alveolar lumen. Neverthless in these lung injury $H_2O$$_2$ localization of IL-1 treated 20 months rats was not different compared to IL-1 treated 4 months rats. After all aging was not a factor to accelate IL-1 induced lung injury. Based on these results, it is suggested that neutrophil infilteration might be an important cause in acute lung injury, and aging is not a factor to change the acute lung injury by oxidative stress.

  • There have been many problems like plasmolysis in the ultrastructural studies with kelps. The present study was conducted with series of different fixatives and buffer solutions and several resins to solve the plasmolysis phenomena. The fixative of 1470 mosmol($3\%$ glutaraldehyde, $2.4\%$ paraformaldehyde in 0.05M cacodylate buffer with 0.2M sucrose and $0.5\%$ caffeine at pH of 7.2) showed the least frequency of plasmolysis. The relationship between the concentration of primary fixatives and the frequency of plasmolysis was not found. With above fixative the possibility of plasmolysis during infiltration period was not identified using ultra-low viscosity resin. Generally, the kelp(Laminaria religiosa) contains a lot of mucilage, which interupts the infiltration of fixative. The possible physiological mechanisms related with mucilage secretion may be unable to fix the region of plasmalemma properly.

Effects of High-salt Diet on the Mouse Adrenal Medulla (고염식이가 흰 생쥐의 부신 수질에 미치는 효과)

  • Exposure to stressful stimuli is known to activate the peripheral sympathetic nervous system and the adrenal gland. In this study, we evaluated the effects of high-salt diet on the mouse adrenal medulla using the tyrosine hydroxylase (TH) immunohistochemistry and the transmission electron microscopic observation. Immunoreactivity for TH was increased after high-salt diet. Especially, the TH immunoreactivity was stronger in 4 days high-salt diet mouse than that of 4 weeks. TH immunoreactivity was mainly present in the cytoplasm and granules of the noradrenaline cells. After high-salt diet, the noradrenaline cells exhibited the ultrastructural alterations consisting of areas of empty cytoplasm, expanded granules, and some damaged mitochondria. These results suggest that high-salt diet may be a factor of stressful stimuli on the mouse.

  • The ultrastructures of spermatogenesis and sperm in Xiphophorus maculatus, ovoviviparous fish were investigated by electronmicroscopy The testis of Xiphophorus maculatus contained numerous testicular sacs, and spermatogenesis was synchronized in these testicular sac. In the case of spermatogonium, the nucleus was comparatively large ellipsoidal, and the nucleolus and mitochondria showed a marked development. The size of primary spermatocyte was smaller than that of spermatogonia, and that of secondary spermatocyte was smaller than that of primary spermatocyte. The chromatin of spermatocyte was highly condensed according to their development. The nucleus with electron-dense was round shape. In spermiogenesis, flagella started to be formed and chromatin was more condensed. The mitochondria were rearranged along the tail. The sperm was formed by loss of cytoplasm. The head of mature sperm was long cone shape and had not acrosome. The microtubules of flagella were arranged 9+2 structure. Also, the sperm has a loop-like structure at the end of a tail.

Proliferation of Toxoplasma gondii Suppresses Host Cell Autophagy

  • Autophagy is a process of cytoplasmic degradation of endogenous proteins and organelles. Although its primary role is protective, it can also contribute to cell death. Recently, autophagy was found to play a role in the activation of host defense against intracellular pathogens. The aims of our study was to investigate whether host cell autophagy influences Toxoplasma gondii proliferation and whether autophagy inhibitors modulate cell survival. HeLa cells were infected with T. gondii with and without rapamycin treatment to induce autophagy. Lactate dehydrogenase assays showed that cell death was extensive at 36-48 hr after infection in cells treated with T. gondii with or without rapamycin. The autophagic markers, LC3 II and Beclin 1, were strongly expressed at 18-24 hr after exposure as shown by Western blotting and RT-PCR. However, the subsequent T. gondii proliferation suppressed autophagy at 36 hr post-infection. Pre-treatment with the autophagy inhibitor, 3-methyladenine (3-MA), down-regulated LC3 II and Beclin 1. The latter was also down-regulated by calpeptin, a calpain inhibitor. Monodansyl cadaverine (MDC) staining detected numerous autophagic vacuoles (AVs) at 18 hr post-infection. Ultrastructural observations showed T. gondii proliferation in parasitophorous vacuoles (PVs) coinciding with a decline in the numbers of AVs by 18 hr. FACS analysis failed to confirm the presence of cell apoptosis after exposure to T. gondii and rapamycin. We concluded that T. gondii proliferation may inhibit host cell autophagy and has an impact on cell survival.
