Inhibitory effect of Fucofuroeckol-A from Eisenia bicyclis on tyrosinase activity and melanin biosynthesis in murine melanoma B16F10 cells

  • Background: The aim of this study was to investigate the in vitro inhibitory effects of Fucofuroeckol-A isolated from Eisenia bicyclis against tyrosinase activity and 3-isobutyl-1-methylxanthine (IBMX)-induced melanin biosynthesis in B16F10 melanoma cells. Result: Among the ethanolic (EtOH) extract of E. bicyclis and its organic solvent fractions, the ethyl acetate (EtOAc) soluble fraction showed a noticeable inhibitory effect on mushroom tyrosinase with an $IC_{50}$ value of $37.6{\pm}0.1{\mu}g/mL$. Repeated column chromatography of the active EtOAc fraction resulted in the isolation of Fucofuroeckol-A. It evidenced more potent tyrosinase inhibitory effect with an $IC_{50}$ value of $11.4{\pm}1.4{\mu}M$ than arbutin ($IC_{50}=1076.6{\pm}44.3{\mu}M$), which was used as a positive control. Lineweaver-Burk plots suggest that Fucofuroeckol-A plays as a noncompetitive inhibitor against tyrosinase. Furthermore, we have evaluated the inhibitory effects of Fucofuroeckol-A on IBMX-induced melanin formation in B16F10 melanoma cells. Fucofuroeckol-A ($12.5-100{\mu}M$) exhibited a significant inhibition of melanin production in the melanoma cells. Conclusion: In the present study, we suggested that Fucofuroeckol-A might prove possibility as a novel inhibitor of melanin biosynthesis in cosmetic applications.

꿀풀과 식물 정유의 주성분인 Thymol의 미백활성에 관한 연구 (Anti-Melanogenic Effect of Thymol, a Major Odorant in Essential Oils of Family Lamiaceae)

  • Objectives : Thymol (2-isopropyl-5-methylphenol), a natural monoterpenoid phenol, is one of the major odorant constituents found in natural essential oils of various herbal plants, such as Thymus quinquecostatus and Thymus vulgaris. Multiple biological activities of thymol, including antioxidative, antimicrobial, and anti-inflammatory effects, have been reported in numerous in vitro studies, and recently it was suggested that thymol may could inhibit oxidization of L-dihydroxyphenylalanine (L-DOPA) to dopaquinone required in melanogenesis pathway, as an antioxidant. Methods : MTT assay was performed to test the cytotoxic effect of thymol in B16F10 cells. Inhibitory effect of thymol to tyrosinase activities were examined using both mushroom tyrosinase and intracellular tyrosinase. Expression level of tyrosinase in B16F10 cells were investigated by western blot analysis. Results : The cell viability was decreased by thymol treatment in dose-dependant manner, leading significant cytotoxicity in 500 and $1000{\mu}M$ thymol-treated groups. In the alpha-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis, administration of thymol significantly decreased extracellular (secreted) melanin content in dose-dependent manner. Cellular tyrosinase activity assay and western blot analysis of intracellular tyrosinase showed that thymol has a strong anti-melanogenic effect by inhibition of tyrosinase activity and by decreasing expression of tyrosinase that contribute to melanin synthesis in the B1610 cells. Conclusions : As the first functional study that prove anti-melanogenic effect of thymol and its underlying mechanism in the living cells, our study suggests the applicability of fragrance as the functional materials of cosmetics or health supplement, not as just an additive.

Prevention of Browning in Mushroom by Onion Extract

  • The inhibitory effect of onion extract on mushroon tyrosinase activity was investigated. The enzyme activity was inhibited by 96.3% with th addition of onion extract. The inhibitor action of onion extract toward mushroom tyrosinase activity slightly increased after heat treatment at 10$0^{\circ}C$ for 10 mn (97.5%) and decreased after incubation of the extract at pH 2.3 for 3hrs (79.9%). However, the inhibitor action of the extract after dialysis decreased to18.8%. The onion extract showed drastic inhibition of the browning in mushroom.

새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과 (Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

위령선 추출물의 항염활성 및 멜라닌 생성 억제효과 (Anti-inflammatory Effect and Inhibition of Melanin Biosynthesis of Clematis mandshurica)

  • Clematis mandshurica (Ranunculaceae) has traditionally been used as a remedy for antidiuretic, antifungal, rheumatic conditions and alleviate pain. We carried out to evaluate the anti-oxidative effect, anti-inflammatory effect and anti-melanogenic effect of ethanol extract and solvent fractions of Clematis mandshurica. The ethanol extract and the dichloromethane fraction of Clematis mandshurica showed an anti-oxidative effect in DPPH assay, the inhibitory activity of nitric oxide (NO) production in lipopolysaccharide (LPS) activated RAW 264.7 cell, and melanin synthesis and tyrosinase activity of B16F10 melanoma cells. They reduced NO production and melanin content in a dose-dependent manner at concentrations of $2.5{\sim}10{\mu}g/ml$. They also suppressed iNOS and tyrosinase protein and m-RNA expressions dose dependently, assayed by western blot analysis and RT-PCR experiment.

파래 추출물의 멜라닌 생성 억제 효과 (Inhibitory Effect of Enteromorpha linza on the Melanogenesis in B16 Melanoma Cells)

  • Melanin is a polymer of phenol which produces hyperpigmentation in the skin. Melanin synthesis is catalyzed by the enzyme tyrosinase. To investigate the whitening activity of the fractions from the ethanol extract of Enteromorpha linza, we studied the inhibitory effect on the tyrosinase activity and melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from E. linza was as strong as that of kojic acid, a positive control. Also, the melanin production was significantly inhibited by the ethylacetate fraction in a dose dependent manner. The ethylacetate fraction showed the highest activity in the fractions and as strong activity as that of arbutin, a positive control. From these results, we suggest that the E. linza extract might be able to apply to cosmetic and medical fields.

쓴메밀 추출물의 미백 개선 효과 (Whitening Effect of Fagopyrum tataricum Extract)

  • 본 연구는 쓴메밀 추출물(FTE)의 미백 효과를 검증하기 위해 DPPH 라디칼 소거능, ABTS 라디칼 소거능, 타이로시나아제 활성 저해 효과 및 세포내 멜라닌 합성 억제 효과를 측정하였고, 단메밀 추출물(FEE)과 그 효능 비교를 하였다. 그 결과로 쓴메밀 추출물은 단메밀 추출물보다 항산화 효과 및 타이로시나아제 활성 저해, 세포내 멜라닌 합성 억제 효과가 높음을 확인하였다. 또한 인체 적용 실험에서 자외선 조사에 의한 인공 색소침착을 유발한 후, 쓴메밀 추출물을 함유한 크림을 제조하여 도포하였고, 8주 경과 후 피부 밝기를 측정한 결과 통계적으로 유의한 수준의 피부 미백 효과를 나타내었다. 이러한 결과들로 쓴메밀 추출물은 미백 개선을 위한 화장품 소재로 이용 가능성이 높을 것으로 기대된다.

백복령 주정 추출물의 멜라닌합성 억제를 통한 미백효과 (Whitening Effect of Poria cocas Ethanol Extract by Inhibition of Melanin Synthesis)

  • 백복령(Poria cocas)은 전통적으로 민간에서 피부미백에 효과가 있다고 보고되어 있다. 지금까지 melanin 합성에 대한 백복령의 직접적인 효과는 과학적으로 잘 연구되어 있지 않다. 따라서 멜라닌 합성에 대한 백복령 주정 추출물(PCEE)의 직접적인 효과를 밝히기 위해, 쥐의 B16F1 세포를 이용하여 DOPA synthesis assay, tyrosinase activity assay, Western blotting for melanogenic proteins [tyrosinase, tyrosinase related protein (TRP)-1 and TRP-2]를 수행하였다. 본 연구에서 PCEE가 3,4-dihydroxyindole-2-carboxylic acid (DOPA) 합성을 차단함으로써 농도 의존적으로 melanin형성을 억제한다는 것이 밝혀졌다. 비록 tyrosinase의 활성은 영향을 받지 않았지만 TRP-1 과 TRP-2의 단백질 발현 수준은 PCEE에 의해 조절되었다. 따라서 이러한 결과는 PCEE가 미백 효능을 가지고 있어 미백 화장품 개발을 위해 이용될 수 있다는 것을 시사하고 있다.

Bacillus coagulans CE-74에 의해 생산된 Biopolymer의 생리적 기능성 (Physiological Function in vitro of Biopolymer from Bacillus coagulans CE-74)

  • 미생물로부터 고분자 물질을 생산하여 기능성 식품보조제로 사용하기 위하여 Bacillus coagulans CE-74에 의해 생산된 biopolymer를 이용해 glucose, bile acid 흡수 지연 효과 및 ACE와 tyrosinase 저해 효과를 조사 검토하였다. 분리된 biopolymer의 소화관내 생리적 기능성을 알아보기 위해 반투막을 이용한 in vitro 실험을 수행하였다. 투석막을 이용하여 biopolymer의 glucose흡수 지연 효과를 살펴본 결과 biopolymer를 2% 첨가한 경우 투석 30분이 경과되었을 때 히ucose retardation index가 43.5%였고 시간이 경과 할수록 그 효과는 감소 하였으며 투석 5시간 이후에는 그 효과가 거의 없는 것으로 나타났다. Bile acid흡수 지연 효과를 살펴본 결과 투석 30 분이 경과 되었을 때 bile acid retardation index는 biopolymer가 1%, 2% 첨가된 경우 각각 34%, 44.2%였으며 시간이 경과할수록 그 효과가 감소하였다. 분리된 biopolymer의 ACE저해효과는 $10\;{\mu}g/{\mu}l$의 농도에서는 97%의 저해를 보였으며, tyrosinase 저해효과를 관찰한 결과 $20\;{\mu}g/{\mu}l$농도에서 48.5%의 저해효과를 나타내었다.

우절 에탄올추출물의 미백활성능과 주름저해 효능평가 (Evaluation of Whitening Activity and Wrinkle Inhibitory Effect of Ethanol Extracts of Nelumbinis Rhizomatis Nodus)

  • 본 연구는 화장품 소재로서 우절 에탄올추출물의 가능성을 확인하기 위한 것이다. 이를 위해 우리는 연근의 마디부분을 70%에탄올로 추출하여 사용하였으며 미백효능 및 주름개선 효과에 대한 생물학적 활성 평가를 수행하였다. 시료의 미백활성을 알아보기 위해 멜라닌세포(B16F10 cells)의 MTT assay를 이용한 샘플의 독성평가와 멜라닌 생성에 영향을 주는 관련 단백질의 발현량을 확인하였다. 시료의 주름활성억제 평가를 위해 collagenase inhibition assay를 수행하였다. 또한 UVB로 유도된 섬유아세포(CCD-986sk cells)의 MTT assay를 이용한 샘플의 독성평가와 matrix metalloprotease (MMP-1) inhibition 및 procollagen synthesis를 평가하였다. 미백활성 평가를 알아보기 위한 western bolt 결과, 멜라닌 생성과 관련된 두 단백질 Tyrosinase related protein-1 (TRP-1, Tyrosinase related protein-2 TRP-2)의 합성이 농도의존적으로 감소됨을 나타내었다. 게다가 우절 에탄올추출물의 collagenase inhibiion 실험의 결과, $500{\mu}g/ml$의 농도에서 대조군인 EGCG와 효능이 비슷한 80% 이상의 효과를 나타내었다. Procollagen synthesis 결과 UVB 자극군에 의해 콜라겐 합성은 68.8%로 감소되었으며 시료의 $25{\mu}g/ml$의 농도에서 80.2%의 콜라겐 합성에 회복능을 보였다. MMP-1 inhibition 실험결과는 $25{\mu}g/ml$ 농도에서 20.2%의 저해능을 나타내었다. 실험 결과는 NRN의 미백 및 주름 억제 효과를 검증하고, 향후 안전한 천연 화장품 재료로 사용될 수 있음을 확인했다.
