A Study on the Depigmenting Effect of Carthamus tinctorius Seed, Cyperus rotundus and Schizonepeta tenuifolia Extracts (홍화자, 향부자, 형개 추출물의 미백효과에 관한 연구)

  • The objective of the present study was to evaluate the skin depigmentation effect of the extracts of three herbs, Carthamus tinctorius seed, Cyperus rotundus and Schizonepeta tenuifolia. Their effects on tyrosinase and melanin synthesis inhibitory action were assessed. We found that the C. tinctorius seed ethanol extracts reduced the tyrosinase activity and melanin formation of B16F10 melanoma cells. The C. tinctorius seed suppressed the expression in microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein 1 (TRP-1), and tyrosinase related protein 2 (TRP-2) in B16F10 melanoma cells. These results show that C. tinctorius seed inhibited melanogenesis on the B16F10 melanoma cell. The underlying mechanism of C. tinctorius seed whitening activity may be the inhibition of tyrisinase, MITF, tyrosinase, TRP-1, and TRP-2 expression. The results suggested that C. tinctorius seed has considerable potential as a natural functional ingredient with a depigmentation effect.

  • To identify inhibitors of melanogenesis, we compared the effects of 5 compounds on mushroom tyrosinase, human melanocytic tyrosinase activity and melanin content. The cytotoxicyty of the components were also tested on cultured human melanoctes. Kojic acid showed marked inhibitory effect both on mushroom and human tyrosinase activity. This action of kijic acid is stronger than that of ascorbic acid. Arbutin inhibited human tyrosinase activity of cultured melanocytes although it had slightly inhibitory effect on mushroom tyrosinase activity. Azelaic acid had no effect on human tyrosinase activity. Melanin production was inhibited significantly by kojic acid and tranexamic acid. MTT assay showed that all of the compounds were non-cytotoxic to melanocytes at the concentrations tested. These results suggest that the effect of kojic acid on cultured meanocytes involve inhibition of tyrosinase activity and melanogenesis without affection the cell number.

  • Kaempferol was isolated and identified from the methanol extract of the flowers of Impatiens balsamina. Kaempferol showed inhibitory activity against mushroom tyrosinase with an $ID_{50}$ of 0.042 mM. Inhibition kinetics, as determined using a Lineweaver-Burk plot, showed kaempferol to be a competitive inhibitor of mushroom tyrosinase with a $K_i$ value of 0.011 mM. The lag phase of tyrosine hydroxylation catalyzed by mushroom tyrosinase clearly increased on increasing the concentration of kaempferol. In addition to its tyrosinase inhibiting activity, kaempferol strongly inhibited melanin production by Streptomyces bikiniensis, in a dose-dependent manner, without inhibiting cell growth. For comparative purposes, the tyrosinase inhibitory activity of kaempferol was also assayed versus quercetin, a positive standard.

  • Three isoflavonoids having tyrosinase-inhibiting activity were isolated from the culture filtrate of Streptomyces sp. 20747. Their structures were determined by UV, EI-MS, 1H-NMR, 13C-NMR to be daidzein, daidzein 7-rhamnoside, and genistein 7-rhamnoside, which were competitive with substrate and had IC50 value of 14, 19, and 16 ng/ml, respectively to mushroom tyrosinase and did not inhibit melanin production of Streptomyces bikiniensis. Soybean meal as well as peptone were found to be a good nitrogen source for tyrosinase-inhibiting isoflavonoids production, susgesting that soybean meal is not the origin of tyrosinase inhibiting isoflavonoids formation in Streptomyces sp. 20747 strain.

  • Inhibitory effect on tyrosinase activity and melanogenesis of the mycelia and mycelial culture broth of Macrolepiota procera were investigated. The methanol fraction of culture broth showed 56.6% of tyrosinase inhibitory activity and its IC50 was 8.78 mg/ml. Using Streptococcus bikiniensis for melanogenesis in vitro, the methanol extract of mycelia showed 94.0% inhibition of melanogenesis.

  • Tyrosinase is known to accelerate the melanin polymer biosynthesis in melanocyte, so tyrosinase inhibitors hinder the melanin polymer biosynthesis and are useful not only for th e material used in cosmetics as skin-whitening agents but also as the remedy for disturbances in pigmentation. During our search for new melanin biosynthesis inhibitors from natural sources, 130 higher plants were tested for the inhibitory effect against tyrosinase activity by the mushroom tyrosinase assay. Among them, Carex humilis ($IC_{50}$, 10vg/ml), Sophora flavescence ($IC_{50},\;20{\sim}50{mu}g/ml$) and Styrax japonica ($IC_{50},\;10{\mu}g/ml$) inhibited the tyrosinase activity strongly.

  • For the use of tannins in the whitening-effect cosmetics, inhibitory effect against tyrosinase activity was determined. Three condensed tannins including gallocatechin, gallocatechin 3',4'-di-O-gallate and epicatechin 3-O-gallate and three hydrolyzable tannins, 1,2,6-tri-O-galloyl-${\beta}$-D-glucose, 2,3-(S)-HHDP-D-glucose and pedunculagin showed 15-29% mild inhibitory effects against tyrosinase activity.

  • Nineteen hydrolysable tannins isolated from the Euphorbia helioscopia (Euphorbiaceae) were tested for the inhibitory effect on mushroom tyrosinase activity in vitro. Inhibitory effect of gallotannin group exhibited more potent than that of phenolcarboxylic acid and ellagitannin groups against the enzyme activity. The inhibitory activity by pentagalloyl glucose on mushroom tyrosinase was more potent ($IC_{50}$/, 4.9 $\mu$M) than that of kojic acid ($IC_{50}$/, 8.7 $\mu$M).

  • Melanogenesis is a physiological process that results in the synthesis of melanin pigments. Tyrosinase is a key enzyme for melanin biosynthesis, and hyperpigmentation disorders are associated with abnormal accumulation of melanin pigments, which can be improved by treatment with depigmenting agents. Among the possible melanin-reducing compounds, tyrosinase inhibitors are most promising for preventing and treating pigmentation disorder and are used as skin-whitening agents in the cosmetic industry. In the present study, the effects of fucoidan on melanogenesis and tyrosinase activity of B16F10 melanoma cells were investigated. Melanin synthesis and tyrosinase activity in B16F10 melanoma cells were decreased in a dose-dependent manner by fucoidan. Melanin production and tyrosinase activity in B16F10 melanoma cells stimulated by a-melanocyte stimulating hormone (a-MSH) were inhibited by fucoidan with a dose-dependent manner compared to control. Fucoidan inhibited tyrosinase activity of B16F10 melanoma cells with a dose-dependent manner as assessed by 3,4-dihydroxyphenylalanine (DOPA) staining. In conclusion, these findings indicate that fucoidan, which inhibit melanin synthesis and tyrosinase activity, is an effective skin-whitening agent.

Inhibitory Efficacy of Black Tea Water Extract on Melanogenesis in Melan-a Cells and Its Action Mechanisms (멜라닌 세포에서 홍차 열수추출물의 멜라닌 합성 저해능과 작용기전)

  • To evaluate the whitening effect of Camellia sinensis water extract (CSWE), CSWE was treated to melan-a cells. Total polyphenol contents and flavonoid contents of CSWE were 102 mg/g and 87 mg/g, respectively. The electron-donating ability of CSWE revealed a dose-dependent response, showing the excellent ability of 82% at 800 ${\mu}g$/mL, and which was higher than the arbutin (48%). The CSWE significantly (p<0.001) suppressed the melanin synthesis and the development of melanocyte dendrites was inhibited in a dose-dependent manner. The CSWE significantly (p<0.001) inhibited both intra-cellular and cell-extracted tyrosinase activities. And inhibitory efficacies of CSWE on both melanin synthesis and tyrosinase activity were significantly (p<0.001) higher than the arbutin. The tyrosinase protein expression was not influenced by arbutin treatment. However, CSWE treatment significantly (p<0.001) reduced it. Both arbutin and CSWE treatment did not influence on mRNA expressions of tyrosinase, tyrosinase-related protein-1 and tyrosinase related protein-2.
