리기다소나무 방부 정각재를 이용한 목재 옹벽의 강도 성능 평가 (Strength Properties of Wooden Model Retaining Wall Using Preservative Treated Square Timber of Domestic Pinus rigida Miller)

  • 국내산 주요 침엽수 소경목의 활용 방안으로 리기다소나무를 이용한 목재 모형 옹벽을 제작하여 강도 성능을 평가하였다. 목재 모형 옹벽은 가로, 세로 11 cm의 CUAZ-2 처리한 리기다소나무 방부 정각재를 사용하여 3개의 Type으로 제작하였다. 각 Type별 옹벽은 횡목 4층 종목 3층 총 7층의 높이 86 cm, 길이 300 cm, 폭 93 cm로 제작 하였으며 기본형인 Type I과 목재 사용량을 줄이기 위해 중간에 긴 종목과 짧은 종목을 번갈아 사용한 Type II, Type II와 동일하게 제작한 후 횡목의 연결부위를 보강목으로 보강한 Type III으로 제작하였다. 각 Type별 옹벽은 수평 재하 시험을 통한 강도 성능 평가와 화상처리를 통한 구조물의 변형을 검토하였다. 수평 재하 시험 결과 최대 하중을 비교해 보았을 때 Type I은 63.17 kN/m, Type II는 57.80 kN/m, Type III은 60.97 kN/m으로 나타났다. 화상 처리를 통해 측정된 전면부의 최상층 변형을 비교해 보았을 때 Type I에 비해 Type II의 변형이 약 1.5배 많은 변형을 보였으며 Type I과 Type III은 비슷한 변형을 보이는 것을 알 수 있었다. 실험 결과 Type III의 경우 목재 사용량을 절감하면서도 양호한 강도성능을 확인 할 수 있었다.

국내 수집종 꽃향유의 정유성분 특성 (Characteristics of Volatile Oil Components in Elsholtzia splendens Nakai Collected in Korea)

  • 이 연구는 국내 8개 지역에서 수집한 꽃향유의 정유성분을 분석하여 화학형을 분류하고, 이들 화학형 성분이 항산화 작용과 식품부패균을 억제하는 효과가 있는지를 알고자 하였다. 정유성분 분석결과 3개의 화학형으로 구분할 수 있었다. 화학형 1의 주성분은 dihydrotaketone (DT type)이었다. 개화기를 기준하여 꽃향유의 개체에 따라 naginatketone (NK type)이 60% 이상을 함유한 것과 elsholtziaketone (EK type)을 60% 이상 함유한 개체로 분류할 수 있었고, 또한 이들 두 성분은 종자세대 후에도 EK type은 elsholtziaketone이 주성분으로 나타났고 NK type은 naginataketone이 주성분인 것이 확인되었다. 따라서 NK type은 제2 화학형으로 그리고 EK type은 제3 화학형으로 구별하였다. 항산화 효과는 $6\;mg/{\ell}$ 농도에서 나타났으며, 같은 농도에서 DT>NK>EK 순으로 항산화능을 보였다. NK type은 Streptomyces와 Bicillus에서, EK type은 Streptomyces와 Pseudomonas에서 높은 항균능력을 보였다.

폐염균 다당류 정제 방법의 개발 및 한국형 폐염균의 수집

  • 한국에서 유행하는 페염균 type을 결정하기 위한 기초 실험을 수행하여 서울대병원과 서울중앙병원의 환자로부터 분리한 88개의 폐염균주를 담즙산에 의한 용해도 및 Optochin에 대한 민감성 등을 측정하고 폐염균의 type을 결정하기 위해서 Double diffusion, Countercurrent immunoelectrophoresis, Rocket immunoelectrophoresis 방법 등으로 폐염균 항혈청과 반응하였다. 또한 ATCC로부터 구입한 폐염균주를 이용하여 배지조성의 변화에 따른 폐염균 생장을 측정하기 위해 CAT broth, Brain Heart Infusion broth, Defined Media에서 3가지 type의 균주(Type I, III, IV)를 접종하고 37$^{\circ}C$에서 배양하면서 1시간마다 550nm에서 흡광도를 측정하였다. 3가지 type (Type I, III, IV)의 폐염균주로부터 협막다당류를 phenol Iysis 추출법으로 폐염균 Capsule 다당류의 분리하여 Total Sugar 함량, Uronic acid 함량, 0-Acetyl 함량, 단백질 함량, 핵산 함량, 분자량분포 Countr-current immunoelectrophoresis 등을 실시하여 폐염균 백신 규격에 부합되는지 확인하였다. 수집한 88개의 폐염균주가 모두 담즙산에 의해 용해되고 6mm Optochin disc를 사용하였을때 성장 저지원이 모두 14mm 이상으로 나타나 감수성이 있어서 폐염균임을 확인하였으며 폐염균의 type 결정에 필요한 방법을 설정하였다. 배지조성에 따른 폐염균의 생장을 측정한 결과 CAT broth 에서는 Type I만이 잘 배양되었고 Type III, IV는 늦게 배양되었으나, Brain Heart Infusion broth에서는 3가지 Type이 모두 잘 배양되었고, Defined Media 에서는 Type I만이 성장되었을 뿐 나머지 두 Type은 배양되지 않았다. 본 실험방법으로 분리된 폐염균 다당류는 폐염균 백신 규격중의 6가지 항목에서 적합하였으며 Statens(Denmark)로부터 구입한 항체와 뚜렷한 침강선을 나타내었다.

The Combined Effect of β2- and β3-Adrenergic Receptor Genotypes on Hyperglycemic Risk in the Korean Population

  • Adrenergic receptors play a major role in thermogenesis and lipolysis in brown and visceral adipose tissues, and have been implicated in the pathogenesis of obesity and metabolic disorders. The purpose of this study was to estimate the effects of $\beta$2-adrenergic receptor ($\beta$2AR) and $\beta$3-adrenergic receptor ($\beta$3AR) genotypes on hyperglycemia and obesity in the Korean population. A representative sample consisting of 530 Korean men and women were measured for height, weight, BMI, WHR, obesity index and body composition. The genotypes of $\beta$2AR polymorphism in codon 27 and $\beta$3AR polymorphism in codon 64 were analyzed by the PCR RFLP method. Serum concentrations of fasting glucose, total cholesterol, HDL cholesterol and triglyceride were determined. The frequencies of $\beta$2AR and $\beta$3AR genotype were: both wild type, 62.5% ; only $\beta$2AR variant type, 12.8% ; only $\beta$3AR variant type, 18.8% ; and both variant type, 5.8% ; the frequency of E and R alleles were 0.098 and 0.137, respectively. Among the physiological parameters, fasting glucose level was significantly higher in subjects with both variant type compared with the three other types (p <0.05), Subjects with both variant type had 12%, 12% and 9.3% increases in serum glucose levels compared with wild type, only $\beta$2AR variant type, and only $\beta$3AR variant type, respectively. When logistic regression analysis was conducted to estimate the risk for hyperglycemia, the subjects were selected for fasting blood glucose concentrations of more than 6.105 m㏖/L (110 mg/dL), and the odds ratios were 1.215 (p=0.636) for only $\beta$2AR variant type,1.659 (p=0.089) for only $\beta$3AR variant type, and 3.078 (p=0.011) for both variant type. These results suggest that the interaction of $\beta$2AR and $\beta$3AR variant genotypes has a strong association with increased glucose levels, and might be a significant risk factor for hyperglycemia among Korean subjects.

척추경나사못을 이용한 유합술과 동반 시술된 극돌기간 삽입기구의 생체역학적 연구 (Biomechanical Analysis of a Combined Interspinous Spacer with a Posterior Lumbar Fusion with Pedicle Screws)

  • Recently, during the multi-level fusion with pedicle screws, interspinous spacer are sometimes substituted for the most superior level of the fusion in an attempt to reduce the number of fusion level and likelihood of degeneration process at the adjacent level. In this study, a finite element (FE) study was performed to assess biomechanical efficacies of the interspinous spacer combined with posterior lumbar fusion with a previously-validated 3-dimensional FE model of the intact lumbar spine (L1-S1). The post-operative models were made by modifying the intact model to simulate the implantation of interspinous spacer and pedicle screws at the L3-4 and L4-5. Four different configurations of the post-op model were considered: (1) a normal spinal model; (2) Type 1, one-level fusion using posterior pedicle screws at the L4-5; (3) Type 2, two-level (L3-5) fusion; (4) Type 3, Type 1 plus Coflex$^{TM}$ at the L3-4. hybrid protocol (intact: 10 Nm) with a compressive follower load of 400N were used to flex, extend, axially rotate and laterally bend the FE model. As compared to the intact model, Type 2 showed the greatest increase in Range of motion (ROM) at the adjacent level (L2-3), followed Type 3, and Type 1 depending on the loading type. At L3-4, ROM of Type 2 was reduced by 34~56% regardless of loading mode, as compared to decrease of 55% in Type 3 only in extension. In case of normal bone strength model (Type 3_Normal), PVMS at the process and the pedicle remained less than 20% of their yield strengths regardless of loading, except in extension (about 35%). However, for the osteoporotic model (Type 3_Osteoporotic), it reached up to 56% in extension indicating increased susceptibility to fracture. This study suggested that substitution of the superior level fusion with the interspinous spacer in multi-level fusion may be able to offer similar biomechanical outcome and stability while reducing likelihood of adjacent level degeneration.

공기 중 음이온과 공간 내 식물용적 변화가 미세먼지 정화에 미치는 영향 (The Impact of Negative Ions and Plant Volume Changes in Space on Fine Dust Purification in the Atmosphere)

  • 본 연구는 공기 중 음이온이 미세먼지(PM10, PM2.5) 정화에 미치는 영향을 파악하고, 식물이 공기 중 음이온 발생과 미세먼지 정화에 미치는 영향을 평가하기 위하여 음이온 발생요인별 음이온 발생량을 측정하고, 각 요인별, 식물 용적별 미세먼지 저감 모형을 구축하여 비교하였다. 음이온 발생요인별 특성은 Type N.I(Negative ion generator; 204,133.33ea/cm3) > Type P30(Plant Vol. 30%; 362.55ea/cm3) > Type C(Control; 46.22ea/cm3)의 순으로 음이온 발생량을 살펴보면 무처리구에 비하여 음이온 발생기 처리구에서 약 4,417배, 식물 배치구에서 약 8배 많았다. 이에 따른 음이온 발생원별 미세먼지 저감 특성은 PM10에서 Type NI가 Type C에 비하여 정화효율이 2.52배, Type P30이 1.46배 높았으며, PM2.5의 경우, Type NI가 Type C에 비하여 정화효율이 2.26배, Type P30이 1.31배 높은 것으로 분석되었다. 식물의 용적별 미세먼지 정화 효율은 Type P20(84.60분) > Type P30(106.50분) = Type P25(115.50분) = Type P15(117.60분) > Type P5(125.25분) = Type P10(129.75분)의 순이었으며, 초미세먼지의 경우 Type P20 (104.00분) > Type P30(133.20분) = Type P25(144.00분) = Type P15(147.60분) > Type P5(161.25분) = Type P10 (168.00분)의 순이었다. 이렇게 음이온의 미세먼지 정화 능력과 식물의 미세먼지 정화능력을 정량적으로 분석하였으며, 향후 미세먼지 정화를 고려한 녹지계획 및 식물식재에 고려해야할 사항을 제안하였다.

간호대학생들의 기본간호실습태도에 대한 주관성 연구 (An Inquiry into Subjectivity of Fundamental Nursing Practice Attitude)

  • The study was designed to identify the attitude of nursing students to fundamental nursing practice through Q-methodology. A Q sample was developed through a review of the literature and interviews. Forty statements made up the finalized Q-sample. The P sample consisted of 25 nursing students in S College. Q statements were written on separate cards and were given to the 25 subjects to sort according to degree of agreement or disagreement. The Q-sort by each subject was coded and analyzed with QUANL PC Program. The analysis discovered three major attitudes, namely "type 1: passive demand", "type 2: active self-confidence", "type 3: practical application". The correlation was .213 between type 1 and 2, .409 between type 1 and 3, .379 between t ype 2 and 3. The results revealed three different types of fundamental nursing practice attitude; 1) Passive demand type: they were not satisfied with fundamental nursing practice time, the number of persons, practice machines. They presented anxiety and worry through fundamental nursing practice. Therefore, they will presenta passive attitude of clinical practice experience. 2) Active self-confidence type: they experienced pride as a nursing student and fascination as a nurse was an acquired recognition. 3) Practical application type: they practiced that fundamental nursing skill was applied their family and oneself. Therefore they had tension through initial fundamental nursing practice but they gained self-confidence and interest through practical study. In conclusion, the researchers suggest that the education program would be more effective if it was planned considering to each types of attitude of nursing students for fundamental nursing practicer fundamental nursing practice

PCR Approach for Detection and Typing of Epidermodysplasia Verruciformis-associated Human Papillomavirus Types

  • Warts, or verrucae, are benign epithelial proliferations of the skin and mucosa caused by infection with human papillomaviruses (HPV). It is now recognized that there are many different HPV types. Especially type3 is most frequently observed in flat wart. Other types, such as type2, 10, 14, 27, 28, 29, 38, and 41 are rarely encounted in flat wart. We describe here a simple and economic method for detection and identification of epidermodysplasia verruciformis-associated HPV. The method is based on polymerase chain reaction (PCR) amplification and restriction analysis. The method has been developed with cloned HPV DNA and DNA from clinical samples. Clinical samples are from either frozen tissue or paraffin-embedded tissue. Genomic fragments were obtained from two different HPV types (3 and 10). The amplification fragments were identified by a form of miniature fingerprinting, with a set of restriction enzymes that gave a unique digestion pattern for each HPV type. We have tested 74 clinical samples. Only type3 among these clinical samples is detected, and one sample is involved in neither type3 nor type10.

접합부 편심을 고려한 단층 래티스돔의 좌굴특성에 관한 연구 (A Study on Effect of the Junction's Eccentricity for Buckling Characteristics of Single-Layer Latticed Dome)

  • In Single-layer latticed domes with rectangular network which is composed of ring of circumferential direction and rafter of longitudinal direction, that is, rib domes, if we use the cross-membered junction's method for the advantage in fabrication and construction, the eccentricity is occurred in the nodal point of crossing members. This paper is aimed at investigating the buckling characteristics for the effect of eccentricity according to rise-span ratios and distance of eccentricity. Analysis method is based on FEM dealing with the geometrically nonlinear deflection problems. The conclusion were given as follows: 1. The maximum decreasing ratio of buckling strength due to the junction's eccentricity is about 60% in models of this paper. 2. In the increasing ratio of buckling strength for rise-span ratio, that of Type 3 models is larger than that of type 2 models. On the other hand, that of Type 2 mode is larger than that of Type 3 for eccentricity-distance. 3. In the viewpoint of the value of buckling strength, that of Type 2 models is larger than that of type 3 models. The effect of the junction's rigidity on buckling strength is not great for overall models. Therefore if we use the cross-membered junction's method for the advantage in fabrication and construction, the method of Type 2 will have the great advantage of that of Type 3.

The Relationship between Papanicolaou Smear Test and Human Papilloma Virus DNA Chip Test in the Uterine Cervix

  • The genotypes of Human Papilloma Virus (HPV) are important in the carcinogenesis of uterine cervical cancer. Diagnosis of uterine cervical cancer screening has been executed using Papanicolau method (Pap) and HPV DNA Chip method. We researched the interrelation of HPV DNA genotypes in single and multiple infections and analyzed the results of Pap and HPV DNA Chip tests at Gunsan Medical Center (GMC). The correlation analysis was surveyed on collected results from 599 patients who have been tested with both Pap and HPV DNA chip tests from November 2004 to May 2010 at GMC. The inconsistency between Pap and HPV DNA Chip tests was 41.1%. The HPV DNA Chip genotype related with high risk cases were type 16 (13.5%), type 52 (10.5%), type 58 (10.1%), and type 18 (3.4%). Those related with low risk cases were type 70 (8.9%), type 6 (1.7%), type 40 (1.2%), type 11 (1.3%), and other types (14.3%). Among the 195 cases of HPV positive status, 161 cases were associated with single infection; 108 (67.1%) cases were related with high risk genotype; 19 (11.8%) cases were low risk genotype; 31 (21.1%) cases were related with other types. 29 cases were associated with double infections; 23 (79.3%) cases were high risks; 5 (17.2%) cases were mixed high and low risks; 1 (3.5%) case was low risk.
