Newcastle disease (ND) is a highly contagious infection of poultry, Two pathology-based techniques, in situ RT-PCR and in situ hybridization (ISH) were applied to formalin-fixed, paraffin-embedded tissues from chickens naturally infected with velogenic ND virus (VNDV). Two pairs of primers and a probe for ISH and in situ RT-PCR, respectively, were selected from highly conserved region of matrix gene of NDV. The ISH experiment was carried out using MicroProbe$^{TM}$ capillary action system within 2 hours. In situ RT-PCR was performed using MicroProbe$^{TM}$ capillary action system and GeneAmp In Situ PCR system. With ISH and in situ RT-PCR, viral nucleic acid was detected in the central nervous system of chickens from infected with neurotropic velogenic Newcastle disease virus (NVNDV), whereas viral nucleic acid was detected in various organs or tissues of chickens from infected with viscerotropic velogenic Newcastle disease virus (VVNDV). In the NVND group, positive signals were characteristically defined in the cytoplasm of neuron, vascular endothelial cells, and perivascular mononuclear macrophages in the central nervous system. One of NVND group, chicken from one farm exhibited positive signals in the bronchial epithelium. The VVND group chickens showed positive reaction in the macrophages, vascular endothelium, and bronchiolar epithelium. Markedly, viral nucleic acid was detected in the macrophages of morphologically normal tissues which were peripheral or located in distant areas from lesions. The central nervous system of chickens infected with VVND virus had positive signals in the vascular endothelial cell, perivascular mononuclear macrophages and some neuron. The number and intensity of the positive cells by in situ RT-PCR were more and stronger, respectively, in comparison with those by ISH. Particularly, positive reaction was detected in macrophages infiltrating in cardiac muscle by in situ RT-PCR, but not obtained by ISH. Therefore, these results demonstrated that ISH is a rapid diagnostic method for detection of NDV and in situ RT-PCR can be used as an efficient method for detection of low viral load infection or subclinical viral infection of NDV.
반합성 베타 락탐 항생물질의 가수분해 및 합성을 촉매하는 효소인 $\alpha$-acylamino-$\beta$-lactam acylhydrolase(ALAH)의 유전자를 Acetobacfer turbidans로부터 클론화하기 위한 연구를 수행하였다. 우선 순수 분리 정제된 효소에 대한 항혈청 (폴리클론 항체)을 제조한 다음 이를 probe로 하여 면역화학적 방법으로 유전자의 선별을 시도하였다. 이러한 용도로 개발된 운반체인 λ gtll에다 A. turbidans의 유전자 단편들을 삽입하여 genomic library를 제조한 후 이 library에서 유전자를 선별한 결과 두개의 positive clone을 얻을 수 있었다. 그러나. 이 두 clone들은 면역화학적으로 서로 다른 반응을 나타내었는데, 그 중 하나는 효소의 항혈청과는 잘 결합하나 융합되어진 베타 갈락토시다아제에 대한 항체와는 잘 결합하지 못하였고(λ gtll dn1), 또 다른 clone 은 이와 반대의 양상을 보여주었다(λ gtll dn2). 더구나 이들 clone을 여러 제한효소들로 분석해본 결과, 유전자가 삽입된 부분인 Eco RI 부위중 하나가 없어진 것을 알 수 있었다. 따라서 A. turbidans의 효소에 대한 유전자가 λ gtll에 클론화 되었으나 이 유전자와 베타 갈락토시다아제의 유전자(lacZ)간에 염기배열상 동위성이 있은 부위가 존재하여 재조합된 λ gtll 파지의 복제과정에서 삭제되어진 것으로 간주되어진다.
선미 반류분포를 계측하기위해 수조에서 흔히 사용하는 5공 피토관의 새로운 캘리브레이션 방법을 제안하였다. 기존의 한 각도(수평 또는 수직)만을 고려하는 1차원 캘리브레이션 방법과는 달리, 본 논문에서 제안한 2차원 차트 캘리브레이션 방법은 수평각과 수직각을 동시에 변화시켜 얻어진 2차원 차트를 이용하여 종전에 큰 각도에서 발생하던 오차를 줄임으로써 계측 정도를 크게 향상시켰다. 또한 종래의 1차원 방법에 맞추어 구성된 계측 시스템을 수정 없이 사용할 수 있도록 상관계수를 정의함으로써 사용의 편의를 도모하였다.
The systems with probe and SIL(Solid Immersion Lens) mechanisms have been researched as the technology to perform NFR(Near Field Recording). Most of them use the flying head mechanism to accomplish high recording density and fast data transfer rate. In this paper, ABS shape of flying head was optimized with the object of securing the maximum compliance ability of OFH. We suggest low different optimization processes to predict the static flying characteristics for the OFH. Two different approximation methods, regression analysis and back propagation neural network were used. And we compared the result of directly connected(between CAE and optimizer) method and two approximated optimization results. Design Optimization Tool(DOT) and ${\mu}GA$ were used as the optimizers.

An experimental study on the flow characteristics was performed for a two-dimensional turbulent wall attaching offset jet at different oblique angles to a surface. The flow characteristics were investigated by using a split film probe with the modified Stock's calibration method. The jet mean velocity, turbulent intensity, wall static pressure coefficient profiles, and time-averaged reattachment point were measured at the Reynolds number Re (based on the nozzle width, D) ranging from 17700 to 53200, the offset ratio H/D from 2.5 to 10, and the inclined angle .alpha. from 0.deg. C to 40.deg. C. The Correlations between the maximum pressure position, minimum pressure position, and reattachment point and offset ratios, and inclined angles are presented.
After the patterning and probe process of wafer have been achieved, the dicing process is necessary to separate chips from a wafer. The dicing process cuts a wafer to lengthwise and crosswise direction to make many chips by using narrow circular rotating diamond blade. But inferior goods are made under the influence of complex dicing environment such as blade, wafer, cutting water and cutting conditions. This paper describes a monitoring algorithm using feature extraction in order to find out an instant of vibration signal change when bad dicing appears. The algorithm is composed of two steps: feature extraction and decision. In the feature extraction, two features processed from vibration signal which is acquired by accelerometer attached on blade head are proposed. In the decision. a threshold method is adopted to classify the dicing process into normal and abnormal dicing. Experiment have been performed for GaAs semiconductor wafer. Based upon observation of the experimental results, the proposed scheme shown a good accuracy of classification performance by which the inferior goods decreased from 35.2% to 12.8%.

In this work, a new non-photolithographic micro-fabrication technique is presented. The motivation of this work is to overcome the demerits of the most commonly used photolithographic techniques. The micro-fabrication technique presented in this work is a two-step process which consists of mechanical scribing followed by chemical etching. This method has many advantages over other micro-fabrication techniques since it is simple, cost-effective, rapid, and flexible. Also, the technique can be used to obtain a metal structure which has sub-micrometer width patterns. In this paper, the concept of this method and its application to microsystem technology are described.

The BEM (Boundary Element Method) is a computational technique for the approximate solution of problems in continuum mechanics. In the BEM both volume and surface integrals transformed into boundary integral equations. So, we applied the ECM (Electrochemical Machining) process to boundary problem, because our focus is only deformed shape. The ECM process is modeled as a two-dimensional problem assuming constant properties of electrolyte, and an incremental formulation is used with automatic mesh regeneration. As a result the final shape is roughly agreed with experimental shape. But, it has an error of exact shape, because a chemically factor is not considered
This paper describes a synthesized photoelastic method developed for the visualization and evaluation of sound pressure distribution of elastic wave in a solid. The visualization of wave stress field is achieved by synthesizing two photoelastic pictures, in which the direction of the principal axis of linear polariscopes differs by $45^{\circ}$. From the analysis of the wave stress distribution using this method, it is possible to evaluate the characteristics of elastic waves in a solid, such as the intensity of stress, directivity and resolution characteristics of the wave emitted from a commercial probe, and characteristics of scattering from various types of defects.
