The tissue water relation parameters were compared to assess the resistance of representative three gardening tree species to water stress. Zelkova serrata showed a strong resistant characteristics to water stress compared to Betula platyphylla var. japonica and Liriodendron turipifera. Turgor pressure at turgor loss point in Z. serrata was -2.54 MPa, whereas those of 5. platyphylla var, japonica and L. turipifera were -2.24 and -2.03, respectively. In addition, Z. serrata showed less reduction in pressure potential to decrease of free water content in the leaf tissue, indicating effective shrinking of the cell wall compared to others. On the other hand, L. turipifera indicated a weak resistance to water stress, which has low turgor potential at turgor loss point and cell wall elasticity. These results suggest that Z. serrata would be suitable for relatively dry conditions of location and humid conditions of location would be suitable for L. dendronas plantation.
To assess resistance of transgenic tobacco plants which overexpress superoxide dismutase (SOD) and ascorbate peroxidase (APX) in chloroplasts to water stress, changes in leaf water potential, turgor potential, stomatal conductance and transpiration rate were measured. Leaf water potential in all plants remained high up to day 4 after withholding water but thereafter decreased markedly. In spite of a remarkable decrease in leaf water potential, some of transgenic plants maintained higher turgor potential compared with control plant on day 12. In particular, the transgenic plant expressing MnSOD showed an outstanding maintenance in turgor pressure by osmotic adjustment throughout the experiment, resulting in high stomatal conductance and transpiration rate. However, among transgenic plants, osmotic potential was reduced more effectively in multiple transformants such as the double transformant expressing both MnSOD and APX, and the triple transformant expressing CuznSOD, MnSOD and APX than single transformants. Consequently, further research is needed to get general agreement on the tolerance of transgenic plants to water stress at different growth stages for each transgenic plant.
Diurnal changes of microclimatic conditions and tissue water relations were measured at two sites where Carpinus laxiflora and C. cordata were allopatrically distributed. The microclimatic conditions at a site where C. laxiflora was distributed produced severe water stress condition during summer months. Daily maximum temperature reached $30.4^\circC$ and the highest vapor pressure deficit was 1.31 KPa when 13 rainless days were continued. During this period soil water content decreased to below the field capacity even at a depth of 20 cm and xylem pressure potential also decreased to 2.04 MPa. However, turgor potential was maintained more than 0.4 MPa. Patterns of stomatal conductance were changed with evaporative demand and soil water availability. On the other hand, microclimatic conditions at a site where C. cordata was distributed were moderate water strees condition compared with those at a site C. laxiflora was distributed. Though soil water content was maintained above field capacity C. cordata showed a remarkable decrease in turgor potential and stomatal conductance throughout the experiment. These results indicate that there is a difference in habitat characteristics between the two species and C. laxiflora is more resistant than C. cordata to water stress.
In order to compare the adjustment of 6 oak species to water stress, the components of water status, tissue elastic modulus, free proline content of leaves and morphological characteristics were determined in pot culture. uercus dentata and . mongolica responded effectively to drought with high root : shoot (R/S) ratio or maintenance of high turgor pressure by large and fast osmotic adjustment and . variabilis with maintenance of high turgor pressure by low elastic modulus under drought. Meanwhile, . aliena and . serrata responded effectively with low omotic potential (Ψo) at full saturation and . acutissima with long root in spite of rigid cell wall and high osmotic potential (Ψo) at full saturation. Proline content in leaves of . dentata, . mongolica and . aliena increased early and rapidly at high leaf water potential (Ψleaf). The results indicate that 6 oak species have adjustment different from each other to water stress.
Plant water status during growth is directly and indirectly associated with seed yield. The objective of the present study was to determine the genotypic differences in leaf water characteristics at an early growth stage of soybean [Glycine max (L.) Merrill] plants through the pressure-bomb technique. Measurements of water potential as well as relative water content (RWC) were made at the third leaf from the fully-expanded top leaf of eight different soybean genotypes grown for 31 to 35 days after field emergence. On the basis of the modified exponential model, pressure-volume (PV) curves were fitted well ($R^2$=0.92** to 0.99** for the curvi-linear region and R=0.67** to 0.96** for the linear region), indicating that a segmented model using PROC NLIN of SAS could be used effectively to estimate the leaf water characteristics. The regression analysis for the pressure-volume (PV) curve revealed genotypic variation in the solute potential at saturation (Ψ$_{s,sat}$ :-10.7 to -14.8 bar), solute potential at incipient plasmolysis (Ψ$_{s,ip}$ : -14.3 to -18.3 bar), RWC at incipient plasmolysis (RW $C_{ip}$ : 83.3 to 91.7%), high integrated turgor pressure from saturation to plasmolysis ( $_1$$^{b}$ : 0.39 to 0.81), and maximum volumetric modulus of elasticity ($\varepsilon$$_{max}$ : 150 to 445 bar).).
The leaf water relations and photosynthetic rate during acute soil drying were compared in potgrown grapevine cultivars, Vitis vinifera cv. Chardonnay and V. vinifera cv. Riesling. Leaf water potential in Riesling decreased significantly from day 2 after water had been withheld, while in Chardonnay leaf water potential for the water-stressed plants was almost identical with that in well watered plants during the first 4 days. Higher stomatal conductance and photosynthetic rate in Chardonnay than Riesling were observed until day 3 after withholding water. Photosynthetic rate in water-stressed Chardonnay was not different from that in control plants until day 3 after withholding water, while that in water-stressed Riesling was reduced markedly from day 2. In Riesling, osmotic potential at turgor loss point was not changed irrespective of watering conditions. However, in Chardonnay osmotic potential at turgor loss point decreased more in the water stressed conditions than in well watered conditions. The osmotic adjustment in Chardonnay under water stress conditions must contribute to the maintenance of higher stomatal conductance and photosynthetic rate than those in Riesling for a significant period of the drying process. Though difference in stomatal conductance between the two cultivars was shown in the process of soil drying, stomatal conductance of both cultivars responded to vapor pressure difference between leaf and ambient air, rather than soil water status and leaf water potential.
Comparative water relations of Quercus acuta and Castanopsis cuspidata var, sieboldii were analyzed to assess their resistance to drought and low temperature stresses from early November to early December, As air temperature decreased both species showed an increased content of osmotically active solute concentration per unit of dry weight (NS/DW), leading to lower osmotic potential of both species at both full turgid state $(OP_{sat})$ and turgor loss point $(OP_{tlp})$ in December than November. No major difference in the ability to adjust osmotically was noticed between the two. This finding suggests that both species must respond adaptively under water and low temperature stresses to maintain turgor pressure in winter season. In addition to osmotic adjustment, a low bulk modulus of elasticity $(E_{max})$ shown in Castanopsis cuspidata var. sieboldii must also play an important role in turgor maintenance during winter season being apt to happen water and low temperature stresses in plants.
This experiment was conducted to investigate the effect of water states (water, osmotic potential and turgor pressure) of fruit vesicle and leaf on soluble solids and organic acid contents of fruits of 'Shiranuhi' mandarin hybrid, M16 A line during the fruit maturing season in plastic house cultivation. The 'Shirauhi' grafted on 'Swingle citrumelo', strong strength of rootstock, produced fruit with lower soluble solids and organic acid content than 'Trifoliate' orange rootstock. The fruits vesicle water potential and turgor pressure measured before dawn in 'Swingle citrumelo' were higher tendency than the 'Trifoliate' orange, but osmotic potential values were lower than the 'Trifoliate' orange. The changes of leaf water potential were very similar to the fruit. The results suggest that in the 'Shirauhi' fruits grafted on two rootstocks changes of soluble solids and organic acid content of the fruit were influenced by the leaf water potential and the osmotic potential of the fruit vesicles, which might be caused by the difference of root distribution between two rootstocks.
This study was to elucidate the relation between the water relations parameters obtained from P-V curves and stomatal closure. The results obtained are as follows: 1) The water potential at incipient plasmolysis obtained from P-V curves was similar to the water potential at critical stomatal closure. 2) The critical stomatal closure of sun leaves appear at -21 bar (-17 bar, shade leaves) in Pinus koraiensis, -20 bar in Pinus rigida, -22 bar in Pinus densiflora, and -24 bar in Larix leptolepis. On a relative water content basis, the critical stomatal closures of sun leaves appear at 85% (82%, shade leaves) in Pinus kordiensis, 77% in Pinus ragida, 85% in Pinus densiflora, and 70% in Larix leptolepis. 3) The incipient stomatal closures of sun leaves appear at -14 bar (-12 bar, shade leaves) in Pinus koraiensis, -10 bar in Pinus rigida, -15 bar in Pinus densiflora, and -6 bar in Larix leptolepis. On a relative water content basis, the incipient stomatal closures of sun leaves appear at 90% in Pinus koraiensis, 93% in Pinus rigida, 90% in Pinus densiflora, and 93% in Larix leptolepis. 4) The leaf conductance was increased by increase in volume-averaged turgor pressure was linearly increased by increase in relative water content.
The water relations parameters such as original osmotic pressure $(\Pi_0)$, osmotic pressure $(\Pi_p)$, at the incipient plasmolysis, the volume of osmotic water $(V_o)$ at the full turgor, the relative water content $(RWC^*)$ at the incipient plasmolysis, the volume of osmotic water$(V_p)$ at the incipient plasmolysis, the volume of symplasmic and apoplastic water$(w_s)$ at the maximum turgor, $V_pV_o, \; V_oW_s, \; W_sD_w, \; V_oF_w$ and the elastic modulus were measured through the pressure chamber (DIK-PC-40 Model) technique with the Williams and Geumgangdaerip soybean cultivar shoots. The original osmotic pressure$(\P $i_{0}$ )$, of the both cultivars shoots were appeared insignificant differences as 8.1 bar and 7.8 bar respectively. The osmotic pressure($\pi$$_{p}$ ) at the incipient plasmolysis was 9.0 bar in Williams and 10.4 bar in Geumgangdaerip. The relative water content(RW $C^{*}$) at the incipient plasmolysis was 86.0 per cent in Geumgangdaerip and 92.6 per cent in Williams. The ratio of volume of osmotic water( $V_{p}$ ) at the incipient plasmolysis to total symplasmic water$(V_o)$ , i.e., $V_p/V_o$ was 83.4 per cent in Geumgangdaerip and 90.4 per cent in Williams. The elastic modulus of Geumgangdaerip shoots showed higher value than Williams$(8.5 $\times$ 10^2bar)$ as $1.6 \times10^5$ bar.r.
