Effect of Maillard reaction with xylose, yeast extract and methionine on volatile components and potent odorants of tuna viscera hydrolysate

  • The aim of this research was to enhance the flavor of visceral extracts from skipjack tuna. Flavor precursors and the optimum condition for the Maillard reaction were determined. The flavor extract was prepared from the tuna viscera using Endo/Exo Protease controlled in 3 factors; temperature, enzyme amounts and incubation time. The optimal condition for producing tuna viscera protein hydrolysate (TVPH) was 60℃, 0.5% enzyme (w/w) and 4-hour incubation time. TVPH were further processed to tuna viscera flavor enhancer (TVFE) with Maillard reaction. The Maillard reactions of TVFE were conducted with or without supplements such as xylose, yeast extract and methionine. The Maillard volatile components were analyzed with gas chromatography-mass spectrometry. Sixteen volatiles such as 2-methylpropanal, methylpyrazine, 2,5-dimethylpyrazine, dimethyl disulfide and 2-acetylthaizone were newly formed via Maillard reaction and the similarity of volatile contents from TVPH and TVFE were virtualized using Pearson's correlation integrated with heat-map and principal component analysis. To virtualize aromagram of TVPH and TVFE, odor activity value and odor impact spectrum (OIS) techniques were applied. According to OIS results, 3-methylbutanal, 2-methylbutanal, 1-octen-3-ol 2,5-dimethylpyrazine, methional and dimethyl trisulfide were the potent odorants contributed to the meaty, creamy, and toasted aroma in TVFE.

어류 프레임 농축액과 시판 사골 농축액의 위생적 및 영양적 특성 비교 (Comparison on Sanitary and Nutritional Characteristics Between Skipjack Tuna Ktsuwonus pelamis Frame and Commercial Beef Bone Extract Concentrates)

  • There is increasing interest in developing flavor-enriched concentrate using byproducts of skipjack tuna Ktsuwonus pelamis as a replacement for commercial beef bone extract concentrate (CBB-EC). This study was compared on sanitary and nutritional characteristics between skipjack tuna frame extract concentrate (ST-EC) and CBB-EC as a control of skipjack tuna frame extract concentrate. The moisture contents of CBB-EC and ST-EC 41.4-4.9% and 45.3%, respectively. The results of viable cell counts and E. coli assays suggested that CBB-EC and ST-EC is a safe and sanitary for use in food. There were no differences in peroxide values among CBB-ECs. The total amino acid contents in CBB-EC and ST-EC were 25.6-29.3 g/100 g and 37.9 g/100 g, respectively, and the major amino acids present glutamic acid, proline, glycine, alanine and arginine. Regardless of kinds of the mineral, mineral contents of ST-EC higher than those of CBB-EC. The major fatty acids were 16:0, 18:0 and 18:1n-9 in CBB-EC, 16:0, 18:1n-9 and 22:6n-3 in ST-EC. These results can be used as background information to develop flavor-enriched concentrates from byproducts of skipjack tuna as replacement for CBB-EC.

Preparation of Tuna Skin Byproduct Film Containing Pinus thunbergii Cone Extract

  • Tuna skin byproduct extract (TSB) was used as a biocompatibility film base material, and its composite film with gellan gum (GG) was prepared. In addition, Pinus thunbergii cone extract (PTCE) was incorporated into the film to provide anti-oxidant and anti-bacteria activities. The tensile strength (TS) of the TSB/GG composite films increased with increasing GG content, whereas elongation at break (E) decreased. TSB/GG film at a ratio of 0.5:0.5 (w/w) showed the most desirable TS and E values. Based on scavenging free radical potentials and disc diffusion method results against growth of bacteria, antioxidant and anti-bacteria activities of films increased with increasing PTCE concentration. Accordingly, this study showed that TSB/GG could be used as a film material while the TSB/GG composite film containing PTCE can be utilized as functional packaging.

Effects of Water Extract of Smoke-dried Skipjack Tuna on Memory in a Scopolamine-induced Amnesia Animal Model

  • Natural products have been used to treat many neurological illnesses such as Alzheimer's disease. In the present study, the effects of the water extract of smoke-dried skipjack tuna (WSST), which is used as a traditional seasoning in Japan, as well as its fractions on acetylcholinesterase (AChE) inhibition in vitro and on memory in scopolamine-induced amnesia mice in vivo were evaluated. Bio-Rad P-2 gel permeation chromatography revealed the presence of 7 peaks and AChE significantly inhibited peak 3 and 5. When in vivo behavioral studies were conducted, a passive avoidance test revealed that treatment with 50 and 100 mg/kg WSST as well as with fraction 3 and 5 improved the loss in memory retention induced by scopolamine. These results suggest that skipjack tuna extract and its fractions improve memory deficits and that these substances are suitable for use in healthy foods designed to improve memory deficits induced by aging and Alzheimer's disease.

가다랑어 훈연조미제품 유래 저분자물질의 생리활성 (Biological Activities of Low Molecular Weight Substances Fractionated from the Seasoned and Smoked Products of Skipjack Tuna (Euthynus pelamis))

  • We studied the anticancer, antioxidative, ACE inhibitory, antithrombic, and cerebral ischemia inhibitory activities, of low molecular weight substances fractionated from the seasoned and smoked products of skipjack tuna (Euthynus pelamis). Low molecular weight substances fractionated from the liquid extract of seasoned and smoked skipjack tuna powder showed 7 peaks on gel permeation chromatography. Anticancer activity was the highest at Peak 3 $(54.23%)$ followed by Peak 4 $(41.58\%)$. The potency order of antioxidative activity was the highest at Peak 3 $(94.14\%)$ followed by Peak 2 $(82.07\%)$ and Peak 5 $(81.24\%)$. ACE inhibitory activity was the highest at Peak 3 $(67.27\%)$ followed by Peak 5 $(45.62\%)$. But there was weak anti thrombotic effect (PT and APTT) in the low molecular weight substances of the seasoned and smoked products of skipjack tuna. Cerebral ischemia inhibitory activity was the highest at Peak 3 $(80.0\%)$ followed by Peak 5 $(40.0\%)$.

막반응기에서 참치 유문수 유래 단백질 분해효소를 이용한 참치 자숙액의 연속적 가수분해 (Continuous Hydrolysis of Tuna Boiled Extract using Proteinase from Tuna Pyloric Caeca in Membrane Reactor)

  • 참치 자숙액 중에 함유되어 단백질을 효율적으로 이용하고자 막반응기에서 연속적으로 효소적 가수분해를 수행하기 위한 최적 조건을 검토하였다. 회분식에서 TPCCE를 이용한 자숙액의 최적 가수분해조건은 pH 9.0, 반응온도는 $40^{\circ}C$, 기질인 자숙액에 대한 효소비 50(w/w) 및 기질농도 $1\%$ (w/v)였으며, 이 때 6시간의 반응시간에 의해 약$70\%$의 가수분해도를 나타내었다. 그리고 TPCCE를 이용하는 것이 시판 단백질 분해효소보다 효과적이었다. 막반응기에서 자숙액을 연속적으로 효소적 가수분해를 수행하기 위한 막반응기의 작동조건의 검토에서, 기질용액을 pH 9 및 $40^{\circ}C$로 조절한 후 자숙액의 반응시간에 따른 가수분해도는 3시간, $1\%$ (w/v) 자숙액에 대한 효소농도는 0,1 g/$\ell$, 자숙액 대 효소비는 100 (w/w), 그리고 기질농도는 $5\%$ (w/v)였으며, 이 때 가수분해도는 약 $60\%$를 나타내었다.

참치 추출물의 항암 및 면역효과 (Antitumor and Immunological Effects of Tuna Extract)

  • 본 연구는 참치의 석유 에텔 추출물로 부터 항암 효과가 있는 활성 성분을 분리 정제하고, 흰생쥐의 임파모 세포인 LI210 과 P388, 인체 장암세포인 HCT-48, HT-29 및 HRT-18를 대상으로 단계별로 얻어지는 성분의 항암 활성올 측정하고, 그 물질의 생체내에서의 작용 기전을 밝히기 위해 여러가지 면역학적 실험을 수행하였다. In vitro배양액에서의 인체 결장암세포인 H HCT-48에 대해, crude extract는 배양액 1ml강 $60\mu\textrm{g}$ 첨가시 증식이 2배로 억제되었으며(1 unit), 이를 column chromatography 수행하여 얻은 fraction D의 1 unit은 $2 22\mu\textrm{g}$이며, 다시 TLC를 하여 얻은 Spot I 의 1 unit은 $9\mu\textrm{g}$으로서 정제 했을때 277배 증가하였다. Spot I 은 GC/ M5에 의해 7가지 성분이 섞여있는 혼합물로서 이 중 4가지 생분은 $C_{14:0},\;C_{16:0},\;C_{17:1},\;및\;C_{18:0}$인 지방산으로 확인이 되었으며, 나머지 3가지는 지방 성분으로 분자식은 확인되지 않았다. 다른 인체 장암 세포인 HT-29과 HRT-18에 대한 crude extract의 1 unit은 $64\mu\textrm{g}\;및\;54\mu\textrm{g}$이었으며 frattion D의 1 unit에 해당하는 양은 두 세포 모두 약 $34\mu\textrm{g}$이었다. 또한, 생쥐의 백혈병 성 임파모 세포인 L1210과 P388의 경우 crude extract의 1 unit에 해당하는 양은 $2-2.5\mu\textrm{g}$이며, fraction D의 경우는 $1.2-1.4\mu\textrm{g}$으로서 참치 추출물의 세포 종식 억제력은 백혈병성 임파모세포에서 인체 장암세포보다 높았으며, 암세포의 종류에 따라 효과의 차이가 있었다. 참치 추출물(fraction D)을 첨가한 배양액에서 배양한 HCT-48 세포획 크기 분포도는 배양시간이 경과함에 따라 세포의 크기가 작아지며, 현미경 사진에서 보면 본래의 형태가 위축되거나, 세포의 주위가 흐트러지면서 사면 현상을 보였다. 참치 석유 에텔 추출물에는 DHA가 18.8%를 차지 하며 EPA는, 3.25% 포함되어 있었다. 대조군 생쥐에 비해 S-180을 접종한 후 참지 추출물을 투여한 생쥐에서 용혈반 형성 세포수와 혈청 단백질 중 immunoglobulin의 relative %가 현저히 증가되었다. 그러나 비장의 자연 살해 세포의 활성에는 변화를주지 않았다. 이상의 결과로 보아, 참치의 석유 에델 추출물중에 in vitro에서 인체 장암세포 및 백혈병성 임파모세포의 증식 억제 및 생체내에서 면역 증강 효과를 나타내는 성분이 존재함을 알 수 있다.

  • The anti-inflammatory effect of tuna heart water extract (THWE) was investigated using lipopolysaccharide-induced inflammatory response in this study. Anti-inflammatory effect was detected by the cell proliferation and the production levels of nitric oxide, pro-inflammatory cytokines such as interleukin-6 (IL-6), IL-$1{\beta}$, and tumor necrosis factor-alpha. As a result, there were no cytotoxic effects on proliferation of macrophages treated with THWE compared to the control. The production of pro-inflammatory cytokines was remarkably suppressed compared with that of the LPS only group. These results suggest that THWE exerts the anti-inflammatory property by inhibiting production of inflammatory factors and may be a potential material for anti-inflammatory therapy.

효소를 이용한 동물성 식품가공부산물의 식품 소재화 탐색 (Study on Utilization of Animal By-products from Food Processing by Enzyme Treatment)

  • 식품 가공의 부산물 중 동물성 부산물을 재활용하여 조미소재로의 개발 가능성을 탐색하였다. 부산물 원료로는 돼지뼈(PB)와 닭뼈(CB), 그리고 참치 혈합육(TDF)을 사용하였다. PB와 CB를 열수로 18시간 추출한 후 단백질과 지방 분해효소를 처리하여 가수분해물을 제조하였다. 각각의 열수 추출물에서 지방의 제거를 위하여 1차로 lipase활성이 있는 pancreatic enzyme을 처리하고 2차로 Flavourzyme을 사용하는 경우 가수분해도(DH)가 PB는 약 70%, CB는 약 80%까지 증가하였다. 지방이 적은 TDF는 단백질 분해효소인 Alcalase와 Flavourzyme으로 분해한 결과 고형분 회수율은 약 22%, 단백질 회수율은 9%이었으며, 효소분해액 중 유리 아미노산 함량은 고형분 중 27%까지 향상되었다. 세 종류 분해액에 가염한 후 관능적인 특성을 조사한 결과 전체적으로 TDF>PB>CB의 순으로 우수하여 참치 혈합육은 조미소재의 원료로 적합한 동물성 부산물로 판단되었다.

향미강화농축물 소재로서 비가열 가다랑어(Katsuwonus pelamis) 프레임의 영양학적 품질 특성 (Nutritional Characterization of the Uncooked Skipjack Tuna Katsuwonus pelamis Frame as a Basic Resource for Flavor-enriching Concentrates)

  • We investigated the nutritional characteristics of the uncooked Skipjack tuna Katsuwonus pelamis frame (U-STF) as a basic resource for preparing flavor-enriching concentrates. The bone rate based on the fish frame (FF) was 53.2% in U-STF, thus lower than those of cooked skipjack tuna frame (C-STF), the uncooked salmon frame (U-SF), and beef leg bone (BLB). The composition of FF had 48.6-58.0% moisture, 18.4-21.3% crude protein, 7.0-15.0% crude lipid, and 12.8-22.1% ash. Compared to the proximate composition of BLB, that of FF was higher moisture and crude protein but lower solids, crude lipids, and ash. The total amino acid contents of FF was 17.6-20.8 g/100 g, which was higher than that of BLB. The levels of calcium and phosphorus of FF were 4.7-8.5 g/100 g and 2.4-4.2 g/100 g, respectively, thus lower than those of BLB. The brix concentration of FF extract was $5.0-8.2^{\circ}$, being highest in U-STF extracts, followed by extracts of U-SF and then extracts of C-STF. Our results indicate that U-STF is an optimal resource for preparing flavor-enriching concentrates.
