IMMUNE RESPONSES OF THE MICE BEARING TUMOR INDUCED BY DMBA(9,10-Dimethyl-1,2-Benzanthracene)

  • This study was undertaken to evaluate the immune responses to sheep red blood cell (SRBC) and potential anti-tumor effect of Bacillus Calmette-Guerin (BCG) in the mice bearing rumor induced by DMBA. The frequencies of tumor appearances were 62% in DMBA-treated mice and 14% in DMBA and BCG-treated group, respectively. Cellular immune response such as delayed-type hypersensitivity (DTH) to SRBCs, natural killer (NK) cell activity and antigen-binding cell (ABC) assay were decreased apparently in the tumor bearing mice compared to the normal controls. Humoral immune responses such as hemagglutinin (HA) and hemolysin (HE) were noted to be reduced in the tumor bearing mice, but the spleen index increased in tumor bearing mice. All the immunological parameters in the DMBA and BCG-group appeared to be higher than those of only DMBA-treated group. These results indicated that DMBA-induced tumor suppressed host immune responses. Also, they imply the idea that BCG enhanced the immune responses of tumor-bearing host and antitumor effects.

A Case of Endobronchial Carcinoid Tumor Treated by Flexible Bronchoscopic Resection (기관지내시경 절제술로 치료한 기관지 카르시노이드 종양 1예)

  • Bronchial carcinoid tumor accounts for less than 5% of all primary lung tumors in adults. Although surgical resection is the treatment of choice, here we report a case of bronchial carcinoid tumor treated with flexible bronchoscopic resection. A 19-year-old-man presented with a history of wheezing with dyspnea for six months. A simple chest x-ray showed no abnormal findings, but a pulmonary function test showed a moderate obstructive lung disease pattern without a bronchodilator response. A computed tomogram of the thorax revealed an enhanced $15{\times}12$ mm nodule in the left main bronchus. Bronchoscopic examination showed a polypoid mass with a stalk in the left main bronchus, which almost completely occluded the left main bronchus. Histopathology of the resected specimen revealed a bronchial carcinoid tumor. We treated the carcinoid tumor with a flexible bronchoscopic resection. During the follow up period of 6 months, the previous tumor didn't relapse. Initial bronchoscopic resection should be considered when bronchial carcinoid tumor can be approached by bronchoscopy.

  • Many cancers arise from sites of chronic inflammation, which creates an inflammatory microenvironment surrounding the tumor. Inflammatory substances secreted by cells in the inflammatory environment can induce the proliferation and survival of cancer cells, thereby promoting cancer metastasis and angiogenesis. Therefore, it is important to identify the role of inflammatory factors in cancer progression. This review summarizes the signaling pathways and roles of C-reactive protein (CRP) in various cancer types, including breast, liver, renal, and pancreatic cancer, and the tumor microenvironment. Mounting evidence suggests the role of CRP in breast cancer, particularly in triple-negative breast cancer (TNBC), which is typically associated with a worse prognosis. Increased CRP in the inflammatory environment contributes to enhanced invasiveness and tumor formation in TNBC cells. CRP promotes endothelial cell formation and angiogenesis and contributes to the initiation and progression of atherosclerosis. In pancreatic and kidney cancers, CRP contributes to tumor progression. In liver cancer, CRP regulates inflammatory responses and lipid metabolism. CRP modulates the activity of various signaling molecules in macrophages and monocytes present in the tumor microenvironment, contributing to tumor development, the immune response, and inflammation. In the present review, we overviewed the role of CRP signaling pathways and the association between inflammation and cancer in various types of cancer. Identifying the interactions between CRP signaling pathways and other inflammatory mediators in cancer progression is crucial for understanding the complex relationship between inflammation and cancer.

Prognostic Significance of 18F-fluorodeoxyglucose Positron Emission Tomography (PET)-based Parameters in Neoadjuvant Chemoradiation Treatment of Esophageal Carcinoma

  • Aims and Background: The purpose of the research was to study the prognostic value of tumor 18F-FDG PET-based parameters in neoadjuvant chemoradiation for patients with squamous esophageal carcinoma. Methods: Sixty patients received chemoradiation therapy followed by esophagectomy and two 18FDG-PET examinations at pre- and post-radiation therapy. PET-based metabolic-response parameters were calculated based on histopathologic response. Linear regression correlation and Cox proportional hazards models were used to determine prognostic value of all PET-based parameters with reference to overall survival. Results: Sensitivity (88.2%) and specificity (86.5%) of a percentage decrease of SUVmax were better than other PET-based parameters for prediction of histopathologic response. Only percentage decrease of SUVmax and tumor length correlated with overall survival time (linear regression coefficient ${\beta}$: 0.704 and 0.684, P<0.05). The Cox proportional hazards model indicated higher hazard ratio (HR=0.897, P=0.002) with decrease of SUVmax compared with decrease of tumor size (HR=0.813, P=0.009). Conclusion: Decrease of SUVmax and tumor size are significant prognostic factors in chemoradiation of esophageal carcinoma.

십전대보탕(十全大補湯) 와송(瓦松) 및 십전대보탕가와송(十全大補湯加瓦松)의 항암효과(抗癌效果)와 면역반응(免疫反應)에 관(關)한 연구(硏究)

  • In order to research the effects of Shipjundaebotang(SDT) and Orostachys Herba(OH) on anti-tumor and immune response, the author performed this experimental study. Experimental groups are divided into five groups, which are solid extract of Orostachys Herba by water(OHW), solid extract of Orostachys Herba by ethanol(OHE), solid extract of Shipjundaebotang (SDT), solid extract of Shipjundaebotang added by solid extract of Orostachys Herba by water(SDT+OHW) and solid extract of Shipjundaebotang added by solid extract of Orostachys Herba by ethanol (SDT+OHE). In these experimental studies, extension of survival days for anti-tumor effect was observed, and de layed type hypersensitivity and rosette forming cell for cell-mediated immune response, hemagglutinin titers and hemolysin titers for humeral immune response, spleenic natural killer cell activity and carbon clearance (K-index) in vitro were measured with mice. The result were summerized as follows: I. SDT, SDT+OHW and SDT+OHE treated groups were significantly recognized to extend the survival days of tumor bearing mice as compared with the control group. 2. Delayed type hypersensitivity was significantly increased in SDT, SDT+OHW and SDT+OHE treated groups as compared with control group. 3. Hemagglutinin titer was increasred in all sample groups as compared with control group, but not significantly. 4. Hemolysin titers was significantly increased in SDT, SDT+OHW and SDT+OHE treated groups as compared with control group, and SDT+OHE treated group showed the increasing effect with significance as compared with the other sample groups. 5. For the effect of roselle forming cell quantitation, SDT, SDT+OHW and SDT+OHE treated groups showed the increasing effect with significance as compared with control group. 6. Natural killer cell activity was significantly increased in SDT+OHW as compared with comrol group, but the other groups, except OHW and SDT+OHW treated groups, revealed the increasing effect as compared with control group, but the significance was not admitted. 7. For the effect of K-index(Carbon clearance), SDT, SDT+OHW and SDT+OHE treated groups showed the increas ing effect with significance as compared with control group. 8. The study didn't show that Orostachys herba had any significance with survival days, anti-tumor effect and immune re sponse.

A study of the tendency of anti-cancer experimental study using herbal medicines (한약(韓藥)을 이용(利用)한 항암(抗癌) 실험연구(實驗硏究)의 경향(傾向)에 관(關)한 고찰(考察))

  • Because there were lots of side effects and tolerances to the existing anticancer therapeutics, the experiment extracting the anticancer effect from medicinal herbs is in progress liviely. Therefore the purpose of this study were to research the tendency and the course of anticancer studies. To research the tendency of anticancer studies, medicinal herbs of fifty three experimental papers were analyzed and to examine the course of studies, anticancer papers in the medical world were used. The obtained results were as follows: Methods of herbal medicinal treatments were elimination the pathogenic factor(祛邪) and supporting healthy energy(扶正) method used. In this study, immediately tumor bearing and immune response were the most important point. The subject of immediately tumor bearing was not in the specific cancer but in the influence on the life span of general cencerous cells. In the experimental study of immune response, the effect on NK cell activity of medicinal herbs most studied. The combined usage of medicinal herbs and anticancer agent mostly intended to know whether it inhibits the tumor cell growth. The serum test and blood cell number test show if medicinal herbs inhibit side effect of anticancer agent. More than 80 percents of used medicinal herbs, there were anticancer activities. However anticancer experimental studies using medicinal herbs two weak points. The one, it was difficult to choose a prescription according to differentiation of symptoms and signs(辨證論) of the Oriental Medicine, because we put to the test not a man but a mouse. The other, as we observed the indirect effect of the whole physiological regulation caused by synergic effects of the complex prescription, we don't understand the detailed mechanism of the herb. Therefore, if the anticancer effect of the herb is proved the experiment, we should research the concrete medical action of medicinal herbs and immunological analysis of herbal medicines to the body.

Expression of HYOU1 via Reciprocal Crosstalk between NSCLC Cells and HUVECs Control Cancer Progression and Chemoresistance in Tumor Spheroids

  • Among all cancer types, lung cancer ranks highest worldwide in terms of both incidence and mortality. The crosstalk between lung cancer cells and their tumor microenvironment (TME) has begun to emerge as the "Achilles heel" of the disease and thus constitutes an attractive target for anticancer therapy. We previously revealed that crosstalk between lung cancer cells and endothelial cells (ECs) induces chemoresistance in multicellular tumor spheroids (MCTSs). In this study, we demonstrated that factors secreted in response to crosstalk between ECs and lung cancer cells play pivotal roles in the development of chemoresistance in lung cancer spheroids. We subsequently determined that the expression of hypoxia up-regulated protein 1 (HYOU1) in lung cancer spheroids was increased by factors secreted in response to crosstalk between ECs and lung cancer cells. Direct interaction between lung cancer cells and ECs also caused an elevation in the expression of HYOU1 in MCTSs. Inhibition of HYOU1 expression not only suppressed stemness and malignancy, but also facilitated apoptosis and chemosensitivity in lung cancer MCTSs. Inhibition of HYOU1 expression also significantly increased the expression of interferon signaling components in lung cancer cells. Moreover, the activation of the PI3K/AKT/mTOR pathway was involved in the HYOU1-induced aggression of lung cancer cells. Taken together, our results identify HYOU1, which is induced in response to crosstalk between ECs and lung cancer cells within the TME, as a potential therapeutic target for combating the aggressive behavior of cancer cells.

Association between Chemotherapy-Response Assays and Subsets of Tumor-Infiltrating Lymphocytes in Gastric Cancer: A Pilot Study

  • Purpose: The purpose of this pilot study was to evaluate the association between adenosine triphosphate-based chemotherapy response assays (ATP-CRAs) and subsets of tumor infiltrating lymphocytes (TILs) in gastric cancer. Materials and Methods: In total, 15 gastric cancer tissue samples were obtained from gastrectomies performed between February 2007 and January 2011. Chemotherapy response assays were performed on tumor cells from these samples using 11 chemotherapeutic agents, including etoposide, doxorubicin, epirubicin, mitomycin, 5-fluorouracil (5-FU), oxaliplatin, irinotecan, docetaxel, paclitaxel, methotrexate, and cisplatin. TILs in the tissue samples were evaluated using antibodies specific for CD3, CD4, CD8, Foxp3, and Granzyme B. Results: The highest cancer cell death rates were induced by etoposide (44.8%), 5-FU (43.1%), and mitomycin (39.9%). Samples from 10 patients who were treated with 5-FU were divided into 5-FU-sensitive and -insensitive groups according to median cell death rate. No difference was observed in survival between the two groups (P=0.216). Only two patients were treated with a chemotherapeutic agent determined by an ATP-CRA and there was no significant difference in overall survival compared with that of patients treated with their physician's choice of chemotherapeutic agent (P=0.105). However, a high number of CD3 TILs was a favorable prognostic factor (P=0.008). Pearson's correlation analyses showed no association between cancer cell death rates in response to chemotherapeutic agents and subsets of TILs. Conclusions: Cancer cell death rates in response to specific chemotherapeutic agents were not significantly associated with the distribution of TIL subsets.

In Vivo Enhanced Indocyanine Green-Photothermal Therapy for a Subconjunctival Tumor

  • Indocyanine green (ICG) is a dye approved for use in clinical diagnostics. ICG remains in the intravascular space following intravenous administration, due to its ability to rapidly bind to the plasma proteins, and its therapeutic potential has been studied in well-vascularized cutaneous tumors. Here we have evaluated the clinical response of a subconjunctival tumor to photothermal therapy (PTT) using an ICG-enhanced near-infrared diode laser and its adverse effects, in a rabbit. 22 male New Zealand white rabbits with subconjunctival tumors were enrolled (control group 6, laser-only group 8, laser-with-ICG group 8). Rabbits in the laser-with-ICG group received ICG (twice, 2 mg/kg each time, intravenously) directly followed by irradiation with a diode laser (λ = 810 nm). Rabbits in the laser-only group were irradiated with the diode laser. ICG angiography, ultrasonography, and pathologic examination were performed to evaluate PTT response at specific time points (0, 2, and 4 weeks after PTT). Two weeks after initial treatment, the eight rabbits treated by laser with ICG showed a 100% response rate. There was no clinical response in both laser-only and control groups. ICG-PTT is a potential and effective palliative therapeutic modality for subconjunctival tumors.
