영지와 표고의 융합체의 면역활성 증강작용 (Immunostimulating Activity by Protoplast Fusants between Ganoderma Iucidum and Lentinus edodes)

  • On the inter-order protoplast fusants of Lentinus edodes and Ganoderma lucidum was the antitumor activity test performed and the fusant P22 was selected. The hot water extract of the cultured mycelia of P22 were purified by DEAE-cellulose chromatographya and the resulting purified fraction was designated as P22A. It was found to be a proteoglycan whose molecular weight was 47 kDa. When examined for immunopotentiation activity, P22A increased the number of colony forming unit in the bone marrow stem cells to 3-folds. It also potentiated the secretion of nitric oxide in activated macrophages to 2-folds. In humoral immune response, it increased the activities of the alkaline phosphatase in differentiated B cells to 1.6-folds and the number of plaque forming cells to 1.8-folds. In cellular immune response, it restored the depressed response of delayed type hypersensitivity in tumor bearing mice to normal level. These results suggest that P22A have potential to restore the decreased immune activity of the tumor bearing mice to normal level.

인진쑥 methanol 추출물의 투여가 암이 유발된 마우스에서 보인 혈액생화학적 및 종양 무게에 미치는 영향 (Effects of Blood Biochemistry and Tumors' Weights of Artemisia capillaris Methanol Extract in Mice Bearing Cancer Cells)

  • The Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and probably the most common plant among the various herbal folk remedies being used in the treatment of abdominal pain hepatitis chronic liver disease, jaundice and coughing in Korea. Recently the biological and pharmacological actions of herb have been studied well such as antibacterial, antidiabetic and antitumor activities. This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillarix extracts against Hepa-1c1c7 and Sarcoma 180 cancer cells on in vivo experimental tests. On in vivo experimental tests using 280 ICR mice the gain of body weight in the control-group mice bearing Sarcoma 180 ascites tumor was 1.5 times more than that of the normal-group mice after 33 days. However, the gain of body weight in all experimental groups administered with Artemisia capillaris extracts was significantly lower than that of the control-group mice (P<0.05). The mean survival times of mice administered with Artemisia capillaris extracts of 25 and 100 mg/kg for 28 days were shown to be 25.39% and 15.39% longer than that of the control-group mice injected with saline (P<0.05). Artemisia capillaris extracts showed the highest tumor inhibition effects, which were 42.4% and 27.2% when intraperitoneally injected with doses of 25 and 100 mg/kg once a day for 28 days in inoculated ICR mice with Sarcoma 180 solid tumor cells (P<0.05). The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.

Antitumor Responses of Adoptively-Transferred Tumor-Specific T-Cell Cultures in a Murine Lymphoma Model

  • The purpose of this study was to establish an in vitro culture method of tumor-specific T cells, and determine the efficacy of the cultured tumor-specific cytotoxic T-lymphocytes (CTL) as an agent of anti-tumor immunotherapy against a murine lymphoma, TIMI.4. Tumor-specific T-lymphocytes derived from C57BL/6 mice (thy-1.2) immune to TIMI.4 were activated by in vitro stimulation with the irradiated TIMI.4 cells, and expanded by restimulation with TIMI.4 in the presence of the concanavalin A-stimulated rat spleen culture supernatant, and splenic antigen-presenting cells. In vitro restimulation enhanced markedly the proportion of $CD8^+$, a predominant surface marker of CTL and the cytotoxic activity in the cultured immune T cell population. The resulting TIMI.4-specific T cells were adoptively transferred into nude mice. The tumor cells residing in the host after 7 days of adoptive transfer to B6.PL (thy-1.1) mice were quantified by use of an antibody directed to the thy-1.2 allele. The TIMI.4 cells in the recipient nude mice were decreased in a dose-dependent manner. Anti-tumor activity of the TIMI.4-specific T cells was also demonstrated by a survival test, where the tumor-bearing nu/nu mice which received the activated T-cells survived about 30% longer than the control mice which received the tumor cells alone. These suggest that adoptive transfer of TIMI.4-specific T cells could be a candidate for effective therapy of the murine lymphoma.

Protective Antitumor Activity through Dendritic Cell Immunization is Mediated by NK Cell as Well as CTL Activation

  • Dendritic cells (DCs) are potent professional antigen-presenting cells (APC) capable of inducing the primary T cell response to antigen. Although tumor cells express target antigens, they are incapable of stimulating a tumor-specific immune response due to a defect in the costimulatory signal that is required for optimal activation of T cells. In this work, we describe a new approach using tumor-DC coculture to improve the antigen presenting capacity of tumor cells which does not require a source of tumor-associated antigen. Immunization of a weakly immunogenic and progressive tumor cocultured with none marrow-derived DCs generated an effective tumor vaccine. Immunization with the cocutured DCs was able to induce complete protectiv immunity against tumor challenges and was effective for the induction of tumor-specific CTL (cytotoxic T lymphocyte) activity. Furthermore, high NK cell activity was observed in mice in which tumors were rejected. In addition, immunization with tumor-pulsed DC s induced delayed tumor growth, but not tumor eradication in tumor-bearing mice. Our results demonstrate that coculture of DCs with tumors generated antitumor immunity due to the NK cell activation as well as tumor-specific T cell. This approach would be used for designing tumor vaccines using DCs when the information about tumor antigens is limited.

Transferrin-Conjugated Liposome/IL-12 pDNA Complexes for Cancer Gene Therapy in Mice

  • Transferrin ($T_{f}$) has been used as a targeting ligand for delivering liposome/interleukin-12 (IL-12) pDNA complexes to cancer cells mostly due to the greater number of transferrin receptors ($T_{f}R$) found on tumor cells than on normal cells. $T_{f}$ was conjugated to liposomes via the reaction of MPB-PE with thiol groups of $T_{f}$ introduced by a heterobifunctional cross-linking agent, N-succinimidyl-3-(2-pyridyldithio)propionate (SPDP). Four days after C26 inoculation when the tumor volume reached ${\sim}100mm^{3}$, tumor-bearing Balb/c mice were injected intravenously with $T_{f}-liposome/IL-12 pDNA$complexes twice a week for 3 weeks. Significant suppression of tumor growth was achieved in the group treated with the $T_{f}-liposome/IL-12 pDNA$ complexes, with a dose of $10{\mu}g$ of IL-12 pDNA showing the highest suppression effect among the tested doses. Similar results were obtained when the therapy was initiated one day after tumor inoculation, although in this case $30{\mu}g$ IL-12 pDNA/$T_{f}-liposome$ complexes showed a significant suppression of tumor growth between 19 and 23 days after tumor inoculation. This result indicates that the transferrin receptor-targeted liposomal system is an efficient delivery agent of therapeutic genes, such as IL-12, in mice and that its potential clinical use warrants further research investigation.

Intratumoral Administration of Rhenium-188-Labeled Pullulan Acetate Nanoparticles (PAN) in Mice Bearing CT-26 Cancer Cells for Suppression of Tumor Growth

  • The feasibility of pullulan acetate nanoparticles (PAN) with ionic strength (IS) sensitivity as a radioisotope carrier to inhibit tumor growth is demonstrated. PAN was radiolabeled with rhenium 188 (Re-188) without any chelating agents. The labeling efficiency of Re-188 into PAN (Re-188PAN) was $49.3{\pm}4.0%$ as determined by TLC. The tumor volumes of mice treated with 0.45 mCi of Re-188-PAN were measured and compared with that of free Re-188 after 5 days of intratumoral injection. For the histological evaluation of apoptotic nuclei of tumor cells, hematoxylin and eosin (H&E), and terminal deoxynucleotidyl transferase biotinylated deoxyuridine triphosphate nick end labeling (TUNEL) staining were performed. The mean tumor volume of the Re-188-PAN-treated group was decreased by 36% after 5 days, whereas that the free Re-188-treated group was decreased by only 15% (P<0.05). The mean number of TUNEL-positive cells in Re-188-PAN-treated tumors at $144.3{\pm}79.9$ cells/section was significantly greater than the control ($26.7{\pm}7.9$ cells/section, P=0.03). The numbers of leukocyte and lymphocyte were decreased in both free Re-188- and Re-188-PAN-treated mice. These results indicated that the intratumoral injection of Re-188-PAN effectively inhibits the tumor growth by prolonging Re-188 retention time in tumor site induced by the IS sensitivity.

Immunomodulatory effect of captopril and local irradiation on myeloid-derived suppressor cells

  • Purpose: This study is to investigate the effect of captopril when combined with irradiation. Materials and Methods: 4T1 (mouse mammary carcinoma) cells were injected in the right hind leg of Balb/c mice. Mice were randomized to four groups; control (group 1), captopril-treated (group 2), irradiated (group 3), irradiated and captopril-treated concurrently (group 4). Captopril was administered by intraperitoneal injection (10 mg/kg) daily and irradiation was delivered on the tumor-bearing leg for 15 Gy in 3 fractions. Surface markers of splenic neutrophils (G-MDSCs) and intratumoral neutrophils (tumor-associated neutrophils [TANs]) were assessed using flow cytometry and expression of vascular endothelial growth factor (VEGF) and hypoxia-inducible factor 1 alpha ($HIF-1{\alpha}$) of tumor was evaluated by immunohistochemical (IHC) staining. Results: The mean tumor volumes (${\pm}$standard error) at the 15th day after randomization were $1,382.0({\pm}201.2)mm^3$ (group 1), $559.9({\pm}67.8)mm^3$ (group 3), and $370.5({\pm}48.1)mm^3$ (group 4), respectively. For G-MDSCs, irradiation reversed decreased expression of CD101 from tumor-bearing mice, and additional increase of CD101 expression was induced by captopril administration. Similar tendency was observed in TANs. The expression of tumor-necrosis factor-associated molecules, CD120 and CD137, are increased by irradiation in both G-MDSCs and TANs. Further increment was observed by captopril except CD120 in TANs. For IHC staining, VEGF and $HIF-1{\alpha}$ positivity in tumor cells were decreased when treated with captopril. Conclusion: Captopril is suggested to have additional effect when combined to irradiation in a murine tumor model by modulation of MDSCs and angiogenesis.

생약복합제제 길경탕 및 가미길경탕의 항암효과 (제 1 보) (Antitumor Activity of the Medicinal Formula Kilkyungtang and Two Modified Kilkyungtangs in Vivo. I)

  • The prescription, Kilkyungtang (KKT), which originally consists of twelve kinds of medicinal plant materials and was used as a decoction for the treatment of malignant tumors and two modified Kilkyungtangs (KKT-1 and KKT-2), supplemented by the additional crude drug to KKT (KKT-1:Houttuyniae herba, and KKT-2:Oldenlandiae diffusae herba) were investigated on their antitumoral properties, in vivo respectively. All KKTs were found to exhibit significant life time-prolonging effects when they were administered orally to Sarcoma-180 bearing ICR mice for 7 days. (ILS was estimated as 20% in KKT, 42% in KKT-1 and 57% in KKT-2). A profound lessening of tumor weights was also observed when KKTs were administered to $B16-F_0$ bearing C57B/6 mice.

영지버섯 생장점 단백다당체 GLB-A, GLB-B의 항암효과 및 면역 활성에 관한 유세포 분석학적 연구 (Flow Cytometrical Analysis of the Antitumor and Immunomodulatory Activities of GLB-A and GLB-B, the Protein-polysaccharide Fractions of the Growing Tips of Ganoderma Lucidum)

  • In the previous study we described the antitumor effect of GLB, a protein-polysaccharide fraction separated from the growing tips of Ganoderma lucidum, against sarcoma 18 0 solid tumor in ICR mice. In this study, we separated an acidic protein-polysaccharide fraction, GLB-A, and a basic protein-polyaccharide fraction, GLB-B, from GLB by differential precipitation, and elucidated their antitumor and immunomodulatory activities. When ip injected at the dose of 50mg/kg/day into the ICR mice, GLB-A and GLB-B inhibited the growth of ip implantated sarcoma 180 cells by 32.4% and 21.0%, respectively. Of these, GLB-A increased the % lymphoblast in the spleen of the tumor-bearing and the normal mice by 20.9% and 123.0%, and the CD4/CD8 ratio by 73.3% and 22.4%, respectively. GLB-A also increased the expression of CD25 (IL-2 receptor alpha ch0ain) in normal mice by 82.0%. These results strongly suggest that GLB-A is a promising candidate for antitumor immunomodulatory medicine.

키토산 유도체인 Sulfated N-acetyl Chitosan의 종양전이 억제효과 (Inhibitory Effects of Tumor Metastasis by Chitosan Derivative, of Sulfated N-acetyl Chitosan)

  • 키토산 유도체 인 sulfated N→acetyl chitosan을 합성하여 이 화합물을 쥐에 이식한 B16/BL6 mela noma에 대하여 폐암전이의 억제효과를 조사하였다. 키토산의 황산유도체는 13C-n.m.r.에 의하여 확인한 결과 3, 6, O-disulfate 엄을 알 수 있었다. SuI fated N-acetyl chitosan을 100mg/kg을 투여하였을 때 B16/BL6의 melanoma cells을 가장 효과적 으로 억제하였고 종양 무게의 증식도 대조군에 비해 억제되었다. 폐암의 전이에 있어서 B16/BL6에 sulfated N-acetyl chitosan을 LV. V로 주사하였을 때 B16/BL6의 melanoma의 전이 세포의 수는 자발적인 전이에 있어서도 용량 의존형(20 ~ 1OOmg/kg) 에 따라 줄어들었다. B16/BL6을 접종한 후 sulfated N-acetyl chitosan을 I.V.로 투여했을 때 전이세 포의 수는 현저히 감소하였다. Sulfated N-acetyl chitosan은 laminin의 종양세포에의 세포접착 능력을 부분적으로 억제하였다. 이와 같은 결과는 chitosan의 유도체인 sulfated N-acetyl chitosan이 세포접착 능력이 실험적으로 자발적인 암전이 모델에 효과적으로 작용하였다.
