Electron Microscopic Study on the Role of Actin Filaments during the Formation of Bile Canaliculi in Isolated Rat Hepatocyte Culture System (흰쥐에서 분리 배양한 간세포의 담세관 형성에 있어서 액틴미세섬유의 역할에 관한 전자현미경적 연구)

  • Bile canaliculi are the structure delivering bile secreted by hepatocytes into the bile passage. Bile secretion is mainly controlled by the cytoskeletal elements, mainly of actin in the microvilli, pericanalicular web. Most studies on the bile secretion have been done in viva situation, however, to control the various parameters in vitro culture system seem to be more useful. To set up an in vitro experimental system, the investigator isolated hepatocytes with an enzymatic method using a mixture of collagenase and hyaluronidase from normal Sprague-Dawley rat liver and cultured. Isolated hepatocytes were round and formed cords in culture. Microvilli covered the whole surface of hepatocytes. Bile canaliculi were formed between hepatocytes and were characterized by the presence of microvilli of various lengths and shapes mainly arising from small surface mounds. Actin filament core in the microvilli and pericanalicular actin web were incomplete. After cytochalasin D treatment, cultured hepatocytes were round but the surface were irregular with surfacen blebs, folds and grooves. Microvilli on the surface were scarce. Bile canaliculi were markedly dilated often with the detached junctional complexes. Bile canaliculi lacks microvilli almost completely and extended into the pericanalirular cytoplasm showing complex vacuolar and tubular structures by transmission electron mciroscopy. Pericanalicular actin web, intermediate filaments were hardly identified. Subsurface actin filaments were scattered scarcely under the cell membranes. These results suggest that hepatocytes isolated from rats can survive and form bile canaliculi in culture and the actin filaments are involved in the formation and/or maintenance of the bile canaliculi.

Histological Examination of Tissue Isolated from Fascia with a View of Meridian System (경락의 관점에서 본 근막 분리조직의 조직학적 연구)

  • The threadlike structures of fascia were examined by tight and electron microscopy. In order to distinguish its tissue organization, we used staining methods including hematoxylin-eosin, Masson's trichrome, Van Gieson's collagen fiber stain and Kluver-Barrera's luxol fast blue for nerve stain. Under the light microscope, the threadlike structures were composed of many collagen fibers and nerve. In higher magnification, they looked like as the bundle of tubular structures. Many myoid cell-spindle nuclei were observed in the tissue, which were taken from the fascia. It was identical with Bonghan duct known as one of meridian network theory. In the early 1960's the North Korean Bong-Han Kim showed the anatomical structures of the acupuncture points, and explained the meridian system as the concrete duct network system. According to Bonghan theory the Bonghan ducts spread throughout the body Because it is believed that the duct could have the role of signal pathway, the theory was reinvestigated in these days. All of the threadlike structures isolated from fascia shows the abundance of collagen fibers. The electron microscope examination (TEM) could confirm the well arranged collagen fiber and nerve. This investigation reveals that superficial Bohghan duct are nerve fiber parallel running with collagen fibers. We conjectured that the intermingled structure of collagen fiber, blood vessel and nerve fiber might have the role of meridian system. And the more, regardless of histological research, the study on collagen fiber as response transmitter in acupuncture treatment are in need.

  • Sirtuin proteins have emerged as important modulators of several age-associated diseases. These include cancer and diabetes, as well as cardiovascular and neurodegenerative diseases. Among the sirtuin family members, SIRT2 mRNA is strongly expressed. To investigate the pathophysiological significance of SIRT2 as a primary regulator of angiogenesis, we focused on the biological role of SIRT2 under hypoxic conditions, examining the gene expression pattern of sirtuin family members in human umbilical vein endothelial cells (HUVECs). SIRT2 was expressed primarily in the cytoplasm, but it was dynamically trans-localized in the nuclear by hypoxia stimuli. Interestingly, both SIRT2 and the pro-angiogenic factor, VEGF, were up- regulated by hypoxia. A Matrigel assay demonstrated that the HUVECs formed a tube-like structure under hypoxia. The SIRT2 inhibitor, AK-1, significantly decreased the tube-forming activity of the HUVECs under either normoxia or hypoxia conditions. These findings suggest that SIRT2 might be a key regulator of angiogenesis.

  • In this study, pervaporation experiments of water, ethanol and IPA (Isopropyl alcohol) single components and water/ethanol, water/IPA mixtures were carried out using zeolite 4A membranes developed by Fine Tech Co. Ltd. Those membranes were fabricated by hydrothermal synthesis (growth in hydrothermal condition) after uniformly dispersing the zeolite seeds on the tubular alumina supports. They have a pore size of about $4{\AA}$ by ion exchange of $Na^+$ to the LTA structure with Si/Al ratio of 1.0, and shows strong hydrophilic property. Physical characteristics of prepared membranes were evaluated by using SEM (surface morphology), porosimetry (macro- or meso- pore analysis), BET (micropore analysis), and load tester (compressive strength). Pervaporation experiments with various temperature and concentration conditions confirmed that the zeolite 4A membrane can selectively separate water from ethanol and IPA. Water/ethanol separation factor was over 3,000 and water/IPA separation factor was over 1,500 (50 : 50 wt%, initial feed concentration). Pervaporation behaviors of single components and binary mixtures were predicted using ACM (activity coefficient model), GMS (generalized Maxwell Stefan) model and DGM (Dusty Gas Model). The adsorption and diffusion coefficients of the zeolite top layer were obtained by parameter estimation using GA (Genetic Algorithm, stochastic optimization method). All the calculations were carried out using MATLAB 2018a version.

Microscopic Characteristics of the Olfactory Organ in the Gluttonous Goby Chaenogobius gulosus(Pisces, Gobiidae), Compared to Sympatric Intertidal Gobies (별망둑 the gluttonous goby Chaenogobius gulosus 후각기관의 해부, 조직학적 특성 및 동소 망둑어과 출현종들과의 비교연구)

  • Using stereo, light, and scanning electron microscopes, we researched the anatomical and histological structure of Chaenogobius gulosus's olfactory organ and compared it to those of sympatric gobies Luciogobius guttatus and Favonigobius gymnauchen. Results revealed the following common characteristics: i) tubular anterior nostril (AN) and flat posterior nostril (PN), ii) a single longitudinal lamella, iii) two accessory nasal sacs (ANS, ethmoidal and lacrimal), iv) abundant sensory epithelium lymphatic cells (LC), v) an eosinophil cell, and vi) a ciliary length a quarter of the knob diameter in the olfactory receptor neuron (ORN). Some characteristics are specific to C. gulosus and different from the other two gobies: i) 0.5~1.0 mm AN and 0.2~0.5 mm PN (vs. 0.2~0.3 mm and 0.2~0.3 mm in L. guttatus; 0.2~0.4 mm and 0.1~0.3 mm in F. gymnauchen), ii) two ANS (vs. absence in L. guttatus; two in F. gymnauchen), iii) abundant LC (vs. low in L. guttatus and F. gymnauchen), iv) low density non-sensory cilia on the lamellar surface (vs. high in L. guttatus; low in F. gymnauchen), and v) a quarter ciliary length to knob diameter ratio in the ORN (vs. mixture of a quarter to equal ratio in L. guttatus; two or three times in F. gymnauchen). From these results, we confirmed the C. gulosus olfactory organ has adapted anatomically and histologically to the sand-rock tidal zone.

  • Fine structures of retina of an ommatidium in dragonfly, having eyes of closed rhabdom type, were studied under light and electron microscopes. The ommatidia consisted of eight retinular cells distributed in a circular pattern and the retinular layer in turn can be divided into three sublayers according to the number of cells in the retina. Each retinular cell has different starting points in the retina and the length of retinular cells is varied greatly; the length of one distal retinular cell shows one half of that of others. In the middle layer, three proximal retinular cells interconnect the adjacent two rhabdoms which are triangular in the appearence of the cross section which in turn consisted of tubular, parallel and lamellated microvilli. The rhabdom is formed by three rhabdomeres, each of which is separated by $120^{\circ}$ between them, but they can be distinguished into two parts according to electron density. Around the outer part of microvilli composing rhabdom, electron density was much less than the inner part of the structure. The microvilli of the inner part appear to be connected to the cytoplasm of retinular cells. Rough endoplsmic reticulum with enlarged cisternae runs through the vacuoles in the outer part of distal retinular cells. Abundant mitochondria concentrated in the vicinity of rhabdom are found at the central part of the retinular cells, while in the area of immediate vicinity of the rhabdom, prominent vacuoles are observed. Above the rhabdom of an ommtidium stands a crystalline cone which is consisted of four cone cells arranged radially along the axis. The crystalline cone is surrounded by cells containing pigment granules. The outermost photoreceptor element of an ommatidium is corneal lens.

In situ posture of anterior body of Metagonimus yokogawai in experimentally infected dog (개의 실험적 요꼬가와흡충증에서 충체의 자세)

  • The in situ posture of anterior body of Metagonimus yokogawai was observed in experimental metagonimiasis of dog. The metacercariae were collected from naturally infected sweetfish by peptic digestion; a total of 8 dogs was orally infected with 10,000 metacercariae respectively. Two dogs were killed on 3 days, 9 days, 4 weeks and 10 weeks after the infection. The postures of worms in histological section of small intestine and of whole worms collected from the fixed intestinal mucosa were examined by light and scanning electron microscope. The results were summarized as follows: 1. The recovery rates of worms were 42.6% on 3 days, 55.0% on 9 days, 33.2% on 4 weeks and 9. 8% on 10 weeks after the infection respectively. 2. In histological sections of small intestine, most of worms were found at intervillous spaces as ovoid sections of posterior body. However, many worms, especially in 3 day-old worms, revealed protruded anterior body in glandular lumens of crypt. Some sections of anterior body were bifurcated or sacculated. 3. The worms collected from fixed intestinal mucosa under dissecting microscope exhibited a variety of postures. Many worms showed flat shapes with a concavity or curvatures. However, in many worms, the anterior body made a single or multiple protrusions. 4. By SEM observation of protruded anterior body, a longitudinal groove was found. 5. The frequency of worms with protruded anterior body decreased in 9 days, 4 weeks and 10 weeks than in 3 day-old worms. The above findings indicated that the anterior body of juvenile and adult M. yokogawai protruded to lumens of glandular crypt by folding their lateral portions to make a reversible tubelike structure. Frequent multiple protrusions were considered to be made to adapt the microniche of glandular crypts of dog intestine.

Characteristics of New Microsporidia S80 Isolated from Silkworm, Bombyx mori L. in Korea (가잠(家蠶)으로부터 분리(分離)된 새로운 Microsporidia S80의 특성(特性))

  • The new microsporidia S80 isolated from, Bombyx mori L. in Korea showed ovoid in the morphology of the spores and the size were measured $2.9{\pm}0.28{\mu}$ in length and $1.7{\pm}0.29{\mu}$ width. No other microsporidian spore like this has not been so far isolated from Silkworm. The length of the polar filament extruded in hydrogen peroxide ($H_2O_2$) at $30^{\circ}C$ was $26{\mu}$ of a round cytoplasm on the top. The spores were partly stained with Giemsa, Safranin-O and Gram as the same staining properties as Nosema bombycis, Microsporidia K 79 and other microsporidian spores. The fine structures were observed under scanning eleceron microscope through ultrathin sectioning. The spore wall was composed of three layers ; the thin exospore of an electron dense rippled layer, the thick electron lucent endospore which was thinning considerably at the polar filament insertion point, and the inner limiting membrane. Polar cap present at the sporeapex, with a long polar filament of 12-13 coils, subtending angle of $60^{\circ}$ to spore axis, which is tubular made up of a multilayered and are a benes core, light ring structure enclosing the dance core, the dark ring structure enclosing the inner light ring structure and the other than and light ring structure bounded from cytoplasm. Lamellate polaroplast occupied the anterior part of the spore, and the two neclei with dense nucleoplasm bounded by a double nuclear envelope were cited in the slight downer middle portion of spore. From the characteristics of the shape, size and fine structures, it is certain to reason the Microsporidia S80 belong to the phylum Microspora, class Microspora, order Microsporida, order Microsporida. The shape of two nuclei cited seems to be genus Nosema, but in the classification for the suborder it should be defined wheather pansporoblasts be formed or not and for the genis especial attempts have been made to define the characters which distinguish the disporous genera in the life cycle. Survey through the infection of the bad cocoons during 1980 to 1982 in South Korea the areas contaminated with new microsporidia were revealed 5 provinces of Kyung-Gi, Kang-Won, Chung-Nam and Chun-Nam. Pathological effects inoculated per os at second instar larvae of silkworm, the LD 50 was $7.1{\times}10^7/ml$ as lower pathogenecity than that of Nosema bombycis Naegeli of $1.2{\times}10_7/ml$. While on the other hand the inoculation of the microsporidia at fourth instar larvae lowerd the whole cocoon weight and cocoon shell weight and significant at 1% level. The microsporidia S80 defined it can not be transmitted transovarially from the result of predictive and collective examination of 21 egg batches from the infected female moth.

Selection of Chemicals for Separation of Copulated Moth of Silkworm, Bombyx mori L. (약제에 의한 가잠교미아의 할애효과에 대하여)

  • To save the labour reguired for separation of copulated moth during egg Production, some of the chemicals available at the market were on screen test for easy separation of copulated moth of silkworm. The obtained results are summarized as following. 1. In a separation ratio of copulated moth along with a treating time, TCTFE(Trichloro-trifluoroethane) completely separated the copulated moth in 10 minutes. The combinations of TCTFE plus Acetone (87.5 : 12.5 V/V) and TCTFE plus Acetic acid (50 : 50 V/V) take 25minutes for the complete separation of copulated moth. Use of Acetic acid solution only makes 100% separation of copulated moth in one hour and foully minutes and non$.$treatment shows only 47.5% of separation in three and half hours. On the other hand. There is no statistical significance between TCTFE Plus Acetone and control in the egg productivity. 2. The combination of TCTFE Plus Acetone (87.5 : 12.5 V/V) does not infuluence the egg productivity of the moth, showing 443 grains for an average number of egg per moth out of which 417 grains are for the number of fertilized eggs while control shows 452 grains for an avarage number of egg/moth and 428 grains for the number of fertilized eggs. However a sing1e use of TCTFE and Acetic acid shows less egg productivity and number of ferilized eggs per moth, respectively as compared to those of the control. In particular, a single use of acetic acid makes an increase of number of non-egg prodncible moth and it seems to be brought due to a chemical damage. 3. In a rearing test of the egg laid by the chemical treated moth, there are no differences among the treatments in all of the useful characters of the larvae; larval duration, survival rate, cocoon yield, single cocoon weight, single cocoon shell weight and cocoon shell ratio. In these regards, it is recognized that TCTFE can be practically used for the separation of copulated moth and the combination of TCTFE and Acetone promote its efiectiveness on the separation of copulated moth.

Gametogenesis and Reproductive Cycle of the Cockle, Fulvia mutica (Reeve) (새조개, Fulvia mutica (Reeve)의 생식세포형성과정 및 생식주기)

  • The structure of gonads, gametogenesis and reproductive cycle of the cockle, Fulvia mutice, were studied mainly by histological observation. The materials were monthly sampled in the southern area of Yeosu from October 1980 to September 1981. F. mutica was monoecious. The gonads were situated between the liver tissues and the outer fibronuscular layers compacted by the connective tissue fibers and muscle fibers beneath the outermost layer of simple cuboidal epithelium. The gonad was composed of a number of the ovarian sacs and the testicular tubules which form the tubular structure. Testicular tubules in the mature stage sometimes contained 'testis-ova' The undifferentiated mesenchymal tissues and the eosinophilic cells were abundantly distributed on the germinal epithelium in the early development stage. With the further development of the ovary and testis, these tissues and cells gradually disapprared. The undifferentiated mesenchymal tissues and the eosinophilic cells are related to the growing of the oocytes and spermatocytes . Early multiplicating oogonium was about $10{\mu}m$ in diameter. As the oocytes grow to $27-34\times50-58{\mu}m$ by increasing cytoplasm, the oocytes connected to the basement membrane by their egg-stalks. The ripe eggs were about $60{\mu}m$ in diameter and they were surrounded by gelatinous membrane. Most male germ cells in mature stage were transformed into the spermatozoa and they formed the sperm bundles. After spawning, undischarged ripe eggs and spermatozoa remained in the ovarian sac and the testicular tubule respectively for some time, then they finally degenerated. Especially the early spent ovarian sacs in May did not contract significantly and then they took part in the secondary maturation within two or three months during the summer season. The monthly changes of the fatness well agreed with the reproductive cycle. The reproductive cycle of F. mutica could be classified into six successive stages : multiplicative, growing, mature, spent, degenerative and recovery stage. It seems that the spawning season is closely rotated to the water temperature, and the spawning occurs from May to October at about $20^{\circ}C$ in water temperature. The peak spawning seasons appeared twice a year between June and July and in September.
