예쁜꼬마선충을 이용한 돼지감자의 수명 연장 효능 (Lifespan Extending Effects of Helianthus tuberosus Linne in C. elegans)

  • Helianthus tuberosus Linne (Compositae) has been widely used as a folk remedy to treat various ailments including fever, bleeding, fracture and contusion. This study was designed to elucidate the lifespan extending activities MeOH extract of the tubers of Helianthus tuberosus Linne (MHT) using Caenorhabditis elegans (C. elegans) model system. In the current study, we found that the lifespan of worms was significantly extended by MHT supplement, dose-dependently. MHT also provided robust protection against various stress environments such as osmotic, thermal and oxidative condition. In addition, elevated antioxidant enzyme activities by MHT resulted in attenuation of intracellular reactive oxygen spices (ROS) levels, suggesting antioxidant capacity of MHT might be associated with longevity properties. Herein, we showed that altered food intake and growth of worms were also involved in the MHT activity. Furthermore, MHT increased body movement in aged worms, indicating possible role for MHT in healthspan.

  • Aconiti Kusnezoffii Tuber has been traditionally used to treat the symptoms of rheumatoid arthritis and joint pain. The main constituents are diterpenoid alkaloids such as benzoylmesaconine, benzoylaconine, mesaconitine, aconitine, and hypaconitine. In Korea, Aconiti Kusnezoffii Tuber is officially defined as the tubers of Aconitum kusnezoffii Reichb., A. ciliare Decasisne, and A. triphyllum Nakai. On the other hand, only the tuber of A. kusnezoffii is to be used in China. In order to identify the botanical origin of Aconiti Kusnezoffii Tuber circulated in Korea, we analyzed 24 samples of Aconiti Kusnezoffii Tuber obtained from local markets for comparative DNA analysis. The sequence analysis of nrRNA ITS 1 was useful to distinguish Aconitum species and revealed that the roots of A. karakolicum were circulated in Korean markets without discretion. HPLC quantitative analysis showed that aconitine was detected at the highest amount in A. karakolicum. Authentic diterpenoid alkaloids were coinjected for quantification of aconitine-type ingredients. All data were statistically grouped by Principal Component Analysis (PCA). This study suggests that both molecular and chemical analyses should be utilized for the standardization and the quality control for Aconiti Kusnezoffii Tuber.

Cloning of celC, Third Cellulase Gene, from Pectobacterium carotovorum subsp. carotovorum LY34 and its Comparison to Those of Pectobacterium sp.

  • Phytopathogenic Pectobacterium carotovorum subsp. carotovorum (Pcc) LY34 secretes multiple isozymes of the plant cell wall degrading enzyme endoglucanases. We have cloned a third cel gene encoding CMCase from Pcc LY34. The structural organization of the celC gene (AY188753) consisted of an open reading frame (ORP) of 1,116 bp encoding 371 amino acid residues with a signal peptide of 22 amino acids within the NH$_2$-terminal region of pre-CelC. The predicted amino acid sequence of CelC was similar to that of Peetobaeterium ehrysanthemi Cel8Y (AF282321). The CelC has the conserved region of the glycoside hydrolase family 8. The apparent molecular mass of CelC was calculated to be 39 kDa by CMC-SDS-PAGE. The cellulase­minus mutant of Pee LY34 was as virulent as the wild-type in pathogenicity tests on tubers of potato. The results suggest that the CelC of Pce LY34 is a minor factor for the pathogenesis of soft-rot.

수확시기와 저장방법에 따른 마의 塊根重과 품질변화 (Tuber Yield and Characteristics of Chinese Yam as Affected by Harvesting Date and Storage Condition)

  • To give some information on the harvest and storage of Chinese yam (Dioscorea opposita), the experiment was done to examine the effect of its harvesting date and storage method on tuber weight, tuberrelated characters during the storageor at the end of storage. The yam tubers harvested 4 times at 20 days interval from Oct. 15 were stored with different storage methods(pit, room and cold temperature) and on Mar.15. all the characters were measured. Fresh weight of the tuber harvestedafter Nov. 5 and stored were greater than that done on Oct. 15 while dry tuber weight did not have any difference between the harvesting dates. The fresh weigth done directly from the field on Mar. 15 was higher compared to the other harvesting dates owing to its higher moisture content. The rate of sound tuber stored at room or cold temperature condition was higher than that at the field at which the rate was greater at the harvest after late nov. compared to the earlier harvests. Although the two storages were similar at sound tuber rates and marketable yield, the tuber weight was more reduced when stored at room temperature than when done at cold temperature . Brightness and value 'b' of chromaticaity realted to the tuber coloring and sugar content were higher but moisture content was lowerwhen harvested on Oct. 15 than the other havesting dates. All the characteristics realted to the marketability after storage were nearly same when stored at room and cold temperature. Value 'a' of chromaticity and sugar content were higher when harvest on Mar. 15 than when harvested before Dec. 15 but the 'b' value and mositure content were reverse results.

현호색속(현호색과)의 신종: 봉화현호색(Corydalis bonghwaensis M. Kim & H. Jo) (A new species of Corydalis (Fumariaceae): C. bonghwaensis M. Kim & H. Jo)

  • 경상북도 봉화에서 현호색속의 신종인 봉화현호색(Corydalis bonghwaensis M. Kim & H. Jo)을 새로이 발견하여 기재하였다. 봉화현호색은 꽃이 흰색이고, 소화경에 털이 없으며, 괴경의 내부는 흰색이고, 주두돌기가 다각형이라는 점에서 남도현호색과 흰현호색과 유사하다. 그러나 봉화현호색은 꽃색이 연노랑에서 흰색으로 변하고, 내화피 정단은 약하게 돌출하며, 잎 열편은 가는 선형이고 2열 종자를 가진 방추형 삭과이다. 이에 반하여 남도현호색은 내화피 정단이 오목하게 함몰하고 잎 열편은 다양하며 2열 종자를 가진 방추형 삭과인 반면에, 흰현호색은 내화피는 오목하게 함몰하고 잎 열편은 타원형이며 1열 종자를 가진 선형 삭과이다. 또한 난장이현호색은 꽃색이 하늘빛 보라색이고 소엽은 타원형이며 꽃이 1-5개 달리고 아래 외화피가 반구형인 점에서 봉화현호색과 뚜렷이 구별된다.

Botrytis cinerea에 의한 초석잠 잿빛곰팡이병 발생 (Occurrence of Gray Mold of Stachys sieboldii Caused by Botrytis cinerea)

  • 2005년 3월경 경남농업기술원 움막저장고에서 저장중이던 초석잠의 덩이줄기에 이상증상이 발생하였다. 병징은 초석잠 덩이줄기 부분이 수침상으로 물러지고 썩으면서 병반부위에 회색의 곰팡이가 많이 생겼다. 분생포자는 무색, 단포자이며 난형 또는 타원형으로 크기는 $5{\sim}16{\times}4{\sim}12{\mu}m$였다. 분생포자경은 갈색으로 격막이 있고 나무가지 모양이며, 폭은 $14{\sim}30{\mu}m$였다. 균사생육과 균핵형성 적온은 $20^{\circ}C$였다. 초석잠에서 병징과 병원균의 균학적 특징, 병원성을 조사한 결과 Botrytis cinerea Persoon:Fries에 의한 초석잠 잿빛곰팡이병으로 동정하였다.

  • Kluyveonyces fragilis로부터 정제한 ${\beta}-fructosidase$ (inulase)를 Tygon 관과 aminoethyl-cellulose에 고정화 시킨후 효소의 일반적 성질을 알아 보았다, Tygon곤에 효소를 고정화 시키는데 있어서는 크로로포름 용액에서 65^{\circ}C의 조건으로 실란화(silanization) 시킨 다음 10 % 글루타르알데히드로 처리하는 것이 매우 중요한 처리 조건인 반면 2 % 글루타르알데히드 처리가 aminoethyl-cellulose에 inulase를 고정화 시키는데 매우 효과적이 었다. Tygon관에 고정화 시킨 inulase는 g matrix당 11.5 units의 inulase 역가로 보여 주었고 효소 역가의 회수율은 22.5 %인 반면 aminoethyl-cellulose에 고정화 시킨 제품은 g matrix 당 39.3 units, 53.4 % 효소 역가 회수율을 보여 주었다. 두 고정화 제품을 사용하여 invertase(기질이 sucrose) inulase (기질이 inulin) 역가에 대한 효소 안정성, pH 효과, 온도 효과 및 기질 효과를 살펴 보았다.

  • 본(本) 시험(試驗)은 벗풀 우점도(優占度)의 지속적(持續的) 증가(增加) 원인인(原因)과 효과적(效果的)인 방제법(防除法)을 밝히고자 수행하였다. 1. Sulfonylurea계(系) 제초제(除草劑) 관행처리시(慣行處理時) 벗풀은 일정(一定) 기간(期間)동안 생육억제(生育柳制) 후(後) 재생(再生)되었다. 2. Pyrazolate에 의한 벗풀의 살초범위(殺草範圍)는 7엽기(葉期) 전개(展開) 이전(以前)이었다. 3. Pyrazolate에 대한 벗풀의 살초범위(殺草範圍)와 괴경(塊莖)의 크기 및 탄수화물(炭水化物) 소비량(消費量)과는 상관(相關)이 없었다. 4. Pyrazosulfuron-ethyl과 pyrazolate 일차(一次) 처리(處理) 후(後) 이들 제초제(除草劑)의 체계처리시(體系處理時) 벗풀 괴경형성(塊莖形成)이 95% 이상(以上) 억제(抑制)되었다.

The Contents of Heavy Metals (Cd, Cr, As, Pb, Ni, and Sn) in the Selected Commercial Yam Powder Products in South Korea

  • Yam (Dioscorea) has long been used as foods and folk medicine with the approved positive effects for health promotion. Although consumption of yam products is increasing for health promotion, reports for the metal contamination in commercial yam powder products to protect the consumers are lacking. In this study, we aimed to assess whether the commercial yam powder products were heavy metal contaminated or not using the yam products from six commercial products from various places in South Korea. The contents of heavy metals (Cd, Cr, As, Pb, Ni, and Sn) in yam powder products were measured and compared to national and international food standard levels. Also, the metal contamination was monitored during the food manufacturing steps. The study results showed that the contents of heavy metals (Cd, Cr, As, and Pb) in yam powder products are similar to those in national 'roots and tubers' as well as in various crops. In comparison to three international standard levels (EU, Codex and Korea), Cd content in yam powder products was lower but Pb content was 5 times higher. Also, Pb, Ni, and Sn may have the potential to be contaminated during food manufacturing steps. In conclusion, the level of heavy metals (Cd, Cr, As, Ni, and Sn) except Pb is considered relatively safe on comparison to national and international food standard levels.

감자 바이러스 무균종묘의 대량생산과 플러그화에 관한 기초 연구 (Production of Plug Plantlets for Mass Propagation Using Stem Cuttings of Virus Free Microtubers in Potato)

  • 감자의 생장점 배양을 통하여 생산된 소량의 소괴경(microtuber)을 경삽하여 균일하게 생장된 플러그우량묘를 대량생산할 수 있는 기술을 개발하기 위하여 삽수를 정단 및 기부부위로 구분하고 NAA 및 IAA 10 mg /1 용액에 2시간 침지하여 cocopeat, vermiculite, perlite, peatmoss 혼용배지를 이용한 삽목과 온로장 처리 ,포장정식후 괴경형성효을에 대하여 시험한 결과를 요약하면 다음과 같다. 1. 감자의 소괴경 유내 줄기를 이용한 플러그 삽목시 고체배지는 cocopeat, vermiculite, perlite, peatmoss를 이용한 혼합배지에서 플러그의 형성율이 양호하게 나타났다. 2. 고온장일조건(2$0^{\circ}C$이상, 16시간의 일장)에서는 지상부생육 및 지하부발근이 양호하였으나, 외부방치구에서는 발근이 부진하고 복지가 발생하여 소괴경이 형성되었다. 3. 지상, 지하부 생체중은 생장조절제 전처리시 NAA 10mg/1 및 IAA 10mg/1 간에 차이가 없었다. 4. 삽수부위별로는 삽목묘의 초기생육은 초장에서는 비슷한 경향이었고, 그 외 엽수, 근수, 근장 등의 형질은 정단부위묘가 양호하였지만, 포장정식 후에는 큰 차이가 없었다. 5. 삽목묘를 봄재배하여 괴경형성여부를 시험한 결과 초장은 70~80cm, 복지수 10여개, 괴경수는 7~8개가 형성되었다.
