Plant growth promoting rhizobacteria influence potato tuberization through enhancing lipoxygenase activity

  • Molecular insights on the role of plant growth promoting rhizobacteria (PGPR) in potato tuberization are reported in the present study. The PGPRwere isolated from the soil collected from potato fields of Highland Agricultural Research Centre, Pyeongchang, Korea and they were identified to the genus level based on the 16S rRNA sequence analysis. These PGPR were heat-killed, filtered and the filtrates were addedindividually at a concentration of $10^7\;cfu\;mL^{-1}$ in MS (Murashige and Skoog's) medium supplemented with 7% (w/v) sucrose to study their influence on in vitro potato tuberization. Tuber initiation occurred early in untreated control, while tuber growth was pronounced in case of PGPR treatments. The control explants showed tuber formation as a result of sub-apical swelling of stolons while several sessile tubers formed directly in the axils of nodal cuttings in case of PGPR treatments, which is an indication of strong induction for tuberization. Theexplants cultured on MS medium supplemented with bacterial isolate 6 (Bacillus firmus strain 40) showed highest average tuber yield (Ca. 12.56 g per treatment) after 30 days of culture, which was 3 folds increase over the untreated control. A significant increase in lipoxygenase (LOX1) mRNA expression and activity of LOX enzyme were also detected in the tubers induced on PGPR treatments as compared to untreated control. This LOX expression level correlated with increased tuber growth and tuber yield. Further studies focused on the role of bacteria cell wall components, growth regulators and signal molecules released by PGPR are under investigation to elicit clues for PGPR-mediated signal pathway controlling potato tuberization.

Purification and Thermal Inactivation of Two Lipoxygenase Isoenzymes from Potato Tubers (감자 Lipoxygenase isozyme의 분리와 열불활성화)

  • Two lipoxygenases (F-I and F-II) were purified from potato tubers by ammonium sulfate fractionation and ion-exchange column chromatographies. The purified isoenzymes were apparently homogeneous on polyacrylamide gel electrophoresis. Both enzymes showed a similar optimum pH of 5.5-6.0. From thermal inactivation experiments with the purified enzymes in the range of 50 to $65^{\circ}C$, D-values of 13.3 min and 4.3 min at $65^{\circ}C$, and z-values of $11.8^{\circ}C\;and\;10.3^{\circ}C$ were obtained respectively for F-I and F-II. By applying absolute reaction rate equation, thermodynamic parameters wire also determined for the activation part of the inactivation process.

Toxicologic Features and Management in Aconitine Intoxication Following Ingestion of Herbal Tablets Containing Aconitum Species (한방환약 복용 후 발생한 Aconitine 중독의 임상독성학적 특성과 환자관리)

  • Purpose: Unrefined tablets prepared from Aconitum tubers are occasionally used in Korean folk medicine. This study defines the potential sources, clinical toxicology, and treatment of aconitine poisoning. Methods: A retrospective survey was conducted in 63 patients in the ED of a tertiary University Hospital with suspected toxicity from an unrefined tablet prepared from Aconitum tubers from 1999 to 2007. Results: A total of 63 cases enrolled included 26 men and 37 women, aged 30 to 86 years. Forty-eight patients ingested aconitine tablets as digestives, 26 tablets on average. After a latent period of 30 to 450 minutes, patients developed a combination of neurologic (87.3%), gastrointestinal (82.5%), cardiopulmonary (41.3%), and other (28.6%) features typical of aconitine poisoning. Initial ECG abnormalities revealed dysrhythmia (61.9%), conduction disturbance (42.9%), and abnormal waveforms (39.7%), with 28.6% of patients having normal ECGs. All patients received supportive treatment or close observation regardless of ingestion amounts. Patients with hypotension or ventricular arrhythmia were treated with inotropic agents or amiodarone. Conclusion: Toxicologic signs and symptoms can occur after the consumption of aconitine tablets, regardless of ingestion amount. The risk occurs because of inadequately processed aconitine roots. This study will provide important data for public education and distribution regulations for Aconitum sp. in Korea.

  • The study tried to examine the characteristics of transcription culture on YouTube through narrative analysis methods. The study found five meaningful features in YouTube's transcription culture. YouTube's transcription culture was first characterized by efficient writing and learning skills. Second, there was a characteristic of a transcription to read and understand text more deeply. Third, it had the characteristics of five strategies to advance my writing. Fourth, YouTubers had time to self-heal and comfort through transcription. Fifth, YouTube's transcription culture has expanded and developed into left-handed writing and digital writing. The characteristics of these YouTubers' transcription culture are expected to enrich the transcription culture that has been handed down for many years.

  • Travel YouTubers who cross the boundary between tradition and novelty communicate by sharing their activities with others through SNS-based media. Media content should not only satisfy individuals, nor should it be too purpose-oriented. YouTube channels should be operated so that users can easily access content naturally, and more diverse methods can be pursued based on usage patterns and satisfaction theory. This study is about the type of perception of Generation Z on travel YouTube titles. As a result of conducting QUANL program analysis on 34 Q samples and 28 P samples from the Q methodological perspective, a total of three types were found. For types with unique characteristics, the first type was named "attention of keywords that draw imagination," the second type was "preferred to stories that stimulate curiosity," and the third type was "image satisfaction reflecting expectations." In addition, considering the characteristics of each type found, the scalability and strategic plan of the activities that Generation Z travel YouTubers want to unfold were presented.

Monitoring on Bolboschoenus planiculmis Restoration in Nakdong River Estuary: Implications for Wetland Restoration Using Shoot Transplantation (낙동강하구 새섬매자기(Bolboschoenus planiculmis) 복원 모니터링: 식물체(shoot) 식재를 이용한 습지복원)

  • Bolboschoenus planiculmis has been acknowledged as a key species in whooper swans (Cygnus cygnus) habitat by providing food for this migratory waterfowl. B. planiculmis wetlands are being degraded by water shortages and salinization caused by anthropogenic activities and climate changes. In 2004, the distribution of B. planiculmis in the tidal flats of the Nakdong Estuary was 2,475,568 m2, and in 2021, the distribution area was 798,731 m2, which decreased by 32.3%. In order to restore the degraded B. planiculmis wetlands, shoot transplantation and seed sowing were tentatively used in three places with different salinity and water levels. The average density per unit area in September at the optimal growth period after planting were A (fresh water level 50 cm) 58±15.65 m-2, B (brackish water level 0~5 cm) 188±63.83 m-2, C (brackish water level 0 cm or less) 188±45.13 m-2. The tubers were observed as A 0 g dw m-2, B 25.32±2.94 g dw m-2, and C 13.39±0.91 g dw m-2. Tubers were distributed in the soil, with only 3.0% at the 10~20 cm depth but 97.0% at the 0~10 cm depth. In contrast, the germination rate of B. planiculmis seeds was observed to be 0%. Results of this study provide technical support for the restoration of B. planiculmis wetland and the improvement in the quality of whooper swans habitat.

  • This study was conducted in order to know the ecology and to establish the control method of Potamogeton distinctus A. BENN. The tubers of Bog pond weed was almostly located at soil depth of 10 cm to 20cm, germination rates and number of formed tubers were different according to transplanting depth of soil. Effects of herbicides application on weeds were different according to leaf-stage of pondweed, and the pondweed were withered to death by absorption of Avirosan into the leaves. It was suitable to keep water depth at 3cm level until about a week after Avirosan application.

  • This study was conducted as part of a phased study aimed at closely examining the mutual ecosystem between creators and viewers through paying attention to the personal media environment that is in the center of rapid changes in the media industry and particularly exploring the areas of activity of the single-person or minority media creators who lead the mobile media environment that could be accessed, viewed and produced anywhere. In particular, this study aimed to not only pay attention to the situation in which the potential to expand professional YouTuber areas encounters the changing desire and demand of content service users who continue to become more evolved as time passes, but also examine the influence of the reputation of professional YouTubers on users' loyalty and subscription intention, examine the moderating effect of sincerity on the relationship between YouTubers and users, and demonstrate such relationship formation process based on concrete data. In the conclusion section, implications that can be drawn from the study results and suggestions for further studies in the future were proposed.

  • This study was carried out to determine optimum harvest time of in vitro multiplied tuber Pinellia ternata(Thunb.)Breit from 1990 to 1992. Tuber yield and its related traits were observed with four harvest times. Plant height was 7.2cm at 6 months and 19.3cm at 24 months after planting. The number of tubers per unit area was significantly increased with the passage of growing time after planting$(78\;tubers/m^2)$, those were increased by 2.3 times in 6 months, 5.6 times in 12 months, 13.3 times in 18 months and 20.0 times in 24 months. The tuber sizes(length, width and weight) were highly enlarged during 12 to 18 months after planting compared with before or after this time. There were smaller tubers and lower tuber yields with shorter growing period, while large tubers above 7.1mm and higher yields with longer growing. It was suggested that the optimum harvest time was 18 months after planting, before over wintering rather than next year of spring, 24months after planting.

  • Pinellia tripartita(Blume) Schott is a herbaceous perennial plant belonging to the Araceae and distributed on Asia including of Korea, Japan, and China. P. tripartita is often used for gardening but has not been developed mass-breeding methods. In this study, we compared the tuber growth in different combinations of mixed soils used six compositions. Tubers used to study was cultured in vitro and divided into two groups. Type I was diameter more than 1cm and the group of Type II was diameter below than 1cm. Enlargement of tubers and growth of aerial parts were measuring the plant height, number of fresh leaves and dead leaves, number of bullets, tuber size, and fresh / dry weight. The size/weight and numbers of tubers from the mixed soil B (coir 68.0%, peat moss 14.7%, perlite 3.0%, vermiculite 7.0% and zeolite 7.0%) were the best grown up for eight weeks. In case of Type I, GI (Growth index) of tuber size and weight were 45% and 101%, respectively. The difference of growth was doubled compared to the bad growth treatment as the mixed soil E(Coir 14.3%, peat moss 14.3%, perlite 42.9%, vermiculite 14.3%, and zeolite 14.3%). These results could be used as the basic information for the similar experimental design for the P. ternata.
