Effects of Application of Compost Made from Citrus Skin and Starch Sludge on Potato Growth

  • The experiment fields consisted of five plots as follows; 2, 4, and $8\;tons{\cdot}ha^{-1}$ citrus skin in combination with starch sludge and pig manure mixing compost (CSSP), $4\;ton{\cdot}ha^{-1}$ fermented pig manure compost (FPMC) treated plot, and untreated control. Plant height and stem diameter were significantly increased by CSSP. Most of all, average tuber weight and tuber yield per plant were significantly increased in 4 and $8\;tons{\cdot}ha^{-1}$ CSSP plots compared to the other plots. Marketable tuber (>50 g fresh weight) yield were superior in order of 4 and $8\;tons{\cdot}ha^{-1}$ CSSP plot, $4\;tons{\cdot}ha^{-1}$ FPMC plot, and $2\;tons{\cdot}ha^{-1}$ CSSP plot.

Optimization for Hot water Extraction Condition of Liriope spicata Tuber Using Response Surface Methodology (반응표면분석법에 의한 맥문동 열수추출 조건의 최적화)

  • Optimal conditions for hot water extraction of Liriope spicata tuber were investigated with changes in solvent ratio(2∼6 fold) and heating time(1∼5 hr) by response surface methodology. The content of extractable solids increased with an increased in solvent ratio, and the highest content showed at heating time of 3 hr. The content of total steroid saponin increased with a decrease in solvent ratio, and increased with an increase in heating time at increasing the solvent ratio. The content of non-reducing sugar containing oligosaccharides at a lower solvent ratio didn’t show changes depending on heating time, while that at a higher ration decreased with an increase in heating time. Optimal extraction conditions using hot water as the limited conditions of 15∼18% extractable solids, 1.5∼2.0% total steroid saponin, 6∼8% reducing sugar, 6∼7% non-reducing sugar and 13∼15 brix were 3 hrs of heating time and 4 fold of solvent ratio.

  • MeOH extracts of sixteen herbal drugs were tested for the effects on bovine adrenal tyrosine hydroxylase and dopamine ${\beta}-hydroxylase$. The MeOH extracts of Paeoniae Radix and Pinelliae Tuber showed 65% inhibition on the tyrosine hydroxylase activity at the concentration of 100 $\mu$g in the enzyme reaction mixture. Those of Paeoniae Radix, Pinelliae Tuber and Evodiae Fructus showed 87, 84 and 62%, respectively, on the dopamine ${\beta}-hydroxylase$ activity.

Genotype Difference of Plant Regeneration from Dormant Bud Culture in Colocasia esculenta Schott.

  • This study was conducted to obtain the basic breeding information of Colocasia esculenta Schott. Effect of supplemental plant growth regulators and genotype difference were investigated on dormant bud tissue for proliferation. The plant regeneration ratio, plant height and root length were the best upon mixed treatment of 0.8mg/L IAA and 2.0mg/L zeatin. Both leaf weight and root weight were heavy upon culture in a dark condition. The leaf and root weights were heaviest in 6Pie sucrose concentrations. In several collected area the heaviest one was Binnangxin and then in the order of Suwon, Wanju and Puan. Genotype differences of tuber diameter and tuber weight were found in Suwon. Tuber weight was found in the order of Suwon (862mg) >Wanju(723mg) >Puan(649mg) >Binnangxin (424mg).

  • We examined the growth characteristics, fresh tuber yield, rotenone content of two yam beans (Pachyrhizus erosus and P. ahipa) introduced from Mexico. P. erosus species showed better adaptable ability than P. ahipa species in seed productivity and tuber yield. Rotenone content in the P. erosus species extracted with chloroform was 3.6 folds much more extracted than ethanol extraction. The order of rotenone content found in crude extract obtained by different solvent extraction from the highest to the lowest was mature seed (484.7 ${\mu}g$) and leaves (17.2 ${\mu}g$) of the P. erosus species, respectively.

Report on Tuber huidongense, a Truffle Species Previously Unrecorded in Korea

  • The fruiting bodies and roots of a Tuber species were collected from the rhizosphere of Quercus dentata in Pohang, Korea. The morphological characteristics of the ascoma and ectomycorrhizal roots were studied, and phylogenetic analyses were performed using the sequences of the internal transcribed spacer, large subunit rDNA, and β-tubulin DNA. Based on the features of the fruiting bodies, the species was identified to be Tuber huidongense, which has not been reported earlier in Korea. The morphological characteristics and phylogenetic analysis of T. huidongense are described in the present study.

Effects of Planting Density and Harvesting Time on Production of Small-size Tuberous Roots in Sweet Potato (소형 고구마 생산을 위한 재식거리와 수확시기의 영향)

  • Small-sized tuber of sweet potato (Ipomoea batatas Lam.) is more preferred by Korean consumers, because it is convenient to eat with hands, and can easily be steamed or roasted in small pan as a healthy snack for a small family. The present study was aimed to investigate the effect of high-density plantings and harvesting times on production of smallsized tuber (50~200 g). Four varieties ('Annobeny', 'Daeyumi', 'Shinzami', and 'Yeonhwangmi') were planted in 2-row zigzag high-density planting ($75{\times}30cm$, A; $75{\times}25cm$, B) and 1-row planting ($75{\times}25cm$, control) covered with black vinyl film on May 23th, 2014. Marketable yields and small-sized tuber yields were compared between 120- and 150-day harvesting. Vine length and fresh weight per plant were significantly decreased in 2-row planting A and B plots compared to the control at 120 days after planting. Weight of tuber per plant, mean weight of tuber and number of tuber per plant were significantly decreased in 120-day harvesting of A and B plots than in the control. Marketable yields per 10a were increased by 17% ($2.4ton{\cdot}10a^{-1}$) and 8% ($2.6ton{\cdot}10a^{-1}$) in 120-day harvesting of A and B plots compared to the control ($2.4ton{\cdot}10a^{-1}$), and those were largely increased by 29% ($4.0ton{\cdot}10a^{-1}$) and 26% ($3.9ton{\cdot}10a^{-1}$) in 150-day harvesting than in the control ($3.1ton{\cdot}10a^{-1}$). Proportions of small size tuber (50~200 g) were increased by about 65% in 150-day harvesting of A and B plots compared to the control (55.3%). Small-size tuber yield and marketable yield were highly increased in 150-day harvesting of 2-row zigzag high-density planting A ($75{\times}30cm$).

  • Using a bioassay for tuber enlargement activity, which was carried out with culture of microtuber of nodal stem segments in vitro, the endogenous substances was detected from leaves of yam(Diosorea alata L., cv. Solo Yam) treated under the short and the long day length condition. Tuber enlargement activity was found in both the aqueous and ethyl acetate phase of extract obtained from leaves of yams. Those activity was found in both the aqueous and ethyl acetate phase of extract obtained from leaves of yams. Those activities increased under the short day length, whereas not increased under the long day length. Guided by bioassay, the active substances in the ethyl acetate phase were purified by charcoal adsorption chromatography. The result obtained indicated that the 40% ethanol fraction contained the most prominent tuber enlargement activity. In seasonal changes of the leaves, tuber enlargement activity remained almost constant during summer(June-August), but after. the middle of September, the activity increased gradually and then reached a maximum in early October. Jasmonic acid (JA) was isolated with the guidance of microtuber test and identified by gas chromatography. The level of endogenous JA in the leaves of plants was more than $290{\mu}g/kg$ at the 10 hr day length. These results seem evidences for the occurrence of the tuber enlargement activity which is formed in leaves under the short days and transmitted to under ground part to induce growth and enlargement of tuber.

  • Aconiti tuber butanol fraction has been recently known to have stimulatory effect on myocardial contractility. In the present study, the possibility that the Aconiti tuber butanol fraction acts directly on contractile proteins of myocardium has been investigated using natural actomyosin extracted from dog heart. It revealed that Aconiti tuber butanol fraction in concentrations from $10^{-2}{\sim}10^{-7}\;gm/ml$ had no stimulatory effect on either the $Mg^{++}$ or $Ca^{++}$-activated adenosinetriphosphatase activity of cardiac actomyosin. And no direct $Ca^{++}$-like action of the drug on cardiac actomyosin was also found. Aconiti tuber butanol fraction in concentrations above $10^{-4}\;gm/ml$, however, was somewhat stimulatory on superprecipitation of actomyosin and markedly inhibited the membrane bound $Na^+-K^+$-activated ATPase activity. In these connections, the positive inotropic action of Aconiti tuber butanol fraction on myocardium thus does not seem to reflect a direct interaction with contractile proteins, but the drug seem to stimulate myocardial contractility through the actions on the membrane transport of $Ca^{++}$.

  • The freezing resistance of Eleocharis kuroguqai was evaluated in artifical and natural freezing conditions in relation to tuber size, depth of tuber burial and soil moisture conditions. Osmotic potential of immature and mature tuber of E. kuroguqai before hardening was -12.1 and -23.5 bar, respectively, but decreased to -61. 0 and -67.1 bar after 120-day hardening period. Percent survival of E. kuroguqai tubers during the winter period was 13% and 67% in dry and moist moisture conditions at burial of 5cm depth, but all tubers were survived in the three moisture conditions employed when buried at 25cm depth. Greater decrease of osmotic potential was observed in tubers buried at 5cm depth than tubers buried at 25cm depth, resulting in greater freezing resistance in the former. Large tuber was more susceptable to freezing temperatures than small tuber.
