Study of the Suppressive Effect and Its Mechanism of Amomum Cardamomum L. on Free Fatty Acid-induced Liver Steatosis (지방간에 대한 백두구 에틸아세테이트 추출물의 억제 효과 및 기전 연구)

  • Through this study, the authors investigated the anti-steatosis effects of the Amomum cardamomum ethyl acetate fraction in free fatty acids (FFAs)-induced human hepatocellular carcinoma HepG2 cells. The ethyl acetate fraction of Amomum cardamomum (ACEA) was extracted with 70% ethanol and then the extract was evaporated using a rotary evaporator prior to sequential fractionation. Human hepatocellular carcinoma were treated with different concentrations of ACEA in the presence and absence of FFAs. To demonstrate the reactive oxygen species (ROS) scavenging activity, DCFDA level was analyzed by using in vitro assay system. Cell viability, lipid accumulation, intracellular triglycerides, malondialdehyde (MDA), liver steatosis related signaling molecules and inflammatory cytokines such as interleukin (IL)-6, 8, tumor necrosis factor-alpha ($TNF-{\alpha}$) were also investigated. As results, ACEA inhibited the FFAs-induced ROS, lipid accumulation, intracellular triglycerides, and MDA in a dose dependent manner. Treatment of human hepatocellular cells with ACEA induced the phosphorylation of 5' adenosine monophosphate-activated protein kinase (AMPK) and carnitine palmitoyltransferase I (CPT1) expression using western blot analysis. ACEA also potently suppressed the FFAs-induced inflammatory cytokines including IL-6, IL-8 and $TNF-{\alpha}$. These results suggest that the ethyl acetate fraction of Amomum cardamoum extract own inhibitory effects of liver steatosis by inhibiting ROS, lipid accumulation, intracellular triglycerides, MDA through AMPK signaling and anti-inflammatory actions.

Assessment of Biochemical Profiles in Premenopausal and Postmenopausal Women with Breast Cancer

  • Objective: The study was conducted to assess biochemical profiles in premenopausal and postmenopausal women having breast cancer. Materials and Methods: A hospital based case control study was carried out at Manipal Teaching Hospital (MTH), Pokhara, Nepal. The analysed variables were age, metabolic profile including total cholesterol, triglycerides, HDL-C, LDL-C, blood sugar, insulin concentration, C-peptide, HbA1c and selenium. Descriptive statistics and testing of hypothesis were used for the analysis using EPI INFO and SPSS 16 software. Results: In premenopausal women, significant differences were noted for total cholesterol (P value <0.001), triglycerides (P value 0.002), HbA1c level (P value <0.001), insulin concentration (P value 0.030), C-peptide concentration (P value 0.001), and selenium (P value <0.001) between cases and controls. Insignificant results were found for HDL-C (P value 0.749), LDL-C (P value 0.933), blood sugar (P value 0.59) and BMI (P value 0.746). Similarly, significant difference in total cholesterol (P value <0.001), triglycerides (P value 0.001), LDL-C (P value <0.001), HDL-C (P value 0.025), blood sugar (P value <0.001), insulin concentration (P value <0.001), c-peptide concentration (P value <0.001), HbA1c level (P value <0.001) and selenium (P value <0.001) were observed for postmenopausal patients and controls. Conclusions: Assessing metabolic changes and their management may be important for control of breast cancer and increased survival.

Effect of Catechins on Serum Lipids in Obese Women (카테킨 음용이 비만여성의 혈중 지질농도에 미치는 영향)

  • This study was developed to test the effectiveness catechin beverage drinking as exercise supplementary food for obese women. The effects of catekin were determined by weight, total cholesterol, triglycerides, High density lipoprotein cholesterol, high density lipoprotein cholesterol. Twenty-seven obese women were participated in this study. Eight women were tea catechin drinking group, Nine women were exercise group, and ten women were tea catekin drinking-combined with exercise group. After catechin drinking or exercise conducted, blood lipid factor were analyzed. body weight and triglycerides low density lipoprotein cholesterol levels were significantly decreased in tea catekin drinking combined with exercise group (p<0.001) and exercise group (p<0.001). total cholesterol were significantly decreased in three groups (p<0.05). It is thought that together catechin drinking and exercise can maximize weight loss of obese woman and reducing of blood lipid factor.

Hypocholesterolemic Response to Karaya Saponin and Rhodobacter capsulatus in Broiler Chickens

  • Dietary karaya saponin and Rhodobacter capsulatus (R. capsulatus) are known to have hypocholesterolemic actions, as reported in our previous studies. This study examined possible synergistic hypocholesterolemic effects of karaya saponin and R. capsulatus in broilers. A total of 150 broilers were allocated into 10 treatments: control, saponin 25 mg, saponin 50 mg, saponin 75 mg, saponin 25 mg+R. capsulatus 0.2 g, saponin 25 mg+R. capsulatus 0.4 g, saponin 50 mg+R. capsulatus 0.2 g, saponin 50 mg+R. capsulatus 0.4 g, saponin 75 mg+R. capsulatus 0.2 g and saponin 75 mg+R. capsulatus 0.4 g. Feed intake and feed efficiency were improved when karaya saponin and R. capsulatus were synergistically supplemented in the diet. Combinations of karaya saponin, especially supplementation of karaya saponin 50 mg+R. capsulatus 0.4 g were shown to have potential hypolipidemic actions in breast and thigh muscle cholesterol and triglycerides, serum cholesterol, low density lipoprotein-cholesterol and triglycerides, as well as improved high density lipoprotein (HDL)-cholesterol (p<0.05). Compared to the control, almost all the treatments significantly increased serum, liver and fecal concentrations of bile acids (p<0.05). Supplementation of both karaya saponin (75 mg) and saponin 50 mg+R. capsulatus 0.4 g reduced palmitic acid (C16:0) and stearic acid (C18:0) in a similar fashion (p<0.05). The ratios of PUFA:SFA or PUFA+MUFA:SFA in the thigh and breast muscle of broilers were greater in karaya saponin and R. capsulatus supplemented groups than in the control group. Thus, our study concluded that supplementation of karaya saponin synergistically with R. capsulatus in the diet of broilers is an effective way to obtain low-cholesterol, low-triglyceride and high HDL-cholesterol enriched poultry meat with a unique fatty acid balance.

Lipid Profiles and Related Factors in Adolescent (청소년의 혈청 지질 분포와 비만, 영양, 운동량의 연관성)

  • Objectives : To assess the prevalence rate of dyslipidemia and the level of related factors in adolescents groups in Seoul and Yangpyong area. Methods : Design; School based survey during May-Jun 1996 in Seoul and Yangpyong county. Subject; 2,453 boys and girls, aged 13 to 19 years; 1,137 Seoul and 1,316 Yangpyong county. Main Outcome Measures; Prevalence rates of raised serum total cholesterol, triglycerides, LDL-cholesterol, decreased HDL-cholesterol, obesity, and levels of energy intake and expenditure Results : Energy intake and fat intake of boys were higher than those of girls and they were higher in Seoul. Energy expenditure per day of boys was bigger than that of gins too. Prevalence rate of obesity was higher in boys of Seoul(15.2%) and girls in Yangpyong county(14.0%). Serum lipid profiles(total cholesterol, triglycerides, LDL-cholesterol) were higher among girls and all prevalence rates of dyslipidemia were higher in boys in Seoul and in girls in Yangpyong county. Especially, girls(29.1%) in Yangpyong had raised serum cholesterol level$(\geq170\;mg/dl)$. In contrast, boys in Seoul had higher level of decreased HDL-cholesterol(46.8%) compared to Yangpyong(23.6%). The relationships between serum lipid profiles and relative weights and sex are highly significant. And the relationship between triglycerides and energy intake is significant(p=0.038). But, the associations between serum lipid profiles and energy expenditure had borderline significances Conclusions : Hypercholesterolemia rates in girls were higher. Obesity prevalence rate was highest in boys of Seoul. Relative weight and sex are significantly related to lipid profiles. Therefore, Korea is in need of preventive strategies for different obesity and gender groups.

Influence of Functional Food Containing Bacillus polyfermenticus SCD on Lipid and Antioxidant Metabolisms in Rats Fed a High-Fat and High-Cholesterol Diet

  • We evaluated the effects of newly developed functional food containing Bacillus polyfermenticus SCD as the main material on the lipid and antioxidant metabolisms of hypercholesterolemic rats. Thirty male SD rats were divided into three groups after a 1-week adaptation period and were fed with a high fat-cholesterol diet (control), or with a high fat-cholesterol diet supplemented with low or high doses ($3.1\;{\times}\;10^6\;cfu/day$ or $3.1\;{\times}\;10^8\;cfu/day$) of B. polyfermenticus SCD and other physiological active materials for 6 weeks. Both doses of B. polyfermenticus SCD significantly reduced hepatic total cholesterol and triglycerides, while increasing the fecal excretion rates of total lipid, total cholesterol and triglycerides. B. polyfermenticus SCD increased the total radical trapping antioxidant potential (TRAP). The erythrocytic glutathione peroxidase activity in the B. polyfermenticus groups was significantly lower than that in the control group. Plasma TRAP levels exhibited a highly significant negative correlation with hepatic total cholesterol while a significant positive correlation was detected between fecal total cholesterol and plasma TRAP. This hypolipidemic and antioxidative effect of B. polyfermenticus SCD seemed to be unrelated to its dosage. These results suggest that functional food containing B. polyfermenticus SCD can improve oxidative stress and hepatic lipid profiles by enhancing the excretion of cholesterol and triglycerides in feces of rats fed with high fat-high cholesterol diet.

Validity for Use of Non-HDL Cholesterol Rather than LDL Cholesterol

  • NonHDL cholesterol values have been suggested as a risk marker for cardiovascular disease. NonHDL cholesterol values were calculated, using a very simple measurement [nonHDL cholesterol=serum total cholesterol-HDL cholesterol]. This formula is very useful as a screening tool for identifying dyslipoproteinemias, risk assessment, and assessing the results of hypolipidemic therapy. The data from the 2009 Korean National Health and Nutrition Examination Survey were used. Analysis was done for 1,992 subjects with lipid panels (Cholesterol, HDL, LDLdirect and Triglycerides) results. We studied the relationship between nonHDL cholesterol and LDL cholesterol. As a result, nonHDL cholesterol values were plotted against the LDL direct and calculated values. The linear regression equation for nonHDL cholesterol and direct LDL cholesterol was $nonHDLchol=23.60+1.03{\times}LDLdirect$ (p<0.0001, $r^2=0.80$) in all subjects. The subjects were classified into triglyceride values. When triglycerides are below 400 mg/dL, the linear fit to LDL direct is found to be $[nonHDLchol=17.34+1.07{\times}LDLdirect]$ (p<0.0001, $r^2=0.88$) and to the Friedewald LDL calculation is $[nonHDLchol=23.10+1.02{\times}LDLcalc]$ (p<0.0001, $r^2=0.82$). For triglycerides above 400 mg/dL, the linear fit equation is $[nonHDLchol=87.57+0.92{\times}LDLdirect]$ (p<0.0001, $r^2=0.50$) and to the LDL calculated, it is $[nonHDLchol=142.70+0.50{\times}LDLcalc]$ (p<0.0001, $r^2=0.32$). This study provides examples of the utility of nonHDL cholesterol concentrations in clinical medicine.

Triglyceride Composition of Kidney Bean (Phaseolus vulgaris L.) (강남콩(Phaseolus vulgaris L.)의 Triglyceride 조성)

  • Lipids of Kidney bean were extracted by the mixture of chloroform-methanol-water (1:2:0.8, v/v) and from these lipids, triglycerides were separated by thin-layer chromatography(TLC) and then reseparated into different fractions by high performance liquid chromatography (HPLC) depending on the polarity of the triglycerides. The triglyceride and fatty acid composition of these fractions were determined by gas liquid chromatography (GLC). From these results, the major triglycerides of Kidney bean lipids were estimated to be $C_{18:2}C_{18:3}C_{18:3}$ (26.6%), $C_{18:2}C_{18:2}C_{18:3}$ (18.5%), $C_{18:3}C_{18:3}C_{18:3}$ (14.9%).

  • The effect of $Fe^{2+}$ and myoglobin on the oxidative stability of triglyceride molecular species was investigated at $25^{\circ}C$. The molecular species of soybean oil triglycerides were analyzed by capillary column gas chromatography and electron impact ionization mass spectrometry utilizing selected ion monitoring. When $Fe^{2+}$ and myoglobin were added to soybean oil triglycerides, the oxidative stability of each molecular species of triglycerides appeared to decrease in proportion to the increase in the number of double bonds present in the acyl residues, and it was affected by degree of unsaturation of fatty acid when the total degree of unsaturation of triglyceride was the same. But the length of the saturated acyl chain had no influence on the stabilization of unsaturated fatty acid present in the same glyceride when prooxidants were added.

  • Phycomyces sp. has been isolated from Meju and its lipid composition was investigated. The lipid content was 18.2 % of dry weight and composed of 69.2 % neutral lipids. 24.3 % glycolipids and 6.5 % phospholipids. It was possible to identify the presence of carotene, hydrocarbons, esterified sterols, fatty acid esters, triglycerides. free fatty acids and free sterols in the neutral lipid fraction. Major components in the neutral lipid fraction were free fatty acids and triglycerides. Twelve molecular species of triglycerides and 8 molecular species of diglycerides were fractionated by argentation TLC. The major fatty acids in total lipids were stearic, linoleic, arachidic and linolenic acids, and those in the free fatty acid fraction were stearic, linoleic and arachidic acids.
