Inhibitory Effects of Ethanol Extract of Purple Solanum tuberosum L. on Obesity and Hyperlipidemia (자색 마령서 에탄올 추출물의 비만 및 고지혈증 억제효능)

  • This experimental study was performed to evaluate the effects of ehtanol extract of Purple Solanum tuberosum L(EPSTL) on obesity and hyperlipidemia in rats induced by high fat diet. Sprague-Dawley rats were divided into normal group, control group, EPSTL (100 mg/kg and 200 mg/kg) treated group. EPSTL was orally administered to the obese rats by high fat diet for 6 weeks. The effect of EPSTL on obesity and hyperlipidemia were examined in vitro and in vivo as follows : EPSTL effectively inhibited the differentiation of 3T3-L1 adipocytes in a concentration dependent manner. EPSTL significantly lowered body weight of rats fe high fat diet for 6 weeks from 4 weeks treatment compared with untreated control. EPSTL significantly reduced the weight of retroperitoneal fat only at 100 mg/kg and epididymal fat pad in rats fed high fat diet for 4 weeks at 100 and 200 mg/kg. EPSTL significantly reduced the level of triglyceride at 200 mg/kg, while EPSTL tended to lower the levels of total lipid and phospholipid in rats fed high fat diet for 4 weeks with no significance. EPSTL significantly attenuated the level of total cholesterol, LDL and atherosclerosis index (Al) as well as increased the level of HDL at 200 mg/kg. EPSTL significantly reduced the levels of total lipid and triglyceride of hepatic tissues in rats fed high fat diet for 4 weeks at 200 mg/kg. EPSTL significantly lowered the level of insulin, while it did not affect leptin compared with untreated control. Taken together, these results suggest that EPSTL can be used for the treatment or prevention of hyperlipidemia and obesity.

Effects of the purified extracts from Lycii Cortex Radicis and ginger on lipid statusand serum cytokine levels in rats fed high fat diet (지골피와 생강 정제 추출물이 고지방식이 흰쥐의 체내 지질과 혈청 Cytokine 수준에 미치는 영향)

  • The present study was to investigate the effects of Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) and ginger (Gin) on body lipid status and serum levels of cytokines. Sprague-Dawley (SD) male rats weighing $193.6{\pm}16.8g$ were divided into five groups, including one low fat (LF) and four high fat groups, i.e. HF-Control, HF-LCR, HF-Gin and HF-LCR + Gin groups. Diets for HF-LCR, HF-Gin and HF-LCR + Gin groups contained purified extracts having 0.2 g LCR tyramine, ginerol and 0.1 g tyramine plus 0.02 g gingerol per kg, respectively. Compared with those of the HF-Control total serum cholesterol level decreased, and HDL-cholesterol level increased in the HF-LCR group and serum triglyceride levels decreased in the three experimental groups fed the purified extracts. Liver cholesterol level was lower in the HF-LCR group than the HF-Control group, but triglyceride levels, which were increased by high fat diets were not changed by significantly by LCR or ginger extracts. Fecal lipid excretion was higher in the HF-LCR and HF-Gin groups, but cholesterol excretion was lower in the HF-Gin group than in the HF-Control group. The activities of liver cytosolic glucose-6-phosphate dehydrogenase and malic enzyme were lower in the HF-LCR + Gin group than in the HF-Control group. Serum adiponectin levels did not differ among the five groups, while leptin level was lower in the HF-Gin group and C-reactive protein levels were lower in the HF-Gin and the HF-LCR + Gin groups than in the HF-Control group. It is concluded that LCR can be utilized as an ingredient for lipid-lowering functional foods in the form of purified extract and addition of small amount of ginger extract would be useful for reducing one of the inflammatory cytokines to help prevent atherosclerosis.

Observational Study of ChondroT's Improvement of Blood Metabolites in High-fat Diet-induced Hyperlipidemia (고지방 식이 유도 고지혈증에 대한 ChondroT의 혈액 내 지질대사에 미치는 영향)

  • Objectives The objective of the study was to investigate effects of ChondroT by improvement of blood metabolites in high-fat diet (HFD)-induced hyperlipidemia rat model. Methods Sprague-Dawley rats were randomly assigned to intact, control, simvastatin, and CT100, CT200 and CT400 (each n=6). For observing cholesterol change, animals were first fed high fat diet for 5 weeks and then high fat diet and drugs for 3 weeks. At the end of the experiment, total cholesterol, triglyceride, high density lipoprotein-cholesterol (HDL-C) and low density lipoprotein-cholesterol (LDL-C) were analyzed by obtained blood collection. Further, amplified leptin, peroxisome proliferator activated receptor (PPAR) and adiponectin DNA were observed by reverse transcription polymerase chain reaction analysis. Results Observing the effect of ChondroT on the change of lipid metabolism in hyperlipidemia-induced rats, triglyceride and total cholesterol were significantly decreased in SV100 group, HDL-C was significantly increased in SV100, CT100 and CT200 groups, and LDL-C was significantly decreased in SV100, CT100, CT200 and CT400 groups, compared to the control group. Leptin level in hyperlipidemia-induced rats was significantly decreased in CT100 and CT200 groups, compared to the control group. The effect of ChondroT on adiponectin level in hyperlipidemia-induced rats was significantly increased in SV100, CT100 and CT200 groups. PPAR level in hyperlipidemia-induced rats was significantly decreased in SV100, CT200 and CT400 groups. Platelete activating factor level in hyperlipidemia-induced rats was significantly decreased in CT100 and CT200 groups. Conclusions Based on these results, it could be suggested that ChondroT has certain effects of improving blood metabolites in HFD-induced hyperlipidemia.

Effects of Dietary Intake of Agarum cribrosum Ethanol Extract on Lipid Level in Diet-induced Obese Rats (구멍쇠 미역 에탄올 추출물의 섭취가 비만유도 흰쥐의 지질농도에 미치는 영향)

  • This research was performed to investigate the effect of Agarum cribrosum ethanol extracts on the levels of lipids in the serum of rats fed a high-fat diet for 10 weeks. Experimental groups were divided into basal diet only (BDG), high fat diet control (HFDCG), high-fat diet and 5% Agarum cribrosum ethanol extract powder (HF5S), and high-fat diet and 10% Agarum cribrosum ethanol extract powder(HF10S) groups. The levels of hematological variables were not significantly different among the four groups. Compared with the control group's serum total cholesterol level of 89.14 mg/dL, the levels of the HF5S and HF10S groups were significantly lowered to 77.26 and 75.47 mg/dL, respectively. Compared with the control group's LDL-cholesterol leve of 27.41 mg/dL, the LDL-cholesterol levels of the HF5S and HF10S groups were significantly lowered to 20.64 and 20.17 mg/dL, respectively. Also, compared to the control group's serum triglyceride level of 98.36 mg/dL, the triglyceride levels of the HF5S and HF10S groups were significantly lowered to 87.94 and 87.39 mg/dL, respectively. These results indicate that dietary intake of Agarum cribrosum ethanol extracts might have beneficial effects on obesity by reducing body weight and improving blood lipid profile.

Effects of Aged Deodeok Extract on Serum Lipid Content in Rats Fed a High-Fat Diet (고지방식이를 섭취한 흰쥐에서 숙성더덕추출물이 혈청지질농도개선 효과에 미치는 영향)

  • This research was performed to investigate the effect of aged Deodeok (Codonopsis lanceolata) water extracts on the levels of lipids in the serum of rats fed a high-fat diet for 10 weeks. Experimental groups were divided into basal diet only (BDG), high fat diet control (HFDCG), high-fat diet and 10% aged deodeok extract powder (HF10S), and high-fat diet and 20% aged deodeok extract powder (HF20S) groups. The levels of hematological variables were not significantly different among the four groups. Compared with the control group's serum total cholesterol level of $339.38{\pm}4.06mg/dL$, the levels of the HF10S and HF20S groups were significantly lowered to $225.38{\pm}5.44$ and $215.02{\pm}4.77mg/dL$, respectively. Compared with the control group's LDL-cholesterol leve of $64.91{\pm}3.67mg/dL$, the LDL-cholesterol levels of the HF10S and HF20S groups were significantly lowered to $54.16{\pm}3.46$ and $46.14{\pm}1.79mg/dL$, respectively. Also, compared to the control group's serum triglyceride level of $103.07{\pm}13.2mg/dL$, the level of the HF20S group was significantly lowered to $48.25{\pm}11.52mg/dL$. These results suggested that dietary supplementation of aged deodeok extract does not have any adverse effect on the hematological variables, while improving the lipid content and reducing hepatic damage of the high-fat fed rats.

Association between Plasma Tocopherol Levels and Related Factors in Middle-Aged Korean Men (한국 성인 남자의 혈장 Tocopherol 수준과 관련 요인 분석)

  • Vitamin E in the body system plays an important role in preventing chronic diseases by decreasing the oxidative stress by free-radicals. However, there are not enough researches on analyzing the primary factors affecting vitamin E levels in the blood in Korean adults. Therefore, the purpose of this research was to examine blood tocopherol levels and the primary factors affecting the status. A complete lifestyle survey was performed on 314 Korean adult men and surveyed their smoking, drinking and exercising habits. The average plasma level of ${\alpha}-\;and\;{\gamma}-tocopherol$ showed similar mutual relations with plasma total cholesterol (TC), triglyceride (TG), or low density lipoprotein cholesterol (LDL-C) levels (p<0.001). Plasma ${\alpha}-tocopherol$ level of the subjects did not show any difference as smoking, drinking and exercising habits changed. However, ${\gamma}-tocopherol$ per TG showed much lower figure in smokers than non smokers (p < 0.05). Amongst diet factors, plasma ${\alpha}-tocopherol$ level showed negative correlations with Vitamin E intake, while ${\gamma}-tocopherol$ level showed positive correlations with Vitamin E intake. Erythrocyte superoxide dismutase (SOD) activity and plasma tocopherol showed negative correlations, and catalase activity and plasma ${\alpha}-tocopherol$ showed positive correlationship. The level of cell DNA damage of Iymphocyte and plasma ${\alpha}-\;or\;{\gamma}-tocopherol$ showed negative correlations. As a result of this research, the factors that affect Korean adult men's plasma ${\alpha}-tocopherol$ level are plasma TG, LDL-C and cell DNA damage in Iymphocyte, while the factors that affect ${\gamma}-tocopherol$ level are plasma TG, LDL-C and vitamin E intake based on multiple regression analysis. These findings implies that the level of different types of tocopherol depends on slightly different factors. A further research is needed on the factors involved in the differentiation of the types of tocopherol.

Effects of Corni Fructus Extract on the Progression of Diabetes and Renal Function in STZ-induced Diabetic Rats

  • This study investigated the effect of Corni Fructus(Cornus officinalis Sieb. et Zucc.) extract on hyperglycemia and renal function in streptozotocin-induced diabetic rats. Male Sprague-Dawley rats were divided into three groups including normal control(NC), diabetic control(DC), and diabetic treatment with Corni Fructus(DCF). Over a 4-week experimental period, Corni Fructus aqueous extract was administered orally at 500 mg/kg BW/day. The final fasting serum glucose, serum urea nitrogen, triglyceride, urinary total protein level, and relative weight of the left kidney in the DCF group were significantly lower than the DC group. Serum insulin level in the DCF group was higher than the DC group by 23%. The renal xanthine oxidase and superoxide dismutase activities in the DCF group were significantly lower than the DC group. The renal catalase activity in the DCF group was significantly higher than the DC group. In conclusion, these results indicated that Corni Fructus can reduce glucose level and prevent or retard the development of diabetic complication via its antioxidative effect and protecting against diabetic renal damage in streptozotocin-induced diabetic rats.

The Effect of Silk Amino Acid Supplementation on the Level of Blood Energy Substrates and Hormones during Prolonged Exercise

  • The silk amino acid supplementation is unknown to affect the release of several hormones related to energy production and metabolism during prolonged exercise. This study examined the effects of silk amino acid supplementation on the level of blood amino acid, energy substrates and hormones level during prolonged treadmill exercise in college taekwondo player. A prolonged treadmill test was carried out 60 min at 65% of maximal heart rate on 8 athletics. Blood samples were obtained form antecubital vein of subjects at rest bed 30 minute before test, after exercise and rest 1 hour. The subjects were supplemented silk amino acid (6,390 mg/day) fur 4 week. The silk amino acid supplementation did not produce significant changes on the levels of blood lactate, ammonia, amino acid, glucose, triglyceride, total cholesterol, HDL, LDL, seratonin and leptin at rest bed 30 minute before test, after exercise and rest 30 minute. The silk amino acid 4 week supplementation did not affect the levels of blood amino acid, energy substrates and hormones during prolonged treadmill exercise.

Effect of Royal Jelly on Therapy and Prevention of Streptozotocin Induced Diabetic Rats (로얄제리가 Streptozotocin 유발 당뇨 흰쥐의 치료 및 예방에 미치는 영향)

  • In order to investigate the preventive and therapeutic of royal jelly on diabetes, the levels of blood glucose and serum lipids as well as the number of blood cells were determined in streptozotocin(STZ) diabetic rats. Rats were divided into seven groups. The RJ group was administered royal jelly and the STZ group was treated with streptozotocin to induce diabetes. To determine the preventive effect, diabetes was induced after administration of royal jelly for 2 weeks in group RS1/RS2. In group SR1/SR2 diabetic rats were administered royal jelly for 2 weeks to investigate the therapeutic effect. After 3 weeks, the body weight was reduced in STZ and SR1 groups and food intake was increased in the STZ, RS1 and SR1 groups. The blood glucose level was similar to the control group in the RJ, RS1 and RS2 groups and there was no effect in the other groups. The total lipid and triglyceride level were lower in the SR1 group as compared to STZ, and the total cholesterol level was higher in the STZ group. The index of atherogenesis was lower in the RJ and SR1 groups compared to the normal group. The number of red blood cells and hemoglobin was higher in the RJ and SR1 groups and the number of white blood cells was higher in the RJ and SR2 groups.

The Status of Serum Lipids and Antioxidant Vitamins in Male Patients with Derebrovascular Disease (남성 뇌혈관질환자의 혈청지질과 항산화비타민 상태에 관한 연구)

  • This study was conducted to investigate serum lipids and antioxidant vitamins status of male patients with cerebrovascular disease. The study consisted of 16 hospital patients(57.8$\pm$13.7 years) at 3 General Hospitals in Taegu who suffer from cerebrovascular disease and 21 healthy adults(55.5$\pm$7.2 years). The results were as follows: The systolic blood pressure of the patients(150.0$\pm$13.7mmHg) was sig nificantly higher than healthy control(126.0$\pm$17.4mmHg). In patients the percentages of drinking alcohol(more than 5 times/week) and smoking were also higher. And the patients had more stress than control. Total calorie, protein, carbohydrate, vitamin B1 and niacin intakes were significantly lower in the patients. The energy nutrients consumption reached the recommended level in all subjects who were investigated. HDL cholesterol(24.99 vs 37.86mg/dl) and vitamin E(12.94 vs 20.88mg/L) level showed significantly lower, but triglyceride level(175.50 vs 117.69mg/dl), total cholesterol/HDL cho lesterol(6.22 vs 4.39) and atherogenic index(5.22 vs 3.39) were significantly higher in the patient group. Therefore, it might be expected that refraining from drinking alcohol and smoking, and that having proper exercise and certain other healthy living habits which can decrease stress in addition suitable consumption of antioxidant vitamins can prevent the occurrence of cerebrovascular disease.
