This study investigated the effect of lemon balm (Melissa officinalis) extract on improving blood triglycerides according to the extraction solvent using 3T3-L1 cells. Lemon balm was extracted with water (MOW100), 70% ethanol (MOE70), 50% ethanol (MOE50), and 30% ethanol (MOE30). To verify its efficacy on improving blood triglycerides, cell viability, lipid accumulation, triglyceride (TG) content, and expressions of protein kinase A (PKA), adipose triglyceride lipase (ATGL), hormone-sensitive lipase (HSL), perilipin, and CGI-58 proteins were evaluated. Cytotoxicity was not evident up to an extract concentration of 1 mg/mL. Lipid accumulation and TG content were decreased in a concentration-dependent manner compared to their levels in the control group. When the MOW100 extract was applied at a concentration of 0.2 mg/mL, an inhibitory effect was evident, with lipid accumulation inhibited by 21.3% and TG content reduced by 32.7%. PKA phosphorylation and ATGL HSL and CGI-58 levels were increased. The data indicate that lemon balm extract obtained using water is more efficacious than extracted with ethanol. The aqueous extract shows potential in triglyceride control through lipolysis and lowering triglyceride levels.
Triglycerides (TG) are implicated in the development of atherosclerosis through formation of foam cells and induction of macrophage cell death. In this study, we report that addition of exogenous TG induced cell death in phorbol 12-myristate 13-acetate-differentiated THP-1 human macrophages. TG treatment induced a dramatic decrease in interleukin-$1{\beta}$ (IL-$1{\beta}$) mRNA expression in a dose- and time-dependent manner. The expression of granulocyte macrophage colony-stimulating factor and platelet endothelial cell adhesion molecule remained unchanged. To identify signaling pathways involved in TG-induced downregulation of IL-$1{\beta}$, we added p38 MAPK, protein kinase C (PKC) or c-Raf1 specific inhibitors. We found that inhibition of p38 MAPK alleviated the TG-induced downregulation of IL-$1{\beta}$, whereas inhibition of PKC and c-Raf1 had no effect. This is the first report showing decreased IL-$1{\beta}$ expression during TG-induced cell death in a human macrophage line. Our results suggest that downregulation of IL-$1{\beta}$ expression by TG-treated macrophages may play a role during atherogenesis.
Hypertriglyceridemia is the main risk factor for atherosclerosis. It is reported that triglyceride (TG) induces macrophage cell death, and is involved in the formation of plaques and development of atherosclerosis. We previously reported that TG-induced cell death of macrophages is mediated via pannexin-1 activation, which increases the extracellular ATP and subsequent increase in potassium efflux, thereby activating the caspase-2/caspase-1/apoptotic caspases, including the caspase-8 pathway. Contrarily, some studies have reported that caspase-8 is an upstream molecule of caspase-1 and caspase-2 in several cellular processes. Therefore, this study was undertaken to investigate whether caspase-8 influences its upstream molecules in TG-stimulated macrophage cell death. We first confirmed that caspase-8 induces caspase-3 activation and poly ADP-ribose polymerase (PARP) cleavage in TG-treated macrophages. Next, we determined that the inhibition of caspase-8 results in reduced caspase-1 and -2 activity, which are upstream molecules of caspase-8 in TG-induced cell death of macrophages. We also found that ATP treatment restores the caspase-8 inhibitor-induced caspase-2 activity, thereby implying that caspase-8 affects the upstream molecules responsible for increasing the extracellular ATP levels in TG-induced macrophage cell death. Taken together, these findings indicate that caspase-8 potentiates the TG-induced macrophage cell death by activating its upstream molecules.
Optimal winterizing condition of com oil was cooling temperature of bleached oil from $105^{\circ}C$ to $40^{\circ}C$. Then, filtering it after keeping $1{\sim}2^{\circ}C$ by lowering the temperature gradually with treating perlite of 0.3%(w/w) amount about bleached oil and stirring. We could measure that triglyceride(TG) that extracted from lipid components from spent perlite(SP) obtained through filtration after winterizing by SACC method is major causing materials of clouding in com oil. The result of separating TG fraction by agentation TLC was that it classified into 4 kinds -U3, SU2, S2U, S3 type and the most were U3 type. From this, it's easy to identify cause of clouding in com oil is TG fraction and most of them form wax materials that can observed. The results were they kept clear appearance at $0^{\circ}C$ generally during 39.6 to 96.5 hours, especially the result of A sample that had the lowest temperature condition while they have some difference by condition of treating temperature.
Secrum lipid and vitamin E levels were determined and smoking, alcohol drinking and exercise habits were asked in 357 healthy male subjects aged 49.4$\pm$6.7 years in Taegu. Average serum levels of total cholesterol, HDL-cholesterol and triglyceride(TG) were 189$\pm$43mg/이, 42$\pm$13mg/dl and 136$\pm$73mg/이, respectively. Serum level of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS) of the subjects was 2.01$\pm$0.73MDA nmoles/ml and that of $\alpha$-tocopherol was 9.53$\pm$3.14ug/ml. The correlation coefficients between $\alpha$-tocopherol and serum lipids were 0.3631 for triglyceride, 0.2993 for cholesterol, and 0.3025 for total lipid. Heavy smokers who smoked more than 20 cigarettes a day had higher levels of TG and TBARS than those who smoke less. Alcohol drinkers had higher levels of TG and TBARS than nondrinkers. Vitamin E level(per ml serum)was significantly higher in the heavy smokers and drinkers, which was reflected by the higher level of serum lipid. When the level of $\alpha$-tocopherol was expressed as $\alpha$-tocopherol/triglyceride, it was negatively correlated with serum TBARS level and was lower in the heavy smokers than in the moderate smokers. It is concluded that vitamin E level as $\alpha$-tocopherol/triglyceride would be better compared to $\alpha$-tocopherol/total cholesterol or $\alpha$-tocopherol/total lipid for the evaluation of vitamin E status in Korean men.
Purpose: The triglyceride-to-high-density lipoprotein-cholesterol (TG/HDL-C) ratio is one of the main predictive indices for cardiovascular disease. This study was examined the relationship between TG/HDL-C ratio and metabolic syndrome (MetS) in male office workers. Methods: Secondary analysis was conducted to determine the risk between the TG/HDL-C ratio and MetS in male office workers. A total of 765 people underwent the 'regular workplace health checkups in 2014'. Among the subjects who were male and responded to the questionnaire and health lifestyle survey, 470 (61.4%) excluding those with missing and/or abnormal values were analyzed. The association between MetS, MetS components, and the TG/HDL-C ratio was examined by a Chi-square test, One-way ANOVA, Turkey post-hoc test and Logistic regression analysis. Results: The number of males with MetS was 70 (14.9%) and the number of MetS components increased with increasing TG/HDL-C ratio (p<.001). Logistic regression analysis with an adjustment for potential confounders revealed a 31.8 times higher odds ratio of the Quartile4 group for MetS than that of the Quartile1 group (p<.001). Conclusion: These results show that the likelihood of MetS, particularly the risk of MetS in the Quartile4, increases with increasing TG/HDL-C ratio.
Triglyceride (TG) is known to be associated with inflammatory disease including atherosclerosis. In a variety of atherosclerosis models, T lymphocytes are localized in the earliest lesions of atherosclerosis. T cell associated cytokines such as $TNF-{\alpha}$ and $IFN-{\gamma}$ have pre-dominant inflammatory effects in chronic vascular diseases. In our previous study, we found that the expression of $TNF-{\alpha}$ and its receptor, $TNF-{\alpha}R$ was increased when Jurkat T lymphocyte cell lines were exposed to TGs. Therefore, experiments were conducted to determine which cell signaling pathway are involved in the increase of $TNF-{\alpha}$ and $TNF-{\alpha}R$ expression by TGs. To identify signal transduction pathways involved in TG-induced upregulation of $TNF-{\alpha}$, we treated TG-exposed Jurkat T cells with specific inhibitors for MEK1, PI3K, $NF-{\kappa}B$ and PKC. We found that inhibition of the MEK1 pathway blocked TG-induced upregulation of $TNF-{\alpha}$. However, the expression level of $TNF-{\alpha}R$ did not change with any signal transduction inhibitor. Based on this observation, we suggest that increase of exogenous TG induces increase of $TNF-{\alpha}$ expression through MEK1 pathway in Jurkat T cells. In addition, it was confirmed that the increase of $TNF-{\alpha}$ and $TNF-{\alpha}R$ expression by TGs occurs via different pathways.
A study was conducted to investigate the effects of deodorizing temperature on physicochemical characteristics, fatty acid and triglyceride (TG) composition, and to estabilish the optimal deodorizing conditions in corn oil. Acid value (AV) of $240^{\circ}C$ treating group was showed 0.065 as the lowest level, the AV range was $0.08{\sim}0.09$ at $250{\sim}270^{\circ}C$. Lovibond total color was 24 as the lowest level, peroxide values (POV) were zero at $240{\sim}270^{\circ}C$ range. But POV was showed 0.42 meg/kg oil at $235^{\circ}C$, according to the optimal deodorizing temperature for the complete removal of peroxides was required more than $240^{\circ}C$. On the other hand, inclose of deodorizing temperature was conducted to the increase of saturated fatty acid content as myristic, palmitic, stearic and arachidic acid etc. Change of TG composition at treated group more than $250^{\circ}C$ was reliable, $S_3$ type TG was appeared. As a result, high temperature deodorizing was induced the undesirable influence at the physicochemical properties in deodorized corn oil.

The present study explored the short-term effects of dietary conjugated-linoleic acid (CLA) on liver lipid metabolism in starved/refed Otsuka Long Evans Tokushima Fatty (OLETF) rats. Male OLETF rats (12 weeks old) were starved for 24 hours, then refed for 48 hours with either a CLA diet [7.5% CLA and 7.5% Safflower oil (SAF)] or a SAF control diet (15% SAF). The results demonstrated a 30% reduction of hepatic triglyceride (TG) concentration in the CLA group when compared to the control group. Liver cholesterol concentration was also 26% lower in the CLA fed rats. The activity of mitochondrial carnitine palmitoyltransferase, the rate-limiting enzyme of fatty acid oxidation, was moderately elevated by 1.2-fold in the livers of the CLA group when compared to the control. In contrast, phosphatidate phosphohydrolase, the rate-limiting enzyme for TG synthesis, was found to be 20% lower in the livers of the CLA-fed rats. Therefore, dietary CLA evidently lowers liver lipid concentrations through a reduced TG synthesis and enhanced fatty acid oxidation in starved/refed OLETF rats.
Crude saponin fractions were isolated using non-ionic resin chromatography from Ginseng Radix Alba (PG) and Gynostemmae Herba+ (GP). These saponin fractions were orally administered to strep- tozotocin (STZ)-induced diabetic rats for 2 weeks and to high-fat diet-induced obese rats for 4 weeks. Treatment with either PG saponin or GP saponin at a dose of 1 mg/kg significantly decreased the plasma glucose level to that of glibenclamide treated or normal groups. The increased plasma triglyceride (TG) level in diabetic rats was decreased by 50% with PG or GP saponin treatment. Combined administration of PG and GP saponins with different ratios (total dose of 1 mg/kg) also had the similar effects on the blood glucose and TG levels with that of PG or GP alone. Treatments with GP (1 mg/kg) or GP (0.5 mg/kg) and PG (0.5 mg/kg) together significantly suppressed the rise in TG levels induced by high-fat diet whereas slightly suppressed the rise in the total cholesterol level. The body weight gain was also decreased both in the two saponin treated groups. These results demonstrate that either alone or mixture of PG and GP have similar degree of effects on hyperglycemia and hyperlipidemia.
