Nucleotide sequence analysis of the 5S ribosomal RNA gene of the mushroom tricholoma matsutake

  • From a cluster of structural rRNA genes which has previsouly been cloned (Hwang and Kim, in submission; J. Microbiol. Biotechnol.), a 1.0-kb Eco RI fragment of DNA which shows significant homology to the 25S and rRNA s of Tricholoma matsutake was used for sequence analysis. Nucleotide sequence was bidirectionally determined using delection series of the DNA fragment. Comparing the resultant 1016-base sequence with sequences in the database, both the 3'end of 25S-rRNA gene and 5S rRNA gene were searched. The 5S rRNA gene is 118-bp in length and is located 158-bp downstream of 3'end of the 25S rRNA gene. IGSI and IGS2 (partial) sequences are also contained in the fragment. Multiple alignment of the 5S rRNA sequences was carried out with 5S rRNA sequences from some members of the subdivision Basidiomycotina obtained from the database. Polygenetic analysis with distance matrix established by Kimura's 2-parameter method and phylogenetic tree by UPGMA method proposed that T. matsutake is closely related to efibulobasidium allbescens. Secondary structure of 5S rRNA was also hypothesized to show similar topology with its generally accepted eukaryotic counterpart.

Spore Dispersion of Tricholoma matsutake at a Pinus densiflora Stand in Korea

  • The spore of Tricholoma matsutake is considered to be the starting point of the mushroom growth cycle, but the mechanism of mycelial development from the spore stage is not yet clarified. In this study, we tried to measure how far the spores of T. matsutake disperse from a fruiting body located at a Pinus densiflora stand in Korea. We established 16 slide glasses coated with glycerin near a fruiting body in four directions separated by four different distance intervals within a mushroom productive stand after removing all other fruiting bodies from three plots. The number of dispersed spores increased with time from the first day (475 $spores/cm^2$) to the fourth day (836 $spores/cm^2$) after the pileus opened. The number of spores dispersed downward was about 1.5 times greater than that dispersed toward the ride. The number of dispersed spores decreased exponentially as the distance from each fruiting body increased. More than 95% of the spores dropped within a meter from the fruiting body, with 75% dropping within 0.5 m. Even so, the number of spores dispersed over 5 m from the fruiting body was more than 50 million when considering the total number of spores produced by a fruiting body is about 5 billion.

Cloning, characterization and expression of glucoamylase gene from ectomycorrhizal basidomycete, Tricholoma matsutake

  • In order to confirm the presence of putative glucoamylase gene in Tricholoma matsutake genome, the genomic DNA was prepared from T. matsutake NBRC30773 strain and was used as template to clone the glucoamylases gene (TmGlu1). We obtained the nucleotide sequence of TmGlu1 and its franking region. The coding region (from ATG to stop codon) is 2,186 bp. The locations of exons and introns were determined from the nucleotide sequences of 3'- and 5'-RACE PCR and RT-PCR products. On the other hand, to investigate the relationship between composition of medium and glucoamylase expression, we checked the expression level of glucoamylase gene by realtime reverse transcription PCR and measurement of glucoamylase enzyme activity. It was found that enzyme activity of glucoamylase was very low in different medium. Expression of glucoamylases gene appeared to not be affected by different carbon source.

송이[Tricholoma matsutake]에서 분리된 균사 배양액의 Tyrosinase 억제효과 (Inhibition Effect of the Extracts of Trichloma matsutake Mycelia on Tyrosinase Activity)

  • 송이의 공시균(KFR1432)과 실험실에서 자체 분리한 균(YxT, WxT, $\textrm{NK}_3\textrm{T}$)의 생장속도의 차이와 FE SEM으로 확인한 미세구조의 차이로 공시균과 자체 분리균이 서로 다른 균임을 확인할 수 있었다. Tyrosinase 활성 억제 실험을 위해 각각 균사체의 열수 추출물, 에탄올 추출물, 배양액을 hexane, chloroform, ethyl acetate로 분획하였다. 그 결과 모든 균의 ethyl acetate 분획층과 수층에서 tyrosinase 활성을 억제하는 것으로 나타났으며, KFR1432의 ethyl acetate 분획층과 YxT의 수층에서 억제율이 100%를 넘어 큰 저해 활성을 보였다. 또한 항산화제 처리 결과 ascorbic acid와 glutathion은 생장을 다소 증가시키거나 별 영향을 주지 못한 반면, tocopherol 의 경우 농도가 높아질수록 생장을 억제하는 것으로 나타났다. Tyrosinase 활성 억제에 있어서는 모든 항산화제 처리구에서 무처리구에 비해 높은 억제율을 보였다.

Analysis of Bacterial Diversity and Communities Associated with Tricholoma matsutake Fruiting Bodies by Barcoded Pyrosequencing in Sichuan Province, Southwest China

  • Endophytes play an important role in the growth and development of the host. However, the study of endophytes is mostly focused on plants, and reports on bacteria associated with fungi are relatively rare. We studied the bacteria associated with fruiting bodies of Tricholoma matsutake picked from seven main T. matsutake-producing areas in Sichuan, China, by barcoded pyrosequencing. About 8,272 reads were obtained per sample, representing 40 phyla, 103 classes, and 495 genera of bacteria and archaea, and 361-797 operational taxonomic units were observed at a 97% similarity level. The bacterial community was always both more abundant and more diverse than the archaeal community. UniFrac analysis showed there were some difference of bacterial communities among the samples sites. Three bacterial phyla, Proteobacteria, Bacteroidetes, and Firmicutes, were dominant in all samples. Correlation analysis showed there was a significant correlation between some soil properties and bacterial community associated with T. matsutake. This study demonstrated that the bacteria associated with T. matsutake fruiting bodies were diversified. Among these bacteria, we may find some strains that can promote the growth of T. matsutake.

Metagenomic Analysis of Fungal Communities Inhabiting the Fairy Ring Zone of Tricholoma matsutake

  • Tricholoma matsutake, an ectomycorrhiza that has mutual relationships with the rootlet of Pinus denisflora, forms a fruiting body that serves as a valuable food in Asia. However, the artificial culture of this fungus has not been successful. Soil fungi, including T. matsutake, coexist with many other microorganisms and plants; therefore, complex microbial communities have an influence on the fruiting body formation of T. matsutake. Here, we report on the structures of fungal communities associated with the fairy ring of T. matsutake through the pyrosequencing method. Soil samples were collected inside the fairy ring zone, in the fairy ring zone, and outside the fairy ring zone. A total of 37,125 sequencing reads were obtained and 728 to 1,962 operational taxonomic units (OTUs) were observed in the sampling zones. The fairy ring zone had the lowest OTUs and the lowest fungal diversity of all sampling zones. The number of OTUs and fungal taxa inside and outside the fairy ring zone was, respectively, about 2 times and 1.5 times higher than the fairy ring. Taxonomic analysis showed that each sampling zone has different fungal communities. In particular, out of 209 genera total, 6 genera in the fairy ring zone, such as Hemimycena, were uniquely present and 31 genera, such as Mycena, Boletopsis, and Repetophragma, were specifically absent. The results of metagenomic analysis based on the pyrosequencing indicate a decrease of fungal communities in the fairy ring zone and changes of fungal communities depending on the fairy ring growth of T. matsutake.

Ergosterol and Water Changes in Tricholoma matsutake Soil Colony during the Mushroom Fruiting Season

  • The purpose of this study is to understand spatio-temporal changes of active fungal biomass and water in Tricholoma matsutake soil colonies during the mushroom fruiting season. The active fungal biomass was estimated by analyzing ergosterol content at four different points within four replicated locations in a single circular T. matsutake colony at Ssanggok valley in the Sogri Mt. National Park in Korea during 2003 to 2005. The four points were the ahead of the colony, the front edge of the colony and 20 cm and 40 cm back from the front edge of the colony. Ergosterol content was 0.0 to 0.7 ${\mu}g$ per gram dried soil at the ahead, 2.5 to 4.8 ${\mu}g$ at the front edge, 0.5 to 1.8 ${\mu}g$ at the 20 cm back and 0.3 to 0.8 ${\mu}g$ at the 40 cm back. The ergosterol content was very high at the front edge where the T. matsutake hyphae were most active. However, ergosterol content did not significantly change during the fruiting season, September to October. Soil water contents were lower at the front edge and 20 cm back from the front edge of the colony than at the ahead and 40 cm back during the fruiting season. Soil water content ranged from 12 to 19% at the ahead, 10 to 11% at the edge, 9 to 11% at the 20 cm back and 11 to 15% at the 40 cm back. Our results suggest that the active front edge of the T. matsutake soil colony could be managed in terms of water relation and T. matsutake ectomycorrhizal root development.

송이 감염묘를 이용한 송이 발생 및 발생환경 분석 (Analysis of Environment and Production of Tricholoma matsutake in Matsutake-infected Pine Trees)

  • 송이는 매우 고가의 버섯이기 때문에 세계 여러 나라에서 인공재배를 시도하고 있다. 지금까지 야외 조건에서 인공적으로 송이 발생을 성공시킨 예는 송이 감염묘를 이용한 방법뿐이다. 국립산림과학원은 2000년부터 송이 감염묘 방법을 다시 연구하여 2010년과 2017년에 각각 송이 발생 재현에 성공하였다. 이 결과는 송이 감염묘를 이용하여 송이가 인공재배 가능하다는 것을 입증한 것이며, 세계 최초로 송이 인공재배가 성공한 사례가 된다. 송이 감염묘 이식 후 6년 6개월만인 2010년 10월에 송이가 처음 발생하였다. 그리고 또 다시 송이 감염묘 이식 후 13년 5개월 또는 15년 5개월이 경과된 시점인 2017년 9월에 송이 5개가 재 발생하였다. 송이 감염묘 소나무와 발생한 송이 사이의 거리는 12 cm (6.6년), 90~115 cm (13.5년), 95 cm (15.5년)으로 나타났다. 송이 버섯 발생은 지온이 $19^{\circ}C$이하로 떨어진 이후 13~16일 사이에서 관찰되었다. 결론적으로 송이 감염묘법은 송이를 인공재배 할 수 있는 현재까지의 유일한 방법으로, 앞으로도 송이 자실체 발생에 영향을 끼치는 환경 인자들을 지속적으로 연구할 필요가 있다.

송이버섯과 동충하초 균사체 혼합배양 추출물의 투여가 Streptozotocin으로 유발한 당뇨 쥐의 혈당에 미치는 영향 (Effects of Extracts from Mixed Culture with Tricholoma Matsutake Mycelium and Cordyceps Militaris Mycelium on Blood Glucose in Streptozotocin-Induced Diabetic Rats)

  • This study was performed to investigate the influence of extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium on hypoglycemia in streptozotocin-induced diabetic rats. Experimental animals were divided into 7 groups : normal control group(NC), diabetes control group (DC), positive control group(PC), non-fermented OCM(oriental medicine & cereal medium) extracts group (UM), crude polysaccharide of non-fermented OCM extracts group (UME), fermented OCM extracts group (UF), crude polysaccharide of fermented OCM extracts group (UFE). NC, DC groups were orally administered saline, PC group was orally administered acarbose. UM, UME, UF, UFE groups were orally administered each extract once a day for 14days. Blood glucose level was lower in the all administering OCM extract groups (UM, UME, UF, UFE) than in the diabetes group(p<0.05), and specially UF, UFE groups were similar to tendency of PC group. ALT, ALP activity in OCM groups were not significantly lowered than PC group(p<0.05). AST activity was not different with PC group. The results of this study show that extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium may have a beneficial effect on the hypoglycemia.
