Bioleaching is the process in which insoluble metal sulfide is oxidized by specialized iron- and/or sulfur-oxidizing lithotrophic bacteria in acidic, metal-rich environments. Most of these processes are carried out by the genus Thiobacillus. Three novel Thiobacillus strains (Thiobacillus thiooxidans AZ11, Thiobacillus thiooxidans MET, and thiobacillus thiooxidans TAS) associated with bioleaching have been isolated from soil and sludge (Korean patent No. 1999-0073060 for T. thiooxidans AZ11, Korean patent No. 1999-0005798 for T. thiooxidans MET, and Korean patent No. 1999-0073059 for T. thiooxidans TAS). A partial sequence of 16S ribosomal RNA gene (16S rDNA) and the entire sequence of 16S/23S intergenic spacer region (ISR) were determined in the three above novel strains and in Thiobacillus ferrooxidans ATCC19859 as a reference strain. When phylogenetic analysis was performed based on G+C contents and sequence alignments, T. ferroxidans ATCC19859 was found to be closely related to previously registered T. ferrooxidans strains in a monophyletic manner, while the three novel T. thiooxidans strains were classified in a paraphyletic manner. Close examination on the base composition of 16S/23S ISR revealed that the 5\` part (nucleotide residues 21-200) was specific for the genus Thiobacillus. On the other end, the 3\` part (nucleotide residues 201-520) showed specificity in T. ferrooxidans strains, but not in T. thiooxidans strains. These results suggest that the proximal and distal halves of 16S/23S could be used as a genetic marker for the identification of the genus Thiobacillus and the species T. ferrooxidans, respectively.

This paper presents a method for point-based 3D building reconstruction using Lidar data and digital map. The proposed method consists of three processes: extraction of building roof points, identification of roof types, and 3D building reconstruction. After extracting points inside the polygon of building, the ground surface, wall and tree points among the extracted points are removed through the filtering process. The filtered points are then fitted into the flat plane using ODR(Orthogonal Distance Regression) in the first place. If the fitting error is within the predefined threshold, the surface is classified as a flat roof. Otherwise, the surface is fitted and classified into a gable or arch roof through RMSE analysis. Experimental results showed that the proposed method classified successfully three different types of roof and that the fusion of LIDAR data and digital map could be a feasible method of modeling 3D building reconstruction.
In this study, samples were collected from the leaves and stems of healthy wild Pistachio trees (Pistacia atlantica L.) from various locations of Baneh and Marivan regions, Iran. In total, 61 endophytic bacteria were isolated and grouped according to phenotypic properties. Ten selected isolates from each group were further identified by partial sequencing of the 16S rRNA gene. Based on the results, isolates were identified as bacteria belonging to Pseudomonas, Stenotrophomonas, Bacillus, Pantoea and Serratia genus. The ability of these isolates was evaluated to phytohormone production such as auxin and gibberellin, siderophore production, phosphate solubilization, atmospheric nitrogen fixation, protease and hydrogen cyanide production. All strains were able to produce the plant growth hormone auxin and gibberellin in different amounts. The majority of strains were able to solubilize phosphate. The results of atmospheric nitrogen fixation ability, protease and siderophore production were varied among strains. Only Ba66 could produce a low amount of hydrogen cyanide. The results of biocontrol assay showed that Pb78 and Sp15 strains had the highest and lowest inhibition effects on bacterial plant pathogens, Pseudomonas syringae pv. syringae Pss20 and Pseudomonas tolaasii Pt18 under in vitro condition. Pb3, Pb24 and Pb71 strains significantly promote root formation on carrot slices. To our knowledge this is the first report of the isolation of endophytic bacterial strains belonging to Pantoea, Bacillus, Pseudomonas, Serratia and Stenotrophomonas genus from wild pistachio trees with plant growth promoting potential and biocontrol activity.
In order to isolate antibiotic producing microorganisms, several actinomycetes were isolated from soil samples. The aerial hyphae of Y-88 strain were gray in color with tree types. Under the microscopic examination, the Y-88 isolate formed a spiral aerial spore mass with a smooth surface and a rectiflexibilis type of spore chain. Y-88 utilized glucose, fructose, arabinose, and sucrose, but not rhamnose, raffinose, mannitol, or inositol. In addition, Y-88 produced melanin on the tyrosine agar and the strain could utilize L-valine, L-phenylalanine, and L-hydroxyproline. Based on these results and the cultural and physiological characteristics described in the Bergey's Manual, the Actinomycetes, Y-88, was identificated as a Streptomyces species. The optimum medium conditions for this antibiotic producing Streptomyces sp. Y-88 was 1.6% soluble starch, 0.6% glucose, 0.6% beef extract, 0.01% $K_2HPO_4$, 0.01% $MgSO_4$$7H_2O$, and 0.01% $ZnSO_4$$7H_2O$ at an initial pH of 4.0, and 25$^{\circ}C$.
The melanocortin 1 receptor (MC1R) gene is related to the plumage color variations in chicken. Initially, the MC1R gene from 30 individuals was sequenced and nine polymorphisms were obtained. Of these, three and six single nucleotide polymorphisms (SNPs) were confirmed as synonymous and nonsynonymous mutations, respectively. Among these, three selected SNPs were genotyped using the restriction fragment length polymorphism (RFLP) method in 150 individuals from five chicken breeds, which identified the plumage color responding alleles. The neighbor-joining phylogenetic tree using MC1R gene sequences indicated three well-differentiated different plumage pigmentations (eumelanin, pheomelanin and albino). Also, the genotype analyses indicated that the TT, AA and GG genotypes corresponded to the eumelanin, pheomelanin and albino plumage pigmentations at nucleotide positions 69, 376 and 427, respectively. In contrast, high allele frequencies with T, A and G alleles corresponded to black, red/yellow and white plumage color in 69, 376 and 427 nucleotide positions, respectively. Also, amino acids changes at position Asn23Asn, Val126Ile and Thr143Ala were observed in melanin synthesis with identified possible alleles, respectively. In addition, high haplotype frequencies in TGA, CGG and CAA haplotypes were well discriminated based on the plumage pigmentation in chicken breeds. The results obtained in this study can be used for designing proper breeding and conservation strategies for the Korean native chicken breeds, as well as for the developing breed identification markers in chicken.
An isolate from holothurians was identified as an eicosapentaenoic acid (EPA)-producing bacterium KMG427, which is characterized by EPA synthesis efficiency, by thin layer and gas chromatographic analyses. The EPA production was maximized to more than 10% of the total fatty acids by incubation at $4^{\circ}C$ after cell proliferation at $20^{\circ}C$. The isolated bacterium was categorized as Gram-negative, rod-shaped, aerobic, and motile with a single polar flagellum. According to phylogenetic analysis based on morphological and physiological specificities as an EPA-producing bacterium, the isolate KMG427 was found to belong to the genus Shewanella. The 16S rDNA of KMG427 was revealed to have 100% of sequence identity to that of S. hanedai CIP $103207^T$. Therefore, the isolate might be classified and identified as Shewanella sp. KMG427.

Objectives : Araliae Continentalis Radix and Angelicae Pubescentis Radix have been used as the same medicinal name Korean and Chinese traditional medicines, respectively. The authentic Araliae Continentalis Radix is described only the root of Aralia continentalis in the Korean Pharmarcopoeia. However, the dried root of Angelica biserrata, Levisticum officinale, or Heracleum moellendorffii also has been distributed adulterants of Araliae Continentalis Radix. To develop a reliable method for identifying Araliae Continentalis Radix from adulterants, we carried out the analyses of universal DNA barcode sequences.Methods : Four plants species were collected from different habitate and nucleotide sequences of matK and rbcL were analyzed. The species-specific sequences and phylogenetic relationship were estimated using entire sequences of two DNA barcodes, respectively.Results : In comparative analysis of matK sequences, we were identified 104 positions of marker nucleotide for Ar. continentalis, 3 for An. biserrata, 4 for L. officinale and 8 for H. moellendorffii enough to distinguish individual species, respectively. Furthermore, we obtained marker nucleotides in rbcL at 42 positions for Ar. continentalis, 5 for An. biserrata and 2 for H. moellendorffii, but not for L. officinale. The phylogenetic tree of matK and rbcL were showed that all samples were clustered into four groups constituting homogeneous clades within the species.Conclusions : We confirmed that species-specific marker nucleotides of matK sequence provides distinct genetic information enough to identify four species. Therefore, we suggest that matK gene is useful DNA barcode for discriminating authentic Araliae Continentalis Radix from inauthentic adulterants.
Studies were conducted on manufacturing techniques by applying microscopy and conservation treatments on the annals of Joseon dynasty storage box at the National Museum of Korea. The results revealed that lime tree wood(Tilla spp.) was used to make the annals of Joseon dynasty storage box. Lacquering techniques were used to coat the box with a layer of lacquer and bone ash and then cover it with traditional Korean paper. After being covered with traditional Korean paper, more layers were applied in the following sequence: mud ashes, black lacquer, pure lacquer, and black lacquer. Before conservation treatments, some components and lacquer layers were missing in addition wood joint were loose. Therefore, conservation and restoration should be conducted based on the identified wood and by observing the shape of the remaining components.
From 2008 to 2017, Korean azalea (Rhododendron yedoense f. poukhanense) showing angular, necrotic leaf spots were found in Jeju and Hongcheon, Korea. The lesions occurred frequently, detracting from the beauty of the glossy green leaves of the plant and causing premature defoliation. Therefore, to identify the fungus associated with the lesions, morphological characterization and molecular phylogenetic analysis of actin (Act), translation elongation factor 1-alpha (EF), internal transcribed spacer (ITS), 28S nrDNA (LSU), and RNA polymerase II encoding the second largest subunit (RPB2) of the two representative isolates were performed. The phylogenetic tree inferred from the neighbor-joining method showed the isolates clustering in the Sphaerulina azaleae group. Therefore, the fungus associated with the angular leaf spots on the Korean azalea was identified as Sphaerulina azaleae.
Network RTK is a precise positioning technique using carrier phase correction data from reference stations within the network, and is constantly being researched for improved performance. However, the study for the system accuracy has been performed but system integrity research has not been done as much as system accuracy, because network RTK has been mainly used on surveying for static or kinematic positioning. In this paper, adequate monitoring system for network RTK is designed as basis research for integrity monitoring on network RTK. To this, fault tree on network RTK is analyzed, and a countermeasure is prepared to detect and identify the each fault items. Based these algorithms, monitoring system to use on central processing facility is designed for network RTK service.
