A Computational Model of Cytosolic and Mitochondrial [$Ca^{2+}$] in Paced Rat Ventricular Myocytes

  • We carried out a series of experiment demonstrating the role of mitochondria in the cytosolic and mitochondrial $Ca^{2+}$ transients and compared the results with those from computer simulation. In rat ventricular myocytes, increasing the rate of stimulation (1~3 Hz) made both the diastolic and systolic [$Ca^{2+}]$ bigger in mitochondria as well as in cytosol. As L-type $Ca^{2+}$ channel has key influence on the amplitude of $Ca^{2+}$ -induced $Ca^{2+}$ release, the relation between stimulus frequency and the amplitude of $Ca^{2+}$ transients was examined under the low density (1/10 of control) of L-type $Ca^{2+}$ channel in model simulation, where the relation was reversed. In experiment, block of $Ca^{2+}$ uniporter on mitochondrial inner membrane significantly reduced the amplitude of mitochondrial $Ca^{2+}$ transients, while it failed to affect the cytosolic $Ca^{2+}$ transients. In computer simulation, the amplitude of cytosolic $Ca^{2+}$ transients was not affected by removal of $Ca^{2+}$ uniporter. The application of carbonyl cyanide 4-(trifluoromethoxy) phenylhydrazone (FCCP) known as a protonophore on mitochondrial membrane to rat ventricular myocytes gradually increased the diastolic [$Ca^{2+}$] in cytosol and eventually abolished the $Ca^{2+}$ transients, which was similarly reproduced in computer simulation. The model study suggests that the relative contribution of L-type $Ca^{2+}$ channel to total transsarcolemmal $Ca^{2+}$ flux could determine whether the cytosolic $Ca^{2+}$ transients become bigger or smaller with higher stimulus frequency. The present study also suggests that cytosolic $Ca^{2+}$ affects mitochondrial $Ca^{2+}$ in a beat-to-beat manner, however, removal of $Ca^{2+}$ influx mechanism into mitochondria does not affect the amplitude of cytosolic $Ca^{2+}$ transients.

New Modeling of Switching Devices Considering Power Loss in Electromagnetic Transients Program Simulation

  • This paper presents the modeling of insulated-gate bipolar transistor (IGBT) in electromagnetic transients program (EMTP) simulation for the reliable calculation of switching and conduction losses. The conventional approach considering the physical property of switching devices requires many attribute parameters and large computation efforts. In contrast, the proposed method uses the curve fitting and interpolation techniques based on typical switching waveforms and a user-defined component with variable resistances to capture the dynamic characteristics of IGBTs. Therefore, the simulation time can be efficiently reduced without losing the accuracy while avoiding the extremely small time step, which is required in simulation by the conventional method. The EMTP based simulation includes turn-on and turn-off transients of IGBT, saturation state, forward voltage of free-wheeling diode, and reverse recovery characteristics, etc. The effectiveness of proposed modeling for the EMTP simulation is verified by the comparison with experimental results obtained from practical implementation in hardware.

  • As a high-speed train enters a tunnel, a compression wave is generated ahead of it due to the piston action of train. The compression waves propagate along the tunnel and reflect at the exit of tunnel. A complex wave phenomenon appears in the tunnel, because of the successive reflections of the pressure waves at the exit and entrance of tunnel. The pressure waves give rise to large pressure transients which impose the fluctuating loads on the running train. It is highly needed that the pressure transients should be predicted to design the train body and to improve the comfortableness of the passengers in the train. In the present study, the pressure transients were calculated numerically for a wide range of train speed and compared with the previous tunnel tests. The calculation results agreed with ones of the tunnel tests, and the mechanism of pressure transients was made clear.

  • In this study, a mathematical model for analyzing the shifting transients of the passenger car with an automatic transmission is proposed. The proposed model comprises a power transmission system and a vehicle system, which are coupled. In order to extract the modeling parameters, on-road car test is carried out. The model is composed of a detailed powertrain, an engine/AT housing, a simplified suspension system, tires and a vehicle body model. On the test, the vehicle accelerations and pitch ratio are measured by using accelerometers and a gyro sensor. The speeds, the brake signal, and the throttle position are taken from sensors which already exist in the vehicle. Considering natural ftequencies, which is calculated from the measured accelerations, and the characteristic equation, vehicle model parameters are identified. Dynamic behaviors during upshift or downshift are simulated using the proposed vehicle model. By comparing and analyzing the simulation result and on-road car test data, the vibration of the Engine/AT housing influences the shifting transients. The effect of model parameters are also studied. Among model parameters, the location of engine mountings influences the vibration of the vehicle body.

  • The torque converter, an important component of automatic transmissions, is a hydrodynamic device which has a great influence on transient characteristics of vehicle during shift. To predict the accurate driving performance in extremely transient state such as shifting process, a detailed analysis of the torque converter is required. In this study, one dimensional performance model of the torque converter based on the concept of mean flow path, was used to analyze the shifting transients and the exact values of equivalent parameters were determined from the experimental results by using BOX program. The dynamic modelings of the components of power transmission systems such as engines, planetary gear systems, clutches and one-way clutches, were carried out. To analyze the shifting transients of tracked vehicle, a simulation program was developed. In the modeling of power transmission systems, the stiffness of shafts was neglected and shifting control logic(TCU) was included. Using the developed simulation program, the driving conditions were simulated and the results of simulation were verified through the experiments on the dynamometer.

OPTIMIZATION OF THE PARAMETERS OF FEEDWATER CONTROL SYSTEM FOR OPR1000 NUCLEAR POWER PLANTS

  • In this study, the parameters of the feedwater control system (FWCS) of the OPR1000 type nuclear power plant (NPP) are optimized by response surface methodology (RSM) in order to acquire better level control performance from the FWCS. The objective of the optimization is to minimize the steam generator (SG) water level deviation from the reference level during transients. The objective functions for this optimization are relationships between the SG level deviation and the parameters of the FWCS. However, in this case of FWCS parameter optimization, the objective functions are not available in the form of analytic equations and the responses (the SG level at plant transients) to inputs (FWCS parameters) can be evaluated by computer simulations only. Classical optimization methods cannot be used because the objective function value cannot be calculated directely. Therefore, the simulation optimization methodology is used and the RSM is adopted as the simulation optimization algorithm. Objective functions are evaluated with several typical transients in NPPs using a system simulation computer code that has been utilized for the system performance analysis of actual NPPs. The results show that the optimized parameters have better SG level control performance. The degree of the SG level deviation from the reference level during transients is minimized and consequently the control performance of the FWCS is remarkably improved.

Transient safety analysis of M2LFR-1000 reactor using ATHLET

  • $M^2LFR-1000$ is a medium-power modular lead-cooled fast reactor, developed by University of Science and Technology of China (USTC), aiming at achieving a reactor design fulfilling the Gen IV nuclear system requirements and meanwhile emphasizing the optimum safety and economics. In order to evaluate the safety performance of $M^2LFR-1000$ reactor core, three typical transients are selected from initiating events, which are unprotected transient overpower (UTOP), unprotected loss of offsite power (ULOHS+ULOF) and increase of feedwater flowrate with primary pumps trip (IFW+PLOF). These three transients presented and discussed in this paper are performed with the code Analysis of THermal-hydraulics of LEaks and Transients (ATHLET), which is developed by Gesellschaft $f{\ddot{u}}r$ Anlagen-und Reaktorsicherheit gGmbH (GRS). The results indicate that the $M^2LFR$ is safe enough under these three transients due to the good inherent safety features of the reactor, without human intervention, the reactor will reach a new steady state under UTOP condition.

겨울철 저온스트레스에 의한 문주란 잎의 항산화효소 활성과 O-J-I-P 곡선 패턴 (Antioxidative Enzyme Activities and O-J-I-P Transients of Crinum asiaticum var. japonicum $B_{AK}$. Leaves under Winter Stress)

  • 본 연구는 환경요인의 변화, 특히 겨울철 저온이 식물체에 미치는 영향을 정량적으로 밝히기 위해, 저온감수성 식물인 문주란을 대상으로 겨울철 일주기에 있어서 항산화효소 활성과 O-J-I-P곡선의 변화를 조사하였다. 겨울철의 superoxide dismutase와 peroxidase의 활성은 여름철에 비해 다소 증가하였다. 특히, peroxidase는 겨울철 새벽과 밤에 높은 활성을 보였으며 초겨울에만 특이적으로 검출되는 isoenzyme들로 관찰할 수 있었다. 또한 겨울철 문주란 잎의 O-J-I-P곡선에서 J, I, P-단계의 형광세기가 여름철에 비해 현저하게 감소하였다. 그리고, O-J-I-P곡선으로부터 산출된 주요 형광변수들 중에 Fm과 $\Phi_{po}$는 겨울철 저온에 노출되는 기간이 길어짐에 따라 각각 $30\%$$50\%$로 감소하였으며, ABS/RC는 초겨울보다는 늦은 겨울에 2배정도 증가하였다.

  • As the speed of train increases, the effects of the pressure wave generated by the train are becoming more important. To calculate characteristics of the pressure wave generated by a high-speed train passing through a tunnel, several methods are simulated. The pressure waves give rise to large pressure transients which impose the fluctuating loads on the train. It is highly that the pressure transients should be predicted to design the tunnel size and to improve the comfortableness of passengers. In this study, the pressure transients were numerically simulated for a wide range of train speed and compared with the previous verified paper. The simulation results were agreed with the paper, and the characteristics of pressure wave made clear.
