Study on Keratinocyte Differentiation and Skin Barrier Function of Adeonphorae Radix Root Extracts (Adenophorae Radix 뿌리 추출물에 의한 Keratinocyte의 분화 및 피부장벽 기능에 대한 연구)

  • We have studied on the keratinocytes differentiation and skin barrier function using Adenophorae radix (A. radix) root extract, which was known to contain triterpenoid, saponin and starch. A. radix root extracts showed the $PPAR{\alpha}$ expression level of Wy-14,643 $0.5-1.0{\mu}M$ in CV-1 cells. The cornified envelop formation (CE) of human keratinocyte cell line (HaCaT) and normal human keratinocyte (NHK) showed a statistically significant increased compared to the control. When HaCaT cells were treated with A. radix root extract, transglutaminase (TGase-1) was significantly increased. As a result of clinical study of the simple cosmetic formulation containing A. radix root extract for about 2 weeks, TEWL values were significantly decreased and water contents were increased. The ceramides, which were obtained from the inner forearm, were also significantly increased statistically. We suggest that the A. radix root extract can be used as a preventive and therapeutic agent for skin diseases such as dry skin and atopy.

Diagnostic Value of Immunoglobulin G Anti-Deamidated Gliadin Peptide Antibody for Diagnosis of Pediatric Celiac Disease: A Study from Shiraz, Iran

  • Purpose: Screening serologic tests are important tools for the diagnosis of celiac disease (CD). Immunoglobulin (Ig)G anti-deamidated gliadin peptide (anti-DGP) is a relatively new autoantibody thought to have good diagnostic accuracy, comparable to that of anti-tissue transglutaminase (anti-tTG) antibody. Methods: Pediatric patients (n=86) with a clinical suspicion of CD were included. Duodenal biopsy, anti-tTG, and IgG anti-DGP antibody tests were performed. The patients were divided into CD and control groups based on the pathological evaluation of duodenal biopsies. The diagnostic accuracy of serological tests was determined. Results: IgA anti-tTG and IgG anti-DGP antibodies were positive in 86.3% and 95.4% of patients, respectively. The sensitivity, specificity, and diagnostic accuracy of the IgA anti-tTG test were 86.3%, 50.0%, and 68.6%, respectively, and those of the IgG anti-DGP test were 95.4%, 85.7%, and 90.7%, respectively. The area under the receiver operating characteristic (ROC) curve was 0.84 (95% confidence interval [CI], 0.74-0.91) for IgA anti-tTG test and 0.93 (95% CI, 0.86-0.97) for IgG anti-DGP test. The comparison of IgA anti-tTG and IgG anti-DGP ROC curves showed a higher sensitivity and specificity of the IgG anti-DGP test. Conclusion: IgG anti-DGP is a reliable serological test for CD diagnosis in children. High tTG and DGP titers in the serum are suggestive of severe duodenal atrophy. The combined use of IgA anti-tTG and IgG anti-DGP tests for the initial screening of CD can improve diagnostic sensitivity.

Effect of Texture Improvement and Shelf Life Extension of Frozen Rainbow Trout Oncorhynchus mykiss Treated with TGase and Polysaccharides (다당류 및 TGase를 활용한 동결 무지개송어육(Oncorhynchus mykiss)의 물성개선 및 저장성 향상 효과)

  • This study investigated the effect of transglutaminase (TGase) and polysaccharide kappa carrageenan on the texture, chemical, and microbiological qualities of refrigerated unmarketable rainbow trout Oncorhynchus mykiss. Gel strength increased substantially in TGase-treated samples, and was adding kappa carrageenan resulted in no significant difference. SDS-PAGE results confirmed that the myosin heavy chain band with a molecular weight of 205-250 kDa was weakened in trout meat treated with 1% TGase, which led to cross-linking reactions between proteins. The volatile basic nitrogen (VBN) increased in all samples during storage at 4℃ for 10 days; however, the samples treated with 0.5% and 1% kappa carrageenan had the lowest VBN. The viable cell count increased in all samples treated with TGase and kappa carrageenan; however, an increase in TGase enzyme and kappa carrageenan concentration successfully hindered total bacteria growth. Thus, adding 1% TGase and 1% kappa carrageenan to refrigerated unmarketable rainbow trout formulations can optimize quality characteristics.

Propolis Inhibits Neurite Outgrowth in Differentiating SH-SY5Y Human Neuroblastoma Cells

  • Propolis is a multicomponent, active, complex resinous substance collected by honeybees from a variety of plant sources. We have studied the effect of propolis on neurite outgrowth of SH-SY5Y human neuroblastoma cells induced to differentiate by all-trans-retinoic acid (RA). Propolis, at a concentration of $3{\mu}g/mL$, had no significant effect on the viability of differentiating SH-SY5Y cells. However, the neurite outgrowth of the differentiating SH-SY5Y cells treated with propolis ($0.3{\sim}3{\mu}g/mL$) for 48 hr was significantly inhibited in a dose-dependent manner. Treatment of RA-stimulated differentiating SH-SY5Y cells with 0.3 to $3{\mu}g/mL$ propolis resulted in decreased level of transglutaminase and 43-kDa growth-associated protein (GAP-43) in a dose-dependent manner. The results indicate that propolis is able to inhibit neurite outgrowth of differentiating SH-SY5Y cells.

Association between Celiac Disease and Intussusceptions in Children: Two Case Reports and Literature Review

  • Association between celiac disease and intussusception has been reported in adult. Although intussusception is common in children, it rarely has been reported in association with celiac disease. Two children, 5 and 7 years old, with celiac disease are reported here, whose initial presentation was intussusception prior to investigation for concomitant failure to thrive. They presented with acute and severe abdominal distention with vomiting, and donuts and pseudo- kidney appearance in abdominal ultrasonography. One patient's intussusception had reducted spontaneously, however the other had required surgery. In investigation for concomitant failure to thrive, tissue transglutaminase levels were very high and duodenal biopsies revealed celiac disease. Thus celiac testing is recommended in children with intussusception and growth failure.

  • Anion-exchange porous hollow-fiber membranes with a thickness of about 1.2 mm and a pore size of about $0.30{\mu}m$ were used as a supporting matrix to immobilize cycloisomaltooligosaccharide glucanotransferase (CITase). CITase was immobilized to the membrane via anion-exchange adsorption and by subsequent enzymatic cross-linking with transglutaminase, the amount of which ranged from 3 to 110 mg per g of the membrane. The degree of enzyme multilayer binding was equivalent to 0.3 to 9.8. Dextran, as the substrate, was converted into seven- to nine-glucose-membered cycloisomaltooligosaccharides (CI-7, -8, and -9) at a maxi mum yield of $28\%$ in weight at a space velocity of 10 per hour during the permeation of $2.0(w/w)\%$ dextran solution across the CITase-immobilized porous hollow-fiber membrane.

Genomic and Proteomic Profiling of the Cadmium Cytotoxic Response in Human Lung Epithelial Cells

  • Microarray and proteomic expression patterns in response to cadmium exposure were analyzed in human lung epithelial cells. Among 35,000 genes analyzed by cDNA microarray, 228 genes were up-regulated and 99 genes were down-regulated, based on a fold change cut-off value of ${\geq}2$. Combining two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time-of-flight-mass spectrometry (MALDI-ToF-MS), 25 of 629 protein spots showed fold changes in expression ${\geq}2$ (17 up-regulated, 8 down-regulated). After comparing the cDNA microarray and proteomic analyses, only transglutaminase 2, translation elongation factor 1 alpha 1, and glyceraldehyde-3-phosphate dehydrogenase showed overlapping signals in the cDNA microarray and proteomic analyses, whereas the remaining differentially expressed proteins showed large discrepancies with respect to mRNA expression.

  • 근육 단백질과 우유 단백질간의 상호작용의 촉매제로써 TGase의 최적화를 위한 온도와 배양시간을 결정하고자 본 실험을 실시하였다. 돼지 등심에서 근원섬유 단백질을 추출하였고 배양온도는 4, $37^{\circ}C$로, 배양시간은 0, 0.5, 2, 4시간으로 설정하였다. 단백질의 열량분석, 점도, 겔 강도, 단백질 밴드의 변화를 측정하였으며, 그 결과 단백질 열량 변화는 각 단백질 별로 열량 변화 패턴이 상이하게 나타났으며 근원섬유와 카제인 염의 혼합액은 각각의 단백질의 피크와 유사하게 나타났고 배양시간과 온도에 따라 차이를 보여 $4^{\circ}C$에 비하여 $37^{\circ}C$에서 열량 변화가 크게 나타났다. 점도의 경우 배양하지 않은 것과 비교했을 때 $37^{\circ}C$에서 2시간 배양했을 때부터 유의적인 차이를 보이며 증가했으나 4시간 배양한 것과는 차이를 나타내지 않았다. 전기영동의 경우에도 $37^{\circ}C$에서 30분 배양한 처리구는 큰 변화를 나타내지 않았으나 2시간부터 저분자의 밴드가 소멸되고 고분자의 biopolymer를 형성되었다.

저염 재구성 햄의 제조시 식염감소를 위한 Microbial Transglutaminase와 우유단백질의 이용

  • 본 연구는 식염첨가의 감소를 MTGase와 우유단백질을 첨가하여 보완하기 위하여 저지방 저염 재구성 햄을 제조하여 이화학적 및 조직학적 성상을 평가하였고, 1.5%의 식염이 첨가된 대조구와 기능성, 조직감 및 관능성을 비교하였다. 제조한 재구성 햄의 pH, 그리고 수분, 지방 및 단백질 함량은 각각 6.07-6.22, 68-72%, 2-5%, 18-22%였다. 색도와 보수력은MTGase와 우유단백질 첨가에 의해 영향 받지 않았다 (p>0.05). 반면에 가열감량은 식염첨가량과 우유단백질에 의해 차이를 보여 식염함량이 증가할수록 가열감량은 낮았고 카제인 염이 유청 단백질에 비하여 가열감량을 줄일 수 있었다 (p<0.05). 조직검사에서는 MTGase에 우유단백질 1%를 첨가한 처리구가 대조구에 비교한 결과, 경도를 제외한 조직검사에서 높은 값을 나타내었다. 관능평가에서는 1.0% 식염에 0.3% MTGase와 1% 우유단백질을 첨가한 재구성 햄이 기능적에서 대조구와 유사하였다. 이러한 결과는 0.3% MTGase에 1% 우유단백질을 첨가하는 것이 식염감소에 의한 재구성 햄의 결점을 보완함을 시사하였고 카제인 염 단백질은 MTGase의 기질로써 유청 단백질보다 더 효율적이었다.

  • 근원섬유 단백질에 카제인염을 첨가함으로써 유화안정성이나 보수력을 증진시킬 수 있으나 가제인염은 열에 안정성이 떨어지는 단점을 가지고 있다. TGase가 이러한 카제인염의 단점을 보완해주는지 알아보기 위해 열량분석기와 전기영동을 이용하여 단백질의 변화를 측정하였다. 열량분석기의 경우 근원섬유 단백질과 카제인 염 단백질의 상호교차결합을 TGase의 첨가유무에 따라 실시하였으며 그 결과 TGase의 첨가는 각 단백질 열량변화가 나타나는 온도를 변화시켰으며 배양시간을 증가할수록 각 단백질별 열량변화의 차이를 보였고 peak의 크기에도 차이를 보였다. 또한 전기영동의 경우 MFP, SC, MFP:SC의 1:1 혼합액을 각각을 TGase 첨가한 것과 하지 않은 것을 비교한 결과, TGase의 첨가는 고분자 polymer를 만들어줌으로써 두 단백질간의 상호작용에 촉매제로써 작용한다는 것을 확인하였다.
