The high expression level of industrial and metabolically important proteins in plants can be achieved by plastid transformation. The CaIA vector, a Capsicum-specific vector harboring aadA (spectinomycin resistance), is a selectable marker controlled by the PsbA promoter, and the terminator is flanked by the trnA and trnI regions of the inverted repeat (IR) region of the plastid. The CaIA vector can introduce foreign genes into the IR region of the plastid genome. The biolistic method was used for chloroplast transformation in Scoparia dulcis with leaf explants followed by antibiotic selection on regeneration medium. Transplastomes were successfully screened, and the transformation efficiency of 3 transgenic lines from 25 bombarded leaf explants was determined. Transplastomic lines were evaluated by PCR and Southern blotting for the confirmation of aadA insertion and its integration into the chloroplast genome. Seeds collected from transplastomes were analyzed on spectinomycin medium with wild types to determine genetic stability. The increased chloroplast transformation efficiency (3 transplastomic lines from 25 bombarded explants) would be useful for expressing therapeutically and industrially important genes in Scoparia dulcis L.

This study, in order for perceiving the mechanical attribute followed by the explosive spalling of high strength concrete material under high temperature and evaluating capacity of endurance of material, targets understanding capacity of endurance of material such as explosive spalling in high temperature, temperature by thickness of clothing, transformation extent, transformation speed and displacement, stocking the maximum load based on the Allowable Stress Design Method. As a result of experimenting the explosive spalling attribute of high strength concrete material, the one possibly causing serious damage is the 50 MPa concrete. In all aspects of 60 MPa concrete, explosive spalling happens. Especially, it is hazardous enough to reveal all the iron bar. All explosive spalling is intensively concentrated on the surface of concrete for the first $5{\sim}25$ minutes, which urges for the explosive spalling protection action. As a result of evaluating the structural safety by the transformation of high strength concrete, while beam assures the fire safety meeting regulation, 60 MPa shows the dramatic increase of transformation, which only counts 84% of safety. In a column, both the concrete exclusion and excessive explosive spalling are concentrated upper part of column, which brings about the dramatic transformation, so it only meets the 50% of safety regulation. Likewise, in 80, 100 MPa concrete which was never experimented considering the condition of domestic structural endurance stocking devices, the faster collapse is expected.
The activation energy to create a phase transformation or for the reaction to move to the next stage in the milling process can be calculated from the slop of the DSC plot, obtained at the various heating rates for mechanically activated Al-Ni alloy systems by using Kissinger's equation. The mechanically activated material has been called "the driven material" as it creates new phases or intermetallic compounds of AlNi in Al-Ni alloy systems. The reaction time for phase transformation by milling can be calculated using the activation energy obtained from the above mentioned method and from the real required energy. The real required energy (activation energy) could be calculated by subtracting the loss energy from the total input energy (calculated input energy from electric motor). The loss energy and real required energy divided by the reaction time are considered the "metabolic energy" and "the effective input energy", respectively. The milling time for phase transformation at other Al-Co alloy systems from the calculated data of Al-Ni systems can be predicted accordingly.
With the continual development of genetically modified (GM) crops, it has become necessary to develop detailed and effective molecular characterization methods to select candidate events from a large pool of transformation events. Relative to traditional molecular analysis methods such as the polymerase chain reaction (PCR) and Southern blot hybridization, next generation sequencing (NGS) technology for whole-genome sequencing of complex crop genomes had proven comparatively useful for in-depth molecular characterization. In this study, four transformation events, including one in Bacillus thuringiensis (Bt)-resistant rice, one in resveratrol-producing rice, and two in beta-carotene-enhanced soybeans, were selected for molecular characterization. To merge NGS analysis and Southern blot-hybridization results, we confirmed the transgene insertion sites, insertion construction, and insertion numbers of these four transformation events. In addition, the read-coverage depth assessed by NGS analysis for inserted genes might provide consistent results in terms of inserted T-DNA numbers in case of complex insertion structures and highly duplicated donor genomes; however, PCR-based methods can produce incorrect conclusions. Our combined method provides an effective and complete analytical approach for whole-genome visual inspection of transformation events that require biosafety assessment.
The genetic transformation of watermelon by Agrobacterium has been known very difficult and a few successful cases have been reported by obtaining the direct shoot formation. However, since this direct shoot formation is not guaranteed the stable transformation, the stable transformation with reproducibility is required by a different approach such as a callus induced manner. The best conditions for inducing the callus from cotyledon and root explants of watermelon were 2 mg/L zeatin + 0.1 mg/L IAA and 2 mg/L BA + 0.1 mg/L 2,4-D, respectively. The GFP expression in the callus was identified and monitored through fluorescent microscopy after transformation with pmGFP5-ER vector. Paromomycin rather than kanamycin was used for selecting the nptll gene expression because it was more effective to select the watermelon explants. Four different callus types were observed and the solid green callus showed stronger GFP expression. The highest frequency of GFP expression in the callus developed from cotyledon was 9.0% (WM8 inbred line), while the highest frequency from root was 8.3% (WM6 inbred line). The WMV-CP was transformed using the method of GFP transformation and the genetic transformation of WMV-CP was confirmed by PCR and Southern blot analysis. Here we present a system for callus induction of watermelon explant and the callus induced method would facilitate the establishment of stable watermelon transformation.

In this paper, we describe the reorientation method of distorted road sign by using projection transformation for improving recognition rate of road sign. RSR (Road Sign Recognition) is one of the most important topics for implementing driver assistance in intelligent transportation systems using pattern recognition and vision technology. The RS (Road Sign) includes direction of road or place name, and intersection for obtaining the road information. We acquire input images from mounted camera on vehicle. However, the road signs are often appeared with rotation, skew, and distortion by perspective camera. In order to obtain the correct road sign overcoming these problems, projection transformation is used to transform from 4 points of image coordinate to 4 points of world coordinate. The 4 vertices points are obtained using the trajectory as the distance from the mass center to the boundary of the object. Then, the candidate areas of road sign are transformed from distorted image by using homography transformation matrix. Internal information of reoriented road signs is segmented with arrow and the corresponding indicated place name. Arrow area is the largest labeled one. Also, the number of group of place names equals to that of arrow heads. Characters of the road sign are segmented by using vertical and horizontal histograms, and each character is recognized by using SAD (Sum of Absolute Difference). From the experiments, the proposed method has shown the higher recognition results than the image without reorientation.
Recently, 3DTV and 3D displays have been released in the market. Accordingly, the production of stereoscopic images has gained much interest. Stereoscopic image being composed of left and right images are currently delivered to viewers without any modifications. The researches on the enhancement of depth perception using high-frequency components and the re-production of natural color by color compensation have been carried out for 2D images. The application of such 2D technologies to 3D stereoscopic images is an aim of this paper. This paper proposes the enhancement of 3D perception by color transformation. For this, we propose a stereo matching method for obtaining a depth map and two color transformation methods such as contrast transformation and background darkening. The effectiveness of the proposed method was verified through experiments.

A vibration response-based detection system was used to investigate the adhesive areas of single-lap joints using a nonlinear transformation approach for deep learning. In industry or engineering fields, it is difficult to know the condition of an invisible part within a structure that cannot easily be disassembled and the conditions of adhesive areas of adhesively bonded structures. To address these issues, a detection method was devised that uses nonlinear transformation to determine the adhesive areas of various single-lap-jointed specimens from the vibration response of the reference specimen. In this study, a frequency response function with nonlinear transformation was employed to identify the vibration characteristics, and a virtual spectrogram was used for classification in convolutional neural network based deep learning. Moreover, a vibration experiment, an analytical solution, and a finite-element analysis were performed to verify the developed method with aluminum, carbon fiber composite, and ultra-high-molecular-weight polyethylene specimens.

Potholes are an important clue to the structural defects of asphalt pavement and cause many casualties and property damage. Therefore, accurate pothole detection is an important task in road surface maintenance. Many machine learning technologies are being introduced for pothole detection, and data preprocessing is required to increase the efficiency of deep learning models. In this paper, we propose a preprocessing method that emphasizes important textures and shapes in pothole datasets. The proposed preprocessing method uses intensity transformation to reduce unnecessary elements of the road and emphasize the texture and shape of the pothole. In addition, the feature of the porthole is detected using Superpixel and Sobel edge detection. Through performance comparison between the proposed preprocessing method and the existing preprocessing method, it is shown that the proposed preprocessing method is a more effective method than the existing method in detecting potholes.

The different cleavage patterns of pYBamy59 plasmid isolated from E. coli $DH5{\alpha}$ and B. longum MG1 by the cell extract of B. longum MG1 suggested that the main reason for its low transformation efficiency was related to the restriction modification (R-M) system. To confirm the correlation between the R-M system and transformation efficiency, in vitro methylation and site-directed mutagenesis were performed in pYBamy59. Sequence analysis of pYBamy59 fragments digested by the cell extract of B. longum MG1 revealed that all fragments were generated by restriction of the sequence recognized by SacII endonuclease. When pYBamy59 from E. coli was methylated in vitro by CpG or GpC methyltransferase, it was protected from SacII digestion. Site-directed mutagenesis, which removed SacII sites from pYBamy59, or in vitro methylation of pYBamy59 showed 8- to 15-fold increases in the transformation efficiency over intact pYBamy59. Modification of the SacII-related R-M system in B. longum MG1 and in vitro methylation in pYBamy 59 can improve the transformation efficiency in this strain. The results showed that the R-M system is a factor to limit introduction of exogenous DNA, and in vitro modification is a convenient method to overcome the barrier of the R-M system for transformation.
