Enhanced Green Fluorescent Protein Gene under the Regulation of Human Oct4 Promoter as a Marker to Identify Reprogramming of Human Fibroblasts

  • Recent studies on nuclear transfer and induced pluripotent stem cells have demonstrated that differentiated somatic cells can be returned to the undifferentiated state by reversing their developmental process. These epigenetically reprogrammed somatic cells may again be differentiated into various cell types, and used for cell replacement therapies through autologous transplantation to treat many degenerative diseases. To date, however, reprogramming of somatic cells into undifferentiated cells has been extremely inefficient. Hence, reliable markers to identify the event of reprogramming would assist effective selection of reprogrammed cells. In this study, a transgene construct encoding enhanced green fluorescent protein (EGFP) under the regulation of human Oct4 promoter was developed as a reporter for the reprogramming of somatic cells. Microinjection of the transgene construct into pronuclei of fertilized mouse eggs resulted in the emission of green fluorescence, suggesting that the undifferentiated cytoplasmic environment provided by fertilized eggs induces the expression of EGFP. Next, the transgene construct was introduced into human embryonic fibroblasts, and the nuclei from these cells were transferred into enucleated porcine oocytes. Along with their in vitro development, nuclear transfer embryos emitted green fluorescence, suggesting the reprogramming of donor nuclei in nuclear transfer embryos. The results of the present study demonstrate that expression of the transgene under the regulation of human Oct4 promoter coincides with epigenetic reprogramming, and may be used as a convenient marker that non-invasively reflects reprogramming of somatic cells.

An Experimental Study on the Performance of Plastic Plate Heat Exchanger (플라스틱 판형 열교환기의 성능에 관한 실험적 연구)

  • Aluminum plate heat exchanger, rotary wheel heat exchanger, and heat pipe heat exchanger have been used (or ventilation heat recovery in the air-conditioning system. The purpose of this research is to develop high efficiency plastic plate heat exchanger which can substitute aluminum plate heat exchanger. Because thermal conductivity of plastic is quite small compared to that of aluminum, various heat transfer enhancement techniques are applied in the design of plastic plates. Five types of heat exchanger model are designed and manufactured, which are plate type, plate-fin type, turbulent promoter type, corrugate type, and dimple type. Thermal performance and pressure loss of each heat exchangers are measured in various operating conditions, and compared each other. Test results show that heat transfer performance of corrugate type, turbulent promoter type, and dimple type are increases about $43\%$, $14\%$, and $33\%$ at the equivalent fan power compared to those of plate type, respectively. On the other hand, the heat transfer performance of plate-fin type decreases $9\%$ because fins can not play their own role.

Tetrapyrazinoindoloporphyrazine Langmuir-Blodgett Films

  • We fabricated tetra(5-n-nonyl-8-tert-butyl-2,3-pyrazino[2,3-b]indolo)porphyrazinato copper(II) (Cu-Pc-$C_8$) Langmuir-Blodgett (LB) films. We further investigated the influence of arachidic acid (AA) as a transfer promoter, as well as the effect of dipping speed, on the deposition of the films on hydrophilic and hydrophobic substrates. In the case of pure Cu-Pc-$C_8$ LB deposition on a hydrophilic substrate, the transfer ratio was close to one for up-stroke depositions, but the previously deposited film was peeled off and re-spread onto water at down-stroke depositions. Whereas the stability of the Cu-Pc-$C_8$ LB films was not improved by AA addition on hydrophilic substrates, the deposition of Cu-Pc-$C_8$ was significantly improved by the presence of AA on a hydrophobic substrate. The AA-assisted deposition had transfer ratio of close to 1 and was essentially stable up to 10-layer depositions. Comparison of the UV-visible spectrum of a Cu-Pc-$C_8$/AA LB film with that of Cu-Pc-$C_8$/AA solution in dichloroethane revealed that the Soret and Q bands for the Cu-Pc-$C_8$/AA LB film were broadened and red-shifted due to the aggregation of phthalocyanines upon assembly in the LB film.

Fabrication and Properties of Organic Semiconductor CuPccp LB Thin Film (유기 반도체 CuPccp LB초박막의 제작 및 특성)

  • A copper tetracumylphenoxy phthalocyanine (CuPccp) thin film was formed on an organic insulator film by Langmuir-Blodgett (LB) deposition for gas sensor fabrication. To increase the reproducibility of film transfer, stearyl alcohol was used as a transfer promoter. The structural properties of the CuPccp layers were optically monitored through attenuated total reflection and polarization-modulated ellipsometry techniques. The average thickness of a single layer of the CuPccp LB film was measured to be 2.5 nm. Despite the role of the transfer promoter, the stability of the layer transfer was not sufficient to ensure homogeneity of the LB film. This was probably due to the presence of aggregates in the molecular structure of the CuPccp LB film. Nevertheless, copper phthalocyanine polymorphism can be greatly suppressed by the LB arrangement, which appears to contribute to the improvement of electrical conductivity. The p-type semiconductor characteristics were confirmed by Hall measurements from the CuPccp LB films.

A Study on the Effect of the Heat Transfer Surface Position to the Condensation heat Transfer (전열면 자세가 응축 열전달에 미치는 영향에 관한 연구)

  • A study has been conducted to investigate the effect of the heat transfer surface position in the range of dropwise condensation, filmwise condensation, and glacial condensation. For dropwise condensation promoter, the heat transfer surface was evaporated by gold. As a result, heat transfer rate is almost same where the position of heat transfer surface is between 45 and 135 degree. It is found that heat transfer rate was reduced as subcooled degree was increased. And it is also found that if the subcooled degree becomes lower, the position of heat transfer surface is more effective. Adversely, if the subcooled degree becomes higher, the effectiveness of surface position is getting relatively lower. Regardless of the position, the transition temperatures from dropwise condensation to filmwise condensation is in the vicinity of 80K.

  • Novel baculovirus vector systems recombined with coding genes of polyhedron promoter, vesicular stomatitis virus G (VSVG), polyA, cytomegalovirus (CMV) promoter, enhanced green fluorescent protein (EGFP), and protein transduction domain (PTD) were constructed. These recombinant baculovirus vector systems were applied into human foreskin fibroblast cells and compared the effects of gene transfer and gene expression of these recombinant baculovirus vector systems with control vector system. From this study, it showed that these novel recombinant baculovirus vector systems were superior efficacy to control vector system in view of gene transfer and gene expression.

Analysis of Promoter Strength of Autographa californica Nuclear Polyhedrosis Virus IE1 Gene by Using Rreconmbinant Baculovirus

  • To analysis a promoter strength of Atographa californica nucler polyhedrosis virus (AcNPV) IE1 gene, an immediate viral gene, ${\beta}$-glactosidase gene as a reporter gene was introduced under the control of the IE1 promoter. The restriction fragment containing IE1 promoter and ${\beta}$-galctosidase gene from pAcIE1-gal were inserter into pBacPAK9 to yield transfer vector pAcNPV-IE1-gal. The pAcNPV-IE1-gal was cotransfected with AcNPV genomic DNA BacPAK6 into Sf9 cells to produce recombinant baculovirus AcNPV-IE1-gal. In addition, recombinant bacvulovirus AcNPV-gal, which express ${\beta}$-galac-tosidase under the control of the polyhedrin promoter, was constrer, was constructed to compared with AcNPV-IE1-gal. The recombinant viruses were respectively infected into Sf9 cells and characterized by the virus titer and expression of ${\beta}$-galactoxidase in Sf9 cells. The promoter strength of IE1 and polyhedrin promoters was determined by the amount of ${\beta}$-galactosidase secreted into medium by viral infection. The titer of AcNPV-IE1-Gal determined by plaque assays in Sf9 cells was similar to that of AcNPV-gal. However, expression level of ${\beta}$-galactosidase by AcNPV-IE1-gal was significantly lower than that by AcNPV-gal. In conclusion, promoter strength of IE1 was approximately 25-fold lower than that of polyhedrin.

  • construct containing reporter gene(pFV4CAT) regulated by carp $\beta$-actin promoter was microinjected into the one-cell stage egg of mud loach (Misgurnus mizolepis), and was successfully expressed, possibly by the integration into the genome. Both mean hatching success and early survival of the microinjected groups were not significantly different with those of control groups (P>0.05). The incidence of transgene was ranged from 7 to 48% based on the PCR and/or Southern blot analyses with the DNA prepared from fin or blood tissue. The spatial and temporal patterns of expression of the pFV4CAT gene, measured by in situ immunohistochemical analysis peroxidase-conjugated anti-CAT antibody, were variable among the experimental individuals. These results suggest that carp $\beta$-actin promoter is effective to express other transgene in mud loach, such that this promoter can be useful in the generation of valuable transgenic mud loach.

  • Recombinant baculovirus was reconstructed with useful genes of polyhedron promoter, vesicular stomatitis virus G (VSVG), polyA, cytomegalovirus (CMV) promoter, enhanced green fluorescent protein (EGFP), and protein transduction domain (PTD). This reconstructed vector was infected into various cell lines and tissues. We investigated gene transfer and gene expression of this reconstructed vector in comparison to other vectors and recognized that this reconstructed vector was higher effective than any other control vector.
