  • The Drosophila auditory system consists of four large basal segments: the arista, the funiculus, the pedicel, and the scape. When an acoustic stimulus is applied to the arista and the funiculus their mechanical vibrations are transmitted to chordotonal neurons in Johnston's organ where mechanoelectric transduction arises. We study the mechanotransduction mechanism in the Drosophila auditory system by using a laser Doppler vibrometer (LDV) and extracellular electrophysiology. We find that large and small peaks appear alternatively and that the antenna vibration is asymmetric depending on whether the pedicel and the scape are fixed. Interestingly, we find that this asymmetric vibration accompanies the alternating neural peak structure. Here, we propose a mathematical model to explain the alternating peak structure by using a model consisting of two opposing neurons that are modeled as strings. Generally, strings have tension only when they are elongated. This property allows the alternating neural peaks for asymmetric antenna motion.

The Effects of Ginseng Components on the Signal Transduction in the Activation of Murine Macrophages (생쥐 대식세포의 활성시 신호전달에 미치는 인삼성분들의 영향)

  • To study the effects of ginseng saponin components on the signal transduction in the ac tivation of murine macrophages, phagocytosis and Intracellular calcium concentration of peritoneal exuded mouse macrophages were examined. The phagocytosis was increased significantly after treatment with total saponin, diol-saponin, $Rg_1$ and $Rg_2$, but triol-saponin was unable to increase phagocytosis. The phagocytosis were increased when H7, a PKC inhibitor, was pretreated and increased significantly by saponin fractions except total saponin. Pertussis toxin, which inactivates G-protein, decreased the phagocytosis. But the phagocytosis was restored to the control level by saponin fractions and the phagocytosis was increased significantly by $Rg_2$ and $Rg_2$. The triol saponin increased phagocytosis approximately by 2-fold as compared with the TMB-8 treated group. Peritoneal exuded macrophages displayed a prominent rise in cytosolic calcium following treatment with triol-saponin, $Rg_1$, $Rg_2$ and $Rg_2$. Incubation of macrophages with PT resulted in an inhibition of cytosolic calcium mobilization, but increased cytosolic calcium mobilization with saponin fraction.

  • One of the major limitations in using adenoviral vector for gene therapy is inefficient infection of host cells. The presence of coxsackievirus and adenovirus receptor (CAR) and ${\alpha}$-integrin on cell surfaces is required for efficient adenovirus infection. In this study, we investigated the effect of trichostatin A, a histone deacetylase inhibitor, on transfection efficiency after transduction of adenovirus mediated p16$\^$INK4a/ gene transfer. In our previous study, p16$\^$INK4a/ tumor suppressor gene transfer in the non-small cell lung cancer cells (A549 cells) by transduction of recombinant adenovirus (Ad5CMV-p16) resulted in significant inhibition of cancer cell proliferation.   • Pheromone biosynthesis in the pheromone gland is triggered from release of pheromone biosynthesis-activating neuropeptide (PBAN) synthesized in the suboesophageal ganglion. PBAN binds to its receptor on the epithelial cell membrane and activates signal transduction pathways for the pheromone biosynthesis. This study reviews sex pheromone, PBAN and its receptor, and pheromone biosynthesis pathway of Maruca vitrata.

PROM1-mediated cell signal transduction in cancer stem cells and hepatocytes

  • Prominin-1 (PROM1), also called CD133, is a penta-span transmembrane protein that is localized in membrane protrusions, such as microvilli and filopodia. It is known to be expressed in cancer stem cells and various progenitor cells of bone marrow, liver, kidney, and intestine. Accumulating evidence has revealed that PROM1 has multiple functions in various organs, such as eye, tooth, peripheral nerve, and liver, associating with various molecular protein partners. PROM1 regulates PKA-induced gluconeogenesis, TGFβ-induced fibrosis, and IL-6-induced regeneration in the liver, associating with Radixin, SMAD7, and GP130, respectively. In addition, PROM1 is necessary to maintain cancer stem cell properties by activating PI3K and β-Catenin. PROM1-deficienct mice also show distinct phenotypes in eyes, brain, peripheral nerves, and tooth. Here, we discuss recent findings of PROM1-mediated signal transduction.
