Effects of Radix Puerariae, Flos Puerariae and Bamboo+Radix Puerariae Water Extracts on the Ethanol-administered Mice (갈근, 갈화, 갈근과죽력의 추출물이 알코올을 투여한 생쥐에 미치는 영향)

  • This study was performed to investigate the effects of Radix Puerariae (RP), Flos Puerariae (FP) and Caules in Liquamen Phyllostachyos+Radix Puerariae water extracts on the alcohol dehydrogenase (ADH). transaminase (GOT, GPT) activities, and two hepatic antioxidant enzyme (SOD, catalase) activities in acute ethanol administered mice, and we have investigated the morphological changes that occur in hepatocytes of the experimental mice. The activities of ADH decreased compared with control group in the A1(66%), C1(57%), C2(54%) groups. The transaminase activites increased in the control groups compared with experimental groups. Ethanol treatment group without the RP or FP administration significantly lowered the activities of hepatic SOD and catalase, whereas MnSOD increased in the A1(27%) and B2(43%)groups, CuZnSOD increased in the B2(25%) and C2 groups. The catalase activites were increased in the A1(270%), A2(478%), B2(487%) and B1(770%) compared with control group, A lot of PAS-positive granules were observed in the A1, A2, C1 and C2 groups compared with the other groups. These results suggested that RP, FP and RP+Bamboo extracts administration may be prevent from liver damage in the alcohol treatment mice.

  • Serum glutamic oxaloacetic transaminase (GOT), latic dehydrgenase (LDH), and alkaline phosphatase (ALP) activities were determined in 5 Korean native cattle to obtain the base line of blood enzyme activities in relation to the stage of gestation. Serum GOT activities showed an increasing tendency with the course of gestation period, serum LDH levels were within normal range through all the stage of pregnancy, and the slight increase in serum ALP activities was acknowledged at the 2nd and 3rd stage of gestation.

A Comparative Study on Effects of Kyungohkgo and Kyungohkgo Ga Nokyong on Growth in Growth Deficiency Rat with Insufficient Nutrition Diet (성장장애 흰쥐에서 경옥고(瓊玉膏)와 경옥고가록용(瓊玉膏加鹿茸)의 성장효과 비교 연구)

  • Objectives This study was designed to evaluate the difference of between Kyungohkgo and Kyungohkgo Ga Nokyong effects of growth Methods We divided male Spraque-Dawley rats into 6 groups. They were Normal group, Growth deficiency rat with insufficient nutrition diet group, Growth deficiency rat with 0.1% Kyungohkgo group, Growth deficiency rat with 0.2% Kyungohkgo group, Growth deficiency rat with 0.1% Kyungohkgo Ga Nokyong group and Growth deficiency rat with 0.2% Kyungohkgo Ga Nokyong group. They were administered for 5 weeks. We measured body weight, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities. Results 1. In body weight, Kyungohkgo 0.1%, Kyungohkgo Ga Nokyong 0.1%, 0.2% groups were showed sighnificantly different than control group. But That groups were not showed significantly different than each others. 2. In serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, we obtained the results of tendency to increase in Kyungohkgo Ga Nokyong group, however these values showed no significantly different. 3. In the counts of RBC, Kyungohkgo group and Kyungohkgo Ga Nokyong group were showed significantly different than control group. But these values showed no significantly different. In the concentration of Hb, Kyungohkgo Ga Nokyong 0.2%group was showed significantly different than control group. Kyungohkgo Ga Nokyong 0.2% group and Kyungohkgo 0.2% group were showed significantly different than control group. 4. The counts of total WBC and its composition showed no significantly different in all treatment groups. 5. The values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities showed no significantly different in all treatment groups. Conclusions So Kyungohkgo Ga Nokyong and Kyungohkgo have an effect of promoting growth of rats. And We know that the effect of Kyungohkgo Ga Nokyong is better than Kyungohkgo.

Effect of Low Atmospheric Pressure on Serum Glutamic Oxaloacetic Transaminase and Lactic Dehydrogenase Activities of Rats (低氣壓이 흰쥐의 血淸 Glutamic Oxaloacetic Transaminase 및 Lactic Dehydrogenase 活性에 미치는 影響)

  • In order to determine the influence of low atmospheric pressure on serum glutamic oxaloacetic transaminase (SGOT) and serum lactic dehydrogenase (SLDH) activities of rats, blood samples were collected from laboratory-conditioned male rats of the Sprague-Dawley strain which were randomly grouped into control and the experimental subjected to a series of one hour-exposure a day to low atmospheric pressure of 500 and 380mmHg up to the the time of 15 day. Results obtained indicated that decompression caused marked alterations in SGOT and SLDH levels when compared to that of the control. The trend of increases or decreases in these enzyme levels were similar in both 500 and 380mmHg exposed rats although the changes were greater in the latter group. Thus, generally all the experimental rats showed temporary steady state to low atmospheric pressure. Changes in enzymatic contents depended on the intensity and extent of the environmental stress under study. The lower the atmospheric pressure the greater is the effect on these serum enzyme levels.

Effect of Cynanchi wilfordii Radix Extract on the Acute Toxicity in Mice and Subacute Toxicity in Rats (백수오(白首烏) 엑스의 마우스 급성독성(急性毒性) 및 흰쥐 아급성독성(亞急性毒性)에 미치는 영향(影響))

  • This study was attempted to investigate the acute toxicity in mice, the subacute toxicity in rats of Cynanchi wilfordii Radix extract, the effect on transaminase activities, hematological parameters, leukocyte parameters in serum of subacute-toxicated rats. In acute toxicity test, the death rate was not observed in 50, 100, 200, 300 mg/kg(i.p.), one tenths to two tenths in 300, 500, 1000, 1500 mg/kg(p.o.) for two weeks. In subacute test, rats were all died in 300 mg/kg(p.o.) during 4 weeks, in 500, 1000 mg/kg(p.o.) during three weeks. The cause of death believed to be stomach ulcer. The activities of S-GOT and S-GPT were significantly increased in all sample-treated groups, when compared with the normal groups. A number of WBC and neutrophil belong to hematological parameter were significantly increased, lymphocyte was decreased in all sample-treated group, when compared with normal group. The hemolytic action on water extract, saponin and alcohol extract showed very low activities.

The Effect of Autoxidized Methyl Linoleate on the Serum Enzyme Activity in the Mouse (Acute Toxicity) (자동산화 Methyl Linoleate가 Mouse혈청의 효소활성에 미치는 영향 (급성 독성))

  • In order to investigate the acute toxicity of autoxidized methyl linoleate(AOML) on the activity of serum enzymes in the mouse, we administered once 0.45ml of AOML to ICR strain mouse by using stomach tube. The following results were obtained: The total lactic dehydrogenase(LDH) activities in the serum of AOML group were generally increased than those of normal group. According to electrophoresis, the activities of LDH, were increased while those of LDH, were decreased. The activities of glutamic oxaloacetic transaminase(GOT), glutamic-pyruvic transaminase(GPT) and ${\alpha}-amylase$ in the serum of AOML group were increased more than those of normal group. The activities of alkaline phosphatase in the serum of AOML group were increased but those of isozyme were not confirmed in the normal and AOML group. In the serum protein of AOML group, albumin was increased, on the other hand ${\gamma}-globulin$ was decreased. At the peripheral blood slide smear, lymphocytes were significantly decreased but neutrophils were increased and the morphological change of erythrocytes was observed. From these results we conclude that the AOML fed to mouse influences on the activity of various serum enzymes and blood cells in the mouse.

Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite (아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

Effect of Natural Drugs on the Life Support of Starving Rats (기아를 일으킨 흰쥐의 생명연장에 미치는 천연약물의 효과)

  • The current study was undertaken to select the crude drug showing life prolonging effect on the starved rats(Sprague-Dawley) and elucidate the reaction mechanism there of. As distilled water extracts prepared from twenty kinds of natural drugs were administered orally to female starved rats, Bupleuri Radix demonstrated the most prominent effect with 34.6% increment in the survival time. To investigate underlying mechanism of life prolonging effect of Bupleuri Radix, the concentrations of adrenocorticosteroids(corticsterone, testosterone and aldosterone) and enzyme activities of superoxide dismutase(SOD), glutamate oxaloacetate transaminase(GOT), glutamate pyrurate transaminase(GPT) were assayed in the serums of rats starved for, 2, 4 and 6 days respectively. The results manifested much elevated values of corticosterone, aldosterone(2 days) and rather decreasing tendency afterwards, specially in the late periods of starvation(6 days). With respect to such a considerable changes according to starvation periods, Bupleur Radix restored these values much near to normal suggesting that Buplerum Radix may play a life prolonging action during starvation probably through reinforcing the homeostatic properties of corticosteroids and some enzyme activities.

Effect of 6-Aminonicotinamide on the Levels of Some Metabolites and Related Enzymes in Rabbit Serum (6-Aminonicotinamide가 토끼혈청내 효소 및 대사물질에 미치는 영향)

  • The effects of an antimetabolite, 6-aminonicotinamide (6-AN) on the levels of enzymes and metabolites in rabbit serum were investigated. The intraperitoneal administration of 6-AN (multiple doses of l5mg/kg body weight) gave tise to a remarkable increase in glucose and cholesterol levels but did not exert any appreciable influence on the concentration of albumin and total protein. Alkaline phosphatase activity was significantly reduced by administration of 6-AN, whereas creatine phophokinase, serum glutamic oxaloacetate transaminase and serum glutamic pyruvate transaminase activities were matkedly enhanced. Nevettheless, the levels of Ca, P, Na, K, Cl and Co were not affeded to any extent by 6-AN.

Effects of Biphenyldimethyl dicarboxylate(DDB) on the Lipid Peroxidation, Oxygen Free Radical Scavenging Enzymes Activities and Hepatic Functions in Ethanol-induced Hepatotoxic Rats (Biphenyldimethyl dicarboxylate(DDB)가 Ethanol 유발 간독성 흰쥐에서의 지질 과산화와 Oxygen Free Radical 제거 효소 활성도 및 간기능에 미치는 영향)

  • In an attempt to define the effects of Biphenyldimethyl dicarboxylate(DDB) on the lipid peroxidation, oxygen free radical scavenging enzymes activities and hepatic functions in ethanol-induced hepatotoxic rats, we studies malondialdehyde(MDA) level and the activities of catalse, superoxide dismutase(SOD), glutamic-oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) in liver of the rats at 24, 48 and 72 hr after the injection of ethanol and DDB. Sprague-Dalwey albino rats weighing 250 to 280gm were injected intraperitoneally with ethanol(2.5 gm/kg ) only and ethanol plus DDB(300mg/kg ). The result obtained can be summarized as follows : 1) The group treated with ethanol showed significantly higher MDA level and lower catalase and SOD activities at 24, 48 and 72hr after the injection as compared with that of control group. 2) The group treated with ethanol showed significantly higher GOT and GPT activities at 24, 48 and 72hr after the injection as compared with that of control group. 3) The group treated with ethanol plus DDB showed significantly lower MDA level and higher catalase and SOD activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. 4) The group treated with ethanol plus DDB showed significantly lower GOT and GPT activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. These results suggest that the excessive oxygen free radicals resulting from the depression of the activities of catalase and superoxide dismutase is an important determinant in pathogenesis of ethanol-induced hepatotoxicity and DDB has antioxidant effects.
