  • 수용액상에서 $Hg^{2+}$에 의한 trans-[Co(3,2,3-tet)$X_2]^+$ (3,2,3-tet = 4,7-diazadecane-1,10-diamine, $X_2\;=\;Cl_2,\;(NO_2)Cl,\;Br_2,\;(NO_2)Br,\;(NO_3)_2)$ 착물의 아쿠아 반응이 연구되었다. 생성물을 확인하고 반응메카니즘을 추정하기 위하여 크로마토그래피를 사용하였고 전자흡수 스펙트럼을 측정하였다. 그 결과 네자리 리간드인 3,2,3-tet가 배위된 여러가지 $trans-[Co(3,2,3-tet)X_2]^+$ 착물은 각각 아쿠아된 trans-[Co(3,2,3-tet)$(OH_2)_2]^{3+}$ 착물을 거쳐 cis-${\beta}$-[Co(3,2,3-tet)$(OH_2)_2]^{3+}$ 착물이 생성되었다. $Hg^{2+}$에 의한 trans-$[Co(3,2,3-tet)Cl_2]^+$ 착물과 trans-[Co(3,2,3-tet)$(NO_2)Cl]^+$ 착물의 아쿠아 반응에 대한 메카니즘을 추정하기 위하여 속도론적 조사를 하였따. 그 결과 $trans-[Co(3,2,3-tet)Cl_2]^+$ 착물은 D(dissociative)-메카니즘으로 진행되었고, trans-[Co(3,2,3-tet)$(NO2_)Cl]^+ $착물은 $I_d$(interchange dissociative)-메카니즘으로 진행되었다. 그리고 입체화학적인 거동을 조사하기 위하여 라세미(R,R:S,S)3,2,3-tet 대신에 키랄성이 R,R인 3,2,3,-tet를 배위시킨 trans-$[Co(R,R-3,2,3-tet)Cl_2]^+$ 착물에 $Hg^{2+}$를 용리시켰을 때 아쿠아 반응에 대한 원편광이색성(circular dichroism) 스펙트럼을 측정하여 그 절대구조를 확인한 결과 ${\Delta}-cis-{\beta}$-[Co(R,R-3,2,3-tet)$(OH_2)_2]^{3+}$ 착물이 생성되었다.

전북지역 유통 가공식품 트랜스지방 함량 (Trans Fatty Acid Content in Commercial Processed Food in Jeon-Buk Area)

  • This study was carried out to examine the proportion of fatty acids and contents of trans fatty acid in commercial processed foods in Jeon-Buk area. Contents of trans fatty acid in bakery products ranged from 0.2% to 1.0% per 100 g food. Especially garlic bread and pastry contained the highest amount. Popcorns contained trans fatty acid from 0.0% to 13.4%. Among them, instant popcorns for microwave contained the highest amount, while trans fatty acid was hardly detected in popcorns for theater. And trans fatty acid detected in popcorns was mostly in the form of 18 : 1 trans. The content of trans fatty acid in pizza was nearly 0.2%. Amounts of saturated fatty acid and unsaturated fatty acid were similar between each type of pizza and frozen pizza. Fritters such as fried chickens and vegetables contained trans fatty acid less than 0.2% per 100 g food.

모유 섭취 신생아 유래 Lactobacillus acidophilus에 의한 Conjugated Linoleic Acid 생성 (Production of Conjugated Linoleic Acid by Lactobacillus acidophilus Isolated from Breast-Fed Infants)

  • 본 연구는 모유를 섭취하고 있는 생후 7일 이내의 신생아로부터 분리 및 선발된 L. acidophilus를 이용하여 다양한 생리활성을 가진 CLA를 천연적으로 생산하는 젖산균을 Screening하고 선정된 L. acidophilus에 의한 CLA의 최적 생성조건을 설정하기 위하여 실시하였다. 9종의 L. acidophilus를 LA가 $200{\mu}g/mL$의 수준으로 각각 첨가된 MRS 액체배지에 접종하여 48시간 배양한 후 L. acidophilus의 균체 세포막 내에서 형성된 cis-9, trans-11 CLA 함량과 배지 내 유리화된 cis-9, trans-11 CLA 함량을 측정한 결과, 생균수는 대부분 $10^8\;CFU/mL$ 수준이었으며 균체 내로 전환된 cis-9, trans-11 CLA는 검출되지 않거나 매우 미량 존재하였다. MRS 배지 내 유리화된 cis-9, trans-11 CLA는 전 시험 균주를 통해 생성되었으며 상대적으로 균체 내 생성량 보다 높게 나타났고, L. acidophilus NB 203과 NB 209의 생성량이 $26-27{\mu}g/mL$으로 가장 높게 나타났다. 동일한 조건으로 8% 탈지유 배지에 실험한 결과 배양 24시간째의 생균수는 $10^8\;CFU/mL$ 수준이었고, MRS 액체 배지에서의 결과와 유사하게 균체 세포막 내로 incorporation된 cis-9, trans-11 CLA는 대부분의 처리구에서 검출되지 않거나 매우 미량 존재하였으며, 반면 탈지유 배지 내 유리 cis-9, trans-11 CLA는 전 시험 균주를 통해 생성되었고 상대적으로 MRS 액체 배지에서의 생성량 보다 높았으며, 그 중 L. acidophilus NB 203 및 NB 209의 배양액 내 cis-9, trans-11 CLA 생성량이 가장 높게 나타났다. 시험 균주 중 cis-9, trans-11 CLA 생성능이 다른 균주에 비해 높은 것으로 나타난 L. acidophilus NB 203 및 NB 209를 8% 탈지유 배지에 LA 첨가량 및 배양시간을 달리하여 최적 조건을 설정하였으며, 두 시험 균주 모두 LA 첨가에 의한 증식억제 현상은 배양 48시간까지 $1.000{\mu}g/mL$ 첨가 수준에서는 전혀 나타나지 않았으며, $3,000{\mu}g/mL$ 첨가 시 미약하게 나타났다. 배양시간대 별 cis-9, trans-11 CLA 생성량은 두 시험 균주 모두 배양 24시간 이전의 대수증식 기에서 주로 증가하였으며, 배양 24시간 이후부터 생성량이 유지되거나 미량 감소하는 것으로 나타났다. LA의 첨가농도 별 cis-9, trans-11 CLA 생성에 있어 두 시험 균주 모두에서 LA 첨가량이 $1,000{\mu}g/mL$ 수준일 때 cis-9, trans-11 CLA 생성량이 가장 높았으나 $500{\mu}g/mL$ 첨가구와 큰 차이를 나타내지 않았으며, 특히 첨가량을 $3,000{\mu}g/mL$ 수준으로 높여도 $1,000{\mu}g/mL$ 수준일 때와 유사하거나 오히려 감소하는 경향을 보였다. 따라서 이 실험 균주의 최적 LA 생성조건은 탈지유 배지에 LA를 $500{\mu}g/mL$ 수준으로 첨가하여 $37^{\circ}C$에서 24시간 배양시로 나타났다.

Isomeric Effects on Volatilization of 1,3-Dichloropropene Fumigant in Soil

  • The fumigant 1,3-dichloropropene (1,3-D) was recently proposed as a direct replacement for methyl bromide ($CH_3Br$) in soil fumigation. This study was conducted to better understand behavior phase partitioning, diffusion and volatilization of 1,3-D as affected by isomer. The Henry's law constant(KH) of cis-1,3-D and trans-1,3-D was 0.058 and 0.037 at $20^{\circ}C$, respectively. $K_H$ of cis form of 1,3-D was higher than that of trans form of 1,3-D. To compare with volatilization of 1,3-D isomer, soil column [70 cm (length)${\times}$12 cm (i.d.)] included a shank injection at 30 cm with 300 kg $ha^{-1}$. Maximum cis-1,3-D and trans-1,3-D concentration reached 57 mg $L^{-1}$ and 39 mg $L^{-1}$ at 30 cm depth at 1h after application. Cumulatively, after 10 days, 51.8% and 43.57% of applied cis-1,3-D and trans-1,3-D was emitted via volatilization, respectively. The total losses of cis-1,3-D were significantly greater than that of trans-1,3-D. Finally, cis-1,3-D and trans-1,3-D, such as isomer are dominant of 1,3-D fates in soil.

Effect of trans-Cinnamaldehyde and High Pressure Treatment on Physico-chemical and Microbial Properties of Milk during Storage Periods

  • This study was carried out to investigate the effect of trans-cinnamaldehyde and high pressure treatment on milk. Cinnamon oil milk was manufactured by high speed homogenization (3,000 rpm) and high pressure homogenization (500 and 2,000 bar) processing UHT milk and trans-cinnamaldehyde of various concentrations (0 to 0.1% (w/v)). Cinnamon oil milk was inoculated with Escherichia coli (6.4 Log CFU/mL) and kept at $7^{\circ}C$ for 10 d to observe the antibacterial effect. The cinnamon oil milk containing 0.05% (w/v) trans-cinnamaldehyde initially began to show an antibacterial effect and Escherichia coli completely died in cinnamon oil milk added 0.1% (w/v) trans-cinnamaldehyde on the 6th day of storage. The result of the TBA value showed that the addition of 0.1% (w/v) trans-cinnamaldehyde was also effective to protect lipid oxidation. In the physical properties of cinnamon oil milk, particle sizes were enlarged in all samples during storage periods and the total color difference of cinnamon oil milk was slightly increased as level of high pressure. The surface tension of cinnamon oil milk treated 2,000 bar was remarkably higher than other samples. It seems that trans-cinnamaldehyde showed antibacterial activity and antioxidation effect at 0.05 and 0.1% (w/v) of concentration. Remarkably, high pressure treatment did not influence its microbial property but slightly affected the physical properties of cinnamon oil milk.

In Vivo Target RNA Specificity of Trans-Splicing Phenomena by the Group I Intron

  • The Tetrahymena group I intron has been shown to employ a trans-splicing reaction and has been modified to specifically target and replace human telomerase reverse transcriptase (hTERT) RNA with a suicide gene transcript, resulting in the induction of selective cytotoxicity in cancer cells that express the target RNA, in animal models as well as in cell cultures. In this study, we evaluated the target RNA specificity of trans-splicing phenomena by the group I intron in mice that were intraperitoneally inoculated with hTERT-expressing human cancer cells to validate the anti-cancer therapeutic applicability of the group I intron. To this end, an adenoviral vector that encoded for the hTERT-targeting group I intron was constructed and systemically injected into the animal. 5'-end RACE-PCR and sequencing analyses of the trans-spliced cDNA clones revealed that all of the analyzed products in the tumor tissue of the virus-infected mice resulted from reactions that were generated only with the targeted hTERT RNA. This study implies the in vivo target specificity of the trans-splicing group I intron and hence suggests that RNA replacement via a trans-splicing reaction by the group I intron is a potent anti-cancer genetic approach.

The Regulatory Effects of Trans-chalcone on Adipogenesis

  • It is noted that chalcone derivatives have characteristic diverse pharmacological properties, and that precise evidence has been growing that they could regulate a tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) induced insulin resistance. The purpose of the present investigation is to elucidate the effects of the identified chalcone derivatives on adipogenesis, and to find the underlying mechanism of action in that case. Consequently, we first investigated whether the chalcone derivatives could affect the identified $PPAR{\gamma}$-induced transcriptional activity on the proliferator-activated receptor response elements (PPRE) at target promoters, and find that trans-chalcone most significantly increased the $PPAR{\gamma}$-induced transcriptional activity. Additionally, we confirmed that there were up-regulatory effects of trans-chalcone during the adipogenesis and lipid accumulation, and on the mRNA of adipogenic factors in 3T3-L1 cells. Next, we examined the effect of trans-chalcone on the inhibition induced by $TNF-{\alpha}$ on adipogenesis. To that end, we noted that the treatment with trans-chalcone attenuated the effect of $TNF-{\alpha}$ mediated secretion of various adipokines that are involved in insulin sensitivity. For this reason, we noted that this study clearly demonstrates that trans-chalcone enhanced adipogenesis, in part, by its potent effect on $PPAR{\gamma}$ activation and by its reverse effect on $TNF-{\alpha}$.

Unusual Fluorescence Spectroscopic Properties of 9-Anthrylethylene Derivatives : Photoisomerization

  • The fluorescence spectroscopic properties of both trans and cis forms of 9-anthryiethylene derivatives such as 9-AnthCH=$CHCO_2CH_3$ [Ⅰ] and 9-AnthCH=CHCN [Ⅱ] as well as 9-AnthCH=>TEX>$CHCH_2O_2CCH_3$ [Ⅲ] and 9-AnthCH=$CHCH_2OH$ [Ⅳ] have been measured in various solvents. In nonpolar solvent, the trans-I and trans-Ⅱ show dual emission spectral bands at 340 nm and 460 nm when exciting with 270 nm while the other trans derivatives show single emission band at 430 or 460 nm. The dual emissions exhibit different excitation spectra, indicating that two emissive states are different from each other. It is interesting to note that the 340 nm emission of both trans-Ⅰ and trans-Ⅱ is enhanced at the expense of the drastic quenching of the 460 nm emission as the solvent polarity increases. The dual emissions are also observed for both cis-Ⅰ and cis-Ⅱ. The solvent dependence of the fluorescence decay times and quantum yields can be correlated with the solvent and excitation wavelength dependences of the trans→cis photoisomerization quantum yields. These results indicate that the 340 nm emission is originated from the $S_2$ state of the cis-form, and the $S_1$ state is the only singlet excited state presenting a large CT (charge transfer) character to facilitate the photoisomerization.

청국장의 발효기간에 따른 trans-Resveratrol과 비소화성 올리고당의 변화 및 항산화활성 (Antioxidant Activities and Changes in trans-Resveratrol and Indigestible Oligosaccharides according to Fermentation Periods in Cheonggukjang)

  • 몇 가지 식물에서 발견되는 항독성 물질(phytoalexin)인 trans-resveratrol(3,5,4'-trihydroxy-trans-stilbene)은 항암, 항산화효과 및 심혈관질환 등에 효과가 큰 물질로 알려져 있으며, 포도, 땅콩 및 오디 등의 발효 시에 증가되어 그 효과가 커지는 것으로 알려져 있다. 한편 stachyose 및 raffinose 등의 기능성 올리고당은 장내 미생물의 생육을 도와 정장작용을 하는 것으로 알려져 있다. 따라서 본 연구에서는 3품종의 대두를 이용하여 제조한 청국장의 발효 중 trans-resveratrol과 기능성 올리고당의 함량 변화를 조사하였으며, 이에 따른 항산화효과를 알아보려고 하였다. trans-Resveratrol 함량은 원료 콩보다는 발효되었을 때 대체로 발효기간이 길어질수록 그 함량이 높게 나타났다. 특히 48시간 발효 후에는 3품종의 콩에서 늘찬, 대원 및 태광의 순으로 각각 $50.06{\pm}0.82$, $39.04{\pm}0.49$, $34.00{\pm}0.54{\mu}g/g$이었으며, 원료 콩에 비하여 4배 이상 증가하였다. Stachyose 함량은 원료 콩에서 $10.84{\pm}0.42{\sim}13.05{\pm}0.13mg/g$, 증자 직후에는 $11.37{\pm}0.03{\sim}12.05{\pm}0.52mg/g$이었으나, 발효 12시간 후에는 $0.16{\pm}0.01{\sim}0.33{\pm}0.02mg/g$으로 원료 콩에 비하여 97% 이상이 줄어들었으며 발효 24시간 후부터는 전혀 검출되지 않았다. 한편 raffinose의 함량은 원료 콩에서 $2.66{\pm}0.09{\sim}3.54{\pm}0.05mg/g$으로 가장 낮았고, 증자 직후부터 발효 24시간 후에는 $10.61{\pm}0.16{\sim}12.66{\pm}0.17mg/g$으로 약 3~4배가량 증가하였으나 48시간에서는 오히려 $8.28{\pm}0.17{\sim}11.83{\pm}0.44mg/g$으로 감소하는 경향을 보였다. 청국장의 발효기간에 따른 항산화효과는 FRAP, DPPH, ABTS assay 모두에서 증자 직후에는 원료 콩에서보다 다소 낮게 나타났으나 대체로 발효기간이 길어짐에 따라 활성이 높아지는 경향이었다. 이와 같은 경향은 trans-resveratrol의 항산화활성이 DPPH, ABTS와 FRAP assay 등에서 $RC_{50}$ 값이 $4.71{\pm}0.36{\sim}8.46{\pm}0.05{\mu}g/mL$인 것을 미루어 볼 때, 발효기간에 따라 유의적으로 증가한 trans-resveratrol의 영향이 일부 있었을 것으로 생각되었다. 그러나 발효 중 기능성 올리고당(stachyose 및 raffinose)의 변화는 청국장의 항산화효과와는 관련이 없는 것으로 나타났다.

근시에서의 라섹과 TransPRK라섹 수술 후 고위수차 비교 (After Lasek Surgeried and TransPRK Surgeried for Myopia, Comparison of High-order Aberrations)

  • Purpose: The aim of this study is to compare total high order aberrations between surgery which uses TransPRK and general laser assisted sub-epithelial keratomileusis. Methods: The patients who had general laser assisted sub-epithelial and TransPRK laser assisted sub-epithelial by visiting ophthalmic clinic in Ulsan from January 2014 to August 2014 was researched. Results: When comparing total high order aberrations before and after operation, it was found that total high order aberration in general laser assisted sub-epithelial group increased to $0.222{\pm}0.078{\mu}m$ from $0.074{\pm}0.019{\mu}m$ while that in TransPRK assisted sub-epithelial group increased to $0.179{\pm}0.045{\mu}m$ from $0.076{\pm}0.032{\mu}m$. When comparing the increment in total high order aberrations between two groups after surgery, it was found that the increments of TransPRK assisted sub-epithelial group were statistically smaller than that of general laser assisted sub-epithelial group (p<0.05). Conclusions: This study made a comparative analysis of total high order aberrations between general laser assisted sub-epithelial and TransPRK assisted sub-epithelial which was introduced recently and the result showed a significant difference. The findings of this study suggest that TransPRK assisted sub-epithelial can be used widely in simply improving eye sight and quality of vision by reducing an increase in high order aberration caused by orthokeratology surgery. It is expected that TransPRK assisted sub-epithelial will be helpful for improving the understanding quality of eye sight which occurs by several methods of vision correction surgery.
