Identification of druggable genes for multiple myeloma based on genomic information

  • Multiple myeloma (MM) is a hematological malignancy. It is widely believed that genetic factors play a significant role in the development of MM, as investigated in numerous studies. However, the application of genomic information for clinical purposes, including diagnostic and prognostic biomarkers, remains largely confined to research. In this study, we utilized genetic information from the Genomic-Driven Clinical Implementation for Multiple Myeloma database, which is dedicated to clinical trial studies on MM. This genetic information was sourced from the genome-wide association studies catalog database. We prioritized genes with the potential to cause MM based on established annotations, as well as biological risk genes for MM, as potential drug target candidates. The DrugBank database was employed to identify drug candidates targeting these genes. Our research led to the discovery of 14 MM biological risk genes and the identification of 10 drugs that target three of these genes. Notably, only one of these 10 drugs, panobinostat, has been approved for use in MM. The two most promising genes, calcium signal-modulating cyclophilin ligand (CAMLG) and histone deacetylase 2 (HDAC2), were targeted by four drugs (cyclosporine, belinostat, vorinostat, and romidepsin), all of which have clinical evidence supporting their use in the treatment of MM. Interestingly, five of the 10 drugs have been approved for other indications than MM, but they may also be effective in treating MM. Therefore, this study aimed to clarify the genomic variants involved in the pathogenesis of MM and highlight the potential benefits of these genomic variants in drug discovery.

Characterization of Hibiscus Chlorotic Ringspot Virus-Derived vsiRNAs from Infected Hibiscus rosa-sinensis in China

  • Lots of progress have been made about pathogen system of Hibiscus rosa-sinensis and hibiscus chlorotic ringspot virus (HCRSV), however, interactions between H. rosa-sinensis and HCRSV remain largely unknown. Hereon, firstly, HCRSV infection in H. rosa-sinensis from Zhangzhou city of China was confirmed by traditional electron microscopy, modern reverse transcription polymerase chain reaction and RNA-seq methods. Secondly, sequence feature analysis showed the full-length sequence of HCRSV-ZZ was 3,909 nucleotides (nt) in length and had a similar genomic structure with other carmovirus. It contains a 5' untranslated region (UTR), followed by seven open reading frames encoding for P28, P23, P81, P8, P9, P38, and P25, and the last a 3-terminal UTR. Thirdly, HCRSV-ZZ-derived vsiRNAs were identified and characterized for the first time from disease H. rosa-sinensis through sRNA-seq to reveal interactions between pathogen ant plant host. It was shown that the majority of HCRSV-ZZ-derived vsiRNAs were 21 nt, 22 nt, and 20 nt, with 21 nt being most abundant. The 5'-terminal nucleotide of HCRSV-ZZ vsiRNAs preferred U and C. HCRSV-ZZ vsiRNAs derived predominantly (72%) from the viral genome positive-strand RNA. The distribution of HCRSV-ZZ vsiRNAs along the viral genome is generally even, with some hot spots and cold spots forming in local regions. These hot spots and cold spots could be corresponded to the regions of stem loop secondary structures forming in HCRSV-ZZ genome by nucleotide paring. Taken together, our findings certify HCRSV infection in H. rosa-sinensis and provide an insight into interaction between HCRSV and H. rosa-sinensis and contribute to the prevention and treatment of this virus.

Effects of Gamiolnyeo-jeon on Lipid Metabolism and Blood Glucose Level in db/db Mice (가미옥녀전(加味玉女煎)이 db/db 마우스 당뇨(糖尿)모델에서 지질대사(脂質代謝)와 항당뇨(抗糖尿) 효능(效能)에 미치는 영향(影響))

  • Objectives : Abnormal regulation of glucose and impaired lipid metabolism that result from a defective or deficient insulin are the key etiological factor in type 2 diabetes mellitus. The our study investigated the effects of Gamioknyeo-jeon (GO) on blood glucose and lipid metabolism improved by it in db/db mice (a murine model of type 2 diabetes mellitus).Methods : The animals were divided into 3 groups: Normal groups were not-treated C57BL/6 mice; Control groups were treated orally with DW in db/db mice; GO groups were treated orally with GO (200 ㎎/㎏/day) in db/db mice. After mice were treated with GO for 5 weeks, we measured AST, ALT, creatinine, BUN, body weight, food intake, blood glucose, insulin and lipid levels (total cholesterol, HDL cholesterol, and LDL cholesterol and atherogenic index(AI) and cardiac risk factor(CRF).Results : Serum AST, ALT, creatinine, BUN levels were not changed by GO do not show any toxic effects. GO groups were decreased in body weight, food intake and blood glucose level among compared to Control groups. Also, GO groups were found to have atherogenic Index and cardiac risk factor as well as lipid metabolism improvement (total cholesterol and LDL cholesterol decrease). Finally, GO groups were increased the insulin compared to Normal and control groups.Conclusions : We suggest that GO may have the control effects of diabetes mellitus by improving blood glucose control and lipid metabolism.

  • Obstetrics and Gynecology include gynecology which is concerned with the treatment for the disease based on physiology and pathology of women, and obstetrics which is concerned with pregnancy delivery. These obstetrics and gynecology can be said to da-te from the birth of human beings. This pap-er has carried on the studies about the deve-loping process of obstetrics and gynecology of Ming and Qing age. The results of this study are as follows: In Ming age, Many Obstetrics and Gynecology books including "Nukecuoyao"("女科撮要"), "Xiaozhufurenliangfang"("校注婦人良方"), "Wanshifurenke"("萬氏婦人科") and "Nukezhingzhizhunsheng"("女科證治准繩") were published Distinction in Ming age we-re equal development on theory and clinic t-aking a serious view of the differentiation of symptoms and signs, fashion of medicine th-ought of reactionism under the influence of "lixue"(理學). The refore Obstetrics and Gyn-ecology were influenced by these points. And for this example, as treatment contents on "Xiaozhufurenliangfang"("校注婦人良方") and the theory about "fetuse-energy"(胎氣) in "Furengui"("婦人規"), theoretic system with a view point's change of women's disease were established on Obstetrics and Gynecology. But it was restricted on a field of diagnosis under the influence of feudal "lixue"(理學), so the the number of obstetrics doc-tors who were mostly men at that time had fallen greatly and maternity who were short of expert medical knowledge appeared. In Qing age, an explosive increase in po-pulation called greater demand on medicine book and generation after generation extre-mely much Obstetrics and Gynecology books including "NukeChanhoubian"("女科産後編"), "Yetianshinuke"("葉天士女科"), and "Shenshinukejiyao"("沈氏女科輯要") were p-ublished, and it is studied that application of "eight extra-channel"(奇經八脈) theory and the study of drug attributive on extra-chan-nel were progressed. Besides, it is studied that existing traditional Obstetrics and Gyn-ecology changed newly under the influence of the school of combination of traditional Chinese medicine and western medicine which was appeared in the late Qing age.

A Study on Treatment for Symptoms of Hypoadrenocorticism and Indicant of Kidney Jeonggyeok(腎正格) (부신피질기능저하증(副腎皮質機能低下症)의 변증논치(辨證論治)와 신정격(腎正格) 적응증(適應症)에 대한 고찰(考察))

  • Objectives : The comparison of symptoms of hypoadrenocorticism with symptoms according to spleen and kidney show how many points of similarity they are. And by analysis of symptoms of hypoadrenocorticism it was examined that Kidney Jeonggyeok help to cure many different symptoms including symptoms of hypoadrenocorticism. Methods : The symptoms of hypoadrenocorticism were compared and analyzed according to standards of the book of korean traditional medical pathology. First, this study was designed to show to found out how many symptoms of hypoadrenocorticism belong to symptoms of kidney qi-deficiency and slpeen qi-deficiency and how similar they are. Second, the symptoms of kidney qi deficiency were compared with the indicant of kidney Jeonggyeok suggested by two of korean traditional doctors to find out kidney Jeonggyeok can help to cure symptoms of kidney qi deficiency. And then on the basis of those, the symptoms of hypoadrenocorticism were compared with the symptoms of kidney qi deficiency. Results : The hypoadrenocorticism seems to be kidney yang(陽) deficiency. But because hypoadrenocorticism doesn't have body temperature decline, symptoms of hypoadrenocorticism are more similar symptoms of kidney qi deficiency than symptoms of kidney yang deficiency. The symptoms of hypoadrenocorticism seems to be correlated with the functions of spleen. But because of vomiting and pigmentation(skin and mucous membrane), they seem to have less to do with the functions of spleen than the functions of kidney. The comparison analysis of indicant of Kidney Jeonggyeok and symptoms of kidney qi deficiency shows that acupuncture stimulation help to cure kidney qi deficiency and hypoadrenocorticism. Conclusions : Symptoms of hypoadrenocorticism are expected to be more similar symptoms of kidney qi deficiency. And Kidney Jeonggyeok is helpful for treatment of Kidney qi deficiency and hypoadrenocorticism.

Two Cases of Endobronchial Actinomycosis that were Cured by Operation and Short Term Antibiotics Therapy (수술 및 단기간의 항생제 투여로 완치된 기관지 방선균증 2예)

  • Endobronchial actinomycosis is a rare chronic suppurative granulomatous infection of the bronchus, and this is sometimes related with a foreign body or a broncholith. The traditional treatment of endobroncial actinomycosis is intravenous antibiotics for 2 to 6 weeks and then oral antibiotics therapy for 6 to 12 months. We report here on 2 cases of endobronchial actinomycosis that were associated with a broncholith and a foreign body, respectively. Surgery followed by short term antibiotics therapy for only 20 days and 34 days, respectively, was effective as treatment for the endobronchial actinomycosis in our cases. After treatment, there were no complications or recurrence during the following period. We suggest that short term antibiotics therapy combined with a surgical operation might be effective as treatment for primary endobronchial actinomycosis, and especially when this illness is combined with a foreign body or a broncholith, as compared with traditional long term antibiotic therapy.

An Empirical Study on Traditional, Complementary and Alternative Medicine Usage among Malaysian Cancer Patients

  • Usage of traditional, complementary and alternative medicine (TCAM) has gained popularity over the past few years. However, very little is known about TCAM use among Malaysian cancer patients. This study aimed to identify the determinants of TCAM usage among cancer patients with determination of relationships between demographic factors, patient satisfaction with conventional treatment, knowledge on TCAM and healthcare professional influence. Patient's perceptions towards TCAM were also determined. A simple random convenient sampling method was used to recruit 354 patients from Hospital Kuala Lumpur between February to April 2013. All were directly interviewed with a structured questionnaire. In this study, 172 respondents were TCAM users. There was no significant differences between demographic background of respondents in the usage of TCAM. Minimal correlation was found between patient satisfaction with the conventional treatment and usage of TCAM (r=0.091). A poor correlation was found between healthcare professional's influence and TCAM usage (r=-0.213) but the results suggested that increase in influence would decrease TCAM usage. Patient TCAM knowledge correlated negatively with the TCAM usage (r=-0.555) indicated that cancer patients are less likely to use TCAM when they have more TCAM knowledge. Healthcare professionals should be fully equipped with the necessary TCAM knowledge while maintaining patient satisfaction with the conventional treatment. They should also intervene on patient TCAM usage where a potential drug interaction or a harmful adverse event can occur.

A study on fundamental basis of four-constitution medicine from the principle of Yeokgyeong (사상의학(四象醫學)의 역철학적(易哲學的) 기초(基礎)에 관한 연구)

  • This study searched fundamental basis of four-constitution medicine from the principle of "Yeokgyeong(易經)" that is scientific foundations of Dongmu(東武) Ijema(李濟馬)'s four-constitution medicine based on system of "Yeokgyeong(易經)" and looked into the principle of our-constitution medicine ontologically. That is to say, the translation of five phase(五行) that represented in "Hwangjenaegyeong(黃帝內經)" regulated that substance of five phase is spleen-earth(脾土). But four-constitution medicine mentions the substance as heart-earth(心土) in place of spleen-earth. Because of it's standpoint, the differences on meanings between spleen-earth and heart-earth on the substance of five-phase becomes motive to interpret scientific system of four-constitution medicine fundamental wrongly. For that reason, the research of this title is needed. The results was summarized as follows. First, in ontological view point of structure of four-constitution, five phase is substance and phenomenon, in other words it includes earth of unrevealed substance and wood, fire, metal and water of self-manifestation of existence. Second, in axiological view point, the four-constitution represent principles and contents of four virtues of human nature. And so the innate four virtues ontologically based on four-constitution of heaven. Therefore a human being is endowed innately benevolence, courtesy, justice, intelligence of four virtues. Third, the concept of greater and lesser of Eum(陰, yin) and Yang(陽, yang) in Dongmu(東武)'s four-constitution medicine is four-constitution in "Yeokgyeong(易經)". Greater principle(太極) and four-constitution is a relation of substance and phenomenon. Fourth, the origin and structure of four-constitution medicine includes the structure and principle of natural philosophical Eumyang and four-constitution, the human-centric theory and sciences of human nature and natural laws and medical experience of traditional oriental medicine and medical principle.

Britanin Suppresses IgE/Ag-Induced Mast Cell Activation by Inhibiting the Syk Pathway

  • The aim of this study was to determine whether britanin, isolated from the flowers of Inula japonica (Inulae Flos), modulates the generation of allergic inflammatory mediators in activated mast cells. To understand the biological activity of britanin, the authors investigated its effects on the generation of prostaglandin $D_2$ ($PGD_2$), leukotriene $C_4$ ($LTC_4$), and degranulation in IgE/Ag-induced bone marrow-derived mast cells (BMMCs). Britanin dose dependently inhibited degranulation and the generations of $PGD_2$ and $LTC_4$ in BMMCs. Biochemical analyses of IgE/Ag-mediated signaling pathways demonstrated that britanin suppressed the phosphorylation of Syk kinase and multiple downstream signaling processes, including phospholipase $C{\gamma}1$ ($PLC{\gamma}1$)-mediated calcium influx, the activation of mitogen-activated protein kinases (MAPKs; extracellular signal-regulated kinase 1/2, c-Jun $NH_2$-terminal kinase and p38), and the nuclear factor-${\kappa}B$ ($NF-{\kappa}B$) pathway. Taken together, the findings of this study suggest britanin suppresses degranulation and eicosanoid generation by inhibiting the Syk-dependent pathway and britanin might be useful for the treatment of allergic inflammatory diseases.

Effects of Herbal Medicine on Tibia Bone Changes in Ovariectomized Rats and SAM P6 Mice (한약이 난소제거 흰쥐와 SAM P6 생쥐의 골소주에 미치는 영향)

  • The effects of herbal medicine on travecular bone area were studied using ovariectomized rat as an animal model of Type I osteoporosis and SAM P6 as that of Type II. We counted red blood cells(RBC), hemoglobin(Hb), and hematocrit(Hct) using Couter`sR method. Each traditional boiling water extract of Achyrathis Radix, Psoraleae Fructus, Rehmanniae Radix Preparat, and Cornii Fructus and a systemic water extract of Astragali Ractiex was given 5g/kg/day, p.o., for 30 days in a group of 4-5 ovariectormized rats. One ml of blood was taken by tail vein at day 0, 7, 14, 21, and 30 days after administration of the extract. The traditional hot water extract of Cervi parvumn Corni (Cervi) was given the same dose as described above for 14 days in a group of 10 SAM P6 mice and systemic water extract of Astragali Radix was administered as the same dose as above for 30 days in 10 SAM P6 mice. Travecular bone area was measured 5 mcm decalcified and stained thin bone slice by image analysis using a digitalizer. In Type I, ovariectomized rats, administration of Astragali Radix, Rhemanniae Radix Preparat, and Corni Fructus decreased in RBC, Hb and Hct. In Type II, administration of Cervi increased in RBC and Hct and that of Astragali Radix was also elevated RBC. In Type I, any administration of herbal medicine used in this study did not elevate travecular bone area significantly except Corni Fructus showed a trend of increase in travecular bone area. However, Type II, Cervi and Astragali Radix increased in both mean and total travecular bone area. Thus, there are significant difference in response of herbal medicine in different types of osteoporosis.
