합성세제 및 계면활성 성분의 독성학적 연구 (Toxicological Studies on Surfactants and Synthetic Detergents)

  • Synthetic detergents and surfactants are in widespread usage as household and industrial detergents. Potential human toxic hazard arises following percutaneous absorption or oral ingestion of solution residues from kitchen and feeding utensils, fruits, and vegetables and contaminated water supplies. A toxicological investigations was performed with the synthetic detergents and surfactants [linear alkyl benzene sulfonate (LAS), ${\alpha}$-olefin sulfonate (AOS), sodium lauryl sulfonate (SLS), sodium lauryl ester sulfonate (SLES)], In acute toxicity, agents were administered subcutaneously into ICR mice. In acute study, after lowering of spontaneous motility, respiratory failure, death appeared, vomitting was often associated with salivation and or retching. No sex difference was observed in LD$\sub$50/ of mice. In subacute toxicity, agents were administered orally into SD rats. Body weight increase was suppressed and there was no adverse effect on food and water consumption. The weight of organs were not changed by agents as compared with control group. No specific change was observed in biochemical and hematologicalor data.

Regulatory Aspect of Risk Assessment and Management

  • Risk assessment is useful tool making good decisions on the risks of certain hazardous com-pound and suggests safe margin through scientific process using toxicological data, statistical tool, exposure value and relevant variants. The goal of risk management is to protect the public health from hazardous compound based on result of risk assessment having reality. For the suggestion of exact man-aging information, risk assessment must be designed to represent a "plausible estimate" of the exposure to the individuals and to minimize uncertainty. Risk assessment methodology and knowledge are expected to change more rapidly than before and up-to-date methodology should be applied in regulatory aspects through the Agency. For the useful application of risk assessment, the communication between the risk assessor and the risk manager is needed before the initiation of the risk assessment and upon its completion. Generally, the risk assessment itself as a practical tool in the regulatory decision making process would be regarded with social economic impact.ic impact.

랫드에서 계피유래활성물질(CB-PH)의 경구투여에 의한 4주간 반복투여독성 시험 (Four Weeks Repeated Toxicity Study of 2-o-Benzoylcinnamaldehyde(CB-PH) by Oral Administration in Sprague-Dawley Rats)

  • Although 'Cinnamon' has been widely used for the food and biophamacy in the world, it's toxicity was not screened completely. Major component of 'cinnamon' is CB-OH and CB-PH. CB-PH has been reported to have antimutagenic effect. To investigate the toxicity of 2-o-Benzoylcinnama-Idehyde (CB-PH), repeated dose (4 weeks) oral toxicity test performed in SD rats. Results of repeated dose oral toxicity tests for 4 weeks (CB-PH; 500, 1000, 2000 mg/kg/day) suggested that the CB-PH treated group showed no significant toxicological findings with body weights, organ weights, hematological and histopathological findings. Therefore, these data indicated that the maximum tolerated dose of CB-PH was 2000 mg above/kg/day in the rats.

  • Mycotoxins are a group of toxicants giving a risk potential to human health in connection with the daily food intake. Food commodities once contaminated with mycotoxins can not be detoxified by any economic means and prevention was suggested as the only measure. In order to minimize the economic loss and health hazard posed by mycotoxins and toxicoses, systematic and toxicological studies on the subject should be undertaken. Most reports in Korea were concentrated on the mycological studies of relatively easy techniques and the confirmation or quantitation of mycotoxins was rarely done. Research topics to be undertaken in future may be exemplifid below: (1) Establishing assay methods for individual or multi-residue of mycotoxins (2) Monitoring of mycotoxins for suspicious food or feed samples in Korea (3) Epidemiological survey of mycotoxicoses (4) Etiological survey of disease outbreaks associated with mycotoxins (5) Accumulation of testing method and data on the toxicity of mycotoxins (6) Legal regulation to control mycotoxins and development of their detoxification / elimination methods

Differences in Gene Expression Profiles Reflecting Differences in Drug Sensitivity to Acetaminophen in Normal and Transformed Hepatic Cell Lines In vitro

  • Acetaminophen (APAP) overdose is known to cause severe hepatotoxicity mainly through the depletion of glutathione. In this study, we compared the cytotoxic effects of APAP on both a normal murine hepatic cell line, BNL CL.2, and its SV40-transformed cell line, BNL SV A.8. Gene expression profiles for APAP-treated cells were also obtained using microarray and analyzed to identify differences in genes or profiles that may explain the differences of susceptibility to APAP in these cell lines. These two cell lines exhibited different susceptibilities to APAP (0-$5,000{\mu}M$); BNL SV A.8 cells were more susceptible to APAP treatment compared to BNL CL.2 cells. A dose of $625{\mu}M$ APAP, which produced significant differences in cytotoxicity in these cell lines, was tested. Microarray analysis was performed to identify significant differentially expressed genes (DEGs) irrespective of APAP treatment. Genes up-regulated in BNL SV A.8 cells were associated with immune response, defense response, and apoptosis, while down-regulated genes were associated with catalytic activity, cell adhesion and the cytochrome P450 family. Consistent with the cytotoxicity data, no significant DEGs were found in BNL CL.2 cells after treatment with $625{\mu}M$ APAP, while cell cycle arrest and apoptosis-related genes were up-regulated in BNL SV A.8 cells. Based on the significant fold-changes in their expression, a genes were selected and their expressions were confirmed by quantitative real-time RT-PCR; there was a high correlation between them. These results suggest that gene expression profiles may provide a useful method for evaluating drug sensitivity of cell lines and eliciting the underlying molecular mechanism. We further compared the genes identified from our current in vitro studies to the genes previously identified in our lab as regulated by APAP in both C57BL/6 and ICR mice in vivo. We found that a few genes are regulated in a similar pattern both in vivo and in vitro. These genes might be useful to develop as in vitro biomarkers for predicting in vivo hepatotoxicity. Based on our results, we suggest that gene expression profiles may provide useful information for elucidating the underlying molecular mechanisms of drug susceptibility and for evaluating drug sensitivity in vitro for extrapolation to in vivo.

독성동태 모델과 데이터의 해석 (Toxicokinetic Models and Data Interpretation)

  • Toxicokinetic studies are intended to provide critical evaluation of drug disposition at toxico-logical doses and help understand the relationship between blood or tissue levels and the time course of toxic events. Relatively high dose levels wed in toxicokinetics, compared to pharmacokinetics, complicates absorption, protein binding, metabolism and elimination processes. In this mini review, frequently wed toxicokinetic models such as linear compartment models, physiological models, and nonlinear kinetic mod-ec are introduced. In addition, optimization of toxicokinetic studies, their role in the drug development process, and prediction oj human toxicokinetics based on animal data by interspecies scaling are briefly discussed.

Establishment of Acceptable Daily Intakes (ADIs) and Risk Assessment for Ephedrine, Menichlopholan, Anacolin, and Etisazole Hydrochloride

  • BACKGROUND: Prior to implementing a positive list system (PLS), there is a need to establish acceptable daily intake (ADI) and maximum residue limit (MRL) for veterinary drugs that have been approved a few decades ago in South Korea. On top of that, chronic dietary exposure assessment of veterinary drug residues should be performed to determine whether the use of these veterinary drugs would cause health concerns or not. METHODS AND RESULTS: To establish the ADI, the relevant toxicological data were collected from evaluation reports issued by international organizations. A slightly modified global estimate of chronic dietary exposure (GECDE) model was employed in the exposure assessment owing to the limited residual data. Therefore, only the ADI of ephedrine was established due to insufficient data for the other veterinary drugs. Thus, instead of ADI, the threshold of toxicological concern (TTC) value was used for the other drugs. Lastly, the hazard index (HI) was calculated, except for etizazole hydrochloride, due to the potential of mutagenicity. CONCLUSION(S): The HI values of ephedrine, menichlopholan, and anacolin were found to be as high as 6.4%, suggesting that chronic dietary exposure to the residues from these uses was unlikely to be a public health concern. Further research for exposure assessment of veterinary drug residues should be performed using up-todate Korean national health and nutrition examination survey (KNHANES) food consumption data. In addition, all relevant available data sources should be utilized for identifying the potentials of toxicity.

Sprague-Dawley 랫드를 이용한 발생독성시험의 기초자료연구 (Historical Control Data for Developmental Toxicity Study in Sprague-Dawley Rats)

  • The background control data were compiled from rat developmental toxicity studies con-ducted at Toxicology Research Center, KRICT during the 1993-1999 period. These data were assembled in order to provide background in formation for the maternal and fetal data collected in 13 developmental toxicity studies using Sprague-Dawley rats. A total of 325 mated females were used in these studies during the seven-year period and overall pregnancy rate of these females was 93.8%. The present background control data included body weights, food consumption, hematological values, and organ weights of pregnant females, caesarean section data, and fetal examination data. These data can be used not only as a historical database for the meaningful interpretation of data from reproductive and developmental toxicity studies, but also as a contribution to biological characterization oj Sprague-Dawley rats.

식품첨가물의 독성자료 고찰과 ADI 평가지침 - 소포제 Polydimethylsiloxane를 사례로 - (Evaluation of Toxicological Data on Food Additives and Guideline for ADI establishment - Polydimethylsiloxane as emulsifier -)

  • 본 연구는 최근 JECFA에서 이슈화된 식품첨가물의 평가 정보를 파악하여 국내에서 선제적 위해관리에 활용할 수있도록 하기 위하여, 제 69차 JECFA 회의에 의제로 선정된 PDMS의 독성자료 검토 및 monograph 작성 내용에 대하여 소개하고자 수행되었다. PDMS의 독성자료 검토는 'Guidelines for the preparation of toxicological working papers for the Joint FAO/WHO Expert Committee on Food Additives'에 근거하였고, 평가 의뢰 시 제공된 자료 및 추가적으로 검색된 자료를 활용하였다. PDMS는 거품제거와 뭉침 방지를 위하여 간장, 제당, 당밀, 유제품, 쨈, 과즙제품, 두부 등의 제조 시 첨가물로 사용된다. 15,000-20,000 Da 이하의 분자량을 가지는 PDMS가 혈관을 통해 좀 더 쉽게 흡수될 수 있다는 문제가 제기되어, 기존에 설정된 ADI 0-1.5 mg/kg bw/day의 검토가 요구되었다. 이에 따라 흡수, 분포, 배설에 대한 자료와 독성시험자료의 검토가 수행되었다. 검토 결과 PDMS는 체내로 흡수가 거의 이루어지지 않고 대부분 배설되었으며, 급성독성, 아만성독성, 만성독성시험에서 특이적인 전신증상이 관찰되지 않았다. 그러나 급성독성, 아만성독성 및 만성독성시험에서 안구독성이 일관되게 관찰되었으며 이 안구독성의 기전이 확실하게 밝혀지지 않았다. 따라서 이전에 설정된 ADI 0-1.5mg/kg bw/day 값에 안구독성에 대한 추가적인 safety factor 2를 적용하여 temporary ADI 0-0.8 mg/kg bw/day로 재설정하였다. 또한 독성시험에서 관찰된 안구독성의 타당성을 설명하기 위한 추가적인 연구결과를 2010년까지 제공하도록 요청하였다.

Improving International Access to the IARC Monographs Database with Linkage to other Sources of Information

  • The IARC Monographs Programme on the Evaluation of Carcinogenic Risks to Humans has reviewed, summarized and evaluated 869 environmental agents and exposures as oj June 2000. This large collection includes all relevant published epidemiological data on cancer in exposed humans and results of bioassays for carcinogenicity in experimental animals. Since 1986. cancer data have been systematically supplemented by summaries of other toxicological data that are relevant to assessments of carcinogenic hazard. These include summaries qf genetic and related effects of chemicals. which have been prepared as Genetic Activity Profiles (GAP) by the U.S. EPA in collaboration with IARC. As the Mono-graphs have proved increasingly valuable and influential worldwide. they have evolved into an encyclopedia on environmental carcinogenic risks to humans. However. the Monographs have historically been prepared only as printed books with limited distribution. and the Monographs Programme has needed to adjust to expectations oj wider availability. Since 1998 the evaluations and summaries have been globally accessible by Internet from IARC (http://www.iarc.fr) and the GAP profiles by Internet from EPA (http://www.epa.gov/gapdb/). with the two web sites linked. Improved EPN/ARC GAP database and software. GAP2000. now link GAP profiles directly to the appropriate IARC web pages for summaries of evaluations of a given compound and its overall IARC classification. During the year 2000. by means of optical character recognition (OCR) technology the entire series of IARC Monographs is being converted to an electronic version. The first edition is now available commercially in CD-ROM format and will soon become available on-line at .
