Resibufogenin induces cardiac arrhythmia

  • Resibufogenin is a single compound isolated from the skin venom gland of the toad (Bufo bufo gargarizans cantor). Formulations containing toad venom have been widely used as complementary and alternative medicines. However, like digitalis, resibufogenin possesses both pharmacological and toxicological activities. Our previous data indicated that resibufogenin induces electro-toxicity, including delayed afterdepolarization and triggered arrhythmias at high concentration, both in cardiac fiber in vitro and in beating heart in vivo.

  • T-2 toxin was given to rats(Spargue Dawley) with a 4mg/kg and 2mg/kg dose p.o. Data was based on the T-2 toxin 4mg/kg treated group except for counting of acidophil bodies. GPT activities increased significantly from 0.5 to 10 hours. GOT activities were also increased at 1 and 5 hours significantly and the relative weights of liver were increased at 0.5 and 5 hours significantly. There were slight necrotic foci in microscopic observation. There was dose-dependent trend for the frequency of the acidophil body between the 4mg/kg treated group and the 2mg/kg treated group.

  • Morinda citrifolia, also known as Great Morinda, Indian Mulberry, or Noni, is a plant belonging to the family Rubiaceae. A number of major chemical compounds have been identified in the leaves, roots, and fruits of the Noni plant. The fruit juice is in high demand in alternative medicine for different kinds for illnesses such as arthritis, diabetes, high blood pressure, muscle ached and pains, menstrual difficulties, headache, heart diseases, AIDS, gastric ulcer, sprains, mental depression, senility, poor digestion, arteriosclerosis, blood vessel problems, and drug addiction. Several studies have also demonstrated anti-inflammatory, antioxidant and apoptosis-inducing effects of Noni in various cancers. Based on a toxicological assessment, Noni juice was considered as safe. Though a large number of in vitro, and, to a certain extent, in vivo studies demonstrated a range of potentially beneficial effects, clinical data are essentially lacking. To what extent the findings from experimental pharmacological studies are of potential clinical relevance is not clear at present and this question needs to be explored in detail before an recommendations can be made.

A Limited Survey of Fumonisin $B_{1}$ Content of Domestic and Imported Corns in Korea

  • Fumonisin $B_{1}$ ($FB_{1}$) is hepatotoxic in all species, and initiator and promotor for hepatocarcinogenesis produced mainly by Fusarium moniliforme. This fungus is commonly natural contaminant of corn and other grains worldwide, and has been associated with animal and human diseases. In these study, natural occurrence of $FB_{1}$ was surveyed in 30 healthy domestic corn kernels in Chonbuk and Kangwon province, harvested in 1994 and intended for human consumption, and 15 imported American and Chinese samples of each, collected from different ship-loading at Inchon harbor for animal foodstuffs. $FB_{1}$ contents were measured by high-performance liquid chromatography (HPLC) with a fiourescence detector. The data revealed that 2 out of 12 corn kernels from Kangwon province with 1.1 and 0.7 ppm, and 2 out of 18 corns from Chonbuk province with 0.5 and 1.3 ppm, respectively. However, there was no detection of $FB_{1}$ in imported corn samples, even though those were visibly moldy.

  • Lethal dose 50% ($LD_{50}$) has been commonly used as a parameter for the estimation of acute toxicity not only in animal experiment, but also in human study. Several methods to estimate $LD_{50}$ had been introduced, but Spearman-Karber and Berens-Karber method have been widely used due to their relative convenience and accuracy. However, $LD_{50}$ values estimated from the two methods showed inconsistency and variation depending on the characteristics of mortality data. In this study, the two methods were comparatively investigated in terms of accuracy and stability for the estimation of $LD_{50}$.

  • Fifty chemicals are currently classified as human carcinogens based on epidemiologic and animal data. Humans are daily exposed to them from various sources of exposure via inhalation, dermal contact and oral ingestion. To reduce cancer risk to man, these human carcinogens should be appropriately regulated and monitored environmentally or biologically for routine human cancer risk assessment. A number of mathematical risk assessment models have been introduced, but any realistic and relevant model system is not available for humans. A mechanistic process for human cancer risk assessment was comprehensively reviewed and problems were also discussed. Here, a new conceptual approach using epidemiology and biological human monitoring was suggested for the most relevant method to study human cancer risk assessment.

Safety Evaluation of Polyethylene Glycol (PEG) Compounds for Cosmetic Use

  • Polyethylene glycols (PEGs) are products of condensed ethylene oxide and water that can have various derivatives and functions. Since many PEG types are hydrophilic, they are favorably used as penetration enhancers, especially in topical dermatological preparations. PEGs, together with their typically nonionic derivatives, are broadly utilized in cosmetic products as surfactants, emulsifiers, cleansing agents, humectants, and skin conditioners. The compounds studied in this review include PEG/PPG-17/6 copolymer, PEG-20 glyceryl triisostearate, PEG-40 hydrogenated castor oil, and PEG-60 hydrogenated castor oil. Overall, much of the data available in this review are on PEGylated oils (PEG-40 and PEG-60 hydrogenated castor oils), which were recommended as safe for use in cosmetics up to 100% concentration. Currently, PEG-20 glyceryl triisostearate and PEGylated oils are considered safe for cosmetic use according to the results of relevant studies. Additionally, PEG/PPG-17/6 copolymer should be further studied to ensure its safety as a cosmetic ingredient.

Inducible Nitric Oxide Synthase Inhibitor form Mela azedarach var. Japonica

  • In bioassay-guided search for inducible nitric oxide synthase (iNOS) inhibitory compounds from higher plants of South Korea, two $\beta$-carboline (2) have been isolated form the cortex of Melia azedarach var. japonica. The structures of these compounds were elucidated on the basis of spectroscopic data. Compounds 1 to 2 showed marked inhibitory activity of iNOS on LPS-and interferon-${\gamma}$-stimulated RAW 264.7 cells.

STUDY ON THE MERCURY CONTENTS IN KOREAN SCALP HAIRS

  • The average value of total mercury contents in scalp hair of Korean male was found to be 1.66+1.10 ppm (S.D.) and that of methylmercury was 1.02+0.72 ppm (61.0% of the total mercury). In case of total mercury contents of female was 1.06+0.46 ppm and that of methylmercury was 0.51+0.27 ppm (48.4% of total mercury level). When compared with data reported by Japanese, the levels were about half. The levels of mercury in scalp hair of male subjects were significantly different with their occupations, but in case of female, the variation was not so much.

Solid-Phase Extraction of Sulfamerazine from Shrimp Residue and Determination by Reversed Phase High Performance Liquid Chromatography

  • The focus of this study was to investigate the suitable analytical methods for measurement of sulfamerazine and its metabolite in shrimp hepatopancreas and tail tissue, in addition to the methods for the optimization of solid-phase extraction cartridge conditions and the elucidation of sulfamerazine concentrations in aqueous buffer using HPLC with UV and EC detectors. Compared with UV detector the EC detector appears to be 10 times more sensitive than that of the UV detector. After the shrimp was exposed to 10 ppm sulfamerazine, the accumulation levels of sulfamerazine and its metabolite in tail tissue, which is edible portion, were considerably lower than 0.1 ppm. The data indicate that sulfamerazine continues to be a candidate for use at levels of sulfamerazine concentration used in aquaculture of shrimp.
