Anti-obestic Effects of Bangpoongtongsungcankamibang(BTSK) in 3%3-L1 preadipocyte cells and the Lipid Metabolism of Obesity induced Rats (방풍통성산가미방(防風通聖散加味方)이 비만유도(肥滿誘導) 백풍(白嵐)와 지방세포(脂肪細胞) 분화(分化)에 미치는 영향(影響))

  • Objective : This experimental study was designed to determine the anti-obestic effects of Bangpoongtongsungcankamibang(BTSK). Method : In vitro, BTSK extracts of various concentration (50,100, 200 $200{\mu}g/ml$)were added examination. The protein and mRNA expression of $C/EBP{\alpha}$ and $PPAR{\gamma}$ receptor was measured by western blot assay and RT-PCR. In vivo, BTSK extracts of various concentration (100, $200{\mu}g/ml$) were orally administered to induced hyperlipidemic rats by poloxamer-407 for consecutive four weeks and serum triglyceride, total cholesterol were measured. This method applied to induced hyperlipidemic rats by triton WR-1339, too. Obesity induced rats by the high fat-diet for six weeks were orally administered BTSK extracts of various concentration (100, $200{\mu}g/ml$) and serum triglyceride, total cholesterol, LDL-cholesterol, triglyceride, LDL-cholesterol, triglyceride, HDL-cholesterol, hydroxy radical, superoxide dismuatse activity were measured. Results : I. In vitro 1. The 3T3-L1 cells' differentiation was significantly decreased by BTSK. 2. expression of $C/EBP{\alpha}$ and $PPAR{\gamma}$ was was significantly decreased by BTSK. II. In vitro 1. BTSK significantly reduced serum triglyceride, total cholesterol, contents in poloxamer-407 treated rat. 2. BTSK significantly reduced serum triglyceride, contents in Triton WR-1339 treated rat. But Total cholesterol did not show a significant change. 3. BTSK significantly reduced body weight gain of rat and adipose tissue mass of rats and serum triglyceride, LDL-cholesterol, contents and significantly increased HDL-cholesterol, HTR(HDL-cholesterol/Total-cholesterol) in rats with obesity induced by the high fat-diet. 4. BTSK reduced blood lipid peroxide, hydroxy radical and increased superoxied dismuatse(SOD) activity.

The Hypocholesterolemic Effect of Platycodi radix Saponin in Rats Fed a Hypercholesterol Diet (길경 saponin이 고콜레스테롤식이를 공급한 흰쥐의 지질대사에 미치는 영향)

  • The objective of this study is to find the effect of improving hyperlipidemia through Platycodi radix saponin. A hypercholoesterol diet and Platycodi radix saponin methanol extract were supplemented to the 6 week old male Spargue Dawley rats for four weeks. Results showed that the serum total cholesterol, triglyceride and LDL-cholesterol level decreased in the group supplemented with Platycodi radix saponin methanol extract(p<0.05), however, the serum total lipid and HDL-cholesterol did not show any difference from the control group according to the supplement of Platycodi radix saponin methanol extract. In the group supplemented with Platycodi radix saponin methanol extract, the total cholesterol decreased by 20%, triglyceride by 36% and LDL-cholesterol by 25% respectively, and there was no difference according to the level of Platycodi radix saponin methanol extract. AI(atherogenic index) and CRF(cardiac risk factor) were low in the group supplemented with Platycodi radix saponin methanol extract. Total lipid, total cholesterol and triglyceride in liver and feces were significantly low in the group supplemented with the Platycodi radix saponin methanol extract compared with the control group(p<0.05). According to this result, it is expected that the Platycodi radix saponin methanol extract supplemented in the hypercholesterol diet can lower atherosclerosis, the cause of cardiovascular disease, by reducing not only serum cholesterol and triglyceride but also liver cholesterol and triglyceride.

  • In Med Dept. Daehan Education Insurance, Total 690 cases(male 376 cases, female 314 cases) were inspected to study the change of serum lipid value. The objects of investigation were total cholesterol, HDL cholesterol, triglyceride, phospholipid and total lipid value. The results are as follows: 1) In the normal subjects, with the relation of serum lipid value and the age. triglyceride, phospholipid and total lipid value showed the increasing tendency by 70 age, but total cholesterol value and HDL cholesterol value showed the increasing tendency by 59 age and the decreasing tendency after 59 age. 2) In the normal subject the difference of serum lipid value between sexes did not make the significant difference of level statistically in all items which can be inspected. 3) The weight change and serum lipid value are very significantly related. Total cholesterol, HDL cholesterol and total lipid value are high in female if the weight is light. But they are high in male the weight is heavy phospholipid value is opposite to the above result. Triglyceride value is high in male regardless the weight, but the significant difference happened in the high weight group. 4) As serum lipid value of hypertension patients is compared with that of the normal subjects, HDL cholesterol, triglyceride, phospholipid and total lipid value are significantly high. 5) The doubtful group of the fatty infittration of liver among the hypertension pafients have very significant difference in serum lipid value compared with not only the value of the normal subject but also that of the obesity group. 6) The doubtless group of the fatty infitration of liver among the hypertension patients have the significant difference in all serum lipid value compared with that of the normal subject, but compared with the obesity group males had the significant difference of HDL cholesterol value and both sexes had the significant difference in total lipid value, but had no significant difference in total cholesterol, triglyceride and phosprolipid value. 7) The ratio of total cholesterol per HDL cholesterol had the significant difference in the doubtful hypertension patients of the fatty infittration of liver and the relative weight, but it had no significant difference compared with the normal subject in the doubtless hypertension patients.

Effect of Lecithin Intake on Lipid Metabolism and Antioxidative Capacity in Rats Fed High Fat Diet (레시틴섭취가 고지방 식이를 섭취한 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effect of lecithin on lipid metabolism and antixidative capacity in 9-week-old rats. Forty-five male Sprague-Dawley rats weighing 249.8 g were blocked into three groups according to their body weight and raised for 8 weeks with experimental diets containing 1% (LM) or 5% lecithin (LH) and control (C) diet. Plasma and liver total lipids, triglyceride, total cholesterol and plasma HDL-cholesterol concenterations, and fecal total lipids, triglyceride, total cholesterol and bile acid excretions were measured. Malondialdehyde (MDA) levels in plasma, liver, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cell and liver, xanthine oxidase (XO) activities in plasma and liver, and total antioxidant status (TAS) in plasma were also measured. Effect of lecithin intake on antioxidative capacity was not significantly different among all the groups. Plasma total lipids, triglyceride and total cholesterol levels were lower in lecithin groups compared to control group, and these three lipid levels of lecithin groups were lowered dose-dependently as dietary lecithin level increased. But liver total lipids, triglyceride and total cholesterol levels were not different among all the groups. Also fecal total lipids, triglyceride and total cholesterol excretions were highest in high lecithin groups compared to two other groups. Thus it is plausible that lecithin intake decreases plasma lipid levels through increasing fecal lipid excretions, and may be beneficial for treatment and prevention of hyperlipidemia, but has no effect on antioxidative capacity.

Studies on the Antihypercholesterolemic Effects of Gamigwaruhaebaekwhanggum - Tang (가미과루해백황금탕의 항고지혈증 효과)

  • Gamigwaruhaebaekwhanggum-Tang (GGHWT) have been evaluated for antihyperlipidemic effects on experimental hyperlipidemic rats and mice induced by Triton WR-1339, com oil and high cholersterol-diet. Especially, GGHWT is formulated with Trichosanthis Fructus, Pinelliae Tuber, Aurantii Immaturus Fructus, Magnoliae Cortex, Allii Macrostemi Bulbuls, Cinnamomi Ramulus and Scutellariae Radix. Oral administration of GGHWT at 500 mg/kg/day for 3 days significantly inhibited the increase of serum triglyceride and LDL-cholesterol, liver triglyceride in hyperlipidemic rats induced by Triton WR-1339. And, GGHWT significantly inhibited the increase of serum triglyceride in hyperlipidemic rats induced by corn oil. Also, administration of GGHWT (500 mg/kg, once daily for 1 week, p.o.) prevented the increase of serum total cholesterol, triglyceride, LDL-cholesterol, liver total cholesterol and triglyceride in 1% cholesterol-diet fed mice. These results suggest that GGHWT is effective for the treatment of hyperlipidemia.

Effect of Cordyceps Militaris on Lipid Metabolism in Rats Fed Cholesterol Diet (동충하초(Cordyceps Militaris)가 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향)

  • The effects of Cordyceps militaris on the weight gain, the food intake, the food efficient ratio, and the lipid concentrations of serum, liver, pancreas, heart and aorta in male rats fed cholesterol diet were studied. Male Sprague-Dawley rats were received 4 types of diets for 4 weeks, respectively : a normal diet(without cholesterol), a control diet(hypercholesterolemic diet), a hypercholesterolemic diet supplemented with 3% fruiting body of Cordyceps militaris(CF), a hypercholesterolemic diet supplemented with 3% mycelium of Cordyceps militaris(CM). The body weight gain of rats fed the control diet were significantly increased compared to those of the rats fed diets with CF or CM. But, the hepatic and pancreatic weights of rats fed diets with CF or CM were heavier than those of rats fed the normal diet. In serum of rats fed the CM diet, the concentrations of total cholesterol, triglyceride, LDL-cholesterol and the atherogenic index were significantly lower than those of rats fed the control diet. Also, serum HDL-cholesterol concentration and HDL-cholesterol/total cholesterol ratio were significantly lower in rats fed the control, CF or CM diets than those in rats fed the normal diet. Whereas, the hepatic concentrations of the total lipid, the total cholesterol and the triglyceride were significantly higher in rats fed 3 hypercholesterolemic diets than those in the rats fed normal diet. The heart and pancreatic concentrations of total cholesterol were the highest in the rats fed diet with the CR among rats fed experimental diets. In conclusion, the CM feeding decreased the concentrations of the cholesterol, triglyceride, and LDL-cholesterol and the atherogenic index in the serum of the rats. But the CF or CM feeding could not decrease the hepatic concentrations of the total lipid, cholesterol, triglyceride in the rats fed hypercholesterolemic diets. Contents of Total Lipids and Their Composition in Colored and Aromatic Rices Cultivars (유색미와 향미 품종의 지방질함량 및 지방질의 조성)

  • The contents of total lipids and their components of brown rice grain were studied for 5 colored rice cultivars, 1 aromatic rice cultivar and 1 normal rice cultivar grown in Korea. The total lipid contents ranged 2.04 to 4.69% based on total dry weight. The Tohoku #149 showed the highest content which was followed by Hansanheukmi and Sanghaehyang-hyeolna. The lowest content were observed in three cultivars, Heuknambyeo, Hyangnambyo and Dongjinbyeo which showed no difference among cultivars. The main classes of the total lipid were triglyceride, diglyceride, free sterol, free fatty acid and sterol esters in all cultivars examined. The ratio of triglyceride in total lipid was high in colored rice and aromatic rice compared to Dongjinbyeo which is normal rice cultivars, but the ratio of diglyceride was high in Dongjinbyeo. The major fatty acid compositions in the cultivars examined were linoleic acid, oleic acid and palmitic acid. There was a varietal difference in fatty acid composition, for example, erucic acid was detected in aromatic and colored rices cultivars, while none of this compound was found in Dongjinbyeo.

  • 1. Objectives The purpose of this research is to find out the Constitutional difference between the amount of Plasma Homocysteine, Total cholesterol, and Triglyceride which are factor of Cerebrovascular disease. 2. Methods We enrolled 132 patients inexperienced Strokes who visited our hospital for a medical examination from April 26, 2004 to June 29, 2005 except patients whose body constitusions were undetermined according to QSCOII. This research investigated the Constitutional difference between the amount of Plasma Homocysteine, Total cholesterol and Triglyceride. 3. Results & Conclusions This research has shown that there is a statistical significance between the average amount of Plasma Homocystein level that is higher in Taeumin. But there is no significant difference between the average amount of Total Cholestrol and Triglyceride in Sasang Constitution.

The Effects of Micro Crystalline Chitin and Palm Oil on the Lipid Metabolism in the Rat (미세결정화키틴과 팜유의 수준을 달리한 식이가 흰쥐의 지방대상에 미치는 영향)

  • The effects of MCC and palm oil at different levels on lipid metabolism were assessed in adolescent rat. Dietary fat levels were 20% and 40% (kcal/kcal) and MCC level were 0%, 2%, and 4% (wt/wt). The experimental period took 8 weeks. During the experiment, weight gain and food efficiency ration were not affected by dietary factors. The weight of thymus however, was lower in high fat groups than in middle fat groups. The contents of total lipid and total cholesterol in plasma and HDL-cholesterol in plasma and HDL-cholesterol were significantly affected by dietary factors. The concentrations of cholesterol in LDL and VLDL , and the triglyceride content of VLDL was higher in high fat groups than in middle fat groups. Adding MCC lowered the LDL triglyceride in the high fat groups. The analysis of plasma fatty acids generally reflected the composition of the dietary palm oil. MCC had significant effects on total lipid and triglyceride of feces, but not on total cholesterol . In conclusion, the level of MCC had little hypolipodemic effects on lipid metabolism in rats fed a high fat diet. the middle fat group that contained 4% MCC showed increased contents of fecal triglyceride than the others, indicating that MCC had an effect on lipid absorption. Therefore, the other physiological functions of MCC need to be tested for their useful applications.

Effects of Extrasct of Pueraria radix on Lipid Metabolism in Rats Fed High Fat Diet (갈근추출물이 고지방식이를 섭취한 흰쥐의 지질대사에 미치는 영향)

  • This study was designed to investigate the effect of Pueraria radix extract on lipid metabolism in the rats fed high fat diet. Male sprague dawley rats were given 5% Pueraria radix extract and fed diet containing 20 weight % fat. The results obtained were summarized as follows. Serum ALT and AST activities were reduced by Pueraria radix extract. Levels of total lipid, total cholesterol and triglyceride in the serum were higher in the HF group than in the control, but showed no significant differences between control and HF Pue group. Liver total lipid, total cholesterol and triglyceride contents were increased by high fat diet, but were lower in the HF Pue group than in the HF group. Fat absorption rate was increased with high fat diet feeding and higher in the HF Pue group than in the HF group.
