  • This study investigated growth and hematological changes in parrotfish Oplegnathus fasciatus (~200 g/fish) reared under different total ammonia nitrogen (TAN) concentrations (0, 4, or 8 mg/L) for 6 weeks. Survival rates of parrotfish in all experimental groups did not significantly differ, as they were all ~100%. Although specific growth rate (SGR), weight gain, and daily feed intake in the high TAN concentration group (TAN8) were significantly lower than in the other two groups, there was no significant difference between the TAN4 group and the control group, (TAN0), indicating that parrotfish have a strong resistance to ammonia toxicity. As for temporal changes of the major blood components, cortisol increased as a result of stress caused by the high ammonia concentration in the TAN8 group. For this reason, the concentrations of energy sources such as glucose and total cholesterol were reduced. However, there was little difference among all experimental groups in concentrations of liver function glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT), and nutrient factors, such as total protein and albumin.

  • We analyzed the concentration of Aspartate Aminotransferase (AST), Alanine Aminotransferase (ALT), Alkaline Phosphatase (ALP), Gamma Glutamyl Transferase (GGT), total protein (TP), albumin, total cholesterol (TC) and triglyceride in sera of patients (n=455) aged 60 years and older with hyperglycemia (${\geq}120mg/dL$ in serum). The average concentration of biochemicals in patient group was compared with those in reference group. Our data showed that the average concentrations of AST and TC in sera of senior patients with hyperglycemia were significantly increased by 27.5% (P<0.05) and 82% (P<0.05) in total group, 38.5% (P<0.05) and 75.0% (P<0.05) in sixty years' group, 16.0% (P<0.05) and 89.7% (P<0.05) in seventy years' group and 27.0% (P<0.05) and 79.5% (P<0.05) in eighty years' group, respectively. Patients with hyperglycemia showed a significant decrease in albumin level by 6.7% (total group, P<0.05), 4.5% (sixty years' group, P<0.05), 8.9% (seventy years' group, P<0.05) and 4.5% (eighty years' group, P<0.05), respectively. In conclusion, the high concentration of glucose in the senior patients with hyperglycemia may be a cause of severe liver function and lipid metabolism disorder.

  • Transforming growth factor $-{\beta}$ is one of the polypeptide growth factors that mediate the activity of mesenchymal cells and regulate wound healing process via cell proliferation, migration and extracellular matrix formation. The purposes of this study is to evaluate the effects of transforming growth factor $-{\beta}$ on the protein synthetic activity of human periodontal ligament cells and human gingival fibroblasts. The cells which were prepared were primary cultured gingival fibroblasts and periodontal ligament cells from humans, and the fourth or sixth subpassage were used in the experiments. Cells were seeded and at a confluent state, 0, 0.5, I, 2.5, 5, 10 ng/ml $TGF-{\beta}$ and $2{\mu]Ci/ml\;[^3H]$ proline were added to the cells and cultured for 24 hours. Then, 1 and 5 ng/ml concentrations were selected and added to confluent cells and cultured for 24 and 48 hours. They were labeled with $2{\mu}Ci/ml\;[^3H]$ proline for 24 hours and a collagen assay was done by the Peterkofsky and Diegelman method. The results were presented as the mean disintegration per minute (dpm) per well and S.D. of four determinations, The results were as follows. : The total protein, collagen and noncollagenous protein synthesis in periodontal ligament cells and gingival fibroblasts were increased dose- dependently by transforming growth factor-p to 2.5-5 ng/ml concentration and decreased at 10 ng/ml concentration. The percent of collagen was slightly changed according to the concentration of transforming growth factor-po The effect of transforming growth $factor-{\beta}$ was not specific for collagen synthesis since it increased the total, noncollagenous and collagenous protein, simultaneously. In the comparison of protein synthetic activity between the human periodontal ligament cells and human gingival fibroblasts, the human gingival fibroblasts had higher activities than the human periodontal ligament cells at all times and concentrations of $TGF-{\beta}$. In the comparison of protein synthetic activity between the 24 hour effect and the 48 hour effect of $TGF-{\beta}$, the 48 hour cultured cells' synthetic activity decreased more than the 24 hour cultured cells at human periodontal ligament cells and human gingival fibroblasts. In conclusion, $TGF-{\beta}$ has important roles in the stimulation of protein synthesis in human periodontal ligament cells and human gingival fibroblasts. Thus, it may be useful for clinical application in periodontal regenerative procedures.

  • This study was performed to elucidate the mechanism of dietary protein level on renal function through lipid and eicosanoids metabolism. Male rats of 337.8$\pm$5.7g body weight were undergone unilateral nephrectomy or sham-operation. The rats were divided into high protein(40% casein), normal protein(15% casein) and low protein(8% casein) diet groups and fed experimental diets ad libitum for 24 weeks. The results are summarized as follows. Serum total lipid, cholesterol and HDL-cholesterol of rats in 15% and 40% casein groups were higher than those of 8% casein group. But serum triglyceride was affected neither by uninephrectomy nor by dietary protein level. Serum thromboxane(TX) B2 and 6-keto prostaglandin F1$\alpha$ increased with increasing dietary protein level. Serum prostaglandin(PG) E2 was not affected by uninephrectomy nor by dietary protein level. Urinary PGE2 and TXB2 excretion tended to be lower in uninephrectomized groups. Renal tissue concentration of TXB2 was lower in uninephrectomized groups and in high protein group. These results suggest the possibility that the effects of dietary protein level on renal function could be due to changes in lipid and eicosanoids metabolism.

  • In the present study, we analyzed the defensin protein deduced from Korean radish (Raphanus sativus L.) seeds. To express the genes in E. coli, we constructed a recombinant expression vector with a defensin gene, named rKRs-AFP gene isolated from Korean radish seeds. Over expressed rKRs-AFP proteins was separated by SDS-PAGE to determine the purity, and protein concentration was determined by the Bradford method. Antifungal activity was assessed by disk assay method against the tested fungi. As a result, when 500 mL of cell culture were disrupted by sonicator, 32.5 mg total proteins were obtained. The purified protein showed a single band on SDS-PAGE with estimated molecular weight about 6 KDa, consistent with the molecular mass calculated from the deduced amino acid sequence. The purified rKRs-AFP protein showed remarkable antifungal activities against several fungi including Aspergillus niger, Botrytis cinerea causing the gray mold disease, and Candida albicans. In field tests using the purified rKRs-AFP protein, the protein showed the reducing activity of disease spot and the mitigating effect of spreading of disease like agrichemicals. The immuno-assay of rKRs-AFP protein showed that the purified protein entirely accumulated at B. cinerea cytoplasm through the hyphal septa shown by fluorescence imaging. There was no fluorescence inside the cell, when the hypha was incubated without the protein. These all results indicate that the recombinant rKRs-AFP proteins can be utilized as a potential antifungal drug to control harmful plant fungal pathogens.
