Antioxidant and Antibacterial Effects of Carnosic Acid on White Sausage (카르노스산에 의한 화이트소시지의 항산화 및 항균 효과)

  • The relevant main constituents of rosemary are compound of a vast number of polyphenolics, including carnosic acid, carnosol, rosmarinic acid, and ursolic acid. Recently, phenolic compounds of plant origin have attracted considerable attention due to their beneficial functional and nutritional effects including antioxidant and antibacterial activity. This study was carried out to investigated effect of carnosic acid on pH, thiobarbituric acid reactive substance (TBARS), volatile basic nitrogen (VBN), 2,2-diphenyl-1-picryhydrazyl (DPPH) radical scavenging activity, and total bacteria counts in white sausage during the storage at room temperature. Addition of carnosic acid was decreased of pH, TBARS, VBN, and enhanced antioxidant and antibacterial activity in white sausage. These activities increased with increasing concentrations of carnosic acid. Furthermore, the potent antibacterial activities of carnosic acid against pathogenic bacteria (Escherichia coli KCCM 11234 and Salmonella enteritidis KCCM 12021) were measured. Carnosic acid showed noticeable inhibitory effects on E. coli and S. enteritidis. In conclusion, carnosic acid might be used as a natural preservative in white sausage.

  • This study aimed to investigate of Andong sikhe by using pure culture inoculation technique and the improvement of storage stability by the addition of stabilizers to the product. Microorganisms were selected for the pure culture inoculation in the fermentation. The changes in chemical composition such as total acidity, sugar content, amino acid and various forms of nitrogen during fermentation were determined. The changes in pH of the product, the enzyme activities and the population of lactic acid bacteria and yeast were also followed in the process of fermentation.

Effect of Texture Improvement and Shelf Life Extension of Frozen Rainbow Trout Oncorhynchus mykiss Treated with TGase and Polysaccharides (다당류 및 TGase를 활용한 동결 무지개송어육(Oncorhynchus mykiss)의 물성개선 및 저장성 향상 효과)

  • This study investigated the effect of transglutaminase (TGase) and polysaccharide kappa carrageenan on the texture, chemical, and microbiological qualities of refrigerated unmarketable rainbow trout Oncorhynchus mykiss. Gel strength increased substantially in TGase-treated samples, and was adding kappa carrageenan resulted in no significant difference. SDS-PAGE results confirmed that the myosin heavy chain band with a molecular weight of 205-250 kDa was weakened in trout meat treated with 1% TGase, which led to cross-linking reactions between proteins. The volatile basic nitrogen (VBN) increased in all samples during storage at 4℃ for 10 days; however, the samples treated with 0.5% and 1% kappa carrageenan had the lowest VBN. The viable cell count increased in all samples treated with TGase and kappa carrageenan; however, an increase in TGase enzyme and kappa carrageenan concentration successfully hindered total bacteria growth. Thus, adding 1% TGase and 1% kappa carrageenan to refrigerated unmarketable rainbow trout formulations can optimize quality characteristics.

Effect of Modified Atmosphere Packaging on Quality Preservation of Mackerel Fillets (고등어 필렛의 품질유지에 미치는 변형기체포장의 효과)

  • In Korea, mackerel is the most preferred red fish commodity and has been increasingly consumed in chillstored fresh state rather than in frozen or salted fish. Modified atmosphere packaging (MAP) technology as a replacement of air with low O2 and high CO2 concentration gas was applied in this study to preserve its freshness. Four MAP conditions of CO2(60):O2(30):N2(10), CO2(60):O2(5):N2(35), CO2(60):O2(0):N2(40), and CO2(30):O2(0):N2(70) were compared in quality preservation effect with air package used as Control. Three hundred grams mackerel fillets packaged in gas barrier tray were stored for duration of 10 days at 5℃. Quality was assessed in total aerobic bacterial count, pH, total volatile basic nitrogen (TVB-N), thiobarbituric acid-reactive substances (TBARS), peroxide value (POV), texture, and surface color. High CO2 concentration MAPs (CO2(60):O2(30):N2(10), CO2(60):O2(5):N2(35), and CO2(60):O2(0): N2(40)) inhibited total aerobic bacteria growth in the fish fillets. MAPs of high CO2 concentration with O2 containment (CO2(60):O2(30):N2(10) and CO2(60):O2(5):N2(35)) showed a low TVB-N content through the storage. The treatments containing O2 above 20% (Control and CO2(60):O2(30):N2(10)) showed more accelerated increases in TBARS and POV than other treatments. The visual appearance was better for fillets in the packages of CO2(60):O2(5):N2(35), CO2(60):O2(0):N2(40), and CO2(30):O2(0):N2(70) than for those of other treatments. The MAPs of CO2(60):O2(5):N2(35) and CO2(60):O2(0):N2(40) are expected to be effective in keeping the freshness of mackerel fillets.

Effects of Enzyme Complex on Odor Emission from Swine Slurry and Swine Buildings (효소복합체가 양돈슬러리 및 돈사 악취발생에 미치는 영향)

  • This study was carried out to investigate the effect of enzyme complex on odor emission from swine slurry and the process of making swine liquid manure. Ammonia and hydrogen sulfide concentrations were significantly decreased by using the enzyme complex of liter per ton level of liquid swine slurry in the manure storage tank according to the time. Characteristics of liquid swine slurry were affected by the enzyme complex, total nitrogen and ammonia nitrogen contents were reduced compare with control. Ammonia and hydrogen sulfide concentrations in the finishing pig building and offensive odor compound on the boundary line of swine farm were significantly decreased by spraying in swine finishing building. In conclusion, the results obtained from this study suggest that using the enzyme complex of liter per ton level of liquid swine slurry for making liquid swine manure may improve the quality of swine liquid fertilizer and reduce odor emission. Also farm scale enzyme complex treatment may improve air quality in finishing pig building and deduce offensive odor compound of swine farm.

  • Polyhydroxyalkanoates (PHAs) are synthesized by numerous bacteria as carbon and energy storage compounds and are raw materials for biocompatible plastics. In this paper, the effect of $CO_2$ concentrations on the growth of C. necator and the accumulation of Poly(3-hydroxybutyrate) (P(3HB)) are investigated by increasing the $CO_2$ concentration in the substrate gas mixture. During 6 d cultivation in a nitrogen-present mineral medium, the $CO_2$ concentration did not affect the growth of the cells, while the Poly(3- hydroxybutyrate) (P(3HB)) content decreased with increasing $CO_2$ concentrations from 1% to 20%. During 4 d cultivation in the nitrogen-limited medium, the P(3HB) accumulation was the greatest at 3% $CO_2$; however, the total amount of accumulated P(3HB) was the greatest at 1% $CO_2$, which decreased with increasing $CO_2$ concentrations. The results indicate that the gas mixture with 1% $CO_2$ is the most effective in both growing the cells and accumulating P(3HB) under our experimental conditions.

Effect of Natural Antimicrobial Extracts on Shelf-life Extension and Quality Improvement of the Flounder Sikhe (천연 항균 추출물의 첨가가 가자미 식해의 품질 및 저장성에 미치는 영향)

  • Flounder sikhe is one of the traditional Korean fermented foods. Because honey, fucoidan, propolis, aronia berry, and horseradish are well known to have natural antimicrobial properties, we investigated their antimicrobial effects on improving the quality of flounder sikhe. Ethanol extracts of propolis inhibited the growth of Bacillus cereus, Staphylococcus aureus, and Listeria monocytogenes. The extracts of aronia berry showed similar effects against Salmonella Typhimurium. We also assessed the total polyphenol and total flavonoid content and 2,2-diphenyl-2-picrythydrazyl (DPPH) radical scavenging activity in the extracts obtained from these natural materials. The contents of polyphenols and flavonoids were the highest in propolis, which also showed a high DPPH radical scavenging activity. Therefore, addition of propolis 0.5% to flounder sikhe showed optimal improvement, with a storage stability of 15 days at pH 5.39, volatile basic nitrogen (VBN) content at 24.73 mg/100 g, and total bacterial count at $1.81{\times}10$ colony forming unit (CFU)/g.

Chemical Composition of Korean Geoduck and Changes in Their Composition during Frozen Storage (코끼리조개의 성분 조성과 냉동 저장 중의 성분 변화)

  • To obtain the principal data for useful treatment and processing of Korean geoduck (Panope japonica A. ADAMS) which inhabit mostly at Dong-Hae coastal area in Korea, changes of $NH_2$-N, TMAO, TMA, total creatinine, protein composition and fatty acid composition in raw and blanched geoduck muscle during storage at $-20^{\circ}C$ were investigated. In addition, its chemical composition variation in the whole year was elucidated. The moisture content in geoduck muscle meat was 78.1% to 82% in the whole year. Particularly, in July its moisture content was maximum as 82% and in September minimum as 78.1%. Crude protein was in the range of 12.3-16.4%, crude lipid the average was 1.5%, crude ash on the average was 1.4%. The abundant fatty acids in geoduck muscle oil were $C_{16}$ : 0, $C_{16}$ : 1, $C_{18}$ : 0, $C_{18}$ : 1, $C_{20}$ : 5, and $C_{22}$ : 6 acids. During storage at $-20^{\circ}C$, content of unsaturated fatty acid such as eicosapentaenoic acid (EPA, $C_{20}$ : 5) and docosahexaenoic acid (DHA, $C_{22}$ : 6)in raw geoduck muscle decreased somewhat and the raw geoduck was slightly oxidized. Trimethylamine (TMA), volatile basic nitrogen (VBN)and $NH_2$-N of raw muscle increased compared to blanched muscle. Trimethylamine oxide (TMAO) was slightly decreased during the storage period. The muscle protein was approximately composed of 37% sarcoplasmic, 29% myofibrillar, 22% alkali soluble, and 12% stroma protein. Among several proteins, myofibrillar protein content decreased mostly, while the alkali-soluble and stroma protein content increased slightly during storage at $-20^{\circ}C$.

Quality Characteristics of Kochujang with Different Ratios of Rice-nuruk (쌀누룩의 혼합비율을 달리한 고추장의 품질특성)

  • The quality characteristics of kochujang added with additives of different ratios of rice-nuruk (at 1:0.7, 1:1.0, 1:1.3) were studied. The kochujang was stored for 8 weeks at $20{\pm}3^{\circ}C$. The brix tended to increase by increasing mixing ratio of rice-nuruk in all treatments. It increased rapidly up to 4 weeks fermentation and then showed a slight change after 6 weeks. The more mixing ratio of rice-nuruk increased, the more salinity of samples decreased. The initial pH value of kochujang was 5.08-5.10., however, the pH value decreased to 4.82-4.83 after an 8 weeks of storage period. The initial acid value was 0.66-0.68 and increased from 0.66-0.68 to 0.87-0.89. It had slight reduction phenomenon in the sixth week. As the aging period passed, chromaticity such as $a^*$- and $b^*$-values decreased gradually except for $L^*$-value. The moisture content of samples decreased gradually by adding rice-nuruk. but increased by passing an aging period. The initial amino nitrogen content of sample was 304-315 mg%. It increased to 399-442 mg% during the aging period. As the aging period passed, the total cell number increased from 6.81 log CFU/mL to 7.01 log CFU/mL in the fourth week and then steadily decreased to 6.66 log CFU/mL in the eight week. Results of sensory evaluation show that, as more rice-nuruk was added, the kochujang received a lower mark. There was no significant difference in flavor in all treatments. Color and sweetness with increasing amounts of rice-nuruk showed the lowest score. Saltiness rice-nuruk at the ratio of 1:1.0 was the highest at 6.22 points. Texture scores decreased with increasing amounts of the rice-nuruk Based on these results, the overall acceptability of the 1:0.7 rice-nuruk treatment which received the highest score is considered excellent.

The Development of HACCP-Based Standardized Recipe and the Quality Assessment of Cook/Chilled Soy Sauce Glazed Mackerel (쿡췰(Cook/Chill)시스템을 이용한 고등어조림의 HACCP 레시피 개발 및 생산과정의 품질평가)

  • The purposes of this study were to develop Hazard Analysis Critical Control Point-based standardized recipe applicable to cook/chilled soy sauceglazed mackerel and to evaluate the qualities related to the product flow of this item. After conducting experimental cooking, preliminary test, and analysis of recipes, critical control points were identified, control methods were determined, and HACCP-based recipe was standardized. At each critical control point, time-temperature profile was recorded and microbiological analysis (total aerobic plate counts, psychrotrophic plate count, coliform, and fecal coliform count), chemical analysis (pH, acid value and volatile based nitrogen (VBN)) and sensory evaluation of the item were done. Time-temperature data showed that the time the menu item had passed through temperature danger zone (5∼60$^{\circ}C$) during all phases was 60 min. At rapid cooling, but after cooling at room temperature, the temperature of this menu item did not drop below the ambient temperature. The results of microbiological test were negative throughout all phases following cooking and the results of chemical analysis did not change significantly in terms of storage periods except for VBN which increased on 7th day significantly(p<0.05). After steam/convection oven reheating and microwave oven reheating, the sensory score of the only appearance decreased significantly related to the storage time of overall quality profiles. But significant differences were not detected according to two reheating methods. In conclusion, this HACCP-based recipe was considered as an effective tool for assuring microbial as well as sensory quality of this cook/chilled item.
