The Effect of L-Carnitine and Isoflavone Supplementation on Weight Reduction and Visceral Fat Accumulation in Overweight Women (과체중 여성에서 L-carnitine과 Isoflavone 보충 섭취가 체중 및 내장지방 감소에 미치는 영향)

  • This study was performed to examine the combined effects of L-carnitine and isoflavone supplementation on weight reduction and body fat distribution in overweight women. Overweight/obese women (body mass index > $23kg/m^2$) who were not diagnosed any type of diseases were included in this study and sixty subjects ($41.1{\pm}1.5$ years, $25.9{\pm}0.3kg/m^2)$ were randomly assigned to a placebo (n=30) or a supplement group (n=30, L-carnitine 300 mg+isoflavone 40 mg/day). We measured anthropometric parameters, abdominal fat distribution by computerizd tomography and blood components before and after the 12 week intervention period. After the 12 weeks of supplementation, subjects in L-carnitine and isoflavone supplement group showed a significant reduction of body weight (p < 0.001), body fat % (p < 0.05), and waist to hip ratio (p < 0.01) whereas placebo group did not show any changes. In a CT-scanned results, total fat area at L4 level was significantly reduced by 8.1% (p < 0.01) with the reduction of visceral fat area (-11.1%, p < 0.001) and subcutaneous fat area (-7.0%, p < 0.05) in the supplement group. The supplementation of L-carnitine and isoflavone showed the significant improvement of HDL-C (p < 0.01) and apoB (p < 0.05) concentrations, however, change values in those markers were not significant compared with those of the placebo group. In addition, a significant increase of adiponectin level (p<0.001) was observed in the supplement group after the intervention. The result of present study demonstrated that supplementation of 300 mg L-carnitine and 40 mg isoflavone per day fur 12 weeks can give beneficial effects on weight reduction and visceral fat accumulation. These potential antiobesity supplement can produce more favorable effects when combined with lifestyle modification.

Effects of Dietary Isoflavone and Casein Phosphopeptide on Hatching Egg Production and Eggshell Quality in Aged Egg-Type Breeder Hens (산란종계 사료 내 이소플라본 및 Casein Phosphopeptide의 첨가가 종란 생산성 및 후기 난각질에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary Ca levels and some feed additives such as isoflavone and casein phosphopeptide (CPP) on eggshell quality and hatching egg production in aged egg-type breeder hens. A total of three hundred and sixty, 56-week-old Hy-Line Brown breeder hens were divided into six groups and fed experimental diets of two levels of Ca (3.3% or 3.6%) either with addition of 0.2% isoflavone, 0.5% CPP or devoid of all for 5 weeks. There were no significant differences in laying performances and settable egg production among the groups. Significant increases (P<0.05) in eggshell strength were observed with increasing dietary Ca and addition of isoflavone, but not with addition of CPP. Fertility and hatchability were not influenced by dietary Ca and addition of isoflavone or CPP. The treatment had few significant effects on tibial proximal compositions and breaking strength. The concentrations of Ca, P, estrogen and calcitonin in serum were not affected by the dietary treatments. These results indicated that relatively high level of dietary Ca in combination with isoflavone had a beneficial effect on improving eggshell quality in aged egg-type breeder hens. But hatching egg production was not affected by dietary isoflavone or CPP.

Physico-chemical Characteristics of Dua-Chungkukjang (두아 청국장의 이화학적 특성)

  • Unlike the traditional preparation method for Chungkukjang, we prepared Dua-Chungkukjang with soybean fermented at $40^{\circ}C$ after the soaking, germination and steaming process, and we studied the physico-chemical characteristics of Dua-Chungkukjang. Reducing sugar content in both traditional Chungkukjang and Dua-Chungkukjang increased sharply after 10 hr of fermentation, but it decreased slightly after that. Total nitrogen content in both traditional Chungkukjang and Dua-Chungkukjang increased slowly as fermentation took place. Total nitrogen content reached 3,500 mg% and 3,700 mg%, and soluble nitrogen compound reached 750 mg% and 1,800 mg% after 40 hr of fermentation, respectively. $\beta$-Glucosidase activity increased continuously as fermentation took place, and it reached 193, 223, 235 and 248 units in Dua-Chungkukjang while it reached 125, 178, 205 and 214 units in traditional Chungkukjang after 10 hr, 20 hr, 30 hr and 40 hr of fermentation. This means that $\beta$-Glucosidase activity in Dua-Chungkukjang was $1.2{\sim}1.5$ times higher than that in the control treatment, traditional Chungkukjang. Total isoflavone content was $101.44{\mu}g$/g and $365.92{\mu}g$/g for 10 hr of fermentation, and $225.98{\mu}g$/g and $441.37{\mu}g$/g for 40 hr in traditional Chungkukjang and Dua-Chungkukjang, respectively. Isoflavone content of Dua-Chungkukjang was 2-3.5 times higher than that of traditional Chungkukjang. Anthocyanin content also increased continuously as fermentation took place, and it reached 5.0 mg/g and 6.9 mg/g in traditional Chungkukjang and Dua-Chungkukjang after 40 hr of fermentation, respectively.

Variation of Major Isoflavone Contents in Soybeans (Glycine max (L.) Merill.) of Collection Area, Diversity, and Seed Coat Color

  • Soybean seeds are important sources of non-nutritive health promoting metabolites. The contents of these metabolites are affected by both genetic and environmental factors. In the present study, the contents of five common isoflavones including daidzin, genistin, glycitin, malonyldaidzin, and malonylgenistin were analyzed in 72 soybeans of different seed coat colors, diversity, and of different origins including China (22), Japan (9), USA (12), India (4), and Korea (25). The average total isoflavone content (TIC) was maximum in Indian soybeans (3302.36 ㎍/g) and minimum in Chinese landraces (1214.95 ㎍/g). The Korean landraces had higher average TIC (2148.05 ㎍/g) than the USA genetic materials (1580.23 ㎍/g) and Japanese landraces (1485.99 ㎍/g). The content of malonylgenistin was in the range of 54.31 - 2385.68 ㎍/g in the entire population, and was the most abundant isoflavone irrespective of origin although there was content variation among individual soybeans. Besides, glycitin was the least concentrated isoflavone, and its content ranged from 0.00 to 79.79 ㎍/g. With respect to seed coat color, green soybeans from all countries displayed the maximum malonylgenistin and TIC contents. Exceptions were those of Korean and Indian origins where black and yellowish-green soybeans presented the highest malonylgenistin and TIC contents, respectively. In multivariate analysis, 92.72% of the variance was explained by the first two principal components, and the soybeans were grouped in to three clusters based on isoflavone contents. Overall, our findings signify the importance of seed coat color and origin as discriminant parameters, and provide wide spectrum of routes for breeding soybean cultivars.

Variation of Anthocyanin, and Isoflavone Contents in Korean Black Soybeans Grown at Different Latitudinal Locations (위도가 다른 재배지역에서 생육한 검정콩의 안토시아닌 및 이소플라본 함량 변이)

  • This experiment was conducted to investigate the variation of anthocyanin, and isoflavone contents in five cultivars and two lines of Korean domestic black soybeans grown at different latitudinal locations, a high latitude, Suwon ($37^{\circ}$16'N) and a low latitude, Milyang ($35^{\circ}$30'N). Delphinidin-3-glucoside (D3G) contents of anthocyanin in Geomjeongkong # 3 and Ilpumgeomjeongkong, cyanidin-3-glucoside (C3G) content in Milyang # 113, petunidin-3-glucoside (Pt3G) contents in Milyang # 113, and Ilpumgeomjeongkong, and total anthocyanins in Milyang # 113 were highest among the seven black soybean cultivars and lines. D3G, C3G, and total anthocyanins in Geomjeongkong # 3, C3G, and total anthocyanins in Ilpumgeomjeongkong grown at high latitude were higher compared to low latitude. Daidzein, glycitein, genistein, and total isoflavone contents in Geomjeongkong # 4 were highest among the seven cultivars and lines. Daidzein contents of isoflavone in Geomjeongkong # 3, Milyang # 112, and Milyang # 113 grown at high latitude were higher compared to low latitude. Glycitein contents in Geomjeongkong # 3, and # 4 grown at high latitude were higher compared to low latitude, while it in Milyang # 113 grown at low latitude was higher compared to high latitude. Genistein contents in most black soybeans except Milyang # 113, and total isoflavone contents in Geomjeongkong # 4 and Cheongjakong grown at low latitude were higher compared to high latitude. The variations of anthocyanin except Pt3G and isoflavone contents seemed to be affected by environmental conditions like different latitude than the genotype because they showed the significant interaction between cultivars and locations.

  • Isoflavones are associated with prevention of chronic diseases such as cancer, osteoporosis, and hypercholesterolemia. The isoflavone contents at different development stages of soybean were anayzed to examine the period of their formation and reaching maximum level. Soybean sedds have been collected from 13th to 21st week after sowing. Total daidzein and genistein contents increased from 115 and 37mg/kg at 13th week, to 795 and 699mg/kg at 21st week, respectively. Total glycitein content showed less remarkable change during seed development than other isoflavones. Thus mature soybean may have maximum preventive effect from chronic diseases since it contains the highest level of isoflavones.

Development of Soybean Snacks and Analysis of Bioactive Compounds (조미 콩 스낵 개발 및 생리활성물질 분석)

  • The purpose of this study was to analyze the bioactive compounds of soybean snacks and to carry out a sensory evaluation of those variations which had various seasoning mixing ratio. Five soybean snacks were developed for these experiments. Proximate composition, the contents of total phenol compounds and isoflavone, and sensory characteristics of the soybean snacks were measured. There were significant differences in amounts of isoflavones and total phenolic compounds of soybean snacks($p$ <0.05). The seasoned soybean snacks had a higher level of daidzein and genistein contents than did control group. The total phenol compound contents of the control group was 210.3 mg gallic acid equivalents/100 g, whereas those for individual soybean snacks(A~E) were 152.3, 160.2, 162.4, 158.4 and 164.6 mg gallic acid equivalents/100 g, respectively. In sensory evaluation, there were significant differences($p$ <0.05) in taste, texture, and overall acceptability of the soybean snacks.

  • In this study, soy-powder yogurt (SPY) with enhanced levels of ${\gamma}$-aminobutyric acid (GABA) and isoflavone aglycone was produced from sprouting high-protein soybeans (HPSs). The fermented steam-HPS sprouts (0 to 4 cm) were fermented (72 h) with Lactobacillus brevis, and the total free amino acids (FAAs) of the formed mixtures were determined to be 79.53, 489.93, 877.55, 780.53, and 979.97 mg/100 mL in the fermented HPS (FHPS), and the fermented steam-HPS with 0 cm (FSHPS-0), 1 cm (FSHPS-1), 2 cm (FSHPS-2), and 4 cm sprouting lengths (FSHPS-4), respectively. The levels of glutamic acid (GA) and GABA were observed to be the highest, 100.31 and 101.60 mg/100 mL, respectively, in the unfermented HPS (UFSHPS-1, 1 cm) and FSHPS-1 sprouts, respectively. Moreover, the total contents of the isoflavone glycoside form decreased proportionally to the increasing total levels of isoflavone aglycones after fermentation in FSHPS-0, FSHPS-1, FSHPS-2, and FSHPS-4. The levels of isoflavone aglycones were detected as 350.34, 289.15, 361.61, 445.05, and $491.25{\mu}g/g$ in FHPS, FSHPS-0, FSHPS-1, FSHPS-2, and FSHPS-4, respectively. While FSHPS-1 exhibited the highest DPPH (63.28%) and ABTS (73.28%) radical scavenging activities, FSHPS-4 contained the highest isoflavone aglycone ratio (81.63%). All in all, the FSHPS-1 mixture prepared in this study exhibited high GABA content and functional prosperity, thereby making it suitable for potential applications in the soy-dairy industry.

Isoflavone Distribution and $\beta$-Glucosidase Activity in Home-made and Factory-produced Doenjang (재래식 및 개량식 된장의 아이소플라본 분포 및 $\beta$-glucosidase 활성 연구)

  • Isoflavone distribution and $\beta$-glucosidase activity in 16 commercially available doenjang samples were determined. Twelve of the samples were home-made doenjang (HMD) with a relatively long fermentation period and 4 of the samples were factory-produced doenjang (FPD) from four different manufactures. Total isoflavones (TI) in the HMD ranged from $370-723\;{\mu}g/g$, while those in FPD ranged from $179-537\;{\mu}g/g$. The isoflavone distribution in HMD was different from those in FPD. Generally, the TI in HMD was higher than those in FPD. The major isoflavone was aglycones, which ranged from 42.98 to 89.96% in HMD and from 35.51 to 93.48% in FPD. Isoflavones in the $\beta$-glucoside forms were not detected in tested FPD samples. Principal components analysis (PCA) of the isoflavone profiles showed that HMD were differentiated from FPD. First principal component (PC1) and second principal component (PC2) expressed 43.6 and 22.9% of the data variability, respectively. $\beta$-Glucosidase activity in doenjang was lower than that in raw soybeans. The results of this study can be used to understand the differences in the isoflavone distribution in traditionally manufactured and factory produced doenjang.

Anti-Platelet Aggregating Effect of Solvent Extracts from Korean Soybean Varieties and Isoflavone Derivatives (품종별 국산콩 추출물 및 Isoflavone 유도체의 혈소판 응집억제작용)

  • Soybean (Glycine max L.) is an increasingly important food source and functional food. Platelet aggregation plays an important role in thrombogenesis and atherosclerosis. Here, we studied the anti-platelet aggregating effects of solvent extracts from Korean soybean varieties and isoflauone derivatives. Nine Korean soybean varieties were extracted by solvents (methanol and buthanol and their extracts was investigated for the inhibition against tile aggregation of washed rabbit platelets induced by collagen or thrombin. Maximal inhibition of buthanol extracts against platelet aggregation induced by collagen was $95\%$ in Black-kong and Jinpum - kong. The potency of their inhibition was in the following order : Black > Jinpum > Bokwang > Hwangkum > Pureun > Malli > Danbaek > Danyeob > Jangsu - kong. The Black - kong only seemed to produce the maximal inhibition against platelet aggregation induced by thrombin. Total isoflavone content measured was Jinpum-kong ($1347.8{\mu}g/g$) and Black-kong ($918.7{\mu}g/g$). Maximal inhibition of isoflavone derivatives against platelet aggregation induced by collagen was $97\%$ in genistein. The potency of their inhibition was in the following order: genistein>daidzein>genistin. The isoflavone derivatives did not affect the platelet aggregation induced by thrombin. However, Black-kong cortex seemed to Produce the optimal inhibition against platelet aggregation induced by collagen. These results suggest that Black-kong and Jinpum-kong may be a good source for antiplatelet agents, and their antiplatelet effect be related to tile content and the chemical structure with the number of -OH group and the attached glycoside in the isoflavone derivative.
