Survey of Yogurt Powder Storage in Ambient Export Countries A Safety Evaluation Standard Compliance and Comparative Analysis

  • Yogurt powder is fermented milk processed in the form of dry yogurt, and has advantages such as stability, storability, convenience, and portability. China and Vietnam are important export target countries because of the increased demand for dairy products. Therefore, we surveyed dairy product standardization in order to establish an export strategy. Lactic acid bacteria counts are unregulated in Korea and Vietnam. In China, lactic acid bacteria counts are regulated at $1{\times}10^6$ colonyforming units (CFU)/mL and detected at $6.24{\pm}0.33\;Log\;CFU/mL$. All three countries have regulated standards for total bacterial counts. In China, total bacterial counts of milk powder are regulated to n=5, c=2, m=50,000, M=200,000 and detected at $6.02{\pm}0.12\;Log\;CFU/mL$, exceeding the acceptable level. Lactic acid bacterial counts appeared to exceed total bacterial counts. Coliform group counts, Staphylococcus aureus, Listeria monocytogenes, and Salmonella species were not detected. Acidity is not regulated in Korea and Vietnam. In China, acidity was regulated to over $70^{\circ}T$ and detected $352.38{\pm}10.24^{\circ}T$. pH is unregulated in all three countries. pH was compared to that of general fermented milk, which is 4.2, and that of the sample was $4.28{\pm}0.01$. Aflatoxin levels are not regulated in Korea and China. In Vietnam, aflatoxin level is regulated at 0.05 ppb. Therefore, all ingredients of the yogurt powder met the safety standards. This data obtained in this study can be used as the basic data in assessing the export quality of yogurt powder.

Effect of Gamma Irradiation on the Microflora of Commercial Ready-To-Use (RTU) Salads during Cold Storage

  • Since ready-to-use (RTU) products are not fully cooked, the shelf-life of the product is comparably short and the products are easily spoiled when contaminated with food-borne pathogens. Low-dose gamma irradiation of 0.5, 1, or 2 kGy effectively reduced the total aerobic bacterial counts in 2 Korean manufactured RTU products by 1.63 to 2.95 log CFU/g during cold storage. Irradiation at 2 kGy reduced the psychrotrophic bacterial counts in most of the samples to below the limit of detection (< log CFU/g). Irradiation at 0.5 kGy completely eliminated Escherichia coli from the commercial RTU samples.

방사선조사에 의한 인삼분말의 저장성에 관한 연구 (A Study on the storage of Ginseng Powder by r-Irradiation)

  • To determine the effect of r-irradiation on the microorganisms destruction and the changes of general constituents of Ginseng Powder, three samples were analyzed at 1st and 3rd month after r-irradiation. The results were as follows: 1. Total bacterial counts were decreased during storage as the irradiation dose increased. It seems that about 300 Krad was satisfactory to the regulation of ginseng powder. ($5.0 \times 10^4 /g$) 2. Coliform group was also decreased during storage as the irradiation dose increased. Coliform group was not detected at the irradiation dose higher than 500 Krad. 3. Total bacterial counts and coilform group were in proportion to the content of moisture in ginseng powder. 4. The contents of moisture, ash, crude protein, crude lipid and total sugar in ginseng powder during storage had nearly no changes. 5. The content of reducing sugar in ginseng powder during storage had the increasing tendencies as the irradiation dose increased. 6. The content of amino-nitrogen in ginseng powder during storage had the decreasing tendencies as the irradiation does increased.

김밥 세균 오염의 원인 규명을 위한 연구 (Prediction of the Cause of Bacterial Contamination in Kimbab and Its Ingredients)

  • The distribution of bacteria in kimbab and its ingredients have been investigated. The total bacterial counts were over 3$\times$106/g(n=30) when the kimbabs were delivered to restaurant and it exceeded the legal level 1$\times$106/g defined by the Code of Food Standard in 1991. The gram-negative coliforms were also detected 1.9$\times$105. In order to study the cause of bacterial contamination in kimbabs, the ingredients used in kimbabs were examined. The bacterial counts were found 104-108/gfor kim (laver), 104-108/g for sausage, 104-106/g for boiled-spinach, 103-107/g for carrot, and 104-106/g for Danmuji, respectively. From these results it could be concluded that the bacterial contamination in Kimbabs was caused mainly by the ingredients such as kim, boiled-spinach, carrot, and sausage. Therefore, this suggested that the sanitary manufacture of kim, the storage at refrigerator temperature of the ingredients for kimbabs, and the proper treatment of equipments should be maintained in order to reduce the bacterial contamination for kimbabs. Furthermore, it should be required to obtain the basal data for establishment of the sanitary standard of kim and kimbab.

Enumeration and Recovery of Bacterial Isolates from Ruminants Fed with Different Dietary Regimes and Their Antibacterial Activity

  • The study evaluated different synthetic and semisynthetic media for maximal recovery of rumen bacteria and expression of their antibacterial activity. Rumen Glucose Cellobiose Agar (RGCA) medium was found to be the best for recovery of rumen bacteria. However, L-10 medium was the best for expression of antibacterial activity of ruminal isolates followed by Easy, M-10, RGCA and M-98-5 medium. The present study recommends the use of L-10 medium as the medium of choice for screening of antibacterial activity of ruminal isolates. Comparative evaluation of bacterial counts on different dietary regimes indicated significant difference between different growth media on a specific diet and between diets on specific growth media within a species. However, there is no overall significant difference between total bacterial counts obtained from rumen liquor of cattle and buffalo with respect to either the feeding regime or growth media. Feeding straw based diet to the animal is the best for high recovery of rumen bacteria.

시판 생수의 성분에 관한 조사 (Survey of Chemical and Bacterial Contents in Commercial Life Water)

  • In order for analysis of the actual condition of commercial life water, 38 samples were investigated during March to December in 1988. The results were as follows 1. All the samples analyzed were suitable for physico chemical criteria drinking water. The mean value of NH$_{3}$-N, NO$_{3}$-N, and KMnO$_{4}$ ,consumption were 0.014, 1.232 and 0.794 respectively. 2. General bacterial counts was detected higher than standard limit by 31.6% of total samples and the positive rate of coliform was 2.6% of total samples.

경남 북부지역 납유원유의 미생물학적 품질에 관한 연구 (A Study on the Microbiological Quality of Raw Milk in the North of Kyeongnam Area)

  • This study was carried out to obtain basic data for the Improvement of microbiological quality of raw milk. Total bacterial, psychrotrophic, thermoduric and spore counts of raw milk samples taken from milk tankers in the nothern part of kyeongnam were investigated for one year from March, 1989. The result obtained were summarized as follow 1. The number of total bacteria in raw milk averaged $4.0{\times}10^6$ CFU / ml and was not affected by seasons 2. The psychrotrophic counts of raw milk averaged $1.5{\times}10^6$ CFU / ml and it was higher in winter than in summer 3. The thermoduric counts of raw milk averaged $5.8{\times}10^4$ CFU / ml and was the lowest in winter 4. The spore counts of raw milk ranging from 3-1, 880/ ml averaged 306/ ml and was the lowest in summer

  • A total of 119 raw milk samples from ten dairy farms were examined for total bacterial count, and 739 quarter milk samples of 118 dairy cows of 14 herds were examined for mastitis. The results obtained were as follows: 1. The mean of total bacterial counts of the 119 raw milk samples was 132,000 per ml. The total bacterial counts of 81 samples (68.1%) were under the standard of 100,000 per ml and those of the 38 samples (31. 9%) were over the standard. The number of bacteria showed a tendency to increase in summer. 2. One hundred and ninety five quarters (26.4%) of 98 cows (52.7%) were proved to be infected with mastitis. Clinical mastitis was found at 7 qtarters (3.5%) of 5 cows (5.0%). 3. Staphylococcus (44.9%) and Streptococcus (26.7%) were two main causative organisms of mastitis. Coliform bacteria (4.6%), Pseuedomonas spp. (4.6%), yeasts (1. 3%) and corynebacterium sp. (0.7%) were also isolated from the infected quarters. 4. The isolates were more sensitive to chloramphenicol ((96.1%), leukomycin (78.8%), streptomycin (75.5%) and tetracycline (72.4%). On the other hand, they were less sensitive to colistin (11.0%), oreandomycin (18.1%), sulfisoxazole (24.6%), penicilline (27.6%), kanamycin (43.3%) and erythromycin (49.7%). Especially the strains of Pseudomonas spp. isolated from the infected quarters were resistant to almost all the drugs examined.

활성슬러지공정 하수종말처리장의 단위공정별 세균 제거효율 (Bacterial Removal Efficiencies by Unit Processes in a Sewage Treatment Plant using Activated Sludge Process)

  • To figure out the removal efficiency of indicator and pathogenic bacteria by unit processes of a sewage treatment plant using activated sludge process, analyses were done for incoming sewage, influent and effluent of primary clarifier, aeration tank, secondary clarifier and final discharge conduit of the plant. A matrix of bacterial items (average of bacterial reduction [log/ml], p value of paired t-test, number of decreased cases of twenty analyses, removal percentage only for decreased cases) between incoming sewage and final effluent of the plant were heterotrophic plate counts (1.54, 0.000, 20, 95.01), total coliforms (1.38, 0.000, 19, 83.94), fecal coliforms (0.90, 0.000, 20, 94.84), fecal streptococci (0.90, 0.000, 20, 98.08), presumptive Salmonella (0.23, 0.561, 7, 99.09), and presumptive Shigella (1.02, 0.002, 15, 92.98). Total coliforms, fecal coliforms, heterotrophic plate counts, and fecal streptococci showed highest decrease through secondary clarifier about 1-log (p<0.001) between 88% and 96%, and primary clarifier represented the significant (p<0.05) decrease. However, final effluent through discharge conduit showed higher total coliforms and fecal streptococci than effluent of secondary clarifier (p<0.05). In addition, final effluent once violated the water quality standard while effluent of secondary clarifier satisfied the standard. Hence some control measures including elimination of deposits in discharge conduit or disinfection of final effluent are necessary.

EFFECTS OF AMMONIATED RICE STRAW FEEDING ON MICROBES AND THEIR FERMENTATION END-PRODUCTS IN THE RUMEN AND CAECUM OF SHEEP

  • Three sheep fitted with cannulas in the rumen and the caecum were used in a $3{\times}3$ Latin square design to study the changes in ruminal and caecal microbial populations and their metabolite composition with ammoniated rice straw feeding. The 3 diets contained either 80% untreated rice straw (UTS) or ammoniated rice straw (ATS) and 20% formula feed. These were a control diet (C), a urea supplemented diet (U) containing urea at 1.1% and an ammoniated rice straw diet (AT). Data were analyzed by analysis of variance and means separated by the Student Neumann Kuel's multiple comparison. AT feeding increased ruminal bacterial counts, in particular cellulolytic bacterial counts (p < 0.05) which were 1.8, 2.4 and 7.0 (${\times}10^6/ml$ ruminal fluid) for C, U and AT, respectively. There was an increasing tendency (p < 0.10) in ruminal fungal population with U; values were 2.0, 5.2, 3.1 (${\times}10^3/ml$ ruminal fluid) for C, U and AT, respectively. Ruminal protozoa counts were not significantly (p > 0.05) altered with diets. Caecal total viable bacterial count with AT was about thrice the value with C. Total VFA concentration in the rumen was significantly increased (p < 0.025) (7.7 mmol/dl for C and 8.2 mmol/dl for AT) and correspondingly, pH lowered when AT was fed. Sheep on AT tended to produce less acetate and more butyrate in the rumen without significance (p > 0.05). Similar to the rumen, total VFA concentrations of 4.4, 3.8 and 5.2 mmol/dl were detected, respectively, for C, U and AT. Caecal ammonia-nitrogen concentrations were about six-fold of that in the rumen, though there were no differences (p > 0.05) among treatments.
