홍삼첨가 고추장의 관능적 특성 및 미생물과 효소력이 변화 (Sensory Evaluation and Changes in Microflora and Enzyme Activities of Red Ginseng Kochujang)

  • In order to improve funtionality of kochujang, red ginseng powder(1, 2 and 5% red ginseng on the total weight basis) was added to the raw material of kochujang. Changes in moisture content, microflora and enzyme activities were investigated in control kochujang(Sunchang kochujang) and red ginseng added kochujang during fermentation at 25oC for 120 days. Kochujang samples fremented for 90 and 120 days were subjected to sensory evaluation test. As the fermentation progressed, the moisture content and titratable acidity were gradually increased whereas pH was decreased. However, red ginseng had no effects on those changes. Bacterial count was maintained at the level of 107 CFU/g during fermentation. The level of yeast count was 104 CFU/g at the early stage of fermentation and increaed to 107 CFU/g after 60 days of fermentation and then maintained at 107 CFU/g. Red-ginseng had no effects on bacterial and yeast counts of kochujang. Amylase activity was gradually reduced during fermentation. Amylase activity was increased until 60 days of fermentation and then reduced by 90 days of fermentation. Acidic protease activity was slightly reduced at the initial stage of fermentation and then rapid increased. Acidic protease activity was at the highest level after 120 days of fermentation. Red ginseng did not alter those enzyme activities of kochujang. There were no differences in sensory properties between kochujang samples fermented for 90 and 120 days of fermentation. The addition of red-ginseng did not affect such sensory properties of kochujang as color, odor, hot taste and salty taste, but overall preference appeared to be improved by red ginseng addition.

감염성 질환들에서 C-반응 단백 값 (C-reactive Protein Level in a Variety of Infectious Diseases)

  • 목 적 : 다양한 감염성 질환에서 CRP, 백혈구 수 및 중성구 분획의 차이를 알아보고자 하였다. 방 법: 1996년 1월부터 2001년 12월까지 가톨릭대학교 대전성모병원에 입원한 환아 중 세균성 질환인 세균성 수막염(19명)과 급성 신우신염(55명), 대표적인 바이러스 질환인 홍역(274명), 수두(38명), 마이코플라즈마 질환인 마이코플라즈마 폐렴(160명), 리케치아 질환인 쯔쯔가무시병(39명) 및 감염이 원인으로 여겨지는 가와사끼병(152명)으로 입원한 환아들을 대상으로 의무 기록지를 후향적으로 조사 분석하였다. 결 과 : CRP 값은 세균성 수막염($17.9{\pm}6.4mg/dL$), 가와사끼병($9.1{\pm}5.6mg/dL$), 급성 신우신염($8.1{\pm}3.3mg/dL$) 순으로 높았다. 마이코플라즈마 폐렴과 쯔쯔가무시병(타세균성 질환)이 $3.2{\pm}2.5mg/dL$, 홍역 및 수두의 바이러스성 질환에서는 $1.0{\pm}0.8mg/dL$를 보였다. 백혈구 수 및 중성구 분획에서도 세균성 질환들과 타세균성 질환 및 바이러스성 질환에서 뚜렷한 차이를 보였다. 가와사끼병의 경우 세균성 질환과 유사한 결과를 보였다. 세균성 질환 및 가와사끼병에서 CRP 값과 백혈구 수 및 호중구 분획 사이에 상관관계가 있었다(P<0.01). 결 론 : 다양한 감염 질환에서 평균 CRP 값과 백혈구 수 및 중성구 분획은 병원체에 따라 뚜렷한 차이를 보였다. 하나의 감염성 질환에서 백혈구 수와 중성구 분획은 연령에 따라 차이를 보였으며, 세균성 질환에서 CRP 값과 호중구 수와는 서로 연관이 있었다.

Therapeutic Effect of Injection-Acupuncture with Bee-Venom (Apitoxin) in Cases of Canine Otitis Externa

  • This study was carried out to determine the therapeutic effect of injection-acupuncture (AP) with bee-venom (apitoxin) in cases of canine otitis externa (COE). Fifteen dogs with naturally-acquired otitis externa were used in this study. The dogs were divided into the following antibiotics group (control group), apitoxin group (experimental group A) and apitoxin combined with antibiotics group (experimental group B). All groups were treated by ear cleaning with normal saline once on day 1. The control group was treated with susceptible antibiotics, and experimental group A was given injection-AP with apitoxin $(100{\mu}g/head)$ at TH17 (Yi Feng), SI19 (Ting Gong), GB03 (Shang Guan) and TH03 (Zhong Zhu) bilaterally. Experimental group B was treated with susceptible antibiotics and injection-AP with apitoxin at the same acupoints as experimental group A. All the groups were treated 3 times/week for 2 weeks. The identity of the causative agents, the changes in the clinical signs, otoscopic findings, bacterial count in the auricular discharges, and total WBC counts and neutrophil/lymphocyte (N/L) ratio in the peripheral blood were investigated in all groups. In bacterial isolation, Staphylococcus spp. combined with Streptococcus spp. was detected higher than other agents. The bacterial cell count in experimental group A was significantly decreased at 2 weeks (p<0.01), and those in experimental group B was significantly decreased at 1 week (p<0.01) and 2 weeks (p<0.01) compared by those of control group, respectively. The changes of clinical score in experimental group B were significantly decreased at 2 weeks (p<0.01) compared by those of control group, but, those of experimental group A was similar to those of control group. The changes of total WBC counts and neutrophil/lymphocyte (N/L) ratio were no significant difference found. In conclusion, injection-AP with apitoxin is an effective treatment for COE and might be an alternative method for treating COE.

재발성 감염 질환의 접근 방법 (Approach to the Children with Recurrent Infections)

  • The major function of immune system is to protect infections. The immune systems are composed of innate and adaptive immunity. In adaptive immunity, the cellular and humoral components interact each other. Neonates and infants are infected frequently, because immune systems are naive and easy to expose to infectious agents. The complete history and physical examination is essential to evaluate the child with recurrent infections. The environmental risk factors of recurrent infections are day care center, cigarette smoke, and air pollution. The underlying diseases such as immunodeficiency, autoimmune diseases, allergy, and disorders of anatomy or physiology increase the susceptibility to infections. In immunodeficiency, infections are characterized by severe, chronic, recurrent, and unusual microbial agents infection. The defects of antibody production are susceptible to sinopulmonary bacterial infections. T cells defects are vulerable to numerous organisms such as virus, fungi, bacteria and etc. The screening tests for immune functions are the quantitative and qualitative measurements of each immune components. A complete blood count with white blood cell, differential, and platelet provide quantitative informations of immune components. Total complement and immunoglobulin levels represent the humoral component. Antibody levels of previously injected vaccines also provide informations of the antigen specific antibody immune responses. T cell and subsets count is quantitative measurement of cell mediated immunity. Delayed hypersensitivity skin test is a crude measurement of T cell function. The long term outcome of children with recurrent infections is completely dependent on the underlying diseases, the initial time of diagnosis and therapy, continued management, and genetic counscelling.

학교 집단급식소 내 식기류 및 집기류의 미생물학적 분석 및 위해요인 평가 (A Microbiological Analysis and Hazard Factor Evaluation of Food Utensils and Fixtures of Food Service Operations in Schools)

  • Objectives: The aim of this study was to evaluate the microbial hazards posed by food utensils and fixtures in food service operations at selected middle and high schools located in Seoul, Korea. Methods: We collected 200 samples of utensils and fixtures including cups, spoons/chopsticks, food trays and tables from five different schools in Seoul. Target microorganisms of this study were divided into two groups: total bacterial count and total coliform as indicators of microbial contamination and Bacillus cereus and Staphylococcus aureus as pathogens of food poisoning. We used selective media to quantify microbial concentration and 16S rRNA PCR assay for qualitative analysis. In addition, intensive interviews with nutritionists were conducted and observations were made to identify factors that may affect microbial contamination. Logistic regression analysis was employed to examine the relationship between the microbial concentration and operation characteristics of each operation. Results: The level of microbial concentration in school B and C were significantly lower than in school A, D and E (p<0.05). Some samples from school A, D and E showed over 3.4 log CFU/100 $cm^2$ (total bacterial count) and 1.0 log CFU/100 $cm^2$ (total coliform), which requires immediate hygienic action. The number of customers per staff member, periodicity of hygiene education for staff and daily operation time of sterilizers were also found to be important factors related with the microbial contamination of food service operations. Conclusions: These results suggested that not only a HACCP (Hazard Analysis and Critical Control Point) approach, but also efforts to assess internal risk factors within operations be needed to reduce the microbial contamination of food utensils and fixtures. This study is expected to provide preliminary data for assessing microbial hazards in food service operations.

초고압 전처리한 찹쌀과 현미의 텍스처 특성과 세균 저감 효과 (Textural Characteristics and Bacterial Reduction in Glutinous Rice and Brown Rice Pretreated with Ultra-High Pressure)

  • 본 연구에서는 건조 쌀에 적합한 가공적성과 세균 저감화를 위하여 백미 찹쌀(동진벼)과 현미 멥쌀(신동진벼)에 초고압 전처리를 하였다. 백미 찹쌀과 현미 멥쌀을 500MPa의 압력 하에서 5분간 초고압 처리하였을 때, 건조 쌀 제조에 있어 중요한 특성인 백미 찹쌀의 찰기와 현미 멥쌀의 경도가 유의적으로 감소하였는데(p<0.05), 이는 초고압 처리법은 백미 찹쌀의 낱알 분리가 힘든 찰진 특성과 현미 멥쌀의 딱딱한 식감을 효과적으로 개선할 수 있다는 점을 확인해 주었다. 쌀 텍스처 단면의 입자를 FE-SEM를 이용하여 관찰한 결과, 초고압 처리한 백미 찹쌀과 현미 멥쌀의 단면의 텍스처가 처리 전보다 부드러워졌음을 확인할 수 있었다. 600MPa의 압력 하에서 5분간 초고압 처리했을 때, 현미 멥쌀에서 생육하는 세균수는 처리 전보다 유의적으로 감소하였지만(p<0.05), 내열성 세균수에는 유의적인 차이가 없었다.

육가공용 향신료의 위생화를 위한 감마선 및 전자선 조사 효과 비교 (Comparison of the Effects of Gamma Ray and Electron Beam Irradiation to Improve Safety of Spices for Meat Processing)

  • 육가공 산업에서 원료로 이용되고 있는 향신료의 위생화를 위하여 대표적인 살균 기술인 감마선과 전자선의 조사효과를 비교하였다. 미생물 오염도를 측정한 결과 대부분의 시료에서 1.5 to 3.8 Log CFU/g의 일반호기성 미생물이 나타났으나, 강황 및 로즈마리에서는 미생물이 검출되지 않았다. 검출된 시료에서는 조사에 의해 효과적인 것으로 미생물이 감소하였으며, 감마선이 전자선보다 효과적으로 미생물을 사멸하였다. 검출된 미생물을 동정한 결과 Bacillus spp. 및 Staphylococcus spp.으로 확인하였다. B. cereus 및 S. aureus에 대한 방사선 감수성 측정 결과 0.19-0.63 kGy로 확인되었으며, 전자선 보다 감마선에 대한 내성이 약한 것으로 확인되었다. Ames test를 이용하여 방사선 조사에 의한 향신료의 유전독성학적 안전성을 평가한 결과, 조사에 의한 돌연변이원성은 관찰되지 않았다. 이상의 결과를 종합하여 볼 때, 향신료의 위생화를 위한 살균방법으로 방사선 조사기술이 효과적으로 판단되며, 감마선(>4 kGy, <6 kGy)이 전자선보다 효과적으로 미생물을 제어하는 것으로 나타났다.

전주 생 밀 막걸리의 산패 원인균의 분리 및 동정 (Isolation and Indenfication of Acid-forming Bacteria from a Fresh Wheat Makgeolli in Jeonju)

  • This research was carried out in order to discover acid-forming bacteria during fermentation of Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. In Makgeolli kept at $25^{\circ}C$, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, whereas the total cell count and bacterial type remained unchanged; the result implies that a succession of bacterial types, including acid forming bacteria, occurred. Two acid-forming bacteria were isolated from acidified Makgeolli and were identified as Acetobacter pasteurianus and Lactobacillus casei. When fresh and heat-treated Makgeolli were inoculated with Acetobacter pasteurianus and/or Lactobacillus casei, the greatest amount of acid was formed in Makgeolli inoculated with Acetobacter pasteurianus and Lactobacillus casei and also in Makgeolli with Acetobacter pasteurianus alone. This result indicates that Acetobacter pasteurianus is the main acidifier; furthermore, it shows the synergy effect in acid formation with Lactobacillus casei.

Effects of bamboo leaf extract on the production performance, rumen fermentation parameters, and rumen bacterial communities of heat-stressed dairy cows

  • Objective: An experiment was conducted to evaluate the effects of bamboo leaf extract (BLE) on the production performance, rumen fermentation parameters, and rumen bacterial communities of heat-stressed dairy cows. Methods: The experiment comprised a 14-day adaptation period and a 21-day experimental period and was conducted in a high-temperature and humidity environment (daily mean ambient temperature = 33.5℃±1.3℃; daily mean relative humidity = 64.9%±0.8%, daily mean temperature-humidity index = 86.2±0.4). Twelve Holstein dairy cows were randomly allocated into two groups. A total mixed ration supplemented with BLE at 0 (CON) and 1.3 g/kg dry matter (DM) were fed, respectively. Feed intake and milk yield were recorded daily. Milk samples were collected on 1, 11, and 21 d of the experimental period to analyze milk performance. Rumen fluid samples were collected on 21 d of the experimental period to analyze rumen fermentation parameters and rumen bacterial communities. Results: Compared with the control group, supplementation of BLE increased milk yield (p<0.01), milk fat yield (p = 0.04), 4% fat-corrected milk (p<0.01) and milk fat content (p<0.01); reduced somatic cell count (p<0.01). No differences in DM intake and milk protein or lactose content were observed between two groups. Supplementation of BLE also increased the rumen total volatile fatty acid (p<0.01), acetate (p<0.01), butyrate (p<0.01), and valerate (p = 0.05) concentrations. However, no significant effects were observed on rumen pH, ammonia nitrogen, propionate, acetate/propionate ratio, isobutyrate, or isovalerate. Furthermore, BLE increased the rumen bacterial abundance and the diversity of the rumen bacterial community. The BLE reduced the Firmicutes/Bacteroidetes abundance ratio and increased the abundances of Butyrivibrio_2 (p<0.01) and Ruminococcus_2 (p<0.01). Conclusion: The BLE supplementation at 1.3 g/kg DM could improve production performance and rumen fermentation in dairy cows during heat stress.

집단 급식용 생계 육에서 Campylobacter의 분리 및 동정 (Isolation and Identification of Campylobacter spp. from Raw Chicken Carcasses in food Service)

  • 집단급식에 공급되는 생계육에의 Campylobacter의 오염특성을 분석하기 위하여 경기지역의 한 공급업체의 단체급식용 생계육에서 Campylobacter를 분리.동정하고 특성을 분석하였다. 생계육에 오염된 일반 세균수는 $10^4$~$10^{6}$ CFU/g로 나타났고 Campylobacter를 분리를 위하여 증균배지와 선택배지를 사용하여 candle method과 microaerophilic chamber를 활용하여 일차적으로 205균주를 분리하였다. Gram staining, catalase, oxidase의 생화학적인 특성으로 28개의 균주를 분리하였고 pA와 pB의 primer를 사용한 PCR로 16개의 Campylobacter spp.를 동정할 수가 있었다. 특히 이들 중 9개의 균주가 42$^{\circ}C$도 뿐만아니고 $25^{\circ}C$에서도 생육이 가능한 것으로 나타났다. 따라서 생계육의 초기 총균수가 평균적으로 $10^{5}$ CFU/g로 높은 것으로 나타났고 저온 생육가능한 Campyobacter도 많이 분포하고 있으므로 infective dose가 낮은 이들에 의한 식중독의 위해를 줄이기 위하여 대량 집단급식시 철저한 위생관리가 요구되었다.
