Pilot Plant Scale Extraction and Concentration of Purple-Fleshed Sweet Potato Anthocyanin Pigment (자색고구마 anthocynin 색소의 대량추출 및 농축)

  • Performance of pilot plant scale extraction and concentration of purple-fleshed sweet potato anthocyanin pigment was tested and the characteristics of pigment extracts and concentrates were investigated. Fifty kilograms of purple-fleshed sweet potato was extracted with 500 L of 1% citric acid in 20% ethanol. As a whole, extraction pattern of the large scale extraction was similar to that of the laboratory scale extraction. The extracted pigment solution was filtered twice with a bag filter and a winding type microfilter and the filtrate was concentrated by a large scale vacuum evaporator at $40^{\circ}C$ and 600 mmHg vac. The mean values of total optical density (TOD) of the extract and the concentrate were 6.53 and 120.45, respectively. Browning index (BI) and Degradation index (DI) of extract were 5.86 and 1.55 and those of concentrate were 5.89 and 1.56, respectively, which indicated that the pigments were not changed or degraded through the extraction and concentration process.

Positive effects of anthocyanin-rich mulberry milk on mental health problems in the working population: an open-label study

  • BACKGROUND/OBJECTIVES: Depression and anxiety are common mental health problems. Anthocyanins from berries might have an inhibitory effect on monoamine oxidase (MAO) enzymes and alleviate various mood and anxiety symptoms. This study examined the effects of a daily supplement of an anthocyanin-rich product on mental health problems. SUBJECTS/METHODS: This study was a secondary analysis from a randomized, 6-week, open-label trial in 300 healthy participants aged 18-60 years who consumed 1 or 2 servings of anthocyanin-rich mulberry milk daily. The General Health Questionnaire-28 (GHQ-28) and Hospital Anxiety and Depression Scale (HADS) were used to monitor mental health problems. In addition, the saliva activity levels of MAO-A, MAO-B, and cortisol were examined at the baseline and after 6 weeks. RESULTS: The total scores of the GHQ-28 and HADS and all their subscales decreased in both groups (all P < 0.05). The cortisol, MAO-A, and MAO-B activities decreased significantly (all P < 0.05), but there were no significant differences between the groups (all P > 0.05). Significant correlations were noted between the decreased activity level of MAO-A enzyme and decreased scores from the GHQ-28 somatic subscale and the HADS depression subscale (all P < 0.05). CONCLUSIONS: Daily consumption of anthocyanin-rich mulberry milk possibly improves mental health problems by reducing depressive and anxiety symptoms in the working population. The suppression of MAO-A activity is a possible underlying mechanism.

Antioxidant Activity of Brown Soybean Ethanolic Extracts and Application to Cooked Pork Patties

  • The brown soybean extract (BE, extracted by distilled water, 50%, 75%, and 95% ethanol) were analyzed for their total phenol, flavonoid, anthocyanin content, and DPPH radical-scavenging activity to determine antioxidant activities. Brown soybean extract with 75% ethanol showed significantly higher DPPH radical scavenging activity, total phenol and anthocyanin content compared to the other treatments (p<0.05). Then, brown soybean extract with 75% ethanol was applied to pork patties at different concentration (0.05%, 0.1%, and 0.2%) and lipid oxidation was evaluated during 15 d of refrigerated storage. Addition of BE significantly increased redness and pH values, respectively (p<0.05). Moreover, TBARS value of pork patties decreased significantly (p<0.05) as BE concentration increased. In sensory evaluation, pork patties with 0.1% BE had significantly higher score than other treatments in flavor and overall acceptability (p<0.05). Consequently, these results indicate that 0.1% BE could be an effective natural antioxidant to inhibit lipid oxidation in pork patties.

  • The black raspberry (Rubus coreanus Miquel) contains anthocyanin, tannins, gallotannin, gallic acid, ferulic acid and phenolics. It brightens the eyes and protects the liver and kidneys. It was effective for anti-aging. Thus, the purpose of this study was to inform the excellence of black raspberry and to screen antioxidant activity to ensure the possibility as a functional material. In this study, bioactive compounds in black raspberry were determined. Additionally, black raspberry was extracted by CM (chloroform:methanol, 2:1, v/v), 70% methanol and 70% ethanol, and were investigated and compared in vitro methods. Total phenol and flavonoid contents were measured to compare each different solvents. In addition, DPPH radical scavenging activity and ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power were determined to measure the antioxidant activity. The results were the highest in 70% ethanol extracts, and the higher the concentration showed a significantly higher antioxidant capacity. When observed the relationship between the study, antioxidant activity of black raspberry was supposed to affect by the anthocyanin, phenol and flavonoid contents.

A comparison of the characteristic properties between soybean (Glycine max [L.] Merrill) seeds with different seed coat colors

  • We profiled the health-promoting bioactive components in nine types of soybean seeds with different seed coat colors (yellow, green, brown, and black) and investigated the effects of different extraction solvents (methanol, ethanol, and water) on their antioxidant activities. The carotenoid and anthocyanin compositions varied greatly by seed color, and the phenolic acids, total phenol, and total flavonoid contents differed by genotype. The carotenoid content was relatively higher in soybean seeds with green and black seed coats than in those with a yellow seed coat while lutein was the most plentiful. The anthocyanin content was considerably higher in the soybean seed with the black seed coat. The results of the DPPH assay showed strong antioxidative activities in the methanol- and water-extracts compared to the ethanol-extract, irrespective of the seed coat colors. Moreover, the soybean seeds with the black seed coat exhibited the highest antioxidant activity among the samples, regardless of the extraction solvent used. Eighteen bioactive compounds were subjected to data-mining processes including principal component analysis and hierarchical clustering analysis. Multivariate analyses showed that brown and black seeds were distinct from the yellow and green seeds in terms of the levels of carotenoids and anthocyanins, respectively. These results help our understanding of the compositional differences in the bioactive components among soybean seeds of various colors, providing valuable information for future breeding programs that seek to enhance the levels of compounds with health benefits.

Comparison of the Antioxidant Activities and Nitrite Scavenging Activity of Black Chokeberry (Aronia melanocarpa) Extracts (블랙 초크베리 추출물의 항산화 활성 및 아질산염 소거 활성)

  • The purpose of this study was to measure the antioxidant activity and bioactivity of black chokeberry (Aronia melanocarpa). The black chokeberry was known to contain many physiologically active substance, such as ascorbic acid, anthocyanin, quercetin. We extracted black chokeberry powder from four solvents such as chloroform:methanol(CM, 2:1, v/v), distilled water (DW), 70% ethanol, 70% methanol. After that, we determined anthocyanin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power and Nitrite scavenging activity of black chokeberry extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of black chokeberry extracts was the highest in 70% methanol (p<0.05). The results suggest that black chokeberry can be used as nutraceutical foods and natural antioxidant.

Variation of Functional Materials and Antioxidant Activity as Affected by Cultivation Environment in Pigmented Rice Varieties (재배환경에 따른 유색미의 기능성물질 및 항산화활성 변이)

  • Production of high quality pigmented rice contained with high content of anthocyanin, and polyphenol was significantly influenced by cultivation environments like transplanting date and cultivation location. This study was carried out to establish an optimum transplanting date and cultivation region to produce maximum content of anthocyanin, polyphenol and antioxidant activity of pigmented rice varieties (black, red colored). Three transplanting times (May 20, June 5, June 20) and four different sites (Miryang, Uiseong, Sangju, Bonghwa) were evaluated with five pigmented rice cultivars. Anthocyanin and total polyphenol index to average temperature during 30 days after heading (DAH) of black, red pigmented rice varieties showed that anthocyanin and total polyphenol contents were decreased by 10% and 9%, respectively, with increasing average temperature of $1^{\circ}C$. The optimum ripening temperature of the 30 DAH for the production of high anthocyanin and total polyphenol was 22 to $23^{\circ}C$ for early maturity black rice, 21 to $22^{\circ}C$ for mid-late maturity of black and red rices, respectively. On the other hand, an estimated heading date of pigmented rices in return according to the optimum ripening temperature of the 30 DAH was ranged Aug. 11 to 17 for early maturity black rice, Aug. 25 for mid-late maturity black, red rice variety in Jecheon, Aug. 27 to Sep. 2 for early maturity type, Sep. 3 to 6 for mid-late type in Daegu. It seemed that Jecheon, Boeun, Mungyeong, and Yeongju were optimum regions for cultivation of pigmented rices. The estimated sowing date of pigmented rices for high anthocyanin and total polyphenol production based on the optimum heading date was May 18 to 26 for early maturity black rice variety, April 11 for mid-late black, red variety in Jecheon, May 23 to 28 for early type, April 9 to 26 for mid-late type in Boeun, respectively.

Comparison of Bioactive Constituents and Biological Activities of Aronia, Blackcurrant, and Maquiberry (아로니아, 블랙커런트, 마키베리의 기능성 성분 및 생리활성 비교)

  • In this study, bioactive constituents and biological activities of aronia, blackcurrant, and maquiberry were investigated. The 60% ethanol extracts were prepared and evaluated for total polyphenol, total flavonoid, and total anthocyanin contents. Biological activities, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities, ferric reducing antioxidant power (FRAP), reducing power, tyrosinase inhibitory activity, and nitrite scavenging activity were also examined. Maquiberry showed the highest bioactive constituents as shown based on total polyphenol (73.66 mg GAE/g), total flavonoid (28.08 mg/g), and total anthocyanin (12.82 mg/g) contents compared to aronia and blackcurrant. Maquiberry also exhibited the highest DPPH and ABTS radical scavenging activity, FRAP, and reducing power at a concentration of 5 mg/mL, which were superior to those of ascorbic acid (a positive control). Therefore, maquiberry could be used as a potential source of antioxidants and functional food substances.

Biochemical Characteristics of Apple Rot Caused by Macrophoma sp. II. Phenolic Compound Content in Infected Fruits (Macrophoma sp.에 의한 사과 부패의 생화학적특성 II. 감염과일의 페놀함량)

  • Changes in levels of phenolic compounds such as total phenols, flavonols and anthocyanins in Macrophoma-infected apples were studied at various developmental stages of apple fruits. The amounts of total phenols in apple fruit flesh and peel drastically decreased as apples became mature. Apple rot resulted in concentration of total phenols somewhat lower than those of healthy apple flesh. The decline in amount of total phenols was distinct in infected fruit flesh of the cultivar Fuji, which was more susceptible to Macrophoma sp. than the cultivar Miller. Higher amounts of total phenols were found in infected than in heathy fruit peel. In the case of the cultivar Miller, increased accumulation of total phenols was pronounced in infected peel. Apple rot resulted in concentrations of flavonols much higher than those of healthy apples. In particular, the drastically increased accumulation of flavonols was detected in infected peel at the first collection on 10 July, when the cultivars tested were completely resistant to Macrophoma sp. Production of anthocyanins was increased considerably by apple rot: anthocyanins in infected fruits of the cultivar Miller increased markedly as compared with their concentration from healthy fruits. These results suggest that the altered phenolic metabolism in apple fruits may be associated with the development of apple rot.

A study on the Development of Grape Vinegar Added Drink Grape Vinegar (포도 식초 첨가 건강음료 개발)

  • To develop the processing methods of grape and increase the use of poor fruits, we researched the health drink made with them after making grape juice and vinegar. The grape heated at $80^{\circ}C$ during 30min was followed by filtration. Grape vinegar was prepared by alcohol and acetic acid fermentation of grape. Optimum initial alcohol concentration for acetic acid fermentation was 6-8%. Acetic acid fermentation of grape vinegar manufactured in onggi took 9 days and was faster than any other type of utensil. As the result of analysis of grape vinegar fermented in the various ratio of 'Campbell Early' and 'Kyoho' grape, the redness and total anthocyanin content and the score of sensory evaluation were higher in vinegar made with 100% 'Campbell Early'. The drink manufactured by adding grape vinegar was developed and adding 10% of grape vinegar to volume of grape juice and water mixture(1:1) was best in sensory evaluation. In this mixture, sugar content was $14^{\circ}$Bx.
