철에 노출된 넙치, Pacalichthys olivaceus의 혈액화학적 변동 (Hemochemical Changes in Olive Flounder, Paralichthys olivaceus Exposed to Various Iron Concentrations)

  • 주요 양식어류인 넙치를 대상으로 $0.1\sim10mg/\ell$ 범위의 철 농도에서 순환식 방법에 의해 50일 동안 사육하면서 혈액화학변동을 조사하였다. 넙치의 혈청 iron과 magnesium 농도는 1.0 mg/$\ell$의 철 농도에서 20일째부터 유의한 증가를 나타냈다. 혈청 calcium농도는 철 농도 1.0 mg/$\ell$에서 는 30일째부터 유의한 감소를 나타냈으나, inorganic phosphate의 뚜렷한 농도변화는 관찰되지 않았다. 혈청 total protein, albumin, glucose 농도는 철 농도 1.0 mg/$\ell$에서 노출 30일째부터 유의하게 감소하였고, total cholesterol 농도는 1.0 mg/$\ell$ 철 농도에서 노출 40일째부터 유의한 증가가 관찰 되었다. 혈청 GOT, GPT, LDH 및 amylase 활성은 철 농도 1.0 mg/$\ell$에서 40일째부터 유의하게 증가하였으나, Al-P의 뚜렷한 변동은 관찰되지 않았다. 이상의 결과는 철에 노출된 넙치 일부의 혈청 무기성분, 유기성분 및 효소활성은 철 농도 1.0mg/$\ell$ 이상에서 적어도 30일째부터는 변동한다는 것을 의미한다.

고품질의 청국장 생산 발효균주 선별 및 최적화 (Selection of Microorganisms and Optimization of Manufacture Process for Cheonggukjang)

  • 본 연구는 청국장의 품질을 향상시키고 최적화시키기 위해 경기도 이천에서 높은 효소가를 가진 균주를 분리 동정하며 선택된 균주들을 이용하여 최적배합비를 산출하고자 하였다. 16S rDNA와 PCR을 이용하여 균주를 동정한 결과, 39개의 균주 대부분이 B. subtilis와 B. licheniformis이었으며 곰팡이는 검출되지 않았고 그 중 5개의 균주가 높은 역가를 나타내었다. 고품질의 장류생산 및 최적화 공정을 확립하기 위해 분리 동정한 균주중에서 amylase, protease, lipase와 cellulase의 활성이 높은 균주를 3개 선택하여 최적화 공정작업에 이용하였고 실험법에 의해 청국장을 제조하였다. 암모니아태질소, 선호도, 아미노태질소 및 항산화가에 근거를 두어 contour map과 numerical 최적화를 한 결과, 50%의 Bacillus sp. SC-l과 50% SC-3을 섞었을 때 최적의 청국장이 만들어졌다.

  • The studies of isolation and physiological characteristics of mesophilic or thermophilic bacteria from thermophilic oxic process (TOP) treating Chinese restaurant wastes were conducted. Chinese restaurant wastes were consist of moisture; 75.8%, solids; 24.2% and ash; 0.49%. The volatile solid was about 99% of total dry solids. In wastes used in this experiment, there was content of crude protein; 4.47%, crude lipid; 3.56%, free sugar, 0.4% , crude starch; 10.34% and crude fiber 3.14%, respectively. And then it has about 4,970 kcal/kg-dry solid of Chinese restaurant wastes. From TOP treating the chinese restaurant wastes, 37 strains of mesophilic or thermophilic bacteria were primarily isolated using medium used for the isolation and among them 6 strains of thermophilic and 7 strains of mesophilic bacteria were selected by testing the activities of amylase, cellulase, protease and lipase. TB-1, TB-9 as thermophilic bacteria and MB-15-1, 15-2, MB23 as mesophilic bacteria having strong enzyme activity were selected among isolated strains. All selected strains reduced nitrate to nitrite and they utilized glucose, manose, manitol, and maltose as carbon source. From these MB15-2 was identified as Bacillus cereus, TB1, Bacillus licheniformis and TB9; Bacillus schlegelii.

Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과 (Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

Effects of Enzyme Supplementation on Growth, Intestinal Content Viscosity, and Digestive Enzyme Activities in Growing Pigs Fed Rough Rice-based Diet

  • The purpose of the present study was to investigate the effects of exogenous non-starch polysaccharides (NSP) enzymes on performance, intestinal content viscosity and digestive enzyme activities of growing pigs fed a rough rice-based diet. A total of 60 crossbred barrows with an initial body weight of 35.16 kg (SD = 0.82) were blocked by body weight and randomly assigned to two treatments with three replications. Each group was fed the diet based on rice with or without exogenous NSP enzymes (2 g/kg of diet). During the 70 days of the feeding trial, all pigs were given free access to feed and water. At the end of the feeding trial, six pigs from each treatment were randomly selected and slaughtered to collect intestinal digesta, intestinal mucosa, and pancreas. The addition of NSP enzymes improved average daily gain (p<0.05) and feed:gain (p<0.05), and decreased viscosity of digesta in the jejunum (p<0.001) and ileum (p<0.01) of pigs. The supplementation of NSP enzymes increased activities of protease (p<0.01), trypsin (p<0.01) and ${\alpha}$-amylase (p<0.05) in duodenal contents. However, digestive enzymes in the pancreas, jejunal and ileal mucosa were unaffected by the supplemental NSP enzymes (p>0.10). The results indicate that the addition of NSP enzymes to rough rice-based diets improved performance of pigs, reduced viscosity and increased digestive activity in the small intestine.

소아의 다발성 치아우식증과 연관된 타액의 생화학적 특성 (BIOCHEMICAL CHARACTERISTICS OF SALIVA TO BE LINKED TO THE MULTIPLE CARIES IN CHILDREN)

  • Saliva is obviously potential medium to protect the dental caries by not only physical clearing effect, but aggregating action of protein with bacteria. Nevertheless, we still do not understand how the dental caries occur and what brings the individual difference in caries prevalence. In the regards of dental caries prevalence, we hypothesized that the composition of salivary protein might be different from caries susceptible group to caries resistant group. The purposes of this experiment were focused on the molecular analysis of salivary proteins from the subjects who were involved in multiple caries. Electrophoretic analysis was done on the whole saliva collected from the children with and without multiple caries. We found 86.2% of subjects with multiple caries has approximately 120 KDa protein band while 30.4% in the healthy subjects. And the concentration of the total protein on the subjects with multiple caries is significantly higher than that of the healthy group. However, it turned out that the difference of the salivary composition does not affect the bacterial adhesion to hydroxyapatite bead. With regards of enzymes in saliva, the activity of ${\alpha}-amylase$ and lactate dehydrogenase does not have any significant difference between both groups. However, the concentrations of $Na^+\;and\;Cl^-$ in saliva from multiple caries group is higher than that of the control group. Taken all together, it may be concluded that 120 KDa protein in saliva may be associated with the process of dental caries, also the high concentration of protein and $Na^+,\;Cl^-$ in saliva may be linked to dental caries development as a cofactors.

Properties of Cheonggukjang Fermented with Bacillus Strains with High Fibrinolytic Activities

  • We previously isolated Bacillus strains with high fibrinolytic activities (FAs) from cheonggukjang prepared by traditional ways. To test their potential as starters for cheonggukjang, soybean was fermented for 72 hr at $37^{\circ}C$ with each isolate and a control lab strain: B. subtilis CH3-25 (BS3-25), B. amyloliquefaciens CH51 (BA51), B. amyloliquefaciens CH86-1 (BA86-1), and B. subtilis 168 (BS168, control, lab strain). Viable cell numbers of all cheonggukjang samples rapidly increased and reached about $10^9$ CFU/g after 6 hr. During 72 hr, the initial pH of 6.3 rapidly increased to 8.1$\sim$8.2 for cheonggukjang fermented with BS3-25 or BA86-1, and 7.3 for those with BA51 or BS168. FAs and protease activities (acid, neutral, and alkaline) rapidly increased in cheonggukjang fermented with BS3-25, BA51, or BA86-1 during the first 12 hr. On the other hand, those of cheonggukjang fermented with BS168 slightly increased during the first 36 hr. There were significant changes in acid and neutral protease activities in cheonggukjang fermented with BA51 or BA86-1 during the 24 hr. Rapid increases of $\beta$-glucosidase activity corresponded well with rapid increases of $\alpha$-amylase and $\alpha$-galactosidase activities in addition to increases in antioxidant activities and the TPCs (total phenolic contents). The highest increase in the TPCs was observed in cheonggukjang fermented with BA86-1 while the least was that fermented with BS168.

Effects of Dietary Fructooligosaccharide on Digestive Enzyme Activities, Intestinal Microflora and Morphology of Growing Pigs

  • One hundred and twenty-eight growing barrows (Jiaxing Black${\times}$Duroc${\times}$Landrace) at an average BW of 20.8 kg were allocated to four treatments for 42 days, each of which was replicated four times with eight pigs per replicate and used to investigate the effects of fructooligosaccharide (FOS) on digestive enzyme activities, intestinal microflora and morphology of growing pigs. The pigs received the same basal corn-soybean meal diet and FOS was added to the basal diet at 0, 2, 4, 6 g/kg diet at the expense of corn, respectively. As compared to control, supplementation with 4 and 6 g/kg FOS significantly improved average daily gain and feed efficiency. Addition of FOS enhanced the growth of Bifidobacterium and Lactobacillus, but inhibited Clostridium and Escherichia coli in the small intestinal and proximal colonic contents. Supplementation with 4 and 6 g/kg FOS significantly improved the activities of total protease, trypsin and amylase in the small intestinal contents. However, FOS had no significant effect on the activity of lipase in the small intestinal contents as well as the digestive enzymes in pancreas. Morphological measurement of jejunal mucosa did show response to consumption of FOS. Villus height and the villus height to crypt depth ratio at the jejunal mucosa were significantly higher with 4 and 6 g/kg FOS supplementation as compared to control.

Effect of Lactic Acid Producing Bacteria on the Performance of Male Crossbred Calves Fed Roughage Based Diet

  • To study the effect of feeding lactic acid producing bacteria on the performance of cattle calves, twenty four, day old male crossbred cattle calves (Bos indicus${\times}$Bos taurus), were distributed into two groups of 12 animals each. The animals were fed on calf starter containing wheat bran and green berseem ad libitum and milk as per requirement upto 8 weeks of age. The diet of calves of Group 2 was supplemented with 500 ml culture of Lactobacillus acidophilus-15. Total duration of the experiment was 31 weeks. There was no significant difference in intake and digestibility of dry matter (DM), organic matter (OM), neutral detergent fibre (NDF), acid detergent fibre (ADF) and crude protein (CP) between the groups. The rumen pH, protozoa numbers, concentration of volatile fatty acids (VFA), ammonia nitrogen ($NH_3-N$), trichloroacetic acid precipitable nitrogen (TCA-ppt N) and activity of microbial enzymes (carboxymethylcellulase, xylanase, amylase and protease) were not affected due to probiotic supplementation. Average live weight gain of the calves was improved (about 10%) and feed:gain ratio was reduced (about 5%) in the animals given Lactobacillus culture. The data indicated that crossbred calves could be reared on a diet devoid of cereal grain and addition of Lactobacillus culture in the diet resulted in an added advantage in growth performance of the animals.

Optimum Water Potential, Temperature, and Duration for Priming of Rice Seeds

  • Experiments were carried out to find out the optimum water potential, temperature, and duration for the priming of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) for better germination at sub-optimal temperatures. Seeds were primed in 0, -0.2, -0.4, -0.6, -0.8, and -1.0 MPa PEG (polyethylene glycol) solutions at $25^{\circ}C$. The optimum water potential for seed priming, the highest water potential at which rice seeds did not germinate, was -0.6 MPa. To find out optimum priming temperature and duration rice seeds were primed in -0.6 MPa PEG solution and 0 MPa (water as a control) for various durations at 15 and $25^{\circ}C$ and the seeds were germinated at 17, 20, and $25^{\circ}C$. Considering germination rate and speed, the optimum priming time in water (0 MPa) was 4 days at 15$^{\circ}C$ and 1 day at $25^{\circ}C$, while 4 days was the optimum priming time in a -0.6 MPa PEG solution, regardless of the priming temperature. Priming reduced the actual time of germination, especially at sub-optimal temperatures. Priming did not affect germination rate in -0.6 MPa PEG solution at 15$^{\circ}C$, but overpriming reduced the final germination rate in water at 15$^{\circ}C$ and in -0.6 PEG solution at $25^{\circ}C$. Total sugars and $\alpha$-amylase activity induced during the seed priming were negatively correlated with the final germination rate and there was no noted relationship with the speed or uniformity of germination.
