The contents and the compositions of total free amino acids were investigated in the rice(Oryza sativa L. cv. Chuncheong) seedlings under low temperatures. Activities of some enzymes associated with the markedly changed amino acid content were also investigaetd. Under low temperature, the contents of soluble protein and the total free amino acids increased, while the content of total nitrogen decreased. Although asparagine+glycine were the most abundant amino acid speceis in the rice seedlings at the control temeprature, low temperature treatment for 3days brought about the decrease in their amount to about 60% level of the control plants. On the other hand, alanine showed the highest increase in the content among all the free amino acids, though glutamine, proline, asprtic acid, valine and tyrosine also increased after low temperature treatment. To eludicate the decrease of asparagine+glycine level under low temperature, the activities of asparagine aminotransferase and asparaginase which metabolize asparagine were investigated in the rice seedlings under low temperature. The activity of asparaginase increased markedly, while that of asparagine aminotransferase decreased under low temperatures. Therefore, it was suggested that asparaginase metabolizes asparagine predominatly in the rice seedlings under low temperatures.
At the different growing stages of Fagopyrum esculentum Moench planted at the green house and out doors, the contents of free amino acids in the whole plants,were determined quantitatively by spot-extract colorimetric method (J. Awapara method) using the chromatograms obtained by the ion-exchange resin and paper chromatographic method. And the contents of carbohydrate in the whole plant were determined by Bertrand method. The results were summarized as follows. 1. Almost same kinds of free amino acids were detected in the both plants. 2. Concentrations of most amino acids was increased progressively during growth; further growth was accompanied by decreased concentrations. 3. Rate of growth and increasing of total amino acid content of green house plant is greater than that of out dorrs plant. 4. At the same stages of gwoth, total free amino acid content of green house plant is higher than that of out doors plant but sugar content is lower. 5. There was rapidly increasing of sugar content and decreasing of total free amino acid content except asparagine during mature of seeds.
This study was conducted to investigate the effects of adding Tenebrio molitor L Powder to broiler feed on fatty acid profiles in broiler breast meat. In total, 180 broiler chicks (1-day old Arbor Acres) were included. The birds were randomly divided into control and treatment groups, with 3 replicate subgroups each(30 birds per subgroup), and fed a diet for 35 days without (control) or with 1% Tenebrio molitor L powder (treatment). Among individual fatty acids, addition of Tenebrio molitor L. powder resulted in slightly higher C18:1n-9, C20:3n-3 and C20:3n-6 contents, and lower C18:2n-6 content compared withcontrols (p<0.05). No remarkable differences in total SFA and total USFA contents were found between groups. In conclusion, inclusion of Tenebrio molitor L. in broiler diets did not improve overall fatty acid profiles.
By using a synthetic medium simulated on the amino acid composition of soybeam, the effect of salt concentrations on the production of volatile organic acid by the strains of Zygosaccharomyces rouxii So-3101, a soy sauce yeast, was studied at the concentrations of 12.5, 18.0, 22.0 and 28.5% NaCl. The growth, consumption of glucose, and production of alcohol, total acid and volatile organic acid, showed the highest values at a concentration of 12.5% NaCl, and those values were decreased with an increase in the salt concentration. The ratio of volatile organic acid to total organic acid was remained at approximately the same level within the range of salt concentrations between 12.5~22.0%, whereas the ratio was decreased at a salt concentration of 28.5%. After incubation for 16 days, 8 volatile organic acids, i.e. acetic, propionic, n-butyric, isobutyric, isovaleric, isocaproic, n-caproic, and heptanoic acids, were detected by gas chromatography. Among the volatile organic acids, acetic acid was produced in the appreciable amiunt and its ratio to the other volatile acids was increased with an increase in the salt concentration.A small amount of isocaproic, propionic, isobutyric and isovaleric acids were produced, and n-caproic, n-butyric and heptanoic acids were detected only at the lower salt concentration.
Amino acids and ribonucleotides were measured for boiled-dried anchovy to determine the effect of 5 kGy γ-irradiation on its quality stability during storage at ambient and cooling (5∼10℃) temperatures for 12 months in a laminated-film package(Polyethylene 10㎛). The anchovy samples contained about 55%(d.b) of total amino acids and about 1%(d.b) of free amino acids. Although there found a significant change(p<0.01) in the content of leucine and lysine immediately sfter irradiation, the overall content of toral amino acids was little changed in stored anchovy even six months after irradiation at cooling conditions. γ-Irradiation caused a significant reduction(p<0.01) in the total content of free amino acids, showing a similar tendency by storage conditions. However, the ribonucleotides. which were 12.00mg/g(d.b) in inosine-5'-monophosphate and 0.38mg/g(d.b) in guanosine-5'-monophospate, were resistant to 5kGy-irradiation. With the lapse of storage time, it was also shown that storage temperature was more influential than irradiation on the contents of amino acids and taste compounds of dried anchovy.
To produce buckwheat sprouts, buckwheats were germinated at $23{\pm}2^{\circ}C$ up to 8 days in total darkness. In proximate analysis, moisture content increased from 13.87% of buckwheat groats to 93.75% of buckwheat sprouts on the 8th day of germination. On dry weight basis, lipid and ash contents increased like as protein content increased from 13.45% to 21.82% while carbohydrate content decreased due to enzyme hydrolysis. Amino acids were rich in glutamic acid, aspartic acids, and lysine, and also the ratio of the essential amino acids to total amino acids increased from 26.84% to 36.84%. Vitamin C did not exist in buckwheat groats but its content has continued to increase as far as 99.56 mg/100 g buckwheat sprouts. Rutin continued to increase by HPLC analysis from 4.71 mg/100 g buckwheat groats to 1,524 mg/100 g buckwheat sprouts. In analysis of organic acids, oxalic, maleic, and citric acids were commonly found in buckwheat sprouts. Fagopyrin was found almost none in buckwheat sprouts. In conclusion, the conversion of buckwheat seeds into sprouts through germination in darkness results in physically different final product with nutritional changes such as higher content of rutin, generation of vitamin C, abundance of the essential amino acids, and the existence of fagopyrin on very little.

The contents of free sugars, organic acids, amino acids, total carotenoids and pectin of pumpkin were analysed. Weight percentage of flesh was 84% of total weight. Rind and funicular attachment of seed were 10% and 3.5%, respectively. Major free sugars in pumpkin were fructose, glucose, sucrose. In flesh, fructose and glucose were the major free sugars, corresponding to 87% of total free sugars . Total sugar content in the flesh was three times higher than that in the funicular attachment of seed. Contents and composition of free amino acids were quite different according to the different parts of pumpkin. Non-volatile organic acids in pumpkin was malic, succinic and fumaric acid. The major acid in the flesh and funicular attachment of seed was malic acid and the contents in these parts were 85.2mg% and 226.5mg%, corresponding to 63% and 70% of the total organic acids, respectively. Eigh-tyseven percent of total carotenoids in the pumpkin was in the funicular attachment of seed, amounting 65.3mg%. However flesh and rind contained 6.6mg% and 3.3mg%, respectively. Water soluble pectin consisted 58% of the total soluble pectin in different parts of pumpkin, and soluble Pectin content was higher in funicular attachment of seed than in flesh. The major neutral sugars of the pectin in flesh and funicular attachment of seed were galactose and glucose consisting 45% and 36.5% of total neutral sugars.

To explore the possibility of using freeze dried loach for instant choo o tang(Korean traditional loach soup), protein qualities and fatty acid composition were evaluated on boiled and steamed loach. Total lipid and ash content were lowered in both heated(boiled and steamed) loaches due to deboning and eviscerating during cooked meat preparation. Profiles of total amino acids were not changed seriously by the type of cooking, but the amount of essential amino acids were comparable in all samples. Two times more free amino acids were quantified in cooked samples compared to raw meat. Available lysine was marginally decreased by cooking, and that caused some measurable change in typsin indigestible substrate(TI) in streamed whole loach. In vitro protein digestibility of the heated loaches was not altered drastically and the protein quality determined as computed protein efficiency ratio(C PER) was similar for the raw, boiled and steamed loach. The ratios of unsaturated to saturated fatty acids changed measurably in heated whole loach. The results shows that heating caused apparent oxidative deterioration of the polyunsaturated fatty acids.
This study was conducted to investigate the presence of biologically active substances such as phenolic acids, fatty acids and organic acids in the medical plants like Kalopanax pictum and Acanthopanacis cortex. Alcohol extracts of K. pictum and A. cortex showed complete inhibition of lettuce seed germination, indicating that these plants contained the biologically active substances. Eleven phenolic acids including protocatechuic acid were identified from K. pictum and A. cortex by GLC, and the contents of total phenolic acid were 1.7917mg/g in K. pictum, and 0.9567mg/g in A. cortex. Polyphenols such as neochlorogenic acid, chi orogenic acid, scopoletin, rutin and kaempferolglycoside which were not detected by GLC were anayzed by HPLC, and among phenolic acids indentified chi orogenic acid seemed to be the major acid in both K. pictum and A. cortex presented in amount of 23.7 and 13.0ppm, respectively. K. pictum contained 5.26mg/g of fatty acids and 27.69mg/g of organic acids, and A. cortex possessed 3.22mg/g of fatty acids and 9.80mg/g of organic acids, linoleic and oxalic acid appeared to be the major fatty and organic acids, representing more than 50% of total fatty acids and 80% of total organic acids.
The fatty and organic acids, and barbaloin in various parts of aloe species dried at different temperatures were analyzed by GC and HPLC. Seven fatty acids and six organic acids were identified and quantified. In the case of fatty acids, generally, the contents of palmitic and eicosanoic acid were abundants, and compared to the total contents of seven fatty acids, Aloe arborescence variant 1 was abundant, but Aloe saponaria was poor. And six fatty acids were distributed in the aloe species with the exception of linoleic acid. The contents of malic, citric and oxalic acids in the aloe species were higher than those of other acids, and compared to the sum of contents of six organic acids, Aloe saponaria was high, but Aloe arborescence variant 1 was low. Therefore Aloe arborescence variant 1 was abundant in total fatty acids, but poor in total organic acids. The contents of fatty and organic acids in the sample dried at $65^{\circ}C$ and $80^{\circ}C$ air circulation were almost similar. The contents of fatty and organic acids in the freeze-dried samples were lower than in the other dried samples. The contents of barbaloin in Aloe arborescence and A. arborescence variant 1 were higher than those of other various samples, and barbaloin was not detected in Aloe saponaria. As the drying temperature was increased, the contents of barbaloin in the various parts of the Aloe vera decreased.
