Regulatory Role of Autophagy in Globular Adiponectin-Induced Apoptosis in Cancer Cells

  • Adiponectin, an adipokine predominantly secreted from adipose tissue, exhibits diverse biological responses, including metabolism of glucose and lipid, and apoptosis in cancer cells. Recently, adiponectin has been shown to modulate autophagy as well. While emerging evidence has demonstrated that autophagy plays a role in the modulation of proliferation and apoptosis of cancer cells, the role of autophagy in apoptosis of cancer cell caused by adiponectin has not been explored. In the present study, we demonstrated that globular adiponectin (gAcrp) induces both apoptosis and autophagy in human hepatoma cell line (HepG2 cells) and breast cancer cells (MCF-7), as evidenced by increase in caspase-3 activity, Bax, microtubule-associated protein light chain 3-II (LC3 II) protein levels, and autophagosome formation. Interestingly, gene silencing of LC3B, an autophagy marker, significantly enhanced gAcrp-induced apoptosis in both HepG2 and MCF-7 cell lines, whereas induction of autophagy by rapamycin, an mTOR inhibitor, significantly prevented gAcrp-induced apoptosis in hepatoma cells HepG2. Furthermore, modulation of autophagy produced similar effects on gAcrp-induced Bax expression in HepG2 cells. These results implicate that induction of autophagy plays a regulatory role in adiponectin-induced apoptosis of cancer cells, and thus inhibition of autophagy would be a novel promising target to enhance the efficiency of cancer cell apoptosis by adiponectin.

Improvement of Leptin Resistance (렙틴 저항성의 개선)

  • Leptin, a 16-kDa cytokine, is secreted by adipose tissue in response to the surplus of fat store. Thereby, the brain is informed about the body's energy status. In the hypothalamus, leptin triggers specific neuronal subpopulations (e.g., POMC and NPY neurons) and activates several intracellular signaling events, including the JAK/STAT, MAPK, PI3K, and mTOR pathway, which eventually translates into decreased food intake and increased energy expenditure. Leptin signal is inhibited by a feedback inhibitory pathway mediated by SOCS3. PTP1B involves another inhibitory pathway of leptin. Leptin potently promotes fat mass loss and body weight reduction in lean subjects. However, it is not widely used in the clinical field because of leptin resistance, which is a common feature of obesity characterized by hyperleptinemia and the failure of exogenous leptin administration to provide therapeutic benefit in rodents and humans. The potential mechanisms of leptin resistance include the following: 1) increases in circulating leptin-binding proteins, 2) reduced transport of leptin across the blood-brain barrier, 3) decreased leptin receptor-B (LRB), and/or 4) the provocation of processes that diminish cellular leptin signaling (inflammation, endoplasmic reticulum stress, feedback inhibition, etc.). Thus, interference of the cellular mechanisms that attenuate leptin signaling improves leptin action in cells and animal models, suggesting the potential utility of these processes as points of therapeutic intervention. Various experimental trials and compounds that improve leptin resistance are introduced in this paper.

Serine 389 phosphorylation of 3-phosphoinositide-dependent kinase 1 by UNC-51-like kinase 1 affects its ability to regulate Akt and p70 S6kinase

  • Phosphorylation of the signaling component by protein kinase often leads to a kinase cascade or feedback loop. 3-Phosphoinositide-dependent kinase 1 (PDK1) signaling pathway diverges into various kinases including Akt and p70 S6 kinase (p70S6k). However, the PDK1 feedback mechanism remains elusive. Here, we demonstrated that UNC-51-like kinase (ULK1), an autophagy initiator kinase downstream of mechanistic target of rapamycin (mTOR), directly phosphorylated PDK1 on serine 389 at the linker region. Furthermore, our data showed that this phosphorylation affected the kinase activity of PDK1 toward downstream substrates. These results suggest a possible negative feedback loop between PDK1 and ULK1.

  • This paper is a research for removing the alternators hunting which came about during the par-allel running. The four-pole six-phase alternator is equipped with the converter made out of twelve diodes to rectify all the waves. The hunting came about during the battery charging due to the hunting inducing point existed in r. p. m band 1575[rpm-1690[rpm] To remove the hunting inducing point We modified the four-pole six-phase alternator into the four-pole twevel-phase and increased the short-circuit ratio by decreasing the field coil pitch and increasing the field coil turns. But this method has two defects the first the alternator structure becomes complicating and the second the second the alternator equipped with the converter used in general purpose lately such as the car alternator can not be run in parallel wlthout modifying. To remove the defects the equalizing line was connected between the same phase of the alterna-tor to flow the synchronizing current which syncronize the phases of generator electromotive forces by which the alternator can be run in parallel without reference to the hunting inducing point and without modifying.

Signal Transducer and Activator of Transcription 3 - A Promising Target in Colitis-Associated Cancer

  • Colorectal cancer (CRC) is the third most common malignancy and fourth most common cause of cancer mortality worldwide. Untreated chronic inflammation in the intestine ranks among the top three high-risk conditions for colitis-associated colorectal cancer (CAC). Signal Transducer and Activator of Transcription 3 (STAT3) protein is a member of the STAT family of transcription factors often deregulated in CRC. In this review, we try to emphasize the critical role of STAT3 in CAC as well as the crosstalk of STAT3 with inflammatory cytokines, nuclear factor (NF)-${\kappa}B$, PI3K/Akt, Mammalian Target of Rapamycin (mTOR), Notch, $Wnt/{\beta}$-catenin and microRNA (MiR) pathways. STAT3 is considered as a primary drug target to treat CAC in humans and rodents. Also we updated the findings for inhibitors of STAT3 with regard to effects on tumorigenesis. This review will hopefully provide insights on the use of STAT3 as a therapeutic target in CAC.

  • Let R be a ring and n a fixed non-negative integer. $\cal{TI}_n$ (resp. $\cal{TF}_n$) denotes the class of all right R-modules of FGT-injective dimensions at most n (resp. all left R-modules of FGT-flat dimensions at most n). We prove that, if R is a right $\prod$-coherent ring, then every right R-module has a $\cal{TI}_n$-cover and every left R-module has a $\cal{TF}_n$-preenvelope. A right R-module M is called n-TI-injective in case $Ext^1$(N,M) = 0 for any $N\;{\in}\;\cal{TI}_n$. A left R-module F is said to be n-TI-flat if $Tor_1$(N, F) = 0 for any $N\;{\in}\;\cal{TI}_n$. Some properties of n-TI-injective and n-TI-flat modules and their relations with $\cal{TI}_n$-(pre)covers and $\cal{TF}_n$-preenvelopes are also studied.

A Study on the Relationship between Sasang Constitution and Clothing Value (사상체질과 의복가치관의 연구)

  • The purpose of this study is to grasp the re-lationship between Sasang constitution of the disposition side and clothing value in grown-up women in order to apply Sasang consti-tution theory to the marketing strategy in clothing industry. The study is based on a questionnaie about Sasang constitution and clothing value. The questionnarie was answered by 519 women. The data ere collected from September 12 to September 30, 1994 and analyzed by cluster analysis correlation analysis ANOVA and fac-tor analysis using SAS. The Findings of this study are as follows: 1. The women were classfied in to 3 clusters by cluster analysis :Sp-Yaung-In type (30.8%) Tae-Em-In type (40.7%) So-em-in(28.5%). 2. Correlations between SO-Yaug In type and each of clothing values showed all signifi-cant and specially social value showed high corelation(0.57163) 3. In Tae-Em-In type all correlations showed significant and esocial value showed the highest correlation of all correlations. 4. In So-Em-In type there was no corre-lation inpolitical value and the order of cloth-ing values showing high correlation was theor-etical social economic and religious value. 5. The aesthetic valu among and consti-tution types showed the lowest correlation of all correlation. By means of the about study results we may know that all women belong to three types think social value of the more important value than any other in clothing value. This is because grown-up women in korea prefer yet clothing of the harmonic and similar type to this of individual type.

Frictional Characteristics of Silicon Graphite Lubricated with Water at High Pressure and High Temperature (고온 고압에서 물로 윤활되는 실리콘그라파이트 재질의 마찰 특성에 관한 연구)

  • Experimental frictional and wear characteristics of silicon graphite materials is studied in this paper. Those specimens are lubricated with high temperature and highly pressurized water to simulate the same operating condition for the journal bearing and the thrust bearing on the main coolant pump bearing in the newly developing nuclear reactor named SMART(System-integrated Modular Advanced ReacTor). Operating condition of the bearings is realized by the tribometer and the autoclave. Friction coefficient and wear loss are analyzed to choose the best silicon graphite material. Pin on plate test specimens are used and coned disk springs are used to control the applied force on the specimens. Wear loss ana wear width are measured by a precision balance and a micrometer. The friction force is measured by the strain gauge which can be used under high temperature and high pressure. Three kinds of silicon graphite materials are examined and compared with each other, and each material shows similar but different results on frictional and wear characteristics.

  • When a safety valve equipped in a nuclear power plant opens in an instant by an accident, a moving shock wave propagates downstream the valve, inducing a complicated unsteady flow field. The moving shock wave may exert severe load to the structure. So, to reduce the load acting on the wall of POSRV, a gradual opening of POSRV is adopted in general. In theses connections, a numerical work is performed to investigate the effect of valve opening time on the unsteady flow fields downstream of the valve. Compressible, two-dimensional Navier-Stokes equations are used with the finite volume method. The obtained results show that sharp pressure rise through moving shock tor the case of instant opening is attenuated by employing the gradual opening of valve. It is turned that the flows for the two cases of gradual valve opening time show the similar to that of highly under-expanded one in jet structure having expansion and compression waves and Mach stem. Also, comparing with the results for the two cases of opening time, the shorter the valve opening is, the pressure gradient at the downstream of the valve becomes softly.

  • Software verification and validation(V&V) is a means to develop high-quality software and assure safety and reliability for software. Also, we can achieve the desired software quality through systematic V&V activities. The software to be applied safety critical system like nuclear power plants is required to setup the V&V methodology that comply with licensing requirements for nuclear power plants and should be performed V&V activities according to it. In this paper, we classified safety-critical, safety-related and non-safety for software according to safety function to be peformed and define V&V activities to be applied software grade. Also, we defined V&V activities, procedures and documentation for each phase of software development life cycle and showed techniques and management to perform V&V. Finally, we propose the V&V framework to be applied software development of SMART(System-integrated Modular Advanced ReacTor) MMIS (Man-Machine Interface System) and to comply with domestic licensing requirements.
