Inhibitory Abilities of Bacillus Isolates and Their Culture Filtrates against the Gray Mold Caused by Botrytis cinerea on Postharvest Fruit

  • Botrytis cinerea, a major phytopathogenic fungus, has been reported to infect more than 200 crop species worldwide, and it causes massive losses in yield. The aim of this study was to evaluate the inhibitory abilities and effects of Bacillus amyloliquefaciens RS-25, Bacillus licheniformis MG-4, Bacillus subtilis Z-14, and Bacillus subtilis Pnf-4 and their culture filtrates and extracts against the gray mold caused by B. cinerea on postharvest tomato, strawberry, and grapefruit. The results revealed that the cells of Z-14, culture filtrate of RS-25, and cells of Z-14 showed the strongest biocontrol activity against the gray mold on the strawberry, grape, and tomato fruit, respectively. All the strains produced volatile organic compounds (VOCs), and the VOCs of Pnf-4 displayed the highest inhibition values. Based on headspace solid-phase microextraction in combination with gas chromatography-mass spectrometry, esters accounted for the largest percentage of the VOCs produced by RS-25, MG-4, Z-14, and Pnf-4 (36.80%, 29.58%, 30.78%, and 36.26%, respectively). All the strains showed potent cellulase and protease activities, but no chitinase activity. RS-25, Z-14, and MG-4, but not Pnf-4, grew on chrome azurol S agar, and an orange halo was formed around the colonies. All the strains showed biofilm formation, fruit colonization, and lipopeptide production, which may be the main modes of action of the antagonists against B. cinerea on the fruit. This study provides the basis for developing natural biocontrol agents against the gray mold caused by B. cinerea on postharvest fruit.

  • $\textrm{T}_{5}$ progeny of one transgenic tomato line (To9) carrying antisense polygalacturonase (PG) cDNA was generated by selfing. Five $\textrm{T}_{5}$ plants were used to analyse in detail. The PG antisense gene was stably inherited through fifth generations. In all five $\textrm{T}_{5}$ plants, expression of the antisense transcripts were detected. In consequence, it led to a reduction of the PG enzyme activity in ripe fruit to between 37% and 65% that of normal. In two plants the expression of endogenous PG gene was inhibited in ripe fruit.

Changes of Sugar Components in Cell Wall Polysaccharides from Tomato Fruits during Ripening (토마토 과실의 성숙중 세포벽 구성다당류의 변화)

  • This study was conducted to understand the characteristics of fruit softening during ripening which causes deep loses in quality of horticultural products during storage and marketing process after harvest. The changes of cell wall components during ripening was investigated. The climacteric rise was between 42 and 49 days after anthesis and then decreased. Ethylene evolution was similar to respiration. The hardness of fruit decreased markedly at this climacteric period and significances of textural parameters among the ripening periods were recognized but the significance between 50 and 55 days after anthesis was not. Sugar components of cell wall polysaccharides were uronic acid, galactose, glucose, arabinose, xylose, rhamnose, mannose and fucose. The contents of arabinose and mannose in alcohol-insoluble solids fraction increased, but other sugars were not changed. In cell wall fraction, the contents of uronic acid, galactose, glucose and arabinose were comparatively high, but galactose, arabinose and ironic acid were decreased markedly during ripening. ironic acid occupied above 75% of total monosaccharide in pectin fraction and decreased markedly during ripening. In acid-soluble hemicellulose fraction, the contents of uronic acid, glucose, galactose and rhamnose were high and they decreased from 50 days after anthesis. The contents of glucose and xylose were high in a alkali-soluble hemicellulose fraction and they decreased markedly at 55days after anthesis.

Partial Purification and Characterization of Superoxide Dismutase from Tomato (Lycopersicon esculentum) Fruit

  • Superoxide dismutase (SOD) from tomato (Lycopersicon esculentum Mill.) fruit was purified by ammonium sulphate precipitation, Sephadex G-100 and DEAE-cellulose column chromatographies. A 22 fold purification and an overall yield of 44% were achieved. The purified enzyme was a homodimer with Mr 37.1 kDa and subunit Mr 18.2 kDa as judged by SDS-PAGE. SOD showed $K_{m}$ values of 25 ${\times}$ 10$^{-6}$ M and 1.7 ${\times}$ 10$^{-6}$ M for nitroblue tetrazolium (NBT) and riboflavin as substrates, respectively. The enzyme was thermostable upto 5$0^{\circ}C$ and exhibited pH optima of 7.8. The effect of metal ions and some other compounds on enzyme activity was studied. $Co^{2+}$ and $Mg^{2+}$ were found to enhance relative enzyme activities by 27 % and 73 %, respectively, while M $n^{2+}$ inhibited the SOD activity by 64%. However, $Ca^{2+}$ and C $u^{2+}$ had no effect on enzyme activity. Other compounds like $H_2O$$_2$ and Na $N_3$ inhibited enzymatic activities by 60% and 32%, respectively, while sodium dodecyl sulphate (SDS), chloroform plus ethanol and $\beta$-mercaptoethanol had no effect on the activity of SOD. of SOD.

Relativeness between Growth and Bio-informations of Aeroponically Grown Tomato as Influenced by Spray Intervals of Nutrient Solution (양액의 분무간격에 따른 분무경재배 토마토의 생장 및 생체정보와의 관련성)

  • This experiment was carried oui to determine the relativeness between growth, yield characters and bio-informations as influenced by the spray and rest time intervals of nutrient solution. Tomato(Lycopersicon esculentum Mill.) were grown in aeroponic system on a misting schedule of continuously 60 sec, 30 sec and 10 sec at 10 min intervals with full strength Yamazaki's solution recommended for tomato production. The results obtained were as follows : 1. Leaf area was highest in the plot of 30 sec spray and 10 min rest while the forest one was the plot of 60 sec spray and 10 min rest. Growth characteristics in terms of dry weight of each organ, number of flower, number of flower setted and fruit dry weight were greater in the plot of 30 sec spray and 10 min rest than the other treatments. 2. The number of flower increased with decreasing dry weight but number of flower sorted was not significantly different among treatment except for the plot of 60 sec spray and 10 min rest. 3. Leaf dry weight and fruit dry weight were highly correlated so that 30 sec spray and 10 min rest plot which is the highest fruit dry weight showed the largest leaf area. Continuously sprayed plot reduced markedly the fruit dry weight compared with leaf area. Optimum spray and rest time of nutrient solution in the range of this experiment was determined as 30 sec spray and 10 min rest. 4. Solar radiation within glasshouse during daytime reduced severely compared with outdoor one and air temperature within greenhouse was higher than the leaf temperature of tomato plant. The changes of environmental factors, solar radiation, temperature were accompanied with the sensitive change of bio-informations of tomato leaf Especially differences of spray intervals of nutrient solution affected greatly to the changes of bio-informations : leaf water potential, stomatal resistance and leaf temperature etc. 5. The changing patterns of leaf growth as influenced by the spray and rest intervals of nutrient solution were closely related to the leaf water potential, stomatal resistance and leaf temperature. Feasibility was demonstrated that measurement of bio-information of tomato leaf as influenced by the change of environmental factors could be expected to the amount of growth and fruit yield.

The Effect of Treatment of Woody Charred Materials on the Growth and Components of Tomato and Chinese Cabbage (목질탄화물의 처리가 토마토와 배추의 생육 및 체내성분에 미치는 영향)

  • A research was conducted to investigate the effect of treatment of woody charred materials such as wood vinegar, charcoal, and mixture of wood vinegar and charcoal on the growth and components of tomato and chinese cabbage. The effect of treatment of woody charred materials on the growth and components of tomato the research findings are as follows. The total number of soil microorganisms increased in the groups treated with woody charred materials compared with the control. The fruit number, fruit length, fruit diameter, fruit weight, hardness and sugar contents of tomato did not show significant difference in all plots treated with woody charred materials, but the plot treated with wood vinegar + charcoal showed relatively higher values in general. Vitamin C contents were shown higher in the plots treated with woody charred materials than the control, and among the treatment plots. The effect of treatment of woody charred materials on the growth and components of chinese cabbage the research findings are as follows. The soil chemical components did not change before and after the treatment of woody charred materials. The total number of soil microorganisms increased in the treatment plots compared with the control, The leaf length, leaf width, inner leaf number, plant weight, and head weight of Chinese cabbage treated with woody charred materials were in general larger than the control. Among the treatment plots, the plot treated with wood vinegar + charcoal showed slightly higher values. The contents of minerals in chinese cabbage such as Ca, Fe, K, Mg and Mn were higher in the treatment plots than the control.

  • A survey of the free amino acids and free sugars in tomato, watermelon, muskmelon, peach and plum was made by means of amino acid autoanalyzer and thin layer chromatography. The results of the survey are summarized as follows. 1) Fifteen amino acids found in fruit were aspartic acid, glutamic acid, alanine, serine, asparagine, lysine, valine, glycine, methionine, histidine, threonine, leucine, isoleucine, proline and arginine, and an unknown was found. 2) Ten kinds of amino acids were detected in tomato, peach and plum, thirteen amine acids in watermelon and muskmelon (edible part), and eleven amino acids in muskmelon (rind). 3) In general, these fruits contained similar amounts of these thirteen amino acids, and although they were not outstandingly high in any one acid they did contain a nutritionally well-balanced mixture. 4) Amino acids found in the greatest amount in fruit were following: glutamic acid and aspartic acid in tomato, asparagine and lysine in watermelon, alanine, serine and aspartic acid in musk-melon, and aspartic acid and serine in peach and plum. 5) Glucose, fructose, sucrose and maltose were detected in all fruit. The contents of glucose and fructose were high, and those of sucrose and maltose were low.

Effects of Ca-Gluconate on Fruit Firmness and Softening Enzyme Activities in Tomato using Hydroponics Systems

  • This study was carried out to investigate the effects of Ca-gluconate (Ca-glu) on fruit firmness and softening enzyme activities of hydroponically grown tomato (Solanium esculentum Mill.). The obtained results revealed that the rate of weight loss was markedly increased from at storage to 5 days after storage (DAS) in control, and was constantly increased until 7 DAS as 4.1% in Ca-glu treatment. Fruit firmness was more rapidly decreased in Ca-glu induced fruit compared to control. Results showed that fruit firmness in control and Ca-glu treated fruit were 0.67 and $0.95kg{\cdot}{\varphi}12mm^{-1}$, respectively. In our investigation, no difference was revealed in Hunter's 'a' value between control and Ca-glu treated fruit. Total carotenoids content of control fruit were rapidly increased while the Ca-glu treated fruit were gently increased. Lycopene content was higher ($63.3{\mu}g{\cdot}g^{-1}\;FW$) in control than Ca-glu treatment ($56.8{\mu}g{\cdot}g^{-1}\;FW$). The activity of Polygalacturonase (PG) was rapidly increased with increasing storage period as from 0.4 to 1.2 units whereas the PG activity of Ca-glu treatment was gently increased from 1 to 7 DAS, and rapidly increased from 7 to 11 DAS. However, the pectinesterase (PE) activity was rapidly increased in control fruit, when the storage period was increased, but interestingly, the Ca-glu treated fruit was slowly increased from 1 to 7 DAS, and rapidly increased 7to 11 DAS. ${\beta}$-galactosidase activity of Ca-glu induced fruit was rapidly increased from 1 to 7 DAS as from 1.6 to 3.0 units, and gently increased from 7 to 11 DAS. ${\beta}$-galactosidase activity of control were higher than Ca-glu treatment.

Effects of High Performance Greenhouse Films on Growth and Fruit Quality of Tomato (기능성 피복재가 토마토 생육 및 품질에 미치는 영향)

  • This study was performed to investigate the effect of high performance greenhouse films on growth and fruit quality of tomato. For this purpose, polyolefin (PO), fluoric, antidrop, antifog and thermal films were compared to normal film, ethylene vinyl acetate (EVA). In spectral irradiance of the films, UV ($300{\sim}400nm$) transmittance was highest in fluoric film and lowest in PO film. PAR (photosynthetically active radiation, $400{\sim}700nm$) transmittance was higher in fluoric, thermal and PO film, and near infrared ray (NIR, $700{\sim}1,100nm$) transmittance was higher in high performance films, compared to the EVA film. Total light transmittance was higher in order of fluoric, antifog, anti drop, PO, thermal, and EVA film. Day air temperature in greenhouse was highest under fluoric film and lowest under EVA film due to the light transmittance, while night air temperature was highest under PO and anti drop film due to the thickness of film. Tomato fruits grown under the high performance films had 0.2 to $0.5^{\circ}Bx$ higher soluble solids and 15 to 30% higher lycopene content, compared to those grown under the EVA film. The results showed that tomato fruit quality such as soluble solids and lycopene content can be heightened in terms of much irradiation and better light quality of high performance films, compared to the nomal film, EVA film.

Effect of NaCl Stress on the Growth, Photosynthetic Rate and Mineral Uptake of Tomato, Red Pepper and Egg Plant in Pot Culture (NaCl 스트레스가 토마토, 고추, 가지의 생육, 광합성 속도 및 무기양분 흡수에 미치는 영향)

  • This study was conducted to investigate the effece of NaCl concentrations on the growth, photosynthetic rate and mineral uptake of tomato, red pepper, and egg Plant in Pot culture. The growth such as plant height, plant fresh and dry weight, root fresh and dry weight and dried matter rate was decreased as NaCl concentrations were increased. Specially, the growth inhibition of tomato and egg plant was shown at over 40 mM NaCl, and that of red pepper at 20 mM NaCl. Yield of tomato and egg Plant was reduced at over 20 U NaCl, that of red pepper at over 10 mM NaCl. Yield reduction was affected by the number of fruit at low concentration and by mean weight and number of fruit at high concentration. Photosynthetic rate, water potential and stomatal conductance were decreased as NaCl concentrations were increased. The higher the concentration of NaCl, the lower the mineral uptake such as T-N, P, K, Ca and Mg, however, the higher the content of Na and Cl.
