Antioxidant Components and Antioxidant Activities of 70% Ethanol Extracts on Suweon-511 and Ilpum Rice (수원 511호와 일품쌀의 항산화성분 및 70% 에탄올 추출물의 항산화활성)

  • To evaluate the potential of a new breed, Suweon-511, antioxidant components and antioxidant activities of the 70% ethanol extracts of Suweon-511 (SWE) and Ilpum (IPE) were measured. Total polyphenol and flavonoid contents were 318.17 and 167.21 $\mu$g/g for SWE, which were higher than that of IPE (222.16 and 100.89 $\mu$g/g). Both $\alpha$- and $\gamma$-tocopherol contents were higher in SWE as well. The total tocopherol contents of SWE (1.31 mg/100 g) was higher than that of IPE (0.86 mg/100 g). The $\alpha$-, $\gamma$- and $\delta$-tocotrienol contents of SWE (0.10, 0.43, and 0.02 mg/100 g) that of IPE (0.09, 0.43, and 0.03 mg/100 g) showed little difference. For $\gamma$-oryzanol, Suweon-511, which contained 76.58 $\mu$g/g, contained ten times as much as I lpum (7.77 $\mu$g/g). The total antioxidant activity of SWE and IPE were 16.00 and 9.73 mg AA eq/g, respectively. For reducing power, at 5 mg/mL, both extracts were 0.18, but at 20 mg/mL, SWE showed 0.61 and that of IPE was 0.58. The DPPH radical, hydroxyl radical, superoxide radical and hydrogen peroxide scavenging activity of SWE were 42.00, 21.59, 23.27, and 17.13% at 5 mg/mL, and for IPE, they were 30.10, 22.37, 25.18, and 18.08%, respectively. These data, as well, did not show big difference between the two breeds. As indicated in the data above, the newly-bred Suweon-511 had higher content of antioxidant components compared to Ilpum, but similar or a little higher antioxidant activity.

Analysis of Vitamin E in Agricultural Processed Foods in Korea (국내 농산가공식품의 비타민 E 함량 분석)

  • Accurate food composition data are essential for calculation of nutrient intake of a population based on its consumption statistics. In the Korean food composition database, there is a lack of reliable analytical data for tocopherols and tocotrienols. Therefore, this study was conducted to provide information on contents on vitamin E in agricultural processed foods in Korea. Tocopherols and tocotrienols were determined by the saponification extraction method followed by high performance liquid chromatography. Analytical method validation parameters were calculated to ensure the method's validity. Samples were obtained in the years of 2013 and 2014 from the Rural Development Administration. The samples included 34 grains and grain products, 14 snacks, 25 fruits, 5 oils, and 11 sources and spices. All vitamin E isomers were quantitated, and the results were expressed as ${\alpha}$-tocopherol equivalent (${\alpha}-TE$). ${\alpha}-TE$ values of cereal and cereal products, snacks, fruits, oils and sauces and spices ranged from 0.03 to 17.53, 1.01 to 12.84, 0.01 to 1.52, 1.09 to 8.15, and 0.01 to $27.53{\alpha}-TE/100g$, respectively. Accuracy was close to 100% (n=3). Repeatability and reproducibility were 2.04% and 4.69%, respectively. Our study provides reliable data on the tocopherol and tocotrienol contents of agricultural and processed foods in Korea.

Antioxidant Compounds and Antioxidant Activities of the Methanolic Extracts from Milling Fractions of Black Rice (흑미의 도정분획 메탄올 추출물의 항산화 성분 및 항산화 효과)

  • The colored rice, particularly black rice, has been well characterized to possess antioxidant properties. Rice bran, a by-product of the rice milling process, contains high levels of several phytochemicals which have antioxidant activities as well as health-beneficial properties. The purposes of this study were to evaluate the antioxidant compounds and antioxidant activities of the methanolic extracts from milling fractions and whole grain of two black rice cultivars. Whole black rice of the two cultivars were milled into rice bran and endosperm using the gradual milling system. These were evaluated for antioxidative activities by DPPH and ABTS radical scavenging activities. To determine the antioxidant compounds in the methanolic extract from the milling fractions, the content of polyphenolics, flavonoids, anthocycanins and ${\gamma}$-oryzanol were measured by spectrophotometric methods and vitamin E analysis was carried out by HPLC. The yield of whole black rice, rice bran, and endosperm were 3.1, 15.3, and 0.9% for Heugjinjubyeo and 2.7, 15.5, and 1.1% for Heugkwangbyeo, respectively. The methanolic extracts from rice bran showed generally higher antioxidant activities than the extracts from whole grain and endosperm. In addition, antioxidant compounds distributed much higher contents in rice bran extract than in the extracts from whole grain and endosperm. A significant correlation was also noted between free radical scavenging activity and polyphenolic compounds. The results of this study show that notable antioxidant activity in black rice bran are considered to have significant health benefits.

  • The objectives of this study were to determine various bio-active components from some brown rice and milled rice of some cultivars, as well as to assess their contribution to the radical scavenging linked-antioxidant activity. DPPH or ABTS radical scavenging activities, and reducing power have been used to investigate the relative antioxidant activities of 70% ethanol extracts. The contents of total polyphenol, total flavonoid, $\gamma$-oryzanol, and vitamin E in the extracts were also measured by spectrophotometric methods or HPLC. The antioxidant components and antioxidant activities of 70% ethanol extracts from brown rice were higher than those from milled rice. In addition, our results showed that the high level of antioxidant compounds was found in brown rice of Hongjinju, and the antioxidant activities of the 70% ethanol extracts from pigmented brown rices, such as Hongjinju and Heugkwang, were comparably higher than the others.

Antioxidant Component and Sensory Evaluation of Mixed Cereals (혼합잡곡의 항산화 물질과 기호도)

  • To improve functionality of rice, we manufactured mixed cereals and mixed cereals with taste materials and evaluated their antioxidant component (ascorbic acid, tocopherols, polyphenols and flavonoids) contents and sensory evaluation. Ascorbic acid content in water extract of the control was 6.68 mg%, and mixed cereals were 18.23 and 21.84 mg%, respectively. Ascorbic acid content in methanol extract of the control was lower than those of mixed cereals too. ${\beta}$-Tocopherol was the only tocopherol detected in the control. However, tocopherols detected in mixed cereals were ${\alpha}$-, ${\beta}$-. ${\gamma}$-, ${\delta}$-tocopherol and ${\alpha}$-, ${\gamma}$-tocotrienol, with a highest content of ${\gamma}$-tocopherol. Total tocophenols content of the control was 1.26 mg%, whereas, those of mixed cereals were 19.24 and 39.32 mg%, respectively. Polyphenols contents in water extract of the control was 2.20 mg%, and mixed cereals were 14.38 and 21.91 mg%, respectively. In addition, polyphenols contents in methanol extract of mixed cereals were higher than that of the control, too. Flavonoids were undetected in water extract of the control, but flavonoids contents of mixed cereals were 42.45 and 32.54 mg%, respectively. In methanol extracts, flavonoids contents of the control and mixed cereals were not significantly different. The sensory parameters including taste, flavor, color, texture and overall acceptance of control were lower than those of mixed cereals. Especially, mixed cereals with taste materials was the highest acceptance.

Changes in GABA Content of Selected Specialty Rice After Germination (발아에 따른 일부 특수미의 GABA 함량 변화)

  • The purposes of this work were to investigate the changes in GABA content of six different rice cultivars along with viatmin E content and antioxidant activity after germination. Brown rice was soaked for 24 hr at 25$^{\circ}C$ and then germinated at 37$^{\circ}C$ for 48 hr. The content of GABA and vitamin E in the rice samples was measured by using spectrophotometeric and HPLC methods, respectively. Antioxidant activity was measured by ABTS (2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid)) and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging methods. GABA and vitamin E contents were significantly increased after germination while no significant change in the antioxidant activity was observed. Among the samples tested, Geunnun cultivar contained the highest GABA content before and after germination. On the other hand, Sinmyungheugchal cultivar showed the highest content of vitamin E and antioxidant activity compared to other rice cultivars. In conclusion, the germinated rice with high GABA content can be used for a functional ingredient in rice processing industry.

Analysis of vitamin E and K contents in sea algae and vegetables frequently consumed in Korea for National Standard Food Composition Database (국가표준식품성분표 개정을 위한 국내 다소비 해조류 및 채소류의 비타민 E 및 K 분석)

  • Vitamin E and K are essential micronutrients required by our body in small amounts for proper metabolic functions; however, the content of vitamin E and K commonly consumed in foods has not been comprehensively defined. In this study, the contents of vitamins E and K in sea algae and vegetables were analyzed and the analytical methods were validated. The α-tocopherol equivalent (α-TE) and vitamin K1 content in sea algae ranged from 0.15 to 1.14 mg/100 g and from 11.91 to 1,629.5 ㎍/100 g, respectively. In addition, α-TE and vitamin K1 of vegetables were detected in the range of 0.02-2.48 mg/100 g and 16.15-979.60 ㎍/100 g, respectively. In particular, β- and γ-tocopherol and α- and β-tocotrienol were detected in several vegetables. The analytical methods were accurate and reproducible. These results provide reliable data on the vitamin E and K contents of foods consumed in Korea for the development of National Standard Food Composition Database.

Effects of genotype and environmental factors on content variations of the bioactive constituents in rice seeds (벼의 유전형질과 재배환경 요인이 기능성물질 함량 변이에 미치는 영향 비교)

  • The composition of crops reveal natural variation according to genetic characteristics and environmental factors such as the cultivated regions. For comparative investigation of the impact of genetic difference and environmental influence on the levels of bioactive components in rice seeds, 23 cultivars including indica, japonica, and tongil rice were grown in two location in Korea (Jeonju and Cheonan) for two years (2015 and 2016). Sixteen compounds consisting of tocopherols, tocotrienols, phytosterols, and policosanols were identified from 368 rice samples and the compositional data were subjected to data mining processes including principal component analysis and Pearson's correlation analysis. Under 4 different environmental conditions (Jeonju in 2015, Cheonan in 2015, Jeonju in 2016, Cheonan in 2016), the natural variability of rice seeds showed that the genetic background (indica vs japonica vs tongil) had more impact on the compositional changes of bioactive components compared to the environments. Especially, the results of correlation analysis revealed negative correlation between α-, β-tocopherols and γ-, δ-tocopherols as a representative genetic effect that did not changed by the environmental influence.

Comparison of Micronutrients and Flavor Compounds in Breast Meat of Native Chicken Strains and Baeksemi for Samgyetang (삼계용 토종닭과 백세미 가슴살의 미량영양소 및 풍미물질 비교)

  • The micronutrients and flavor compounds of three new native chicken strains (A, C, and D) being developed in a breeding program (Golden Seed Project) were compared with a commercial native chicken strain (H) and the Baeksemi (W, white semi broiler). After 100 male chicks in each strains were reared for 5 weeks, the breast muscles from randomly selected 40 birds were analyzed at 8 replications. Native chicken strain A had greater amounts of α-tocopherol and α-tocotrienol compared with strain W. Native chicken strains showed higher contents of vitamin B12 than Stain W. Stain H strain had the highest values of cholesterol content and strain D did the lowest. There was no different content of most minerals between native chickens (A, C, D, and H) and W, but Cu were more contained in native chicken strains compared with W. In terms of nucleotide-related flavor compounds, Stain A had the lowest content of hypoxanthine and strain D had highest inosine monophosphate. Native chicken strains had higher contents of umami-related free amino acids (glutamate and aspartate) than W. Among native chickens, strain A had more amounts in the contents of taurine, tasty flavor compounds, and certain vitamins, despite of the relatively low growth productivity. This result will provide information to select a strain with characteristic meat quality in a chicken breeding program.

Antioxidant Compounds and Activities of Methanolic Extracts from Oat Cultivars (품종별 귀리 메탄올 추출물의 항산화 성분 및 항산화 활성)

  • Oats (Avena sativa L.) were evaluated in recent years as a promising crop for increasing the nutritional quality of foods, due to their abundance in many bioactive compounds. The objectives of this study were to determine antioxidant compounds and antioxidant activities of nine oat cultivars and investigate the relationships between antioxidant compounds and antioxidant activities in oat cultivars. The contents of total polyphenolics and vitamin E in oat cultivars were analyzed by spectrophotometric methods and normal phase HPLC, respectively. 2,2'-Azino-bis-(3-ethylbenzothiazoline- 6-sulphonic acid) (ABTS) radical and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities, reducing power, and chelating effects were used to compare the relative antioxidant activities of oat cultivars. In this study, total polyphenolic and vitamin E contents of oat cultivars ranged from 83.41 to 130.59 mg gallic acid equivalents/100 g and 0.54 to 1.90 mg/100 g, respectively. High levels of ABTS radical (116.14 mg Trolox equivalent antioxidant capacity (TEAC)/100 g) and DPPH radical (38.58 mg TEAC/100 g) scavenging activities and reducing power ($A_{700}=0.922$) were found in Daeyang. Significant correlation was evaluated between total polyphenolic contents and their antioxidant activities, namely ABTS and DPPH radical scavenging activities and reducing power. A low correlation was observed between vitamin E contents and their antioxidant activities. The results of this study could have an impact on oat consumption by enhancing consumer awareness of the health benefits of oats.
