Effect of Viscosity of Binder and[ Storage Times of Pelleted Seed on Physical and Germination Characteristics of Tobacco Seeds. (종자피복용 binder 점도와 피복후 저장기간이 종자의 발아에 미치는 영향)

  • In order to improve the sowing practice, pelleted seeds of tobacco NC 82(Nicotiana tobacco L.) were manufactured in use of binders at the different levels of viscosity, and their physical properties according to pellet size and biological activity in seed germination were investigated while storage time elapsed. Proper range of the binder viscosity for the pellet formation was 20.3-37.2 m.pas. At the high level of viscosity(45.7 m.pas) took longer time to form the pellet and pelleting was not easy. The high binder viscosity and large pellet size revealed higher level in hardness of the pelleted tobacco seeds, which caused the longer splitting time of pellets in water. High level of binder viscosity(37.2 m.pas) in pelleted seeds led to decrease the biological activity of tobacco seed germination by the storage at 4 t over 30 days. But at the level of 20.3m.pas in binder viscosity, the germination of pelleted seeds was little affected by the long storage time to 120 days.

The Water Content and Treatment Period of SMP for Enhanced Seedling Emergence in Tobacco Seeds. (담배 종자의 입묘율 향상을 위한 SMP 수분함량과 처리기간)

  • The purposes of this study are to investigate water content and treatment period in Solid Matrix Priming(SMP; method for enhanced rapid and uniform seedling emergence in tobacco seeds). The higher water content made the faster imbibition speed of tobacco seed. But the radicle emergence shorten the period of water equilibrium. The range of water content in SMP treatment within the limits of no radicle emergence of tobacco seed was 28∼32% to dry weight of tobacco seed. The rate of radicle emergence and seedling emergence was higher in water content 30% and 40% treatment plot than that in 50% and 60% treatment plot. But it was not different between SMP treatment and non-SMP treatment. In water content 40% during 9 days treatment, germinative energy was highest, and T50 and mean germination time(MGT) were the shortest. The germination speed was higher in SMP treatment than that in non-SMP treatment. It was not different on the germination rate between non-SMP treatment and SMP treatment.

Some Management Practices Affecting Outcrossing and Seed Production in Burley Tobacco (Nicotiana tabacum L.) (연초 버어리종의 자연교잡율과 종자생산에 관련된 몇가지 요인)

  • In this study effects of isolation distance, transplanting time of maternal plants, and bagging of flower head with the gauze-cloth bag on the outcrossing of burley tobacco (Nicotiana tabacum L.) were investigated. Also the effect of fertilizer level and control of the number of capsules per plant on seed production and quality were examined. A male sterile line. produced 0.3 to 3.8 capsules Per plant when it was Planted with normally flowering tobacco with the average outcrossing of 7.2 plants, ranging from 2 to 18 out of 20 plants. With the farther the isolation distance between maternal plants and pollen donor plant, the lower the outcrossing occurred. Outcrossing occurred even at the isolation distance of 312 m. When the maternal plants were transplanted 35 days after transplanting the pollen donor ones, the outcrossed plants were not decreased significantly. The bagging of the flower head with the gauze-cloth bas (#0.9∼ 1.0 mm) decreased the outcrossed plants significantly, but couldn't prevent the outcrossing completely. The seed amount per plant was higher in the highly fertilized cultivation. The number of seed capsules per plant affected significantly on seed yield and quality. When the seed capsules was controlled by 30 or 50 capsules per plant, the weight of 1,000 seeds and germination rate were higher than those with 70 or 90 capsules per plant.   • Relative content(% weight) of proteins, carbohydrates, lipids, and crude ashs in seed of korean green tea plant(Camellia simecis L.) are not different from those in seed of sunflower and safflower. However, the Camellia seed contains much higher crude saponin content(12.2%) than that of sesame(0.29%) or peanut(0.63%). It also contains 82% unsaturated fatty acids including oleic acid and contains tocopherol $(22\;{\mu}g/g,\;{\alpha}-form\;only)$ that is significantly less than of other oil-seed.

  • Ginseng seeds were gathered from 3,4,5 and 6 years of age and were classified into four qroups (below 4mm, 4∼5mm, 5∼6mm and above 6mm in across sieve). They were sown in seedling bed and some characters were investigated in each qroup of seed size. 1. The distribution of seed size of below 4mm, 4-5mm, 5-6mm and 6mm were 23.7%, 60.8%, 12.4% and 4.5%, respectively. 2. The ratio of seed coat dehiscence was not affected by seed size but emergence ratio and emerging vigor were superior in large seed. 3. The large seed showed superiority in stem length, stem diameter, leat and also in root length, root diameter and root weight. but diseased root was not affected by seed size. The effect of age(seed harvest) was not significant on all those characters.

  • A CAB cDNA clone(pKGCAB) encoding the light harvesting chlorophyll a/b binding protein of the semi-shade plant, Korean ginseng(Panax ginseng C. A. Meyer) was isolated by the one-way path random sequencing of ginseng cDNA library clones and transgenic tobacco plants(Nicotiana tabacum NC82) were produced by the transformation of this ginseng CAB gene in use of Agrobacterium tumefaciens LBA4404. The CAB gene showed type 1 structure of LHCP-II, 84% similarity in nucleotide sequence and 92% in amino acid sequence to that of Nicotiana tabacum CAB40, respectively. Seed germination and initial growth of the transgenic tobacco plants transformed with the cDNA fragment were accelerated under low light intensity compared with those of normal tobacco plant, that may result from the higher light sensitivity of the transgenic plants than that of the normal.

Seed Production Status in Ginseng Plantations (농가포장에서의 인삼종자 생산실태)

  • To get the basic information about ginseng seed production, yield of seed and seed quality in 27 fields were investigated. Yield of seed ranged from 4 to 10 litres per 100 kan (180cm x 90cm). Ratio of under 4mm seed is 12%, 66% for 4 to 5mm and 22% for over 5mm. Seed weight and ratio of over 4mm seed showed the decreasing tendency with the increase of seed yield per unit area. The seed yield harvested from the selected mother plants was lower than that from all plants, but seed weight and ratio of over 4mm seed were high in fields using the selected mother plants. It showed a positive correlation between length and width of seed, but no correlation between thickness and length or width. Optimum yield for high quality seed appeared under the 7 to 8 litres per 100 kan.

Resistance to Potato Virus Y Conferred by PVY Replicase Gene Sequence in Transgenic Burley Tobacco (감자바이러스 Y 복제 유전자로 형질전환된 버어리종 연초의 PVY에 대한 저항성 특성)

  • The complementary DNA (cDNA) of potato virus Y- vein necrosis strain (PVY-VN) replicase gene (Nlb) was transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Out of 25 putative transformants regenerated, 3 were resistant to PVY-VN, one highly resistant plant with no symptom until seed harvest time and the other two with mild chlorotic spot symptoms at late stages after infection. No symptom was observed in the highly resistant plant, while mild vein necrotic symptoms were developed on suckers of the moderately resistant plants after seed harvest time, In the first generation (T1) via self fertilization, resistance to susceptibility frequency in transgenic plants from the highly resistant transformant was about 3 : 1, while it was lowered much (about 1:2 and 1:19) in T1 of the moderately resistant transformants. In the second generation (T2) of the highly resistant plant, resistance frequencies were similar to T1, but resistance levels varied greatly and appeared to be decreased.   • In order to improve the efficiency of F$_1$ hybrid seeds production(KF 114), some experiments were carried out in a greenhouse this year. Mother plant(MSNC567) and pollen plant(NC) were grown in some pots (30 x 30cm, WxH). The gathered pollens were mixed with celite, pollen deluent dusts and stored in refrigerator at l$0^{\circ}C$ before pollination. To establish the critical range of the mixed rate and the storage period of pollens, the change in a percent of capsule set, number of seed and seed weight per capsule, weight of 1000 seeds and germination percent of seed resulted of pollination with pollen deluent dusts were investigated. The results are as follows; Percent of capsule set showed the mixed rate 1:5 resulted in no difference and those of 1:10 and 1:20 decreased about 5 ~15% when compared with trials using pollen alone. There was no difference among duration of storage in a same mixed rate. Numbers of seed per capsule showed a significant difference among the mixed rates and among durations of storage. Numbers of seed per capsule were decreased about 55 ~ 90 % as the mixed rate increase, about 2 ~ 28 % as the duration of storage increase. Weight of seed per capsule decreased about 16 ~ 23 % as the duration increase and decreased about 3 ~ 89 % as the mixed rate increase when compared with trials using pollen alone. Weight of 1000 seeds showed non significant difference among duration of storage but were higher than trials used pollen alone as the mixed rate increase.

  • Influence of fungi living on the endocarp surface of depulped seeds of Panax ginseng on the dehiscence was investigated with a fungicidal treatment of the seeds and sterilization of the sand at the beginning of stratification. 1. Isolation frequency of the fungi living on the endocarp surface of de-pulped seed was reduced and hardness of the endocarp did not change significantly in seed treated with a fungicide Captan wp. 50%. A significant negative correlation $(r=-0.984^**)$ was found between the frequency of fungi isolation and the hardness of the endocarp. 2. Increase of water content in the seed treated with the fungicide was delayed 20days compared to the untreated. 3. Growth of the embryo and dehiscence of the seed was suppressed by the fungicide treatment. The length of the embryo was inversely proportional to the hardness of the seed. It is suggested that the fungi facilitate the softening of the endocarp thereby enhancing the supply of oxygen and water necessary for the embryo development, therefore, accelerate the growing of embryo and cause the dehiscence.
