세균성마름병에 대한 담배의 저항성검정 방법 (A Screening Method on Resistance of Tobacco Plants to Bacterial Wilt)

  • Three kinds of inoculation methods, capillary, root cutting and dipping were compared for an efficient way to screening the resistant tobacco variety against bacterial wilt, Ralstonia solanacearum. The pricking a capillary tube contained the pathogenic bacterial suspension(10$^{7}$ cfu/$m\ell$) to an axillary bud of each tobacco plant showed different resistance well between varieties. The less period was required in inoculating work and in disease development for the inoculation method used with capillary tube than for two other inoculation methods tested also.

Bacillus subtilis Protoporphyrinogen Oxidase 유전자 형질전환 담배의 Oxyfluorfen 처리에 대한 생리 · 생장반응 (Physiology and Growth of Transgenic Tobacco Plants Containing Bacillus subtilis Protoporphyrinogen Oxidase Gene in Response to Oxyfluorfen Treatment)

  • Bacillus subtilis protoporphyrinogen oxidase 유전자를 Agrobacterium을 매개체로 이용하여 전이 시킨 후 cauliflower mosaic virus 35S promoter하에서 발현케 한 형질전환 담배를 유기하였다. 이러한 형질전환 담배의 diphenyl ether계 제초제 oxfluorfen에 대한 생리적 반응과 여러 환경 조건에서의 생장 반응을 재배종 담배와 비교하였다. Oxyfluorfen의 처리에 의해 나타나는 세포내 구성물질의 누출과 지질과산화작용은 재배종 담배에서보다 형질전환 담배에서 더 작게 이루어졌다. 형질전환 담배의 생장을 여러 온도, 광도 및 수분 조건에서 조사한 결과, 저광도와 포화수분 조건에서의 생장이 재배종 담배에 비해 다소 저하되는 현상이 나타났을 뿐 기타 조건에서의 생장은 재배종 담배의 생장과 거의 동일하였다. 따라서 B. subtilis protoporphyrinogen oxidase 유전자를 전이시켜 cauliflower mosaic virus 35S promoter하에서 발현케 한 형질전환 담배는 oxyfluorfen에 대해 비교적 높은 저항성을 나타내지만 형질전환에 따른 생장 변화는 크게 일어나지 않음을 알 수 있었다. 한편 이러한 형질전환 담배의 oxyfluorfen에 대한 저항성 기작에 대해서도 논의하였다.

향초와 L. A. Burley 21의 상호접목이 담배의 생육 및 화학적 특성에 미치는 영향 (Growth and Chemical Components of Reciprocally Grafted Tobacco Cultivars between Hyangchio and L. A. Burley 21)

  • To investigate the effect of reciprocal grafting between Hyangchio (H) and L. A. Burley 21 (L.A), one a higher producer of nicotine and low yield, the other, a low producer of nicotine and high yield, growth of various parts of each variety and chemical constituents of these parts were evaluated. The results were as follows : The growth of H/H graft was depressed when compared to hyangchio and H/L. A. and L. A. Burley 21 showed most vigorous growth plants having L. A. Burley 21 top. The amount of total alkaloids were low in leaves and stems of plants having L. A. Burley 21 roots (L. A., L. A./L. A., H/L. A.). Plants having Hyangchio roots (H, L. A./H) were high in total alkaloids. The contents of reducing sugar were high in plant having Hyangchio top compared to L. A. Burley 21 tops, but there were not diffences in contents of ether extracts among all treatments.

  • Effects of aqueous ammonia soaking to three annual plants (hemp woody core, tobacco stalk and corn stover) awere investigated to focus on the enzymatic saccharification characteristics change by this treatment. At two different levels of treatment ($90^{\circ}C$-16 h and $45^{\circ}C$-6 days), higher temperature treatment led to more enzymatic saccharification of cellulose to glucose by commercial cellulase mixtures (Celluclast 1.5L and Novozym 342 from Novozyme Korea). Difference among annual plants were significant. corn stover was the best response to enzymatic saccharification of cellulose and xylan by comercial enzymes all treatment conditions but tobacco stalk was the worst response to all of them. chemical composition or physical structure difference may brought this difference.

  • The present study was conducted to compare the efficiency of individual plant selection for resistance to bacterial wilt with 30 dihaploid lines derived by anther culture and Nicotiana africana method in Fl and F2 generation from a cross between Bright Yellow 4 (BY4) and NC95. F2 dihaploid lines were selected from bacterial wilt disease resistant plants screened under the naturally infested filed conditions. The populations of FB - ADH and FB MDH derived from F2 individual plants with bacterial wilt resistance showed higher resistance to the disease than the populations of Fl - ADH and Fl - MDH, respectively, and no difference for the disease resistance appeared by the haploid deriving method within a generation.

  • 본 연구에서는 담배식물의 생장과 glutathione reductase와 catalase의 활성에 미치는 p-CA, BA 및 SA의 영향을 연구하였다. 담배식물의 생장에 미치는 p-CA, BA 및 SA의 영향을 알아보기 위하여, $10^{-5}$ mM p-CA, BA 및 SA가 각각 함유된 MS배지에서 배양한 후 20일부터 50일까지 10일 간격으로 담배의 생장을 비교하였다. 3가지 물질 모두에 의해 50일에서 생장이 가장 잘 되였다. Glutathione reductase의 활성에 미치는 효과를 측정한 결과, BA의 활성은 대조구보다 감소하였으며, p-CA 역시 활성이 감소하였고, SA의 활성은 증가하였다. 또한 모두 40일에서 가장 활성이 높았다. Catalase의 활성에 미치는 효과를 측정한 결과, SA의 활성은 증가하였으며, BA와 p-CA의 활성은 감소하였다. 또한 모든 실험구에서 40일에 활성이 가장 양호하였다. 결론적으로 p-CA와 BA는 담배식물의 생장을 촉진시켰으며, 50일에서 생장이 가장 양호하였으나 항산화 효소의 활성을 억제하였다. SA는 담배식물의 생장을 억제시켰으나 항산화 효소의 활성은 촉진시켰으며, 40일에서 가장 활성이 양호하였다.

Enhanced bacterial resistance in transgenic tobacco expressing a BrRZFP1 encoding a C3HC4-type RING zinc finger protein from Brassica rapa

  • C3HC4-type RING zinc finger proteins essential in the regulation of plant processes, including responses to abiotic stresses. We previously isolated and examined the C3HC4-type RING zinc finger protein (BrRZFP1) from Brassica rapa under abiotic stresses. To elucidate the role of the BrRZFP1 transcription factor in gene regulation, we transformed tobacco plants with the BrRZFP1 gene. Plants were regenerated from 82 independently transformed callus lines of tobacco and analysed for transgene expression. Transgene integration and expression was confirmed by Southern and RT-PCR analyses, respectively. T2 plants displayed more tolerance to the bacterial pathogens Pectobacterium carotovorum and Ralstonia solanacearum, and the tolerance levels were correlated with BrRZFP1 expression levels. These results suggest that the transcription factor BrRZFP1 is an important determinant of stress response in plants and its overexpression in plants could increase biotic stress resistance.

  • Effects of some mineral nutrients on the chlorine absorption by the plants, on the chemical constituents and physical properties of leaves were investigated tinder the paddy field and pot conditions. The chlorine content of cured leaf grown in paddy field was high in iron and manganese application groups and highest in combined application of iron and manganese. Lime application inhibited the absorption of chlorine and increased the yield and quality of cured leaf, and inhibited the absorption of iron and manganese those causing the grey leaf. Lime application reduced the leaf chlorine content and rate of muddy grey leaf by increasing the soil pH in acid soil.

  • Tobacco (Nicotiana tabacum) cultivars including NC 82 and KF 114, and Datura stramonium, Physalis floridana, Gomphrena globosa, and Chenopodium spp. were added to the previous host plants tested for the further examination on the biological characteristics of tobacco mosaic virus (TMV) strains isolated from tobacco (TMV-Common), tomato (TMV-Tomato), and pepper (TMV-Pepper), In TMV-Tomato and TMV-Pepper, different symptoms were noted in P. floridana (no symptom development), and NC 82 (local lesion production on the inoculated leaves) from TMV-Common with which systemic mosaic symptoms were developed. Local lesions were developed in KF 114, D. stramonium, G. globosa, and Chenopodium spp by TMV-Common and TVM-Tomato, while no symptom was observed in KF114 and G. globosa. Also the number and size of local lesions were smaller in KF 114 than Xanthi-nc tobacco (local lesion host) infected with TMV-Tomato. Systemic necrosis was induced in Xanthi-nc and KF 114 when infected with TMV-C at high temperature, but not with the other strains.

담배(Nicotiana tabacum) 세포배양에 의한 Glyphosate-저항성 개체의 재분화 (Regeneration of Glyphosate-Resistant Plant from Tobacco (Nicotiana tabacum) Cell Culture)

  • From the single cell cultures of haploid tobacco (Nicotiana tabacum L. cv. NC 2326) glyphosate-resistant plants were regenerated. After treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and then inoculated onto the LS medium supplemented with 1 mM glyphosate, the single cells survived formed colonies and calluses in 65 and 95 days after culture, respectively, and then whole plants were regenerated in 0.1 mM glyphosate-containing medium from the selected calluses. There was no difference in fresh weight and shikimate content between the selected and normal haploid calluses. When sprayed with 0.1 mM glyphosate, the shikimate contents in the regenerated and normal plants were 0.659 and 20.816 mol/g fr. wt., while that in other normal plants which were not sprayed was 0.921. In addition, the calluses induced from the regenerated plants grew without showing any retardation when treated with glyphosate. These results indicate that the secelcted calluses and regenerated plants are resistant to glyphosate.
