미생물 유래 Dykellic Acid가 담배 녹색배양세포의 생장 및 Superoxide Dismutase 활성에 미치는 영향 (Effects of Dykellic Acid Derived from Microorganism on the Cell Growth and Superoxide Dismutase Activity in Tobacco Photomixotrophic Cultured Cells)

  • 미생물에서 분리한 dykellic acid가 식물세포에 미치는 생리적 영향을 평가하기 위하여, 담배 광혼용영양배양세포(photomixotrophic cultured cells, PM세포)에 dykellic acid를 다양한 농도로 처리하여 세포의 생장과 단백질 함량 및 superoxide diamutase (SOD)활성에 미치는 영향을 조사하였다. 담배 PM세포는 0.7 mg/L 2,4-D, 0.3 mg/L kinetin, 30g/L sucrose, 200 mM NaCl를 함유한 MS배지에서 $25^{\circ}C$, 광조건에서 현탁배양 (100 rpm) 하였다. 계대배양시 화합물을 처리한 후 12일째의 세포생장 억제와 배지의 이온 전도도를 측정한 세포막손상의 결과는 일치하였으며, 화합물은 세포생장을 강하게 억제시켰다 ($IC_{50}$/, 약 20 $\mu$M). Dykellic acid 처리농도가 증가함에 따라 단위세포 무게당 단백질 함량과 SOD 비활성도를 현저히 증가시켰다. 이상의 결과로 dykellic acid는 식물세포 생장을 저해하는 활성을 가지고 있으며, 담배 PM세포는 적은 양으로도 천연물의 생리활성을 평가할 수 있는 유용한 생물소재임이 확인되었다.

감자바이러스 Y 및 역병 저항성 연초 버어리종 신품종 KB 110의 육성 및 농경적 특성 (Breeding of Burley Tobacco KB 110 Resistant to PVY and Black Shank and its Agromomic Characteristics)

  • The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.

내병성 버어리종 신품종 "KB 101"의 육성경과 및 특성 (Performance of "KB 101" a New Burley Tobacco Resistant to Bacterial Wilt and Black Shank)

  • KB 101 is a bacterial wilt(Pseudomonas solanaceamm E.F. Smith) and black shank (Phytophthora nicotianae Breda de Haan Var. nicotianae Waterhouse) resistant cultivar of burley tobacco (Nicotiana tabacum L.) KB 101 was developed by the Korea Ginseng&Tobacco Research Institute, and released in 1987. KB 101 was developed from a single plant selection in the F2 generation derived from the double cross, [(Burley 21X Burley 37) X (Burley 64X Ky 16)]. Burley 37 and Burley 64 were the source of resistance to bacterial wilt and black shank. Yield trials were conducted in the Fs through F6 generations at the four Exp. Stn. of Korea Ginseng &Tobacco Research Institute as JB 7705-1. On-farm yield trials were conducted in the F7 through F9 generations at the 45 locations of burley tobacco growing area from 1984 to 1986 as KB 101. KB 101 has an erect growth habit similar to that of Burley 21: plant size is larger and has more leaves than those of Burley 21. It is late maturing cultivar that flowers approximately 3 days later than Burley 21. The physical characteristics and chemical composition of KB 101 were similar to those of Burley 21.

오리엔트종 연초의 생육 및 화학특성에 미치는 이식시기의 영향 (Growth and Chemical Properties of Oriental Tobacco as affected by Transplanting Time)

  • Oriental tobacco (KA 101) was transplanted from Mar. 25 to May 5 with 10 days interval in 1984 and 1985, and its agronomic characteristics, chemical properties ware compared to determine the Proper transplanting time of aromatic tobacco In Korea. As the transplanting was delayed, days to flowering of plant was shortened, length and width of largest leaf, leaf area index tended to decrease. Yield was highest for the Apr. 5th transplanting followed by Mar. 25th planting, then decreased as the planting date was delayed. Quality by price decreased as the transplanting was delayed later than Apr. 25. Delaying transplanting increased nicotine, total nitrogen and ash content, but decreased reducing sugar and petroleum ether extract of cured leaves, The content of volatile organic acids such as 3-methyl pentanoic acid was lower when it was transplanted later than Apr. 25th. Neophytadiene content increased as the transplanting was delayed, but there were no trends with the content of alcohols, aldehydes esters and ketones. Several quality indices including the ratio between the content of volatile organic acids plus petroleum ether extract and ash content plus pH was higher for the Apr. 5th transplanting.

  • This experiment was conducted to investigate the effects of fertilization level, additional dressing time and form of nitrogen on the growth and formation of Alkaloid in flue-cured tobacco. The results are abstracted as follows. Length and width of harvested leaves, area of a leaf, leaf area per plant, number of harvested leaves, weight of a leaf, and leaf weight of a plant were larger in the $NO_3$-N plot than $NH_4$-N plot. Length, width and area of the middle leaves increased by additional dressing on 7 days before topping, but the dry weight Per unit leaf area of those was decreased. Leaf shape was broaden in additional dressing plots in comparison with 100% of basal dressing plot, but there is no difference among the additional dressing plots. And the smallest leaf shape index was shown at upper leaves in 100% of basal dressing Plot. Leaf weight of a Plaint was the highest in 12kg/10a Plot among $NO_3$-N form and in 12kg/10a plot among $NH_4$-N form. Total Alkaloid content was higher in $NH_4$-N plot between fertilization forms, and highest in 15kg/10a plot among fertilization levels and at the position of middle and upper leaves in the plot of 7 days before topping among additional dressing times.

  • 향끽미종잎담배인 소향의 시비량을 달리하여 엽면수지량과 생육형질과의 관계를 조사한 바, 주당 및 단위 엽면적당의 엽면수지량은 질소수준간에 유의차를 보여 3k9/ 10a수준에서 많았고 인산 및 가리의 영향은 유의성이 인정되지 않았다. 주당엽면적수지량은 초장. 최대영장과 폭. 주당건엽중 및 생명중과 평균엽면적 과는 각각 정 (+). 엽후와는 부(-)의 상관이 나타났다. 단위엽면적당 엽면수지량은 단위엽면적중 및 건물율과는 정 (+), 평균엽면적과는 동일 질소수준에서 부(-)의 상관이 나타났다.

  • 곤충탈피호르몬의 비스테로이드계 길항제인 RH 5849는 엽침적법으로 처리했을 때 담배거세미나방(Spodoptera litura)의 전 유충 발육단계에 걸쳐서 거의 비슷한 $LC_{50}$ 값을 보여주었다(18.1~26.5 ppm). 4령 유충에서 섭식저해율은 처리농도에 따라 비례하였고, 4.2ppm 으로 엽침적처리된 양배추 엽편을 먹은 3령 유충의 체중증가는 처리 후 48시간 이후부터 정체되었다. 양배추와 담배의 전 식물체에 대한 RH 5849의 침투성 약효발현이 관찰되었는데, 약 20ppm(mg/kg soil)이하의 $LC_{50}$ 값이 양배추에서는 15일, 담배에서는 30일 가량 지속되었다.

카드뮴(Cd) 하에서 기내 배양된 담배의 생장과 Rubisco/Rubisco Activase에 대한 Ethylsalicylic acid의 영향 분석 (Effects of Ethylsalicylic Acid on Growth and Rubisco/Rubisco Activase in Tobacco Plant Cultured under Cadmium Treatment in vitro)

  • 카드뮴(Cd)에 의해 유도되는 담배의 생장과 rubisco/rubisco activase에 미치는 ESA (ethylsalicylic acid)의 효과 및 이에 대한 변성제의 효과를 연구하였다. 담배기내 배양에 대한 ESA의 최적 농도를 찾기 위해, $10^{-6}$-10 mM ESA를 처리하여 배양시킨 결과, $10^{-4}$ mM ESA에서 생장이 가장 높게 나타났다. 최적농도인 $10^{-4}$ mM ESA와 0.2 mM $CdCl_2{\cdot}2.5H_2O$를 사용하여, 대조구, Cd 처리구, ESA 처리구 및 Cd와 ESA 혼합구에서의 담배의 생장을 측정한 결과, ESA 처리구의 생장이 가장 좋았으며, Cd 처리구의 생장이 가장 저조하였다. Rubisco/rubisco activase의 함량과 활성을 측정한 결과, 두 가지의 함량과 활성 모두 Cd 처리구가 가장 낮았으며, ESA 처리구에서는 Rubisco와 rubisco activase의 함량이 감소되었고, 활성은 증가하였다. Guanidine-HCl 처리를 제외한 L-cysteine, urea, thiourea, ${\beta}$-mercaptoethanol, EDTA에 의해 rubisco의 활성이 억제되었으며, L-cysteine, urea, thiourea, guanidine-HCl 처리구에서는 rubisco activase 활성이 변화가 없었으나, ${\beta}$-mercaptoethanol과 EDTA는 활성을 증가시키는 것으로 나타났다. 결론적으로, ESA는 rubisco의 함량을 억제시키고 활성은 촉진시키며, rubisco activase의 함량은 촉진시키고 활성은 억제시켰다. 또한 Cd에 의해 저해된 rubisco와 rubisco activase의 활성이 변성제에 의해 저해되었으며, Cd에 의해 저해된 ESA의 회복이 변성제에 의하여 상실되었다.

Salicylic Acid as a Safe Plant Protector and Growth Regulator

  • Since salicylic acid (SA) was discovered as an elicitor of tobacco plants inducing the resistance against Tobacco mosaic virus (TMV) in 1979, increasing reports suggest that SA indeed is a key plant hormone regulating plant immunity. In addition, recent studies indicate that SA can regulate many different responses, such as tolerance to abiotic stress, plant growth and development, and soil microbiome. In this review, we focused on the recent findings on SA's effects on resistance to biotic stresses in different plant-pathogen systems, tolerance to different abiotic stresses in different plants, plant growth and development, and soil microbiome. This allows us to discuss about the safe and practical use of SA as a plant defense activator and growth regulator. Crosstalk of SA with different plant hormones, such as abscisic acid, ethylene, jasmonic acid, and auxin in different stress and developmental conditions were also discussed.

담배의 생장과 광합성에 미치는 카드뮴의 독성과 이에 대한 칼슘의 효과 (Cadmium Toxicity and Calcium Effect on Growth and Photosynthesis of Tobacco)

  • 담배의 생장과 광합성 색소 및 효소에 대한 카드뮴의 독성과 이에 대한 칼슘의 영향을 연구하였다. 카드뮴에 의한 생장과 엽록소 수준에서의 억제 현상은 칼슘에 의해 회복되었다. 특히, 엽록소의 함량은 카드뮴만 처리한 것 보다 카드뮴과 칼슘을 같이 처리하였을 때 현저하게 증가하였다. 또한 rubisco의 활성과 함량은 카드뮴을 처리한 것 보다 처리하지 않았을 때 현저하게 감소하였으며, 카드뮴의 이러한 효과는 칼슘에 의해 환원되었다. Rubisco activase의 활성과 함량도 rubisco와 같은 경향을 보였다. Rubisco에서 카드뮴과 칼슘에 의해 유도되는 이와 같은 변화는 rubisco activase에 의해 기인됨을 의미한다.
